Enzymatic Characteristics of steroid $\Delta^1$-dehydrogenase from Arthrobacter simplex

  • Steroid $\Delta^1$-dehydrogenase purified from hydrocortisone-induced cells of Arthrobacter simplex converted various 3-ketosteroids into their corresponding $\Delta^1$-dehydrogenated products. The transformation efficiencies depend upon the chemical structure of the steroids, especially length of the side chain at 17 position and hydroxyl groups at 11 and 17 positions. The Km values for androstenedione, the most favorable substrate examined, and hydrocortisone were 74 ${\mu}M$ and 294 ${\mu}M$, respectively. The optimum temperature and pH of the enzyme reaction were 35$^{\circ}C$ and pH 9, respectively, and the enzyme was relatively stable at the range from 20 to 35$^{\circ}C$ and from pH 5 to 10 after one hour of incubation. The enzyme activity was markedly inhibited in the presence of $Cu^{2+},\;Fe^{3+},\;Hg^{2+},\;Mo^{6+}$ ions, and somewhat inhibited by $Zn^{2+}$ and $Fe^{2+}$. $\alpha,\alpha'$-Dipyridyl that inhibits 9$\alpha$-hydroxylase and accumulates 1,4-androstadiene-3,17-dione from sterols revealed no inhibitory effect on this enzyme. EGTA showed inhibitory effect. $\beta$-Estradiol competitively inhibited the enzyme activity. Chemical modifications of the enzyme were attempted with several reagents. p-Hydroxymer-curibenzoate showed inhibition of the enzyme activity and protection of the substrate. This suggests that cysteine residue may be involved in the active site of the enzyme.

Conformational Switch of the Strained Native Serpin Induced by Chemical Cleavage of the Reactive Center Loop

  • The native conformation of serpins (serine protease inhibitors) is strained. Upon cleavage of the reactive center loop of serpins by a protease, the amino terminal portion of the cleaved loop is inserted into the central ${\beta}-sheet$, A sheet, as the fourth strand, with the concomitant release of the native strain. We questioned the role of protease in this conformational switch from the strained native form into a stable relaxed state. Chemical cleavage of the reactive center loop of ${\alpha}_1-antitrypsin$, a prototype serpin, using hydroxylamine dramatically increased the stability of the serpin. A circular dichroism spectrum and peptide binding study suggests that the amino terminal portion of the reactive center loop is inserted into the A sheet in the chemically-cleaved ${\alpha}_1-antitrypsin$, as in the enzymatically-cleaved molecule. These results indicate that the structural transformation of a serpin molecule does not require interaction with a protease. The results suggest that the serpin conformational switch that occurred during the complex formation with a target protease is induced by the cleavage of the reactive center loop per se.

2nd Nanotube Formed Surface Observation of the Ti-25Ta-xZr Alloys Using ATO Technique

  • The purpose of this study was to investigate $2^{nd} $nanotube formed surface observation of the Ti-25Ta-xZr alloys using ATO(anodic titanium oxide) technique. Ti-25Ta-xZr alloy was anodized in 1M $H_3PO_4$ electrolytes containing 0.8 Wt. % NaF at room temperature. After formation of nanotube was achieved out, nanotube was eliminated, and then anodization was carried out repeatedly. The microstructures, phase transformation, and morphology of nanotubular Ti-25Ta-xZr alloys and process of nanotube growth by using ATO method was examined by optical microscopy (OM), X-ray diffraction (XRD), and field emission scanning electron microscopy (FE-SEM). The ${\alpha}$ phase and ${\beta}$ phases were affected to form the second nanotube morphology of Ti-25Ta-xZr alloys.

Production of D-Xylonic Acid from Hemicellulose Using Artificial Enzyme Complexes

  • Lignocellulosic biomass represents a potentially large resource to supply the world's fuel and chemical feedstocks. Enzymatic bioconversion of this substrate offers a reliable strategy for accessing this material under mild reaction conditions. Owing to the complex nature of lignocellulose, many different enzymatic activities are required to function in concert to perform efficient transformation. In nature, large multienzyme complexes are known to effectively hydrolyze lignocellulose into constituent monomeric sugars. We created artificial complexes of enzymes, called rosettazymes, in order to hydrolyze glucuronoxylan, a common lignocellulose component, into its cognate sugar ${\small{D}}$-xylose and then further convert the ${\small{D}}$-xylose into ${\small{D}}$-xylonic acid, a Department of Energy top-30 platform chemical. Four different types of enzymes (endoxylanase, ${\alpha}$-glucuronidase, ${\beta}$-xylosidase, and xylose dehydrogenase) were incorporated into the artificial complexes. We demonstrated that tethering our enzymes in a complex resulted in significantly more activity (up to 71%) than the same amount of enzymes free in solution. We also determined that varying the enzyme composition affected the level of complex-related activity enhancement as well as overall yield.

주조 및 불연속 석출물 미세조직을 가지는 Mg-Al 합금의 인장 특성 및 열전도도 (Tensile Properties and Thermal Conductivities of Mg-Al alloy with As-Cast and Discontinuous Precipitates Microstructures)

  • The objective of this study was to investigate the tensile properties and thermal conductivities of Mg9.3%Al alloy in as-cast state and heat-treated state consisting of fully discontinuous precipitates (DPs), respectively. The fully DPs microstructure was obtained by solution treatment at 405℃ for 24 h, followed by furnace cooling to RT. The as-cast alloy showed a partially divorced eutectic β(Mg17Al12) phase particles formed along the α-(Mg) cell boundaries. The DPs had various apparent (α+β) interlamellar spacings, which is related to different transformation temperatures during the furnace cooling. The DPs microstructure exhibited better tensile strength than the as-cast one, resulting from the higher value of elongation in response to its more homogeneous microstructure. It is noticeable that the DPs microstructure had 12.4% higher thermal conductivity in average than the as-cast one between RT and 200℃. The XRD analyses revealed that the lower Al concentration in the α-(Mg) matrix may well be responsible for the better thermal conductivity of the DPs microstructure.

  • Galectins are ${\beta}$-galactoside binding lectins that contain one or more carbohydrate recognition domains. As a consequence of sugar-binding properties, galectins exhibit a variety of interactions with glycoproteins, thus playing important roles in various pathological processes. A number of studies have shown roles of galectins in cancer. Galectin-7 is a prototype member of the galectin family implicated in epithelial stratification and cell migration. It can act as a potent dual regulator in different types of cancer. Galectin-7 may contribute either to neoplastic transformation and tumour progression through regulation of cell growth, cell cycle, angiogenesis, apoptosis and cell migration or may have a protective effect in cancer depending on the tissue type. A perusal of the literature indicates particular roles of galectin-7 in carcinomas and melanomas, while contributions await greater exploration in other types of cancers including sarcomas and leukemia. This review collectively summarizes available literature on expression and roles of galectin-7 in different cancers.

On Some Distributions Generated by Riff-Shuffle Sampling

  • The work presented in this paper is divided into two parts. The first part presents finite urn problems which generate truncated negative binomial random variables. Some combinatorial identities that arose from the negative binomial sampling and truncated negative binomial sampling are established. These identities are constructed and serve important roles when we deal with these distributions and their characteristics. Other important results including cumulants and moments of the distributions are given in somewhat simple forms. Second, the distributions of the maximum of two chi-square variables and the distributions of the maximum correlated F-variables are then derived within the negative binomial sampling scheme. Although multinomial theory applied to order statistics and standard transformation techniques can be used to derive these distributions, the negative binomial sampling approach provides more information and deeper insight regarding the nature of the relationship between the sampling vehicle and the probability distributions of these functions of chi-square variables. We also provide an algorithm to compute the percentage points of these distributions. We supplement our findings with exact simple computational methods where no interpolations are involved.

The Role of Negative Binomial Sampling In Determining the Distribution of Minimum Chi-Square

  • The distributions of the minimum correlated F-variable arises in many applied statistical problems including simultaneous analysis of variance (SANOVA), equality of variance, selection and ranking populations, and reliability analysis. In this paper, negative binomial sampling technique is employed to derive the distributions of the minimum of chi-square variables and hence the distributions of the minimum correlated F-variables. The work presented in this paper is divided in two parts. The first part is devoted to develop some combinatorial identities arised from the negative binomial sampling. Aspartate계 아미노산 대사 효모 유전자 HOM6의 cloning 및 구조분석 (Molecular cloning and restriction endonuclease mapping of homoserine dehydrogenase gene (HOM6) in yeast saccharomyces cerevisiae)

  • Synthesis of threonine and methionine in yeast, Saccharomyces cerevisiae shares a common pathway from aspartate via homoserine. HOM6 gene encodes homoserine dehydrogenase (HSDH) which catalyzes the inter-conversion of beta-aspartate semialdehyde and homoserine. The level of HSDH is under methionine specific control. A recombinant plasmid (pEK1: 13.3kb), containing HOM6 gene, has been isolated and cloned into E. coli by complenemtary transformation of a homoserine auxotrophic yeast strain M-20-20D (hom6, trp1, ura3) to a prototrophic M20-20D/pEK1, using a library of yeast genomic DNA fragments in a yeast centromeric plasmid, YCp50(8.0kb). Isolation of HOM6has been primarily confirmed by retransformation of the original yeast strain M20-20D, using the recombinant plasmid DNA which was extracted from M20-20D/pEK1 and subsequently amplified in E. coli. Eleven cleavage sites in the insery (5.3kb) have been localized through fragment analysis for 8 restriction endonucleases; Bgl II(2 site), Bgl II(1), Cla I(3), Eco RI(1), Hind III(2), Kpn I (1), Pvu II(1) and Xho I(1).

조립자규소 성형체의 효과적 질화가열법에 관한 연구(Densification of Silicon Nitride 3보) (Effective Nitridation of Compacts of Coarse Silicon Particles)

  • To find out the optimum heating profile for the nitridation of compacts of graded silicon grains (max 53$mu extrm{m}$) two batches with the addition of MgO and $Mg(NO_3)_3$$cdot$$6H_2O$ to silicon particles were isostatically pressed into compacts. They were nitrided under some different nitriding schedules. The properties such as bulk densitis microstructures and formed phases were measured and observed. The following results were obtained ; 1) About 10% unreacted silicon remained in specimen which was nitrided at 1, 350$^{\circ}C$ for 240hrs. 2) One of the step-heating processes 1, 150$^{\circ}C$-1, 390$^{\circ}C$ for 65hrs are then $1, 390^{\circ}C$for 50hrs was the low temperature but with that at high temperature. 3) High pressure(10.5kgf/$cm^2$) of nitrogen at 1, 390$^{\circ}C$ accelerated the $\alpha$$ ightarrow$$\beta$ transformation of silicon nitride. 4) Magnesium nitrate was superior to magnesium oxide in the role of nitriding aid and the formation of uniform microstructures.
