  • The potentials of some Korean wild plants as a phytoremediator for cleaning heavy metal pollution were measured. Several plant species, Ambrosia trifida, Brassica juncea, Rumex crispus, and Abutilon theophrasti screened previously for phytoremediator were treated with cadmium and copper solution. In order to know the growth response to heavy metal stress, the plants were cultivated in hydroponic system containing heavy metals with different concentration. To know the effects of heavy metals on emergence and seedling growth, seeds of 4 species were sown in the pot and watered with heavy metal solution adjusted pH to 6.5, 5.5, and 4.5. A proposed species as potential phytoremediator, A. trifida, showed tolerance to $20{\mu}mol/L$ Cd and $80{\mu}mol/L$ Cu in nutrient solution without apparent growth reduction, and up to $100{\mu}mol/L$ Cd and $400{\mu}mol/L$ Cu without critical visual injury. Up to 311mg/kg of Cd and 369mg/kg were accumulated in dried aerial part in A. trifida. In contrast, A. theophrasti showed injury at $400{\mu}mol/L$ Cu. Significant differences were shown in Cu accumulation among the four species. A. trifida had much higher concentrations of Cd in the shoot, whereas R, crispus accumulated higher concentrations of Cd in the shoot. Testing plant species showed reduced emergence rate with heavy metal treatment. When pH was lowered, the emergence and seedling growth were affected severely with heavy metal. We can suggested that A. trifida was the most proper species for phytoremediation in heavy metal-polluted regions.

  • To establish taxonomic system of morphologically similar species of small-spored Alternaria, phylogenetic analysis of internal transcribed spacer (ITS 1, ITS 2 and 5.8S rDNA) and mitochondrial small subunit (mt SSU) rDNA sequences and URP-PCR fingerprinting analysis from 11 species ofAlternaria were performed. Phylogenetic analysis of ITS and mt SSU rDNA sequences revealed that 10 out of 11 species of the smallspored Alternaria were phylogenetically identical with a bootstrap value of 100%. A. infectoria only was phylogenetically differentiated from the other species. The results suggest that the 10 small-spored Alternaria species are very closely related evolutionally and the markers can not be used for differentiation of the smallspored Alternaria species. URP-PCR fingerprinting analysis from eleven species of smallspored Alternaria using 10 URP primers showed that it was possible to differentiate the species, although genetic similarities were found among the species. The Alternaria sp. from common pokeweed could be distinguished from other species by URP-PCR analysis, and it was considered as a new species. A. infectoria could be easily distinguished from the other 10 species by phylogenetic analysis of ITS and mt SSU rDNA sequences and the URPPCR fingerprinting analysis.

  • 1) The purpose of the present study is to investigate the effects of culture filtrates of Fusarium oxysporum f. vasinfectum which is known to produce fusaric acid (wilt toxin) on the germination of sesame seeds and the growth of sesame seedlings. 2) Culture filtrates of Fusarium oxysporum 1. vasinfectum used in this study strongly or weakly inhibited the germination and bring about necrosis accompanying black discoloration of sesame seeds. 3) Varietal difference of sesame in the germination response on the culture filtrates of Fnsarium oxysporum f. vasinfectum is not shown in this study. 4) This study reveals that differential five strains of Fnsarium oxysoprum f. vasinfectum used in this study differ greatly in the toxicity of culture filtrates inhibiting the germination of sesame seeds. 5) In the seedling bed added with culture filtrates of Fnsarium oxysporum f. vasinfectum, the growth of shoot as well as root system of sesame seedlings are notably inhibited and necrotic black discoloration appear on both shoot and root system. But in the seedling beds added with weaker concentration of culture filtrates $/(10\%)$ the growth of shoot is slightly promoted. 6) In culture of sesame seedlings with Knop's solution containing 1 to 3 per cent culture filtrates, the growth of shoot as well as root system are slightly retarded" and till the time of development of the third leaves the whole stem and leaf petiole tissue are weakened so that they become thread like accompanying brown discoloration, interveinal light brown area appear in the second leaves, and the third leaves curl from both sides towards the middle with necrotic brown discoloration, especially symptoms of injury on the third leaves are nearly similar that of the leaves of wilted sesame in the field. 7) A pararell relationship is not found between toxicity of culture filtrates and pathogenicity of five differential strains of Fnsarium oxysporum f. vasinfectum used in this study.
