카라기난 필름 및 카라기난 코팅 종이포장지의 유지투과 특성 (Lipid Penetration Characteristics of Carrageenan-Based Edible Films)

  • 카라기난필름과 카라기난 코팅종이의 유지투과도를 측정하였으며 현재 외식산업분야에서 널리 사용되고 있는 Polyethylene (PE) 코팅종이를 대체할 수 있는 가능성을 조사하였다. 카라기난필름과 카라기난 코팅종이의 유지투과도는 매우 낮은 것으로 나타났으며, 카라기난 중에서는 ${\kappa}$-카라기난의 유지투과도가 가장 낮았다. 카라기난이 $4{\sim}5\;kg/ream\;(278m^2)$ 정도 코팅된 종이의 유지투과도는 PE 코팅종이의 유지투과도와 비슷한 것으로 나타났으며, 카라기난 필름의 유지투과도는 카라기난 코팅종이의 10배정도 낮은 것으로 나타났다. 카라기난필름과 카라기난 코팅종이의 유지투과도는 시간에 따라 지수적으로 증가하는 것으로 나타났으며, 카라기난이 4kg/ream 이상 코팅된 종이는 유지투과도가 낮아 외식산업에서 유지함량이 많은 식품의 포장에 사용될 수 있을 것으로 사료된다.

한약복합물 HT008이 흰쥐 관절염 모델에 미치는 효과 (Effect of New Herbal Mixture, HT008 on Arthritis Rat Model)

  • Objectives : This study was conducted to investigate the effect of HT008, a multi-herbal mixture consisting of 3 herbs, Eleutherococcus senticosus, Angelica sinensis, and Scutellaria baicaleinsis on arthritic model in rats. Methods : The anti-inflammatory and analgesic activities were observed by utilizing the following models: carrageenan-induced edema of the hind paw of rats, acetic acid-induced writhing response in mice. The perimeter of the paw was measured before injection and then at 1, 2, 4, 6 h after injection of 1% $\lambda$-carrageenan. The HT008 at five dose levels (10, 30, 100, and 300 mg/kg) and distilled water given 30 min to treatment groups and control group, before $\lambda$-carrageenan injection. In the writhing test, the mice received 0.7% acetic acid solution in normal saline injected intraperitoneally at a dose of 10 ml/kg. The number of writhes was counted staring 10 min after injection. Results : HT008 at four dose levels (30, 100, and 300 mg/kg) significantly decreased the carrageenan-induced rat paw edema perimeter. E. senticosus and S. baicaleinsis extracts reduced acetic acid-induced writhing response in mice. Also A. sinensis extracts significantly decreased the carrageenan-induced rat paw edema perimeter. Conclusions : These results show that HT008, a multi-herbal mixture has both anti-inflammatory activity and analgesic effects in vivo arthritic model, and suggest that HT008 could be a good therapy to treat human osteoarthritis.

Luteolin 5-O-glucoside from Korean Milk Thistle, Cirsium maackii, Exhibits Anti-Inflammatory Activity via Activation of the Nrf2/HO-1 Pathway

  • Luteolin 5-O-glucoside is the major flavonoid from Korean thistle, Cirsium maackii. We previously reported the anti-inflammatory activities of luteolin 5-O-glucoside in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. In this study, we determined the anti-inflammatory mechanisms of luteolin 5-O-glucoside through the inhibition of nitric oxide (NO) production in vitro and in vivo. Results revealed that luteolin 5-O-glucoside dose-dependently inhibited NO production and expression of iNOS and COX-2 in LPS-induced RAW 264.7 cells. Luteolin 5-O-glucoside also significantly inhibited the translocation of $NF-{\kappa}B$, the activation of MAPKs, and ROS generation in LPS-induced RAW 264.7 cells. In addition, protein expressions of Nrf-2 and HO-1 were also upregulated by luteolin 5-O-glucoside treatment. Moreover, luteolin 5-O-glucoside inhibited ${\lambda}-carrageenan-induced$ mouse paw edema by 65.34% and 48.31% at doses of 50 and 100 mg/kg body weight, respectively. These findings indicate potential anti-inflammatory effect of luteolin 5-O-glucoside particularly by downregulating $NF-{\kappa}B$ and upregulating HO-1/Nrf-2 pathway.

전침자극이 통각과민에 미치는 효과 (Effects on Hyperalgesia of Electroacupuncture)

  • This study aims to examine the effects on nociceptive neuron excitability by application of electroacupuncture in rat with inflammation. It used 20 rats for experiment, divided them into control group, electroacupuncture group (EA group), caused hyperalgesia by injecting ${\lambda}$-carrageenan into hindpaw and conducted treatment three times for experimental period. Change of NFR(reaction time, RMS) showed no significant differences among EA group showed significant differences compared to control group from 48 hours. This study showed that EA group had an effect on nociceptive neurone excitability. Therefore, it is considered that electroacupuncture for pain control will be very desirable.

Effects of the Constituents of Gardenia Fructus on Prostaglandin and NO Production

  • The fruits of Gardenia jasminoides Ellis have been previously reported to possess anti-inflammatory activity. In this study, the constituents including geniposide, geniposidic acid, genipin and crocin were evaluated for their effects on prostaglandin and NO production in an attempt to establish anti-inflammatory cellular mechanisms. Among the constituents tested, only genipin significantly inhibited cyclooxygenase-2-mediated $PGE_2$ and inducible nitric oxide synthase-mediated NO production from lipopolysaccharide-treated RAW 264.7 cells at 10-100 ${\mu}$M. Genipin also inhibited nuclear transcription factor-${\kappa}B$ activation. Moreover, genipin showed in vivo antiinflammatory activity on ${\lambda}$-carrageenan-induced paw edema in mice (10.4-29.9% inhibition at 20-100 mg/kg, i.p.). All of these results suggest that genipin may contribute to anti-inflammatory activity of the fruits of G. jasminoides and an inhibitory action on prostaglandin and NO production is, at least, the part of anti-inflammatory mechanism of genipin.

Temporal and spatial variation in the distribution of life history phases of Chondrus crispus (Gigartinales, Rhodophyta)

  • Thirty populations of Chondrus crispus Stackhouse from Nova Scotia were collected during the years 1993 to 2011. Taken from estuaries, wave exposed open coasts, high intertidal rock pools and shallow subtidal habitats, the populations were evaluated for relative abundance of tetrasporophytic and gametophytic life history phases. Over 2,800 thalli were characterized using the resorcinol-acetal test to distinguish the kappa- and lambda-carrageenan containing fronds of gametophytes and tetrasporophytes, respectively. These populations had $77{\pm}5%$ gametophytes (mean ${\pm}95%$ confidence interval), with most populations having gametophyte : sporophyte ratios ranging from 2 : 1 to 9 : 1. No population had a dominance of tetrasporophytes, although two populations had 1 : 1 ratios. A meta-analysis of our data along with previously published accounts showed no significant changes in gametophyte dominance with respect to hypothesized gradients of wave exposure, salinity, or water depth. Significant changes occurred in ratios at five sites where replicate sampling occurred in different years. We conclude that C. crispus in Maritime Canada has a natural ratio of 3 : 1 or greater in stable conditions, and that lower ratios represent recovery from disturbance in which bare substratum is created that is subsequently colonized by carpospores from remaining gametophytic thalli.

한약복합추출물 HT008의 제조방법에 따른 항염증 효능 및 성분함량 비교연구 (Comparative Study on Anti-inflammatory Effects and Compound Contents of Multi-herbal Extracts HT008 produced by Different Manufacturing Methods)

  • Objectives : The biological activities and compound contents of herbal medicine vary depending on manufacturing processes. In this study, we compared anti-inflammatory effects and compound contents of three kinds of multi-herbal extract HT008 produced by different manufacturing processes in order to determine chemical and biological equivalence. Methods : HT008 was produced by three different manufacturing methods: 1. Freeze dried extract of Eleutherococcus senticosus, Scutellaria baicalensis and Angelica sinensis (HT008 FD), 2. Spray dried extract of E. senticosus and S. baicalensis combined with reflux extract of A. sinensis (HT008 SD), 3. Spray dried extract of E. senticosus and S. baicalensis combined with supercritial fluid extract of A. sinensis (HT008 SF). Anti-inflammatory effects were evaluated using acetic acid induced pain model and ${\lambda}$-carageenan induced paw edema model. Compound contents were evaluated by HPLC quantitative analysis of standard compounds of HT008, eleutheroside E, baicalin, z-ligustilide. Results : HT008 FD, HT008 SD and HT008 SF significantly decreased acetic acid induced pain index and ${\lambda}$-carrageenan induced paw edema volume compared with that of control group. There was no significant difference in efficacy among the HT008 FD, HT008 SD and HT008 SF. Standard compound contents of HT008 FD, HT008 SD and HT008 SF were quantified within the range of Korean pharmacopoeia or other research. Conclusions : Three different manufacturing methods of multi-herbal extracts have been developed without noticeable difference in the efficacy or compound contents. The results might be used to establish manufacturing process and industrialization of herbal extracts.

Anti-Inflammatory Activity of the Total Flavonoid Fraction from Broussonetia papyrifera in Combination with Lonicera japonica

  • To establish the anti-inflammatory activity of the total flavonoid fraction of the root barks of Broussonetia papyrifera (EBP) and a new formula, the ethanol extract of the root barks of B. papyrifera was fractionated with ethylacetate, yielding the hydrophobic prenylated flavonoid-enriched fraction. EBP and the ethanol extract of the whole Lonicera japonica (ELJ) plant were then mixed at a ratio of 1:1 (w/w) to give a new preparation (BL) in the hope of obtaining an optimal formula with a higher anti-inflammatory activity. Evaluation of the effects of these preparations on A23187-treated rat basophilic leukemia (RBL-1) cells revealed that EBP potently inhibited 5-lipoxygenase (5-LOX), while ELJ showed weak inhibition. Additionally, the mixture (BL) clearly showed stronger inhibitory effects against 5-LOX than either preparation alone. These preparations also inhibited cyclooxygenase-2-catalyzed $PGE_2$ and inducible nitric oxide (NO) synthase-catalyzed NO production by lipopolysaccharide-treated RAW 264.7 cells. When tested against arachidonic acid-induced mouse ear edema, EBP showed strong inhibitory activity at doses of 5-200 mg/kg when administered orally, but BL had obviously stronger inhibitory effects. When tested against ${\lambda}$-carrageenan-induced paw edema in mice, BL showed a potent and synergistic anti-inflammatory effect. In addition, in the acetic acid-induced writhing test, BL was found to have strong analgesic activity at 50-400 mg/kg. Taken together, these results indicate that each of these preparations exert anti-inflammatory activity in vitro and in vivo. In particular, BL showed stronger anti-inflammatory activity than EBP, and these anti-inflammatory effects were partially related to the inhibition of eicosanoid and NO production. BL may be useful for the treatment of human inflammatory disorders.

It used 24 rats for experiment, divided them into control group, electroacupuncture group(EA group), Meloxicam group(ME group), combination of electroacupuncture with Meloxicam group(EA+ME group), caused hyperalgesia by injecting ${\lambda}$-carrageenan into hindpaw and conducted treatment three times for experimental period. Noxious flexion withdrawal reflex(NFR) and somatosensory evoked potential(SEP) were measured immediately after induction, at 24 hours, 48 hours and 72 hours after induction. Change of NFR(reaction time, RMS) showed no significant differences among EA group, Meloxicam group, and EA+Meloxicam group, but all treatment groups showed significant differences compared to control group from 48 hours. In NFR threshold, there were significant differences between EA+Meloxicam and other groups. In SEP amplitude, there were significant differences between EA+Meloxicam and control group from 48 hours. This study showed that EA+Meloxicam gel had an effect on nociceptive neurone excitability. Therefore, it is considered that appropriate combination of anti-inflammatory drug with electroacupuncture for pain control will be very desirable.

염증유발 백서에서 전침자극과 미세전류자극의 항-통각과민 효과 (Anti-hyperalgesic Effects of Electroacupuncture Combination of Microcurrent Stimulation in Rat with Induced Inflammation)

  • Purpose: The purpose of this study were to examine the anti-hyperalgesic effects of combination of electroacupuncture and microcurrent on anti-hyperalgesia in local hyperalgesia zone. Methods: It used 24 rats for experiment, divided them into control group, electroacupuncture group (EA group), microcurrent group (MC group), combination of electroacupuncture with microcurrent (EA+MC group), caused hyperalgesia by injecting ${\lambda}-carrageenan$ into hindpaw. Thickness of hindpaw, mechanical pain threshold (MPT), thermal pain threshold (TPT), noxious flexion withdrawal reflex (NFR) and somatosensory evoked potential (SEP) were measured immediately after induction, at 24 hours, 48 hours and 72 hours after induction. The electrical stimulation was given once a day for three days, 20min per session. Results: Change of thickness, MPT, and TPT showed significant difference in all groups compared to control group. In particular, there were remarkable difference in EA+ME group. In particular there were remarkable differences in EA group and EA+MC group. Change of NFR(% threshold, % reaction time, % RMS) and SEP showed mainly significant differences in EA group and EA+ME group compared to control group. In particular, there were remarkable difference in EA+ME group. Conclusion: The above results suggest that appropriate combination of microcurrent with electroacupuncture for pain control will be very desirable.
