Sequential Separation of Lysozyme and Ovalbumin from Chicken Egg White

  • Lysozyme was trapped from $2{\times}$ diluted egg white using Amberlite FPC 3500 ion exchange resin (1 g/10mL of egg white). The lysozyme bound to the resin was recovered using 0.1 N glycine-NaOH buffers, pH 9.0, containing 0.5 M NaCl. After separating lysozyme, the pH of the egg white solution was adjusted to 4.75 and centrifuged to remove interfering proteins. The supernatant was collected, added with 2.5% citric acid and 5.0% ammonium sulfate combination to precipitate egg white proteins, except for ovalbumin. After centrifugation, both supernatant (S1) and precipitant were collected. The precipitant was dissolved with 4 volumes of distilled water, and then 2.0% ammonium sulfate and 1.5% citric acid combinations added, stirred overnight in a cold room, and centrifuged. The resulting supernatant (S2) was pooled with the first supernatant (S1), desalted using an ultrafiltration unit, heat-treated at $70^{\circ}C$ for 15 min, and then centrifuged. The supernatant was collected as an ovalbumin fraction and lyophilized. The separated proteins were confirmed using Western blotting. The yield of lysozyme and ovalbumin was > 88.9% and > 97.7%, respectively, and the purity of lysozyme and ovalbumin was > 97% and 87%, respectively. The results indicated that the protocol was simple, and separated lysozyme and ovalbumin effectively.

  • The study was carried out to determine the frequency distribution of trees by diameter classes according to the method of Bruce and Reineke's alignment with the data of pitch pine (Pinus rigida Miller) and Italian poplar (Populus euramericana Guinier) which are prepared for the purpose of yield table construction. The results obtained are summarized as follows: 1. All through the tree species studied, when their diameters were larger, their distribution range became wider, while the percentage of their distribution frequency became lesser. 2. As for frequency percentage of trees in an average D.B.H. of stands, it became smaller when average D.B.H. of the stand is larger: the percentage of pitch pine stand is 36.2 and that of Italian poplaris 29.0. 3. In case of smaller D.B.H. under the average, the larger their D.B.H. became, the lesser the cumulative frequency percentages became.

Purification and Characterization of a Fibrinolytic Enzyme Produced by Bacillus amyloliquefaciens HC188 (Bacillus amyloliquefaciens HC188이 생산하는 혈전분해 효소의 정제 및 특성)

  • A bacterium producing a fibrinolytic enzyme was isolated from Cheonggukjang. The bacterium was identified as a strain of Bacillus amyloliquefaciens by 16S rDNA analysis and designated as B. amyloliquefaciens HC188. The optimum culture medium appeared to be one containing 0.5% (w/v) maltose and 0.5% (w/v) soytone. Bacterial growth in the optimal medium at $37^{\circ}C$ reached the stationary phase after 27 h of incubation and the fibrinolytic enzyme showed optimum activity at 24 h. The enzyme was purified by 20-80% ammonium sulfate precipitation, CM Sepharose fast flow ion exchange chromatography, and Sephacryl S-200HR column chromatography. Its specific activity was 38359.3 units/mg protein and the yield was 5.5% of the total activity of the crude extracts. The molecular weight was 24.7 kDa and the amino acids of the N-terminal sequence were AQSVPYGVSQIKAPA. The fibrinolytic enzyme activity had an optimum temperature of $40^{\circ}C$ and an optimum pH of 8.0, and the enzyme was stable in the ranges $20-40^{\circ}C$ and pH 6.0-8.0. Enzyme activity was increased by $Ca^{2+}$ and $Co^{2+}$ but inhibited by $Cu^{2+}$, EDTA, and PMSF. It is suggested that the purified enzyme is a metallo-serine protease.

Biological Hydrogen Production from Mixed Organic Waste of Food and Activated Sludge by Pre-treatment (음식물쓰레기와 전처리한 폐활성슬러지의 혼합비율에 따른 생물학적 수소생산)

  • In this study, Bio-hydrogen is produced from organic waste mixtures containing food waste and waste activated sludge (WAS). The effects of different operational factor on hydrogen production, including various solubilization methods for pretreatments of WAS, pH and different ratios of food waste and WAS, were investigated. The highest hydrogen production values are obtained as 4.3 mL $H_2/g$ $VS_{consumed}$ in the case of applying the mixed pre-treatments of alkali and ultrasonic. The pH value in bio-reactor increased from 4 to 8 after the ultrasonic treatment with alkali and the hydrogen yield touched its highest value in the pH range of 5.0 to 5.5. Similarly, the hydrogen production reached the level of 13.8 mL $H_2/g$ $VS_{consumed}$ using the same pre-treatment method from the mixture of food waste and WAS. The ratio of 2 : 1 produced a maximum amount of hydrogen of 5.0 L $H_2/L/d$. The amount of volatile fatty acids(VFAs) including acetate, propionate and butyrate, were also varied considerably. Propionate decreased consistently with rising of hydrogen while butyrate comparing to acetate relatively increased in the effluent.

  • Thiamine derivatives of thiamine tetrahydrofurfuryl disulfide (TTFD) and thiamine propyl disulfide (TPD) were obtained in a good yield and purity by use of thiothiamine $(SB_1)$ and also described in details for the preparing sodium tetrahydrofurfuryl thiosulphate (Bunte's salt) with and without KI. The optimum reaction conditions for the preparation of TTFD was set in which the Box-Wilson plan was applied. The reaction conditions are as follows; 1. pH value of aqueous solution of thiol type of $B_1{\cdot}HCl;12.09$ 2. Quantity of Bunte's salt (to $B_1{\cdot}HCl\;20G$); 35.01g 3. Reaction temperature; $15.59^{\circ}C$ Studies on ultraviolet absorption were made at various pH and showed that the absorption maxima are shifted with change of pH. The absorption maxima are at $244{\sim}246nm,\;234{\sim}235nm$ in TTFD, and $245{\sim}246nm,\;233{\sim}235nm$ in TPD. The structure was proved by the infrared spectral evidence. Quantitative determination was studied.

Comparison of Milk Composition and Blood Metabolites Between High and Low Milk Producing Cows (젖소의 고능력우와 저능력우간의 우유 성분 및 혈중 대사물질 특성 비교)

  • The objective of this study was to estimate the effects of daily milk yield, somatic cell count(SCC), days in milk(DIM), and parity on the compositions of milk and blood in high or low producing dairy cows. To divide the high or low producing group, there were some restrictions in this study. 235 Holstein dairy cows had a average daily milk yield of 23.2 $\pm$ 6.8 kg were grouped into two classes with low producing(average daily milk 17kg) or high producing(average daily milk 29 kg). The other restrictions were two parities(first and second parity), two SCC groups(under $l{\times}10^5$cells/ml, and $l{\times}10^5$ to $7{\times}10^5$ celis/ml), and three DIM groups(under 80, 81 to 180, and 181 to 305DIM). The blood urea nitrogen(BUN), milk urea nitrogen(MUN) and glucose between two group with high and low somatic cell count were not affected by parity, DIM and SCC. But there were significantly different on BUN and glucose between high and low milk producing(p< 0.01), also was different on glucose between parities(p < 0.05). White blood cell(WBC) and lymphocyte were affected(p< 0.05) by SCC level, protein percent was also affected by DIM(p< 0.01). The least square means of protein in second parity was a 1.3 times higher than that in first parity(p < 0.05), and it showed a higher level in the low producing group than the high producing group(p < 0.0l). WBC and lymphocyte were lower in the $1{\sim}7{\times}10^5$ celis/ml than those under $1{\times}10^5$ celis/ml(p< 0.05). Neutrophil was a higher level in first parity than that in second parity(p < 0.05). Only protein and total solid were affected by parity, the other compositions were not affected by parity, DIM, SCC and milk yields. The results suggested that significant differences were in the blood components such as glucose, WBC, lymphocyte and neutrophil between high and low producing cows. The results also show that more studies are required to clarify the factors and markers related to milk yield, quality and mastitis.

Apparent metabolizable energy, growth performance and carcass traits of Japanese quail fed select modern grain sorghum varieties

  • Objective: This study was performed to determine the apparent metabolizable energy (AMEn) content of tannin-free red/bronze, white/tan and U.S. No. 2 varieties of grain sorghum for feeding Japanese quail and validate their nutrient profile by evaluating effects on performance and carcass traits with full-substitution of corn. Methods: Experiment 1 determined the AMEn content of red/bronze, white/tan, and U.S. No. 2 grain sorghum varieties fed to mixed-sex Japanese quail (Coturnix japonica) (n = 314) at 3 and 6-weeks of age. Analyses were based on a 2×4 factorial treatment design with age and grain types defining the treatments, and a randomized complete block experiment design with cage and trials defining the block. AMEn values were validated by evaluating the performance and carcass traits of Japanese quail (n = 644) from 1 to 40 days of age in Experiment 2 with birds were fed 1 of 4 complete diets. Statistical analyses were conducted on performance data and select individual carcass trait measurements. Results: Determined AMEn values at 3-weeks of age were 3,524±122.03 (red/bronze), 3,252±122.03 (white/tan), and 3,039±123.44 (U.S. No. 2) kcal/kg. At 6-weeks of age, determined AMEn were 3,373±297.35 (red/bronze), 3,279±297.35 (white/tan), and 2,966±298.64 (U.S. No. 2) kcal/kg. Carcass traits showed live body weight (p = 0.0409) and hot carcass weight (p = 0.0234) were greatest in U.S. No. 2; however, carcass yield (p<0.0001) was lowest. No significant differences were observed among treatments for feed intake, feed conversion ratio, breast weight and breast yield (p>0.05). Conclusion: These studies demonstrated that tannin-free grain sorghum varieties may be a potential alternative to corn in quail diets while maintaining growth performance and carcass parameters.

The Effect of UV Intensity and Wavelength on the Photolysis of Triclosan (TCS) (광반응을 이용한 Triclosan 분해에서의 UV 광세기와 파장의 효과)

  • We investigated the effect of hydroxyl radicals on the photolysis of triclosan (TCS), which is a potent broad-spectrum antimicrobial agent. TCS degradation during the initial reaction time of 5 min followed a pseudo-first order kinetic model ai all light intensities at a wavelength of 365 nm and at the low light intensities at a wavelength of 254 nm. The photodegradation rate significantly increased with decreasing wavelength and increasing the UV intensities. The activity of hydroxyl radicals was suppressed when methanol was used as the solvent instead of water. An increase in the photon effect was observed when the UV intensity was higher than $5.77{\times}10^{-5}$ einstein $L^{-1}min^{-1}$ at 254 nm, and lower than $1.56{\times}10^{-4}$ einstein $L^{-1}min^{-1}$ at 365 nm. The quantum yield efficiency for the photolysis of TCS was higher at 365 nm than at 254 nm among the above mentioned UV intensities. Dibenzodichloro-p-dioxin (DCDD) and dibenzo-p-dioxin were detected as intermediates at both UV intensities of $1.37{\times}10^{-4}$ and $1.56{\times}10^{-4}$ einstein $L^{-1}min^{-1}$ at 365 nm. Dichlorophenol and phenol were also detected in all cases. Based on our findings, we presented a possible mechanism of TCS photolysis.

Evaluation of the Quality of Beef Patties Formulated with Dried Pumpkin Pulp and Seed

  • The objective of this study was to investigate quality attributes of beef patties formulated with dried pumpkin pulp and seed mixture (PM). Four different meatball formulations were prepared where lean was replaced with PM as C (0% PM), P2 (2% PM), P3 (3% PM) and P5 (5% PM). Utilization of PM decreased moisture and increased ash content of the patties. Incorporation of 5% PM (P5) increased the pH value of both uncooked and cooked patties compared to C group. Increasing levels of PM increased water-holding capacity. No significant differences were found in cooking yield and diameter change with the addition of PM. Incorporation of PM increased fat and decreased moisture retention of the samples. $a^*$ values were decreased with PM addition, where $L^*$ values did not differ among treatments and $b^*$ values were similar in C, P3 and P5 samples. Textural properties were mostly equivalent to control samples with the incorporation of PM even at higher concentrations. The addition of PM did not significantly affect any of the sensory scores tested. These results indicated that utilization of PM presents the opportunity to decrease the amount of meat besides to improve healthier profile without causing negative changes in physical, chemical and technological quality of beef patties.

Studies on chitobiase from Aeromonas salmonicida YA7-625 (Aeromonas salmonicida YA7-625가 생산하는 Chitobiase의 순수분리와 특성)

  • Chitobiase from Aeromonas salmonicida YA7-625 was purified from culture broth through ammonium sulfate precipitation, chitin affinity adsorption, hydroxylapatite column chromatography and gel filtration, with 47.2% yield and 31.5 fold purity. The molecular weight of purified chitobiase was 15,000 daltons, and the chitobiase showes maximum activity at the condition of at $40^{\circ}C$ and pH 6.0. The effects of various metal ions and inhibitors show thatcystein, glutamic acid, serine, tryptophan, and tyrosine residues seem to be concerned in chitobiase activity.
