Characteristics of Newly Bred Shiitake Strain "Chunbaegko" (표고 신품종 "천백고"의 육종 및 특성)

  • New Shiitake (Lentinula edodes) strain "Chunbaegko" was bred by Di-mon method. Bed-log cultivation of "Chunbaegko" was performed. Fruit-body production of "Chunbaegko" was most at spring and autumn. Optimal temperature of fruit-body formation was $14{\sim}22^{\circ}C$ Baegwhako, the best quality fruit-body, is produced during spring. The diameter of pileus is ca. 52 mm. The total amount of fruit-body production during 4 years(one generation) was 140 $kg/m^3$ log.

Degree of Heterosis and Heterobeltiosis in F1 Hybrids of Eggplant (가지 F1세대의 잡종강세 정도)

  • This experiment was performed to obtain the basic information on heterosis and heterobeltiosis of quantitative characters in the eggplant by crossing of varieties. The heterosis and heterobeltiosis were estimated in Fl generation. The results were as follows, based on the graphical analysis, the characters, plant height, stem diameter, fruit weight, leaf length, leaf width, and fruit number per plant(yield) were assumed to inherit dominance, and fruit length and fruit diameter were found to inherit incomplete dominance. The degree of positive heterosis of plant height, stem diameter, fruit weight, fruit length, fruit diameter, leaf length, leaf width, and fruit diameter, leaf width, leaf length, fruit weight, and stem diameter showed negative heterobeltiosis.

  • Watermelon and melon are economically important Cucurbitaceae crops. Recently, the development of molecular markers based on the construction of genetic linkage maps and detection of DNA sequence variants through next generation sequencing are essential as molecular breeding strategies for crop improvement that uses marker-assisted selection and backcrossing. In this paper, we intended to provide useful information for molecular breeding of watermelon and melon by analyzing the current status of international and domestic research efforts on genomics and molecular markers. Due to diverse genetic maps constructed and the reference genome sequencing completed in the past, DNA markers that are useful for selecting important traits including yield, fruit quality, and disease resistances have been reported and publicly available. To date, more than 16 genetic maps and loci and linked markers for more than 40 traits have reported for each watermelon and melon. Furthermore, the functional genes that are responsible for those traits are being continuously discovered by high-density genetic map and map-based cloning. In addition, whole genome resequencing of various germplasm is under progress based on the reference genome. Not only by the efforts for developing novel molecular markers, but application of public marker information currently available will greatly facilitate breeding process through genomics-assisted breeding.

Breeding of New Synthetic Egg Production Line in Domestic Chicken by Intlroducing Sex Linked Gene. I. Production of the Autosexing Breed (성 감별 유전자를 도입한 다산계계통 신품종 육종에 관한 연구 I. 반생유전계통 조성)

  • This study was carried out to build up new synthetic egg Production lines which had sex linked gene for feather color sexing and had also superior combining ability for producing the best commercial chicks. In order to make autosexing layer line, the commercial layers which had Z$^{s}$ Z$^{s}$ and Z$^{s}$ W were mated. Among progeny, the chicks which had homozygote of silver gene and non-silver gene were selected for making dam and sire lines. Afterwards the closed flock breeding method was utilized to improve general performances of the each line. The performances of egg production in synthetic line were 161 day for age at sexual maturity, 219 eggs for total egg number to 60 weeks of age, 84% for hen-day egg production and 619 for average egg weight. There was no difference in egg production between new synthetic lines and imported breeds. In the analysis of genetic trends, the estimates of genetic parameter in the autosexing lines were similar to those of the general population of layer breeders. This results indicated the consistency of genetic variation from this selection.

Development of an SNP set for marker-assisted breeding based on the genotyping-by-sequencing of elite inbred lines in watermelon (수박 엘리트 계통의 GBS를 통한 마커이용 육종용 SNP 마커 개발)

  • This study was conducted to develop an SNP set that can be useful for marker-assisted breeding (MAB) in watermelon (Citrullus. lanatus L) using Genotyping-by-sequencing (GBS) analysis of 20 commercial elite watermelon inbreds. The result of GBS showed that 77% of approximately 1.1 billion raw reads were mapped on the watermelon genome with an average mapping region of about 4,000 Kb, which indicated genome coverage of 2.3%. After the filtering process, a total of 2,670 SNPs with an average depth of 31.57 and the PIC (Polymorphic Information Content) value of 0.1~0.38 for 20 elite inbreds were obtained. Among those SNPs, 55 SNPs (5 SNPs per chromosome that are equally distributed on each chromosome) were selected. For the understanding genetic relationship of 20 elite inbreds, PCA (Principal Component Analysis) was carried out with 55 SNPs, which resulted in the classification of inbreds into 4 groups based on PC1 (52%) and PC2 (11%), thus causing differentiation between the inbreds. A similar classification pattern for PCA was observed from hierarchical clustering analysis. The SNP set developed in this study has the potential for application to cultivar identification, F1 seed purity test, and marker-assisted backcross (MABC) not only for 20 elite inbreds but also for diverse resources for watermelon breeding.

The Status and Prospect of Poplar Research in Korea (포플러 연구현황과 전망)

  • Populus species have been as a model species in tree breeding and we have enormous varieties resulting from the poplar breeding because of their fast growth performance and short rotation age. New varieties developed in Korea are common italian poplar(P euramericana, I-214, I-476), P euramericana“Eco 28”(Italian poplar No.1) and p. deltoides“Lux”(Italian poplar No.2), which were introduced from foreign countries. As hybrid polars, Hyun-Sasi(p. alba ${\times}$ P. glandulosa No.1, No.2, No.3, No4.), P. nigra x P. maximowiczii and P. koreana x P. nigra val. italica, were developed, and P. davidiana was selected as the result of selection breeding The total plantation areas covered with the new varieties are 935,162ha that include 745,773ha of P. euramericana, 184,636ha of P. alba x P. glandulosa, and other new varieties are 4,735ha. The new poplars are contributed to increase farmer's income as well as bare land tree-planting in Korea. The technologies associated with the poplar species were developed, such as the determination of optimum site for new the poplar species, the crossing method between incompatible poplar species, and the vegetative mass propagation. In the future, poplar species will be considered for phytoremediation species at contaminated areas such as landfill sites or with lives stock's waste water as well as wood production, a shade tree like road-side tree and public park tree.

A New Potato Cultivar "Jayoung", with High Concentration of Anthocyanin (Anthocyanin 함량이 높은 감자 신품종 "자영")

  • Jayoung was bred within the potato breeding program of Highland Agriculture Research Center (HARC), National Institute of Crop Science. It was originated from a cross made between Atlantic as a female and AG34314 as a male parent in 2003. Jayoung was evaluated as a clone number Daegwan 1-104 for the growth and tuber characteristics, yielding ability, and resistance to major potato diseases in the regional yield trials from 2006 to 2007. Finally, this clone was renamed as Jayoung and was presented to Korean Seed & Variety Service in 2007 for registration as a new potato cultivar. Jayoung has a oval shape, shallow eye depth, dark purple skinned and fleshed tuber characteristics. Its mean tuber yields from the regional yield trials was 37.3 MT/ha, and it showed the highest yield in spring cropping. Jayoung showed high resistance to both common scab (Streptomyces scabies) and potato leaf roll virus (PLRV), however it was susceptible to late blight (Phytophthora infestans). The dry matter content of Jayoung was fairy high 20.3%, therefore it is suitable for both chip processing and table usage. Its anthocyanin content was the highest in autumn cropping as 43.7 mg/100 g fresh weight and its mean anthocyanin content was 4.7 times higher than that of Jasim.

Analysis of Genetic Relationship of Pear (Pyrus spp.) Germplasms Using AFLP Markers (AFLP 표지를 이용한 배 유전자원의 유연관계 분석)

  • Amplified fragment length polymorphism (AFLP) marker was utilized for evaluation of genetic diversity of 60 pear germplasms. Twenty selective AFLP primer pairs generated a total of 522 polymorphic amplification products. From UPGMA (unweighted pair-group method arithmetic average) cluster analysis by using polymorphic bands, the pear germplasms were divided into four clusters by similarity index of 0.691. The first cluster (I) included European pears belonging to Pyrus communis and wild species such as P. nivalis and P. cordata. The second cluster (II) included Ussurian pea pears belonging to P. betulaefolia and P. fauriei. The third cluster (III) included pea pears belonging to P. calleryana and P. koehnei. Most of germplasms belonging to P. pyrifolia and P. ussuriensis, and interspecific hybrids were included in the fourth (IV) cluster. Therefore pear germplasms originated from East Asia were closely related to P. pyrifolia and P. ussuriensis. Similarity values among the tested pear germplasms ranged from 0.584 to 0.879, and the average similarity value was 0.686.

A New Potato Cultivar "Hongyoung", with Red Skin and Flesh Color, and High Concentrations of Anthocyanins (껍질과 속색이 붉고 Anthocyanin 함량이 높은 감자 "홍영")

"Hongyoung" showed high resistance to common scab (Streptomyces scabies) and potato leaf roll virus (PLRV), however it was susceptible to both potato virus Y (PVY) and late blight (Phytophthora infestans). The dry matter content of "Hongyoung" was low to medium 17.7%, therefore it is suitable for table use. Its anthocyanin content was the highest in autumn cropping with 31.8 mg/100g fresh weight and it contained 3.6 times higher anthocyanin than the control, Jasim.

Characterization of a new commercial strain 'Guseol' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 느타리 품종 '구슬'의 육성 및 그 특성)

  • To develop new variety of oyster mushroom, 63 intra-specific hybrids between the strain Suhan and #Nongi201 were developed using hyphal anastomosis technique in 2004. The Po2008-275 hybrid between the dikaryon strain 04-154(Suhan x #Nongi201) and the monokaryon strain derived from ASI2487 were developed using hyphal anastomosis in 2008. The Po2008-275 was shown the best cultural characteristics, selected to be a new variety and named as 'Guseol'. The new commercial strain, 'Guseol' had dark grey pilei and grows well under spring and autumn conditions in Korea. The fruiting bodies of 'Guseol' were of an excellent quality in that not only the stipe was thick and long but also the pileus was small and hard. The optimum temperatures for mycelial growth and fruiting body development were $25{\sim}30^{\circ}C$ and $10{\sim}16^{\circ}C$, respectively. Time period required for the initiation of the first fruiting body was about 3 to 5 days depending on the temperatures. The shape of fruiting body was thin funnel shape. Fruiting body production per box($43{\times}43{\times}12cm$) was about $1545{\pm}400.9g$ which was almost 137% quantity compared to that of parental strain 04-154. Relatively low temperature incubation ($11^{\circ}C$) resulted in the development of better quality of 'Guseol' mushrooms. When two different media including potato dextrose medium and mushroom complete medium were compared, the mycelial growth of this mushroom were much faster in mushroom complete medium. Similar results were observed with other variety '#Chunchu2'. Analysis of the genetic characteristics of the new commercial strain 'Guseol' showed a major DNA profile as that of the parental 04-154 when primer URP 1, primer URP 2 and primer URP 5 were used, but different to '#Chunchu2' that was used as a control. This new variety of the dark grey oyster mushroom had smart and high quality image that corresponds well to "health food". We therefore expect that this new strain will satisfy the consumers demand for variety and excellent mushrooms.
