A Study on the Acute Toxicity and Accumulation of Mercuric Chloride to Goldfish (Carassius auratus) (Mercuric Chloride가 붕어(Carassius auratus)에 미치는 급성독성 및 축적에 관한 연구)

  • This experiment was performed to study the acute toxic effects of mereuric chloride(HgCl$_{2}$) in goldfish (Carassius aurams). In the study, Lc$_{50}$ values were determined for each 24, 48, 72 ,and 96-hr and mercury bioassay during the tests were performed. in five parts gill, kidney, liver, muscle and egg. The results isummarized were as follows 1. In the range of acute lethal toxicity for goldfish was 0.15-1.50 mg/l and the Lc$_{50}$ values decreased with exposure time. 2. The Lc$_{50}$ values of HgCl$_{2}$ for goldfish at 24, 48, 72 and 96 hour were 0.64, 0.53, 0.46, and 0.39 mg/l, respectively. 3. In the 400 $\mu$g/l treatment group, the average levels of mercury residues in the gill, kidey, liver, muscle and egg during 96 hour were 107.02 $\mu$g/g, 8.65 $\mu$g/g, 6.87 $\mu$g/g, 0.61 $\mu$g/g, and 0.82 $\mu$g/g, respectively. 4. The mercuny residues of gill was up to 12-175 times greater than mercury contents found in the other tissues.

  • The clastogenic effects of the diesel emission particle extract (DEPE), mercuric chloride and lead acetate were examined by the mouse bone marrow micronucleus test. DEPE had a potent clastogenic effect by intraperitoneal injection with dose-response between 100 and 300mg/kg b.w.. Mercuric chloride and lead acetate also gave a clastogenic effects but mercuric chloride only had a dose-response between 1 and 3mg/kg b.w.. When DEPE was administrated with mercuric chloride or lead acetate, the frequency of micronucleated cells was slight but not significant increase in comparision to a single treatment with DEPE alone.

  • This study investigates the effects of aspect ratio (transport length-to-width) on diffusive-convection for physical vapor transport processes of $Hg_2Cl_2-NO$ system. For a system with the temperature difference of 20 K between an interface at the source material region and growing crystal interface, the linear temperature profiles at walls, the total molar fluxes at Ar = 2 are much greater than Ar = 5 as well as the corresponding nonuniformities in interfacial distributions due to the effect of convection. The maximum total molar flux at the gravitational acceleration of 1 $g_0$ is greater twice than at the level of 0.1 $g_0$, where g0 denotes the gravitational acceleration on earth. With increasing aspect ratio from 2 to 5, a diffusive-convection mode is transited into the diffusion mode, and then the strength of diffusion is predominant over the strength of diffusive-convection.

  • Electron microscopic studies were made to investigate changes in the fine structure of the liver, kidney and gills of Carassius carassius L. following exposure to 1 and 2.5 ppm of $HgCl_2$. The following results were obtained: 1. In the mercury-treated liver cells, an increase in the number of lysosomes were noticed. These lysosomes appeared to be of two types; round ones containing some crystalline structures and others with phagocytosed glycogen granules and mitochondria. Also observed were mitochondrial swelling where the matrix appeared less electrondense, and segregation of the nucleoli in the nucleus. 2. In the kidney, mercury treatment resulted in thickening of the basement membrane of the glomerulus, and appearance of vacuoles and cytoplasmic bodies in the proximal convoluted tubule. The vacuoles seemed to be formed from mitochondria. Nuclear shrinkage was also noticed at 2.5 ppm of $HgCl_2$. 3. Many large and small lysosomes appeared in response to mercury in the epithelial cells of the gill lamella. Also the lamellar membrane became fuzzy in appearance. 4. It can be concluded from these results that mercury-induced changes in the fine structure are associated with activation of detoxication processes and impairment of energy metabolism.

A Carbazole-Attached NO2S2-Macrocycle Exhibiting Hg2+ and Cu2+ Selectivity

  • A synthesis and cation-induced fluorescent behavior of the carbazole-attached $NO_2S_2$-macrocycle (L) is described and structurally characterized by single crystal X-ray analysis. The photoluminescence spectrum of L in 80% $CH_3CN/CH_2Cl_2$ displays a peak maximum at 431 nm (blue emission). In the metal-induced fluorometric experiment, L showed a drastic chelation-enhanced fluorescence quenching (CHEQ) effect only with $Hg^{2+}$ and $Cu^{2+}$. In ESI-mass study, a 1:1 stoichiometry for complexation of L with $Hg^{2+}$ was confirmed, suggesting the unique sensing behavior of the proposed ligand L due to the selective complexation affinity for $Hg^{2+}$. The observed results indicate that L is a promising turn-off type fluoroionophore for $Hg^{2+}$ and $Cu^{2+}$ detections. Additionally, the $Ag^+$ complex of the precursor macrocycle was prepared and its solid structure was crystallographically characterized.

  • An oxygen-sensitive fumarate reductase has been purified from the cytosol fraction of the Enterococcus faecalis RKY1 grown anaerobically on a defined medium containing glycerol and fumarate. A major portion of the purification was performed with employing Triton X-100 and reducing agents by Phenyl-sepharose CL-4B DEAE-sepharose and Dephadex G-150 The final activity was 0.42 unit/mg. The deduced molecular mass of active band was 66 kDa. The optimal pH and temperature for the activity were 7.0 and 38$^{\circ}C$ respectively. The enzyme activity was not affected by 1mM metal ions such as bacl2 $.$2H2O HgCl2 MnCl2$.$4H2O ZnCl2 CuCl2$.$2H2O Mgcl2$.$6H2O FeSo4$.$7H2O and by EDTA. Partially purified enzyme ws yellow in color ; spectroscopic study indicated the presence of flavins as a cofactor.

  • In order to further elucidate the process of electron solvation in liquids, the medium effect, as the difference between the free energy of $H^+$ in aqueous and non-aqueous states (${\Delta}{\Delta}G_0$), of THF-water mixtures has been investigated. (${\Delta}{\Delta}G_0$) were determined by electromotive force masurements of the cell Pt$H_2Q$, Q, HCI, THF, $H_2O$|KC1 | $Hg_2Cl_2$|Hg(Pt), where $H_2Q$ and Q are hydroquinone and quinone respectively. The effect of dielectric constant on the difference of free energy and the absorption energy of solvated electrons have been studied. For the consideration of these effect the polymerization of water in THF has been studied. Near infrared spectrum of O-H stetching energy has been used to measure the extent of water aggregates. The expermental results indicate that at least in some composition of binary mixtures the electrons or other ions are solvated preferentially with one component of solvents.

Determination of Hg22+ Ions Using a Modified Glassy Carbon Electrode with 2,2':6':2''-Terpyridine

  • A glassy carbon electrode (GCE) modified with 2,2':6':2”-terpyridine (2,2':6':2”-TPR) using a spin coating method was applied for the highly selective and sensitive analysis of a trace amount of $Hg_2^{2+}$ ions. Various experimental parameters, which influenced the response of the 2,2':6':2”-TPR modified electrode to $Hg_2^{2+}$ ions, were optimized. The linear sweep and differential pulse voltammograms for the 2,2':6':2”-TPR modified electrode deposited with Hg show a well-defined anodic peak at +0.65 V (vs. Ag|AgCl). After a 25 min preconcentration time in an $Hg_2^{2+}$ ion solution (0.1 M acetate buffer, pH 5.0), differential pulse voltammetry(DPV) with 2,2':6':2”-TPR modified electrode shows a linear response between $1.0\;{\times}\;10^{-6}M\;and\;2.0\;{\times}\;10^{-7}M$. The least-square treatment of these data produce an equation of I[${\mu}A$] = 0.031 + 0.005C with r = 0.980(n = 5). The detection limit of this electrode with linear sweep voltammetry and differential pulse anodic voltammetry were $2.0\;{\times}\;10^{-6}M\;and\;8.0\;{\times}\;10^{-8}M$, respectively. The presence of Pb, Fe, Cd, Ti, Ni, Co, Mg, Al, Mn, and Zn did not interfere in the analysis of the $Hg_2^{2+}$ ion. The 2,2':6':2”-TPR modified GCE has been successfully applied in determination trace amounts of Hg in a human urine sample.

  • This research is to investigate the changes in Mercury content of fishes, washed with times by the flowing water or immersed into the acetic acid solutions with different densities (1%, 3% and 5%), after contaminating them artificially on purpose, in the solutions with inorganic mercury. The results were summarized as follow: 1. Average values of mercury content in sand-fish and sardine, as a dried ones, which had not been treated by the mercurial solutions, were 0.027 ppm and 0.020 ppm, respectively. 2. According to the changes, with times, in contained by the flowing water, after immersed into 1 ppm $HgCl_2$ solution for 24 hours, The sand-fish washed for 9 minutes had the least of Mercury, which is about 25% of contained quantity of Mercury of those not cleansed. While the sardines cleansed for 3 minutes had the least of Mercury, which is about 77% of contained quantity of Mercury of those not cleansed. 3. According to the changes in contained quantity of the samples treated by the acetic acid solutions with different densities, the sand-fish treated by 3% acetic acid solution had the least value of remains, which was reduced by 64%, in comparision with that of fish cleansed for 1 minute, while the sardines, treated by 1% acetic acid solution had the least amount of remains, which was reduced by 16% in comparision with that of fish, cleansed for 1 minute.
