Rancidity Estimation of Perilla Seed Oil using NIR Spectroscopy and Multi-variate Analysis Techniques (근적외선 분광기법과 인공신경망을 이용한 식용유지의 산패 분석)

  • 대부분의 가정과 요식업체, 식품가공업계에서 이용하고 있는 식용유지는 저장 및 가공과정 중에 산패가 빈번하게 일어나게 된다. 기존에는 유지 산패를 측정하기 위해 산가, 과산화물가 등을 측정하는 이화학적인 적정방법을 이용하였는데 실험자의 숙련도에 따라 결과의 오차가 발생할 수 있고, 반복실험으로 인한 시간과 비용이 많이 소모되는 등 여러 제약사항을 포함하고 있어 식용유지의 산패를 실시간 비파괴적으로 분석할 수 있는 기술의 개발에 많은 관심이 모아지고 있다. 따라서, 본 연구에서는 식용유지의 저장조건에 따른 산패정도를 비파괴적으로 평가하기 위한 근적외선 분광분석과 인공신경망 분석기술을 개발하여 그 실효성을 평가하였다. 식물성 식용유지인 들기름을 특정 온도에서 일정한 시간동안 저장하면서 이화학적 적정방법을 통해 산가와 과산화물가를 측정하였으며 동일한 시료의 근적외선 투과스펙트럼을 획득하였다. 수집된 정보를 이용하여 유지 산패 예측 모델을 개발하기 위해 다변량 분석기법 (주성분 회귀분석, 최소자승 회귀분석과 인공신경망 분석)을 적용하였다. 분석 결과, 인공신경망 분석모델이 산가 ($R^2_{tra}:0.9037$, $R^2_{val}:0.8175$, $R^2_{test}:0.8555$)와 과산화물가 ($R^2_{tra}:0.9210$, $R^2_{val}:0.9341$, $R^2_{test}:0.8286$)의 예측 성능이 가장 우수한 것으로 확인되었다. 본 연구의 결과들은 농산물과 식품의 성분 측정뿐만 아니라 다른 산업분야에서도 유용하게 활용될 수 있을 것으로 기대되어진다.

Comparison of Autoxidative Stability and Minor Compounds in Oils Extracted from Bran and Germ of Keumkang Wheat and Dark Northern Spring Wheat (금강밀과 dark northern spring밀의 기울과 배아에서 추출한 기름의 자동산화 안정성 및 미량성분 비교)

  • Autoxidative stability of wheat bran and germ oil extracted from Keumkang wheat (WBG-K) or Dark Northern Spring wheat (WBG-DNS) at $50^{\circ}C$ in the dark was compared by peroxide values (POV) and fatty acid composition by gas chromatography. Changes of minor compounds were monitored by HPLC. WBG-K showed significantly higher linoleic but lower oleic acid content than WBG-DNS. WBG-DNS contained more phospholipids but less tocopherols and carotenoids than WBG-K. POV of oils was increased during storage, with no significant difference in the oxidation rates between two oils. Tocopherols, carotenoids, and phospholipids in both oils were degraded during the autoxidation. Total phospholipids content showed the highest correlation with the degree of oxidation in WBG oils. The results clearly showed that both fatty acid composition and contents of tocopherols, carotenoids, and phospholipids co-affected the autoxidation of WBG oil.

The Study on the Oxidative Stability of Mixed Rapeseed oil with Palm oil (팜유 혼합유채유의 산화안정성에 관한 연구)

  • Two kinds of simple and interesterfying mixed rapeseed oil were prepared according to add 30%, 50% and 70% of coconut and palm oil, then the physicochemical propecties such as the tedency of oxidation, foaming activity were Investigated. The initial AV and POV of interesterifyung mixed rape-seed oils were higher than those of simple mixed oils, but the tendencies of oxidation were similar. The intial value of IV interesterfied rapeseed oils was added in simple mixed rapeseed oil, Induction period were increased about 2.4~4 times. Induction period of intereterifyng mixed oils added palm ell increased about 2~3.2 times. While the foaming Buantity of interesterifing mixed rapeseed oils with palm oil were lower about 28~40% than those of the simple mixed rapeseed oils.

Effects of N-nitrosoethylurea on the Activities of Antioxidant Enzymes from Rat Liver Cell (N-nitrosoethylurea가 쥐 간세포의 항산화효소의 활성에 미치는 영향)

  • N-nitrosoethylurea (NEU) -induced changes of lipid peroxide content, aldehyde metabolic enzyme activities and antioxidant enzyme activities were examined in cultured rat liver cell. Aldehyde metabolic enzymes tested in this investigation were alcohol dehydrogenase and aldehyde dehydrogenase. Several antioxidant enzymes tested were glutathione transferase, superoxide dismutase, glutathione reductase and catalase. When the cell was exposed with various concentrations of NEU, lipid peroxide content increased about 2.5 fold with 6.25 mM NEU. Maximun 2.3 times higher alcohol dehydrogenase activity was found after NEU treatment. About 2 times higher aldehyde dehydrogenase activity could also be observed. Only slight increases of glutathione transferase and catalase activities occurred with NEU treatment. In addition mnximun 1.5 times higher superoxide dismutase activities and 3 times higher glutathione reductase activities were found after NEU treatment. Therefore, it is likely that the increases of superoxide dismutase and glutathione reductase could contribute in a antioxidative process against NEU toxicity.

  • The antioxidative action of petroleum ether, chloroform, chloroform-methamol (2 : 1, v/v), methanol and methanol-water extracts of Panax ginseng roots in the autoxidation of mixed methyl esters of unsaturated fatty acids (MEUFA) was investigated in vitro. All of the extracts of Panax ginseng roots possessed antioxidative activity and inhibited the formation of hydroperoxides in the autoxidation of mixed methyl esters of unsaturated fatty acids (MEUEA). The induction period in the autoxidation of MEUFA was extended by the addition of each extract. The antioxidative activity was more prominent with chloroform and chloroform-methanol (2 : 1, v/v) extracts than with other extracts. The antioxidative activity of each extract estimated by Olcott's oven test and the measurement of peroxide value (POV) showed a similar tendency. From the results obtained, it was conclude that extracts of Panax ginseng roots had remarkable antioxidative activity in the autoxidation of MEUFA in vitro.

Development of an Automatic Packer Using Vacuum Packaging and its Effects on the Rice Quality

  • 본 연구에서는 배아미, 현미, 가능성쌀들 및 진공을 요하는 식품의 품질을 보존하기 위해서 연속식 자동진공포장기를 국내 최초로 개발하였다. 배아미의 진공포장 성공률은 92.6%이었고, 실패율 7.4%에서 진공포장기 자체에서 불완전 테이핑의 밀봉작업으로 진공이 서서히 풀리는 경우가 5.5%, 그리고 진공하기 전 자동비닐포장기에서 포장시 봉지 윗부분 밀봉이 불완전해 1.9%가 진공이 풀렸다. 이에 개발한 자동진공포장기의 진공포장성공률은 약 94.5%수준이었다. 진공포장기로 진공시간에 따른 진공압력을 측정하였고, 배아미 2, 3, 5 kg를 진공포장하는데 소요되는 시간은 각각 6, 8, 11초이었다. 진공포장으로 처리한 배아미와 진공포장 처리하지 않은 배아미를 각각 3재월간 저장실험하여 백도, 함수율, 과산화물가, 산가를 저장기간별로 분석하였다. 그 결과, 백도는 진공처리가 2 감소, 비처리가 4 감소하였고, 함수율은 진공처리가 0.8%감소, 비처리가 1.3%감소, 과산화물가는 진공처리가 최대 1.89 meq/kg까지 서서히 증가하였고, 비처리가 3.45 meq/kg까지 급속히 증가하였다가 서서히 감소하였다. 산가 역시 진공처리가 최대 0.71 mg/g까지 서서히 증가하였고, 비처리의 경우는 0.98 mg/g까지 급속히 증가하였다. 그리고 현미의 진공포장에서도 산가 및 과산화물가 변화가 배아미 경우와 비슷하였다. 또한 진공포장 제품에 인위적으로 쌀벌레들을 넣은 경우 15일 지나면 산소부족으로 모두 사멸하였다. 따라서 개발한 진공포장기로 진공포장한 경우 배아미 및 현미의 품질을 잘 보존할 수 있었다.

젖당과 사멸 살모넬라 함유 사료가 Salmonella typhimurium 인공 감염 육계 병아리의 생산성과 면역 반응에 미치는 영향

  • Effects of dietary lactose or killed Salmonella on the performance, immune response and anti-oxidant system was studied in chicks innoculated with Salmonella typhimurium. In 27 days of age broiler, dietary lactose decreased performance, while dietary lactose and killed Salmonella elevated plasma peroxidase activity and IL-1 level in supernatant of PBMC stimulated with LPS. When broiler chicks innoculated with Salmonella, performance, activities of erythrocyte MnSOD and plasma peroxidase were enhanced after 7 days of the innoculation. Dietary lactose and killed Salmonella increased activity of erythrocyte MnSOD, plasma peroxidase, proliferation of PBMC stimulated with LPS and IL-1 level in the supernatant after 15 days of the innoculation. The result indicated that dietary lactose and killed Salmonella have modulated innate immune response and antioxidant system in broiler chicks innoculated with Salmonella typhimurium.

  • Relative retarding or pro-oxidant effect of 0.02% BHT, 0.01% citric acid and 0.1 ppm $CuSO_4$ $5H_2O$, on the peroxide value and the free fatty acid value development of two groups of edible soybean osils was studied. One group of the oils was stored in a dark place at $40{\pm}0.5^{\circ}C$, and the other group was irradiated 3 hours daily, with direct sunlight. The results of the study were as follow; 1. Peroxide value and free fatty acid value were, in general, more rapidly promoted in the case of the irradiated oils than in case of the oils stored in the dark place. 2. Peroxide value and free fatty acid value were rapidly increased in the case of the sample in addition to $CuSO_4$ $5H_2O $ in both caies. 3. Synergistic effect of citric acid on BHT was strong pronounced in both cases.

A Study on the Mutagenicity of Autoxidized Soybean Oil (자동산화 대두유의 돌연변이원성에 관환 연구)

  • Commercial soybean oil, which did not contain any antioxidant, were autoxidized at $60{\pm}2^{\circ}C$ for 79 days, and the changes of peroxide value(POV), thiobarbituric acid value(TBAV), conjugated dienoic acid(CDA) content, and fatty acid composition of the oil were studied during the 79 day-storage period. The samples with POV S of 0, 150, 300, 500, 450, 400 and 300 meq./kg oil were used for the test of mutagenic activity. The Ames test was carried out with and without metabolic activation. Bacterial strains used in this study were the histidine auxotrophic strains of S. typhimurium TA100, TA1535, and TA102 for the detection of base pair, and TA98 and TA1537 for frame shift mutations. Each series of samples was disso1ved in tetrahydrofuran(inhibitor free) and tested at doses ranging from 0.05 to 5 mg/plate. The autoxidized soybean oil increased significantly(p<0.05) the number of $His^+$ revertant colonies in cases of TA 98 with S9 mix, TA100 without S9 mix, 1535 and 1537 with and without S9, respectively. The samples having the highest peroxide values showed the strongest mutagenicity. It seemed that the amount of hydroperoxides in the oils was closely related to the mutagenic activity of the respective oils.

Effects of Heating Time and Storage Temperature on the Oxidative Stability of Heated Palm Oil (가열시간과 저장온도가 가열팜유의 산화안정성에 미치는 영향)

  • Effects of heating time and storage temperature on the oxidative stability of heated palm oil were studied. Palm oil was heated at $150^{\circ}C$ for 0, 1, 10 or 20 min and stored at 4, 20 or $65^{\circ}C$. The oxidative stability of the sample was evaluated by determining peroxide value of the oil and measuring the volatiles in the headspace of the sample. Significant difference in the peroxide or volatile formation was observed (p<0.05) in heated palm oils between samples stored at 4 or $20^{\circ}C$ and those stored at $65^{\circ}C$. Pentane, hexanal, heptane and total volatiles increased with heating time, while storage temperature did not significantly affect their formation, indicating that heating time played more important role in volatile formation in the heated palm oil than storage temperature. However, adverse results were observed for the formation of peroxide. The interaction effect of heating time and storage temperature on the oxidative stability of heated palm oil was also observed.
