Enzyme Activity and Gene Expression of Cytochrome P450 Involved in Capsidiol Biosynthesis in Solanaceae Plants (가지과식물에서 Capsidiol 생합성에 관여하는 Cytochrome P450 유전자의 발현과 효소활성)

  • Enzyme activity and expression of cytochrome P450 gene involved in the pathway of capsidiol biosynthesis were compared in five different solanaceae plants such as red pepper, green pepper, tobacco, potato and egg plant. Base on genomic DNA and/or RT-PCR results, four solanaceae plants such as red pepper, green pepper, tobacco and egg plant possess P450 gene in the genome and specifically expressed by elicitor treatment. However, potato was appeared to have neither P450 nor cyclase gene in the genome. P450 genes did not show any expression in the plants under normal condition, but showed highly specific expression under elicitation condition in various organs and tissue such as leaf, root, stem and culture cells.

Relationship between the Expression of VEGF, HIF-$1{\alpha}$ and Stage in Non-Small Cell Lung Carcinoma (비소세포성 폐암에서 VEGF, HIF-$1{\alpha}$의 발현과 병기의 상관성)

  • 목적 : 비소세포 폐암에서 HIF-$1{\alpha}$, VEGF의 발현과 예후와 관련된 여러 임상적 표지자 및 병리학적 표지자와의 상관관계를 조사하고자 하였다. 방법 : 이들의 예후인자로서의 의미를 알아보고자 외과적으로 절제한 44예의 비소세포 폐암종을 대상으로 VEGF, HIF-$1{\alpha}$에 대한 면역조직화학염색을 시행하였다. 결과 : WHO 분류에 의한 조직학적인 형태인 편평세포암종 28예, 선암종 16예의 비세포성 폐암이 이 연구에 포함되었다. 비소세포성 폐암 24예와 16예에서 VEGF 및 HIF-$1\alpha$이 발현되었다. 결론 : 이 연구에서 VEGF 발현과 병기와의 상관관계, HIF-$1{\alpha}$의 과발현과 병기 사이에서 통계적으로 유의한 상관관계를 볼 수 없었다. 또한 HIF-$1{\alpha}$의 과발현과 VEGF의 발현 사이에 통계학적으로 유의한 관련성이 없었다.

Proteomic analysis of dehydroascorbate reductase transgenic potato plants (Dehydroascorbate reductase 과발현 형질전환 감자 식물체의 단백질체 분석)

  • Ascorbic acid (AsA) is a strong antioxidant/reducing agent that can be converted to dehydroascorbate (DHA) by oxidation in plants. DHA, a very short-lived chemical, is recycled to AsA by dehydroascorbate reductase (DHAR). Previously, DHAR cDNA was isolated from the hairy roots of the sesame plant, and DHAR-overexpressing transgenic potato plants were generated under the control of the CaMV35S promoter (CaMV35S::DHAR). An increase in transgene expression and ascorbate levels were observed in the transgenic plants. In the present study, proteomic analysis revealed that transgenic plants not only accumulated DHAR in their cells, but also induced several other antioxidant enzyme-related proteins during plant growth. These results suggest that DHAR is important for stress tolerance via induction of antioxidant proteins, and could improve stress tolerance in transgenic potato plants.

Expression of VEGF, p53, Apaf-1 and Caspase-9 in Head and Neck Squamous Cell Carcinoma (두경부 편평세포암에서 VEGF, p53, Apaf-1 and Caspase-9의 발현)

  • 배 경 편평상피암은 두경부의 악성종양 중 가장 흔하며, 임상적인 경과가 불량하다. 따라서 나쁜 예후를 가지는 환자군을 조기에 선별하여 더 적극적인 치료의 시행을 결정짓는 표지자의 필요성이 대두된다. 우리는 일련의 두경부 편평세포암 검체에서 몇몇 분자 표지자의 예후적 유용성을 평가하고자 하였다. 방 법 23예의 두경부 편평세포암 검체를 대상으로 VEGF, p53, Apaf-1, caspase-9의 발현과 몇몇 임상병리학적 지표들간의 연관성을 면역조직화학염색을 통해 조사하였다. 결 과 환자군은 남성이 더 많았으며 평균연령은 63.7세였다. 1기가 5예, 2기가 2예, 3기가 8예, 4기가 8예였다. 평균생존기간은 37.3개월이었다. VEGF단백 발현은 종양의 크기와 통계적으로 유의한 연관성을 보였다. 이와 더불어 VEGF단백 발현은 병기, 그리고 림프관 침습과 연관적인 경향성을 보였다. 그러나 VEGF단백 발현과 생존기간과는 관련성이 없었다. 또한, Apaf-1과 caspase-9의 단백발현은 다른 임상지표, 환자의 생존기간과는 관련이 없었다. 결 론 VEGF단백 발현은 두경부 편평세포암 환자에서 나쁜 임상경과를 예측할 수 있게 하는 표지자로서의 역할을 할 수 있다. 또한 본 연구는 두경부 편평세포암에서 별로 연구되지 않은 Apaf-1과 caspase-9의 발현상태를 밝힌 점에서 의의가 있다.

The Expression of Solute carrier family members Genes in Mouse Ovarian Developments (생쥐의 난소 발달과정에서 Solute carrier family 유전자들의 발현양상)

  • Granulosa cells, which surround the oocyte within the ovarian follicle, play an essential role in creating conditions required for the development of oocytes and follicles. The solute carrier family (SLC) is comprised of influx transporters of steroidal hormones, various drugs, and several other substrates. The differential expression of selected DEGs was confirmed using in situ hybridization analysis. SLC23A3 and SLC39A10 were highly expressed in the ovary. The SLC39A10 gene was expressed in the primordial follicle stage, but SLC23A3 was expressed in the growing follicle stage. Contrastingly, the expression of SLC23A3 was increased in granulosa cells at the growing follicle stage. The differential expressions of SLC23A3 and SLC39A10 between the primordial and primary follicles were additionally confirmed by using follicle isolations. The gene expression profile from the present study may provide insight for future studies on the mechanism(s) involved in primordial-primary follicular transition and suggestions to promote follicular development in ovarian dysfunction.

  • Autoimmune regulator gene (Aire) is expressed in the thymus and controls the expression of peripheral self-antigens, known as promiscuous genes. Aire and promiscuous genes are involved in T cell tolerance and autoimmunity in the thymus. Here, we identified Aire-expressing fibroblastic reticular cell (FRC), which was derived from mouse lymph node and also expressed in insulin promiscuous antigen. The expression of insulin was increased in cultured FRC over-expressed with Aire. These data suggest that Aire regulates promiscuous gene expression in FRC, and that this function might be under peripheral selection control.

  • Haptoglobin (Hp) is an acute phase protein, and its plasma level increases consistently in response to inflammation. To investigate the biological role of Hp in lung epithelial cells, the gene expressions of cyclooxygenase-2 (COX-2) and inflammatory cytokines were analyzed using human Hp gene-transfected A549 cells. Western blot analysis showed that COX-2 expression was markedly increased in Hp DNA-transfected cells (stable transfection and transient transfection) compared with that in vehicle DNA-transfected cells. When the Hp-expressing cells were treated with $1{\mu}g/ml$ of LPS or 100 U/ml of $IL-1{\beta}$ for 24 hr, the COX-2 expression was synergistically up-regulated. ACP-based PCR data demonstrated the Hp decreased SPARC expression, but increased IL-4 and S100AI expressions. These findings suggest that the Hp acts as a pro-inflammatory mediator in lung inflammation.

TFAP2C Promotes Cell Proliferation by Upregulating CDC20 and TRIB3 in Non-small Cell Lung Cancer Cells (비소세포폐암 발달 과정에서 TFAP2C에 의해 발현되는 CDC20과 TRIB3의 원암유전자 기능에 관한 연구)

  • Non-small cell lung cancer (NSCLC) has the infamous distinction of being the leading cause of global cancer-related death over the past decade, and novel molecular targets are urgently required to change this status. We previously conducted a microarray analysis to investigate the association of transcription factor activating enhancer-binding protein 2C (TFAP2C) with NSCLC and revealed its oncogenic roles in NSCLC development. In this study, to identify new biomarkers for NSCLC, we focused on several oncogenes from the microarray analysis that are transcriptionally regulated by TFAP2C. Here, the cell division cycle 20 (CDC20) and tribbles pseudokinase 3 (TRIB3) were subsequently found as potential potent oncogenes as they are positively regulated by TFAP2C. The results showed that the mRNA and protein levels of CDC20 and TRIB3 were down-regulated in two NSCLC cell lines (NCI-H292 and NCI-H838), which were treated with TFAP2C siRNA, and that the overexpression of either CDC20 or TRIB3 was responsible for promoting cell viability in both NSCLC cell lines. In addition, apoptotic levels of NCI-H292 and NCI-H838 cells treated with TFAP2C siRNA were found to be suppressed by the overexpression of either CDC20 or TRIB3. Together, these results suggest that CDC20 and TRIB3 are positively related to NSCLC tumorigenesis and that they should be considered as potential prognostic markers for developing an NSCLC therapy.

Clustering and Leaf Ordering for Gene Expression Profiles (유전자 발현 데이터에 대한 클러스터링과 리프오더링 연구)

  • 계층적 클러스터링(hierarchical clustering)은 유전자 발현 데이터를 분석할 때 일반적으로 사용하는 방법이다. 계층적 클러스터링의 결과물은 유전자 발현 데이터의 덴드로그램이다. 이 덴드로그램에서 인접한 리프 노드들간의 유사도는 높아지게 하고 멀리 떨어진 노드들간의 유사도는 낮아지게 하기 위해서, 리프 노드들을 재배열하는 과정을 리프오더링이라고 한다. 본 논문에서는 전체 리프 노드들을 대상으로 하는 리프오더링 알고리즘들을 변형하여 각 클러스터별로 리프오더링을 하는 접근방식을 제안하고, 기존의 리프오더링 알고리즘을 사용했을 때의 결과와 제안하는 접근방식을 사용했을 때의 결과를 비교 분석하였다.

  • 주어진 셀이나 조직에 발현된 단백질 프로파일의 구조적인 분석을 다루는 단백질체학(Proteomics) 연구에 있어서, 질병에 대한 마커 단백질(marker proteins)을 도출(identification)하는 것은 핵심 논점 중 하나이다. 수십 개의 샘플로부터 추출한 셀이나 조직 내에는 수많은 단백질이 포함되어 있으며, 존재하는 단백질의 질병에 의한 발현량(expression level) 변화 및 임상 특성에 의한 영향을 분석하기 위해서 데이터베이스와 데이터 마이닝 기술의 활용이 효과적이다. 본 논문에서는 질병 일 임상 특성에 따른 단백질의 발현량 변화를 분석하기 위한 OLAP 데이터 큐브(Data cube)의 응용 방법과 단백질 데이터의 분석에 적합한 척도(measure)를 제안하고, 유효성을 보인다.
