Effect of cooking methods on the phytosterol content in nine selected vegetables

  • Phytosterol contents in nine vegetables such as paprika (red, yellow, and orange), kohlrabi, bamboo shoot, cherry tomato, cucumber, Chinese chive, and corn were analyzed by gas chromatography. Individual contents of ${\beta}$-sitosterol, campesterol, and stigmasterol in fresh and cooked vegetables (boiling, grilling, stir-frying, deep-frying, steaming, roasting, and microwaving) were determined and compared. Total phytosterol content of paprika, cucumber, Chinese chive and cherry tomato ranged from 23.19 to 46.51 mg/kg (0.002-0.005%) of fresh weight of raw vegetables. Total phytosterol content variation (%) was obtained as follows: [(the content of phytosterol after cooking) - (the content of phytosterol before cooking)] / (the content of phytosterol before cooking) ${\times}100$. Total phytosterol content was found to be high in raw kohlrabi at 138.99 mg/kg fw (0.01%), in corn at 302.86 mg/kg fw (0.03%), and in bamboo shoot at 443.15 mg/kg fw (0.04 %). Total phytosterol content variation (%) in orange paprika ranged from 27.5 to 267.3 while that in cherry tomato ranged from -11.0 to 337.5. Generally, high content variation of total phytosterol was found in stir-fried and deep-fried vegetables. Therefore, higher phytosterol levels were obtained from cooked vegetables than raw vegetables. We suggest that these data will be useful to investigate cooking methods for increased intake of phytosterols.

  • The ethanolic extracts of marine green, brown and red algae collected from Karachi coasts of Arabian Sea afforded a new enol-derivative of N-acylsphingosine named as coelarthenol (1) from Coelarthrum muelleri, two new glucose-derivatives named: botryenal (2) and botryenol (3) from Botryocladia leptopoda, ${\alpha}-tocopherol$ quinone (4) from Codium iyengarii, ${\beta}-sitosterol$ and hexadecanoic acid from Stokeyia indica. The known constituents (4, ${\beta}-sitosterol$ & hexadecanoic acid) have not been reported so far from their corresponding sources and the structures were determined through spectroscopic methods, whereas, the structures of new constituents (1-3) were elucidated with the aid of selective HMBC experiments. The phytotoxicity of 4 was also monitored.

  • There has been no specific test available for identifying the sesame oil among common edible oils. As the contents of sesamin and the ratio of sterols allowed the estimation for the genuine sesame oil, the author investigated to establish some instrumental methods for verification of genuine sesame oil and its distribution in the market. The sesame oil was saponified and the sesamin and sterols were isolated from the unsaponiable fraction by Florisil column chromatography. The individual components were determined by gas- chromatography and sesamin standard (purified sesamin) was obtained by silicagel column chromatography. The gas- chromatographic condition using Flame Ionization Detector supported on 10% OV-101 with di-(2-ethylhexyl) sebacate as an internal standard was suitable, and quantitation of sesamin and sterols, including campesterol, stigmasterol and ${\beta}-sitosterol$ was carried out. The results of this study showed that contents of sesamin in genuine sesame oil were 0.3-0.5% and the ratio of stigmasterol to compesterol was 0.3-0.6 and ${\beta}-sitosterol$ to campesterol 3.0-3.8. The 50 samples from the markets in Seoul were composed of 70% genuine sesame oil, and others were mixed with palm oil, rape seed oil and soybean oil.

Isolation of Anti-Herpes Simplex Virus Type 1(HSV-1) Component from Thujae orientalis Semen (백자인(栢子仁)으로부터 항Herpes 바이러스 1형(HSV-1) 물질의 분리)

  • In order to search for anti-Herpes simplex virus type 1(HSV-1) agents, we screened 80 specimens of Korean traditional medicine by SRB assay. The methanol extracts of Thujae orientalis Semen (Cupressaceae) showed strong anti-HSV activity among samples tested. From the butanol fraction of Thujae orientalis Semen anti-HSV-1 agent was isolated by chromatographic separation using Amberlite XAD-4 and Sephadex LH-20. The structure was elucidated by spectroscopic methods, and was identified as ${\beta}-sitosterol$ (compound I). Compound I exhibited anti-HSV-1 activity with $EC_{50}$ of 0.6 mg/ml and $CC_{50}$ of 5.99 mg/ml, respectively.

홍화유 Tocopherol 분석, Phytosterol 추출 최적화 및 CLA를 함유한 재구성지질 합성

  • 홍화유를 35$^{\circ}C$, 8$0^{\circ}C$ 추출조건 온도에서 각각 1, 3, 6시간 추출하여 HPLC 사용하여 $\alpha$,${\gamma}$,$\delta$-tocopherol 정량하였다. 정량결과 $\alpha$-tocopherol 함량이 ${\gamma}$-tocopherol 함량 보다 높게 나타났으며 $\delta$-tocopherol은 검출되지 않았다 총 tocopherol 함량은 35$^{\circ}C$ 추출조건에서 보다 8$0^{\circ}C$ 추출조건에서 높게 나타났으며 8$0^{\circ}C$ 6시간 추출조건에서 0.23%라는 함량을 나타났다. 또한 반응표면분석에 의하여 홍화유 추출조건에 따른 campesterol, stigmasterol, $\beta$-sitosterol 및 total sterol의 추출 최적화를 모니터링하였으며, 추출온도(35~75$^{\circ}C$, X$_1$), 추출시간(1~11시간, X$_2$), 시료의 전처리 가공 온도(60~10$0^{\circ}C$, X$_3$)를 변수로 한 중심합성계획으로 추출조건을 최적화하였다 추출온도, 추출시간 및 시료의 전처리 가공 온도가 각각 54.69$^{\circ}C$, 4.02시간, 98.37$^{\circ}C$ 이었을 때 campesterol은 0.3852% 추출율을 보였으나 stigmasterol, $\beta$-sitosterol 및 total sterol 반응표면분석 결과 유의차가 없었다. 한편, 추출된 홍화유와 conjugated linoleic acid (CLA)를 1:3 ㏖ 비율로 혼합한 후 고정화효소인 IM60 lipased와 1, 2, 3, 6, 24시간 동안 합성하여 재구성지질을 생성하고 GC에 의해 지방산 조성을 분석한 결과, 총 CLA의 함량은 각각 2.134%, 6.524%, 8.606%, 21.590%, 31.791%로 증가됨을 알 수 있었다.

  • Twelve constituents were isolated from the MeOH extract of the root of Brassica campestris L. ssp rapa. They were identified as linoleic acid methylester (1), palmitic acid (2), ${\beta}-sitosterol$ (3), 1-methoxyindole-3-acetonitrile (4), indole-3-acetonitrile (5), linolenic acid (6), goitrin (7),4-hydroxycinnamyl alcohol (8), coniferyl alcohol (9), p-coumaroylglucose (11) and feruloylglucose (12), on the basis of spectral data respectively.

Topoisomerase I and II Inhibitory Activities and Cytotoxic Constituents from the Barks of Tilia amurnesis

  • Eight compounds, squalene (1), friedelin (2), ${\beta}$-sitosterol (3), ${\beta}$-sitosterol-3-O-glucoside (4), ${\alpha}$-tocopherol (5), betulinic acid (6), trilinolein (7) and 1-O-(9Z,12Z-Octadecadienoyl)-3-nonadecanoyl glycerol (8), were isolated from the barks of Tilia amurensis. Their chemical structures were identified by comparing their physicochemical and spectral data with those published in the literature. These isolated compounds were examined for their inhibitory activities against topoisomerase I and II. Compound 7 showed significant inhibition of DNA topoisomerase I and II activities, with percent decreases in activity of 87 and 95%, respectively at a concentration of $100\;{\mu}M$. Compound 6 exhibited cytotoxicity against the human colon adenocarcinoma cell line (HT-29), the human breast adenocarcinoma cell line (MCF-7) and the human liver hepatoblastoma cell line (HepG-2), with $IC_{50}$ values of 20, 59 and $16\;{\mu}M$, respectively.

Inhibitory Effect on TNF-${\alpha}$-Induced IL-8 Production in the HT29 Cell of Constituents from the Leaf and Stem of Weigela subsessilis

  • Twelve compounds were isolated from the MeOH extract of the leaf and stem of the Korean endemic plant Weigela subsessilis L. H. Bailey. Their chemical structures were elucidated on the basis of physicochemical and spectroscopic data and by comparison with those of published literatures. These compounds were identified as three sterols, ${\beta}$-sitosterol acetate (2), ${\beta}$-sitosterol (3), daucosterol (11), eight triterpenoids, squalene (1), ursolic acid (4), ilekudinol A (5), corosolic acid (6), ilekudinol B (7), esculentic acid (8), pomolic acid (9), asiatic acid (10), and one iridoid glycoside, alboside I (12). This is the first report pertaining to the isolation of these compounds from Weigela subsessilis L. H. Bailey. In addition, three compounds 7, 9, and 12 were found to display a strong inhibitory effect on the production of IL-8 in the HT29 cells stimulated by TNF-${\alpha}$.

Studies on the Oil Soluble Constituents of Korean Ginseng -Part 1. On the Composition of Ginseng Sterols- (한국인삼(韓國人蔘)의 유용성성분(油溶性成分)에 관한 연구(硏究) -제1보 Sterol 성분(成分)에 관하여-)

  • Korean ginseng sterols were obtained from the non-saponifiable matters of etherial extract of the root. The composition of sterols have been determined by gas liquid chromatography and thin-layer chromatographic analysis. The results showed that contents of sterols were ${\beta}-sitosterol$, 77.87% by method of triangulation and 82.72% by methode of planimetry and stigmasterol,15.39% by methode of triangulation and 13.92% by methode of planimetry and campesterol, 6.74% by method of triangulation and 4.36% by methode of planimetry.
