Crude extract of Coptis japonica root-stem was evaluated for antifungal activity against Phytophthora capsici, Fusarium oxysporum, Colletotrichum dematium, Colletotrichum truncatum, Botrytis cinerea, Botryosphaeria dothidea and Alternaria porri, and antifungal active compound from the extract was identified. In addition, the usefulness of the extract for some plants disease control was investigated. Crude extract of C. japonica root-stem exhibited antifungal activity against P. capsici, F. oxysporum, C. dematium, B. cinerea, B. dothidea and A. porri. Antifungal activity of the substance isolated from C. japonica root-stem was similar to a standard chemical berberine-Cl. Red-pepper fruit rot, sesame stem rot and welsh-onion alternaria leaf spot were effectively controlled by the crude extract of C. japonica root-stem. Phytotoxicity was not observed in the red-pepper and welsh-onion leaves and red-pepper and strawberry fruits with exogenous foliage application of the crude extract. Seeds germination and radicle growth of red-pepper and sesame were inhibited by the crude extract of C. japonica root-stem. 4.24g of yellowish compound per 100g of C. japonica root-stem was obtained. The compound was identified as berberine-Cl by HPLC.
The antibacterial protein gene, nuecin was expressed in Sf9 cells using baculovirus expression vector system (BEVS). The antibactetial activity of mature nuecin against Pectobacterium carotovorum subsp. carotovorum, Ralstonia solanacearum and Pseudomonas tolaasii was significantly high, demonstrating that nuecin had a wider antibacterial spectrum on gram negative and positive bacteria. The result appears to be superior to other antibacterial peptide, attacin. The nuecin was purified by SP-sepharose and Mono Q HR ion-exchange chromatography, and then by Superdex 200 HR 10/30 column. The purified nuecin is quite stable at 80$\^{C}$ and 100$\^{C}$ for several hours of incubation and in a wide pH range (pH 2-12).
These experiments were conducted for decrease of injury by continuous cropping in the peanut fields of Chonbuk Wangkungarea. The continuous cropping field for four years was used in this experiment. Italian ryegrass and rye were cultivated andlime materials were distributed for improvement of soil fertility. The results were as follows; 1. Forage crops were cultivatedand lime materials were distributed on the continuous cropping field of peanut. The organic matter content of the expermentalplot cultivating Italian ryegrass was only 1.25%. The organic matter content of soil cultivated Italian ryegrass after distributedmagnesium lime was 1.37% and that of soil cultivated Italian ryegrass after distributed gypsum was 1.30%. It was highcontent comparing to that of soil distributed lime materials only. The organic matter content of soil cultivated rye after distributed gypsum was 1.77%. 2. The phosphate content of soil cutivated Italian ryegrass was 332ppm. The phosphate content ofsoil cultivated Italian ryegrass after distributed magnesium lime was 34Oppm and that of soil cultivated Italian ryegrass afterdistributed gypsum was 31 2ppm. The phosphate content of soil cultivated rye only was 386ppm. The phosphate content ofsoil cultivated rye after distributed gypsum was 41 8ppm. This phosphate content was lower than that of soil distributed limematerials only. 3. The phytotoxin content of soil cultivated Italian ryegrass after distributed magnesium lime was decreased to17.7% and that of soil cultivated Italian ryegrass after distributed gypsum was decreased to 25.3%. The phytotoxin content ofsoil cultivated rye after distributed magnesium lime was decreased to 12.0% and that of soil cultivated rye after distributedgypsum was decreased to 12.8% comparing to the phytotoxin content of soil distributed lime materials only. Italian ryegrasswas effective to decrease phytotoxin among the forage crops and gypsum was effective among the lime materials. 4. Abacterial wilt and a late spot of peanut which were known as, main reason of continuous cropping failure were surveyed.lnccidence of a bacterial wilt was 3.4% in the plot cultivated Italian ryegrass only and that was 2.9% in the plot cultivated ryeonly. lnccidence of a bacterial wilt was 2.5% in the plot cultivated Italian ryegrass after distributed magnesium lime and thatwas 2.3% in the plot cultivated rye after distributed gypsum. Inccidence plot cultivated forage crops was lower than that of plotdistributed lime materials. 5. Inccidence of a late spot was high in the plot cultivated forage crops ony, but it was low in the plotcultivated forage crops after distributed lime materials comparing to that of the control plot. 6. The growth and yield of peanutwere bad in the plot cultivated forage crops only comparing to the control plot distributed lime materials only. These resultswere same in the plot cultivated rye after distributed lime materials, but the growth and yield were grown up in the plotcultured Italian ryegrass after distributed lime materials.
This study was conducted to control the seed borne pathogens (Alternaria spp.) of carrot and to examine the seed germination rate by using 13 environment friendly agricultural materials instead of conventional chemicals for organic cultivation. The growth inhibiting effects on pathogens showed the different responses according to each agricultural material and effective 7 materials against seed borne pathogens were selected. Among 7 materials, the carrot seeds sterilized with plant extracts, Tanger Stop and Land Saver were not germinated at all. The germination rate of seeds sterilized with other materials showed the similar levels with reference chemical (Benlate-T) and non-treated seeds. Infection rates of seeds sterilized with seaweed extract, Bellopper for controlling A. radicina and plant extract, Ssial-100 for A. dauci were similar or low infection rate compared with reference chemical. The germination rate and root length of seeds sterilized with Bellopper and Ssial-100 was similar or superior to those of chemical sterilized seeds. The optimal condition seemed to be a little bit different depending on the concentration of materials. As a result, the sterilization of carrot seeds by using the environment friendly materials could be effectively utilized as a technology to inhibit the infection of seed-borne pathogens.
In 2018, symptoms of black rot on apple (Malus pumila var. dulcissima KOIDZ) cv. Fuji were observed in Yeongcheon-si, Korea. The fruit decay symptoms consisted of purple pimples spots, black rot around the seed cavity (calyx end), mummified fruit. To isolate the causal agent, small fragment (2 to 3 mm) of decayed tissue from the lesion margin were placed on WA or PDA. Fungal colonies on PDA produced dense white aerial mycelium, becoming dark gray with age. Pycnidia and conidia were observed under a light microscopy. The shapes of conidia were aseptate, ovoid, rounded at both ends, and $21.7-28.3{\times}9.9-15.3{\mu}m$. Based on morphological and cultural characteristics, this fungus was identified as Diplodia seriata. To confirm its identity, two loci (ITS and ${\beta}$-tubulin) were sequenced for molecular identification. BLAST searches indicated 100% identity with D. seriata. A pathogenicity test was conducted with isolates on Fuji apples. The apples were inoculated with mycelial plugs (5 mm) from 7-day-old cultures of the putative pathogens. All inoculated apples developed rot symptoms identical to the original symptoms, from which D. seriata were reisolated, fulfilling Koch's postulates. This study is the first report of black rot caused Diplodia seriata on apple.
This study was carried out to investigate the feasibility for the use of environmental-friendly materials and the effective recycling of spent mushroom compost(SMC) after cultivation of Button Mushroom, Agaricus bisporus. SMC of white button mushroom contained diverse microorganisms including fluorescent Pseudomonas sp., Bacillus sp., Tricoderma sp. and Actinomycetes. These isolates showed the extensive antifungal spectrum against plant pathogen. Among of the isolates, fungal pathogen such as Alternaria brassicicola, Phytophtora melonis, Phytophthora capsici and Colletotichum gloeosporioides strong showed strong antagonistic activity. 45.8% of the isolates were actively colonized on the pepper root and 5.8% showed rhizosphere competent of >$5{\times}10^2cfu\;root^{-1}$. The plant growth promotion ability of the collected isolates were tested in pot experiments using red pepper seedling. Among them, 62.7% showed pepper growth promoting ability and growth of pepper root showed superior to the control. The germination of pepper treated with aqueous extracts of non-harvest SMC completely inhibited at concentration of more than 33%. The sterilization of SMC resulted in higher inhibition of germination and early growth of pepper. These results suggest that spent mushroom compost(SMC) of Button Mushroom may have adequately the feasibility for the use with environmental-friendly materials.

The pathogenic fungi associated with blight of leaf and stem in peony were leaf spot (Alternaria sp.), powdery mildew (Erysiphe aquilegiae) and rust (Cronartium flaccidum). The infection of leaf spot and powdery mildew begins from late April to midMay and rust was infected in early June. Blight time of aerial part in peony started from late May and the ratio of blight on leaf and stem was more than 50% in late Aug. Yields of root by the incidence time of blight of leaf and stem were 69.1% in late June, 65.4% in late July and $87.6{\sim}92.7$% in August and September. The number of root of more than 10mm in root diameter blighted in late June and July was much lower than in August, but the paeoniflorin content in the former was much higher than the latter.
This study was carried out to isolate antagonistic bacterium against Sclerotium cepivorum causing Allium fistulosum white rot. Total of 146 strains were isolated from A. fistulosum roots. The isolates were screened for antagonism to S. cepivorum and the isolated strain No. AL-1 was selected among these bacteria. It was identified as Serratia plymuthica based on morphological and physiological characteristics according to the Bergey's mannual of systematic bacteriology and 16S rDNA sequences methods. Serratia plymuthica AL-1 showed broad spectrum of antifungal activities against plant pathogenic fungi Alternaria altrata, Colletotrichum gleosporioids, Phoma sp., Rhizoctonia solani, Sclerotinia sclerotiorum, Stemphylium solani, Fusarium oxysporium niveum but not inhibited Didymella bryoniae. When S. plymuthica AL-1 cultivated in the TSB medium containing 1% colloidal chitin, the high molecular fraction (>10 kDa) have chitinase activity (3.2 units/ml) and the low molecular fraction (<10 kDa) have not chitinase activity. Oppositely, after heat treatment (80℃ for 30 min) of the cultivation supernatant, the high molecular fractions have not antifungal activity but the low molecular fractions have antifungal activity.
Sooty leaf blight was found on Dendrobium sp. in several farmers' fields located in Namji-eup, Changnyeong-gun, Gyeongnam province, Korea in 2001. Symptoms of the disease appeared on leaves. Sooty leaf spots were started with amphigenous, subeircular to irregular spots, with light grayish brown to black color with definite margin lines on the upper surface of leaves. Infected leaves became defoliated and whole plants eventually were died. The infection rates of the disease in the surveyed area reached up to 64.8%, in the early September. Conidiophores of the causal fungus were dark grayish brown in color, densely fasciculate, straight, curved to sinuous, branched, $5{\sim}10$ septate and $63{\sim}164{\times}3.2{\sim}4.9{\mu}m$ in size. Conidia were pale to olivaceous in color, obclavatecylindric, straight to slightly curved in shape, $1{\sim}5$ septate and $46{\sim}98{\times}3.2{\sim}3.9{\mu}m$ in size. The optimum temperature for mycelial growth of the fungus was $25^{\circ}C$. The fungus was identified as Pseudocercospora dendrobii on the basis of its mycological characteristics. This is the first report on sooty leaf light of Dendrobium sp. caused by Pseudocercospora dendrobii in Korea.
Pear cultivar 'Supergold' (Pyrus pyrifolia var. culta Nakai) was originated from the cross between 'Chuwhangbae' and 'Manpungbae' with the aims of improving the fruit quality of 'Chuwhangbae' cultivar at Pear Research Station of National Institute of Horticultural & Herbal Science, Rural Development Administration in 1994. 'Supergold' was preliminarily selected in 2002 and named in 2008. The tree shows a vigorous growth habit and semi-spread characters like as 'Manpungbae'. Furthermore, it has sufficient flowers and carries abundant pollen grains, so it can also be used as a pollinator. 'Supergold' is highly resistant to black leaf spot (Alternaria kikuchiana) in the field condition. The optimum harvest time is around Sep. 11th, which is ahead of 'Whangkeumbae' about 5 days in the harvest period. The fruit shape is oblate and fruit skin color is greenish-white at harvesting time. The average weight of fruit is 570 g, and the soluble solids content is $13.6\;^{\circ}Brix$. The flesh is very soft and juicy, and renders good eating quality. Shelf life is about 6 months under the cold storage condition. To determine the self-incompatibility (SI) genotype of 'Supergold' pear cultivar, it was crossed with other cultivars of which SI genotypes have already known. The result of cross-pollinations of 'Supergold' with other cultivars showed relatively high rates of fruit set from 64.5% to 91.0%, except for the cross with pollens of 'Nijisseiki' that represented only 28.8% of fruiting rate. Although sometimes the stigma of 'Supergold' crossed with 'Hayatama', 'Chojuro', and 'Nijisseiki' showed malformed pollen tube tips, 'Supergold' is generally supposed to have cross-compatibility with all other pollen donor cultivars. It is considered that the S-allele of 'Supergold' is $S_3S_4$, which is based on the result of PCR-RFLP.
