Stylopine is a major component of the leaf of Chelidonium majus L. (Papaveraceae), which has been used for the removal of warts, papillomas and condylomas, as well as the treatment of liver disease, in oriental countries. Stylopine per se had no cytotoxic effect in unstimulated RAW 264.7 cells, but concentration-dependently reduced nitric oxide (NO), prostaglandin E$_2$ (PGE$_2$), tumor necrosis factor-a (TNF-$\alpha$) and interleukin-1$\beta$(IL-1$\beta$), and the IL-6 production and cyclooxygenase-2 (COX-2) activity caused by the LPS stimulation. The levels of inducible nitric oxide synthase (iNOS) and COX-2 protein expressions were markedly suppressed by stylopine in a concentration dependent manner. These results suggest that stylopine suppress the NO and PGE$_2$ production in macrophages by inhibiting the iNOS and COX-2 expressions. These biological activities of stylopine may contribute to the anti-inflammatory activity of Cheli-donium majus.
Objectives This study was conducted in in vitro environments to investigate the anti-inflammatory effect and expression mechanism of Gyejigeojakyakgabuja-tang (GJBT). Methods In the experiment, an inflammatory response was induced with lipopolysaccharide in macrophage RAW 264.7 cells treated with GJBT, and the production of nitric oxide (NO), prostaglandin E2 (PGE2), and cytokines were measured, and inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) were measured. The intracellular gene and protein expression levels of cytokines were measured. Results In vitro, GJBT significantly reduced intracellular NO, PGE2, and cytokine production in a concentration-dependent manner. The intracellular iNOS, COX-2, and Cytokine gene expression levels were significantly decreased, and the intracellular iNOS, COX-2, and cytokine protein expression levels were significantly decreased in a concentration-dependent manner. Conclusions These results show that GJBT has an anti-inflammatory effect.

Smilacis Chinae Radix has been used as an anti-inflammatory agent. This study was performed to anti-inflammatory and MAP kinase signaling pathway in vitro. Experimental studies were obtained by measuring the Cytotoxicity, production of NO, PGE2, TNF-$\alpha$ and protein level of catalase, SOD, MAP kinase, The results were summarized as follows: Smilacis Chinae Radix was not cytotoxic effects against Raw264.7 and HEK293 cells. Concentration of $100{\mu}g/m{\ell}$ Smilacis Chinae Radix inhibited the production of NO in the Raw264.7 cell stimulated with LPS. All concentrations of Smilacis Chinae Radix not significantly inhibited the production of PGE2 in the Raw264.7 cell stimulated with LPS. All concentrations of Smilacis Chinae Radix did not inhibit the production of TNF-$\alpha$ in the Raw264.7 cell stimulated with LPS. Smilacis Chinae Radix has not effect of blocking NF-${\kappa}B$ into nucleus in LPS-induced macrophage Raw264.7 cell. Smilacis Chinae Radix has the effect of Cytoprotection through activation of ERK and inhibition of p38 and JNK. Accordingly the results show Smilacis Chinae Radix could induce anti-inflammation and Cytoprotection effects against In vitro, but it needs more research on the precise mechanism of such effects.

Eucommia ulmoides OLIVER (EU) is a traditional Korean herbal used for the treatment of rheumatoid arthritis (RA). In the present study, the molecular pharmacology basis of its anti-inflammatory effect is revealed in this work, EU was studied in lipopolysaccharide (LPS)-activated macrophage cells (RAW 264.7) as an established inflammation model. After activation, nitric oxide (NO) production and iNOS mRNA were measured by using a colorimetric assay (Griess reagent), and reverse transcription polymerase chain reaction (RT-PCR), respectively. The change in the content of $PGE_2$, $TNF{\alpha}$, and IL-6 was concurrently monitored by ELISA. In results, we found that in the concentration range without showing cytotoxicity, EU produced a remarkable anti-inflammatory effect and showed a dose-dependent inhibition of LPSinduced NO production. Compared with indomethacin, EU has more potency and a specific action of NO inhibition, $PGE_2$, IL-6, and TNF-${\alpha}$ inhibition. These results suggest that EU may be a suitable herbal medicine to yield the greatest anti-inflammatory activity for food additives and medicine.

Objectives : Allergic disease has been well known as an IgE-dependent immunologic response. Recently, interest about the late inflammatory reaction has grown up as well as early allergic reaction characterized by IgE and mast cell. The purpose of this study was to find the anti-inflammatory effect of Gamiyunjo-tang(GMYJT) in allergic reaction. Methods : The experiment was performed using Raw 264.7 cells pretreated with GMYJT extracts. In this study, we observed the toxicity of cells by MTT analysis and measured the production of LPS-induced NO, $PGE_2$, IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ at a concentration of 50, 100, 200 and $400{\mu}g/ml$. Results : No toxicity of GMYJT (50, 100, 200, $400{\mu}g/ml$) on RAW 264.7 cells was found after 24 hours incubation. LPS-induced NO production was reduced after treatment with GMYJT (100, 200, $400{\mu}g/ml$)(P<0.05). $PGE_2$ was reduced after treatment with GMYJT (100, 200, $400{\mu}g/ml$)(P<0.05). IL-$1{\beta}$ did not decrease at any dose. IL-6 decreased at 200, $400{\mu}g/ml$(P<0.05). TNF-${\alpha}$ production decreased only at $400{\mu}g/ml$(P<0.05). Conclusions : These data suggest that GMYJT has anti-inflammatory effects in late allergic reaction.
The chloroform and the ethyl acetate fractions from the roots of Acanthopanax chiisanensis exhibited a significant inhibition of prostaglandin E$_2$ (PGE$_2$)production in rat peritoneal macrophages stimulated by the protein kinase C activator, 12-O-tetradecanoylphorbol 13-acetate (TPA). Hyperin was isolated as an active principle from the ethyl acetate fraction. It sup-pressed not only $PGE_2$ production but also nitric oxide (NO) production in vitro in a concentration dependent manner. their $IC_{50}$, being 24.3 and $32.9{\;}{\mu}M$, respectively. Hyperin also caused a significant inhibition of increase in acetic acid-induced vascular permeability in mice in vivo.
Objectives: This study was to investigate the effect of high frequency electro-acupuncture (EA) at $ST_{36}$ acupoint on the Freund's Complete Adjuvant (FCA)-induced arthritis in rats. Methods : Arthritis was induced by intradermal injection of FCA into base of tail. Experimental groups were divided into 4 groups; Normal, Control, $ST_{36}$ and Non-Acupoint (NA). $ST_{36}$ group was treated by 120Hz EA at $ST_{36}$ acupoint. Body weight, paw edema volume and ankle joint thickness were measured after treatment. And we investigated the effects of 120Hz EA via WBC count, segmen neurophil, lymphocyte, PGE2 assay and NADPH-d histochemistry. Results and Conclusions : The mean of body weight and ankle joint thickness of $ST_{36}$ group was increased compared with control group. The present study was designed to evaluate effects of the commonly used NSAIDs(acetaminophen, aspirin, and ibuprofen) on human periodontal ligament cells. Human periodontal ligament cells were grown from a cell line provided by Kyungpook National University. Effects of NSAIDs on the proliferation of human periodontal ligament cells were assessed using MTT assays. And then $PGE_2$ concentrations were determined by ELISA and the changes of cellular configuration were found by electron micrograph. The results were as follows; 1. The MTT assay demonstrated that the commonly used NSAIDs(acetaminophen, aspirin, and ibuprofen) had not significant cytotoxic effect on human periodontal ligament cells. 2. NSAIDs inhibited the $PGE_2$ synthesis of human periodontal ligament cells compared with the control group. These inhibitory effects had no significant differences with NSAID type and concentration. 3. Electron micrographs of human periodontal ligament cells treated with NSAIDs showed more narrow and irregular shape.
To investigate the effects of antioxidant activities and hair loss prevention of extracts from Platycodon grandiflorum, we've prepared chloroform (CF) and ethylacetate fractions (EA) extracted from P. grandiflorum. In the results of DPPH radical scavenging assay, the two fractions showed dose-dependent antioxidant activities. Furthermore, in the ABTS assay, the two fractions exhibited the inhibitory effect over 90% at 10, 50, 100, 200 mg/mL. To investigate the inflammation inhibitory effect, we used RAW264.7 cells, these extracts were inhibited inflammatory reaction by suppressing the production of nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) in dose-dependent manner. In the assay of HaCaT cells' proliferation, the 24 hr treatment of the extracts significantly accelerarted cell proliferation in the range of concentrations used. The two fractions inhibited the proliferation of Malssezia furfur, the bacteria induce the dandruff. Finally, the CF could significantly inhibit the DHT production at 1, 10, 50, $100{\mu}g/mL$, but EA showed the inhibitory effect at the concentration over $50{\mu}g/mL$. The overall results of this study suggest that the chloroform (CF) and ethylacetate fractions (EA) from P. grandiflorum could be a useful raw material for the hair loss prevention products.

Objectives We have studied to know effects of Bangkeehwangkee-tang on the monosodium iodoacetate (MIA) induced osteoarthritis in rats. Methods Osteoarthritis was induced by injection of monosodium iodoacetate into knee joint cavity of rats. Rats are devided into 4 groups; Sham, control, Bangkeehwangkee-tang, Shinbaro. Sham group was injected by normal saline into knee joint cavity only. Control group was taken no treatment and Experimental groups were taken extracts of Bangkeehwangkee-tang and Shinbaro by orally once a day for 4 weeks. The content of TNF-$\alpha$, IL- $1{\beta}$ and IL-6 in synovial fluids were analysed. COX-2 in chondrocytes and blood PGE2 concentration were analysed. Histopathological examination was performed by Safranin-O fast green staining and Mankins score checking. Results The content of TNF-$\alpha$, IL-$1{\beta}$ in experimental groups were decreased compared with control group. COX-2 in chondrocytes and blood PGE2 concentration in experimental groups were decreased compared with control group. Histopathologically, osteoarthritic scores of experimental groups were decreased compared with control group. But the content of IL-6 in synovial fluids was not significantly compared with control group. Conclusions According to these results, it can be suggested that Bangkeehwangkee-tang has anti-arthritic effects on the MIA induced osteoarthritis in rats.
