Biopolymer is a versatile material used in food processing, medicine, construction, and soil reinforcement. 𝛽-glucan is one of the biopolymers that improves the soil water content and ion adsorption in a drought or toxic metal contaminated land for plant survival. We analyzed drought stress damage reduction in sweet potatoes (Ipomoea batatas L. cv. Sodammi) by measuring the growth and major protein expression and activity under 𝛽-glucan soil amendment. The result showed that sweet potato leaf length and width were not affected by drought stress for 14 days, but sweet potatoes grown in 𝛽-glucan-amended soil showed an effect in preventing wilting caused by drought in phenotypic changes. Under drought stress, sweet potato leaves did not show any changes in electrolyte leakage, but the relative water content was higher in sweet potatoes grown in 𝛽-glucan-amended soil than in normal soil. 𝛽-glucan soil amendment increased the expression of plasma membrane (PM) H+-ATPase, but it decreased the aquaporin PIP2 (plasma membrane intrinsic protein 2) in sweet potatoes under drought stress. Moreover, water maintenance affected the PM H+-ATPase activity, which contributed to tolerance under drought stress. These results indicate that 𝛽-glucan soil amendment improves the soil water content during drought and affects the water supply in sweet potatoes. Consequently, 𝛽-glucan is a potential material for maintaining soil water contents, and analysis of the major PM proteins is one of the indicators for evaluating the biopolymer effect on plant survival under drought stress.
The anti-oxidant activities and ${\beta}$-glucan contents of wild mushrooms collected from Songnisan, Soi-myeon, Joryeongsan, and Ganghwado, Korea, were investigated. Among the wild mushroom extracts, Pycnoporus sanguineus (OK1071), Piptoporus soloniensis (OK1090), and Daedalea dickinsii (OK1094) extracts showed the highest DPPH and $ABTS^+$ radical scavenging activities. The nitrite scavenging activity and total polyphenol content of the extract of P. sanguineus (OK1071) was 74.2% and 37.7 mg GAE/g, respectively, and these values were much higher than those of the other mushrooms analyzed in this study. The Pearson's correlation coefficients of the DPPH radical and nitrite scavenging activities with the total polyphenol contents of the wild mushroom extracts were r = 0.758 (p < .01) and r = 0.951 (p < .01), respectively. The ${\beta}$-glucan contents of Lenzites betulina (OK1040), Trametes versicolor (OK1044), and P. sanguineus (OK1071) were 47.8%, 43.9%, and 41.8%, respectively, which were higher than those of the other mushrooms. Therefore, the wild mushrooms analyzed were confirmed to have excellent antioxidant activities and high ${\beta}$-glucan contents. Accordingly, the fundamental data provided in this study can be used to isolate useful compounds from these wild mushrooms.
The amount of $\beta-glucans$ in 12 edible mushroom species was determined and their water-solubility was assessed. A large variability in $\beta-glucans$ content was observed in the mushroom species, ranging from 4.71 to $46.20\%$ on a dry basis. Gyrophora esculenta, Lentinus edodes, Coriolus versicolor, Ganodenna lucidum, and Flammulina velutipes had high levels of $\beta-glucans$ Soluble $\beta-glucans$ content, which plays a key role in the physiological effects of mushrooms, also varied greatly according to the mushroom species, ranging from 2.12 to $19.66\%$. Water-solubility of $\beta-glucans$ in the edible mushrooms, as a percentage of total $\beta-glucans$ content varied from 42.55 to $73.35\%$.

This study was carried out to investigate the component and immunoregulatory effects of Sparassis crispa stipe. Herein, S. crispa was divided into the pileus and stipe to compare their ingredients (${\beta}-glucan$), antioxidant activity (in vitro), and the immunoregulatory function (cytokines, leukocytes, and spleen weight). The ${\beta}-glucan$ content in each part showed about 1.8 times higher content in the stipe than that in the pileus. The stipe also showed a higher total phenol content and antioxidant activity than the pileus. The cytokines $TNF-{\alpha}$, IL-2, and IL-10 have adjusted in the S. crispa extract-injected groups. In addition, the number of leukocytes was also significantly elevated in the rats administered with the S. crispa stipe extract. These results suggest that the stipe of S. crispa has great potential as an ingredient in functional foods.
In order to breed barley lines with reduced viscosity of wort, a fractured starch mutant of naked barley cultivar, Nubet, was obtained from the M2 seeds mutated by the diethyl sulfate treatment. Seeds of this fractured starch mutant were opaque and the endosperm consists of angular, irregular and fractured starch. The mutant was caused by single recessive mutation and assigned by the symbol fra. The gene was located on chromosome 4, distal in long arm by linkage recombinations using translocation homozygote lethal test set. The linkage value between the fractured starch mutant and 72-4a, 72-4d were 26.0$\pm$4.9, 34.2$\pm$3.1 percent respectively. In addition to the reduced seed size, fewer kernels per spike and higher tillering ability, lower $\beta$-glucan viscosity and higher lysine content of the grain were associated with this mutant. $\beta$-glucan viscosity of the Nubet grains increased from 3 weeks after anthesis to matury and most of the viscose substances appeared to be stored in the middle of the endosperm tissue. Since the mutant grains showed better milling property as compared to Nubet, it can be used as breeding resources to develope new barley cultivars for maltins and milling purpose.

This study was conducted to investigate the physiological activities of the ethanol extracts from 10 different strains of Pholiota nameko. In addition, the ${\beta}$-glucan and polyphenol contents from these strains were also measured. Total polyphenol contents from all the strains were more than 40 mg% with the highest content of $61.50{\pm}0.59$ mg% and ${\beta}$-glucan contents were 30% with the highest content of $37.20{\pm}1.12%$. The highest inhibitory activities on ${\alpha}$-amyloglucosidase activity and nitric oxide production were $13.78{\pm}0.56%$ and $56.59{\pm}7.11%$, respectively. However, ethanol extracts from all the strains have little effects on DPPH radical and nitrite scavenging rates and the activity of angiotensin I converting enzyme. The cytotoxic activity of ethanol extracts from all the strains on A549 cell was shown upto 30% with the highest effect of $47.96{\pm}8.46%$ treated at 1 mg/mL concentration.
Objectives This study was conducted to investigate the beta-glucan, ginsenoside content, antioxidant activity and safety of modified Kyungohkgo added to Sparassis crispa and Hericium erinaceum. Methods The marker compounds contents, antioxidant activity and safety of modified Kyungohkgo were tested. The contents of beta-glucan and ginsenoside Rb1, Rg1, and Rg3 marker compounds were measured, the antioxidant activity was measured using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, and a safety test was conducted via single dose toxicity assessment. Results Analyzing the contents of marker compounds showed 351.75 mg/g of beta-glucan, 0.0327 mg/g of ginsenoside Rb1 and 0.0802 mg/g of ginsenosai Rg3. In the DPPH free radical scavenging activity, the inhibition concentration 50% of modified Kyungohkgo was 0.2880%. The scavenging activity of modified Kyungohkgo was 5.49% activity at 0.05% concentration, 89.66% activity at 0.5% concentration, 94.68% activity at 1% concentration, and 96.06% activity at 5% concentration. In the single dose toxicity test of modified Kyungohkgo, a dose of 2,000 mg/kg B.W. was set at its highest capacity and observed after oral administration to female and male rats. No toxicological findings were recognized. It was observed that the resulting lethal dose can be set to 2,000 mg/kg B.W. or higher for both females and males. Conclusions The results of the experiment on modified Kyungohkgo showed that the marker compounds contents were beta-glucan and ginsenoside Rb1 and Rg3, that antioxidant activity was observed through the DPPH free radical scavenging activity, and safety was confirmed through the single dose toxicity assessment.

Objectives This study was conducted to investigate the beta-glucan & ginsenoside content, antioxidant activity, anti-inflammatory effect and safety of herbal medicine mix. Methods The marker compounds contents, antioxidant activity and safety of herbal medicine mix were tested. The contents of beta-glucan and ginsenoside Rg3 were measured, the antioxidant activity was measured using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, anti-inflammatory and a safety test was conducted via single dose toxicity assessment. Results Analyzing the contents of marker compounds showed 362.3 mg/g of beta-glucan, and 0.4184 mg/g of ginsenoside Rg3. In the DPPH free radical scavenging activity, the IC50 of herbal medicine mix, was 0.146%. The scavenging activity of herbal medicine mix was 88.28% activity at 0.5% concentration, and 90.61% activity at 5% concentration. In the lipopolysaccharides (LPS) anti-inflammatory test, the herbal remix showed a significant decrease in tumor necrosis factor-alpha (TNF-𝛼) and interleukin-6 (IL-6) compared to the LPS-induced group. In the single dose toxicity test of herbal medicine mix, a dose of 2,000 mg/kg body weight (BW) was set at its highest capacity and observed after oral administration to female and male rats. No toxicological findings were recognized. It was observed that the resulting lethal dose can be set to 2,000 mg/kg BW or higher for both females and males. Conclusions The results of the experiment on herbal medicine mix showed that the marker compounds contents were beta-glucan and ginsenoside Rg3, that antioxidant activity was observed through the DPPH free radical scavenging activity, anti-inflammatory effect was observed through TNF-𝛼 and IL-6 measurement, and safety was confirmed through the single dose toxicity assessment.

This study investigated the isoflavone content, total phenol content, antioxidant activities (DPPH radical scavenging and oxygen radical absorbance capacity) and ${\beta}$-glucan content of defatted soybean extracts by bioconversion. Soybean was fermented with Lentinula edodes using submerged liquid fermentation system. Defatted soybean powder prepared by hexane (HDS; hexane defatted soybean) and ethanol (EDS; ethanol defatted soybean). The major components of non-fermented HDS (NFHDS) and EDS (NFEDS) were glucoside, such as daidzin, glycitin and genistin. During the bioconversion processing, isoflavone glucoside converted into aglycone such as daidzein, glycitein and genistein. The highest total isoflavone contents of fermented HDS (FHDS) were $2577.96{\mu}g/mL$, and the lowest total isoflavone contents of NFEDS were $428.27{\mu}g/mL$. The highest total phenol contents of fermented EDS (FEDS) was 42.34 mg GAE/g. DPPH radical scavenging and ORAC value were 31.30 to 59.92% and 247.48 to $786.36{\mu}M\;TE/g$ in non-fermented defatted soybean and fermented soybean, respectively. ${\beta}$-Glucan contents were 0.09 to 0.11% in non-fermented defatted soybean and fermented soybean, respectively. These results indicate that fermented soybean could be used as natural antioxidants for the development of functional foods.

Sparassis (S.) crispa is an edible mushroom abundant in dietary fiber and ${\beta}$-glucan. The aim of this study was to prepare extracts and residues of the fruit bodies of S. crispa fermented with Lactobacillus (L.) brevis and Monascus (M.) pilosus and to measure the remaining dietary fiber and ${\beta}$-glucan. Dried powder of S. crispa containing 64.4 g/100 g total dietary fiber (2.6 g/100 g soluble and 61.8 g/100 g insoluble dietary fibers) and 24.0 g/100 g ${\beta}$-glucan was used as the starting material for the extraction. Raw and fermented S. crispa were extracted with hot water and three kinds of aqueous ethanol (50, 70, and 90%, v/v), respectively. A hot water extract from S. crispa fermented with M. pilosus had greater soluble dietary fiber content (19.3 g/100 g) than that from raw S. crispa with 14.6 g/100 g soluble dietary fiber or that from L. brevis-fermented S. crispa with 8.2 g/100 g soluble dietary fiber. The yield of the extract was 16.6% of intial weight of dried S. crispa. After hot water extraction of S. crispa fermented with M. pilosus, residues containing 90.5 g/100 g total dietary fiber (1.3 g/100 g soluble and 89.2 g/100 g insoluble dietary fibers) were obtained, and the yield was 69.6% of intial weight of dried S. crispa. The residue (31.0 g/100 g) contained more ${\beta}$-glucan than raw S. crispa or M. pilosus-fermented S. crispa (24.4 g/100 g). The resulting hot water extract and residue from S. crispa fermented with M. pilosus would be suitable for use in preparing liquid and powdered health functional foods, respectively.
