Development of a Functional Mortar for Restraining Surface Algal Growth

  • Proliferation of algae on the surface of concrete or mortar in aquatic habitat has a negative impact on maintenance of concrete-based structures. Growth of algae may decrease stability of structure by bio-deterioration. In this study, we developed a functional mortar for restraining bio-deterioration by using $Cu^{2+}$ ion. The mortar contains soluble glass beads made of $Cu^{2+}$ ion, which can dissolve into water slowly. Mortars prepared with different ratio of glass beads (0, 2, 5, 10, and 15%) were placed in a culture medium with algae and incubated over a month period. Water chemistry, chlorophyll-a, and extracellular enzyme activities were measured. The incubation was conducted in both freshwater and seawater conditions, to assess applicability to both aquatic conditions. Overall, mortar with Cu glass exhibited lower chlorophyll-a content, suggesting that the functional mortar reduced algal growth. DOC concentration increased because debris of dead algae increased. Cu glass also decreased phosphatase activity, which is involved in the regeneration of inorganic P from organic moieties. Since, P is often a limiting nutrient for algal production, algal growth may be inhibited. Activities of ${\beta}$-glucosidase and N-acetylglucosaminidase were not significantly affected because carbon and nitrogen mineralization may not be influenced by the Cu glass beads. Our study suggests that functional mortar with Cu glass beads may reduce the growth of algae on the surface, while it has little environmental impact.

Hesperidin 대사에 의한 대금음자(對金飮子)와 평위산(平胃散)의 처방해석 (Scientific Analysis of Daekumeumja and Pyungwesan by the Metabolism of Hesperidin)

  • To analyze scientifically the polyprescription principle of Daekumeumja(對金飮子), which has been used for alcoholic damage, and Pyungwesan(平胃散), which has been used for indigestion, the transforming rate of hesperidin of these polyprescriptions to hesperetin was investigated. The transforming rate of the former was higher 3 times than that of the latter. The transforming rate of hesperidin of Aurantii nobilis Pericarpium was inhibited by Magnoliae Cortex, but was activated by Glycyrrhizae Radix. The activity of trypsin was inhibited by Glycyrrhizae Radix and Daekumeuja. However, Aurantii nobilis Pericarpium, Atractylodis Rhizoma, Magnoliae Cortex and Pyungwesan did not inhibited it. When human intestinal microflora were cultured with the media containing Daekumeumja, Pyungwesan and herbal medicines consisting of them, Aurantii nobilis Pericarpium and Magnoliae Cortex inhibited the activity of ${\beta}$-glucosidase. These results suggests that the therapeutic effect of Daekumeumja may be better than that of Pyungwesan on alcoholic damage and the therapeutic effect of Pyungwesan may be better than that of Daekumeumja on indigestion, although these prescriptions are consisted of the same herbal medicines.

Comparison of Dyes for Easy Detection of Extracellular Cellulases in Fungi

  • To evaluate which dye is effective in a plate assay for detecting extracellular cellulase activity produced by fungi, four chromogenic dyes including remazol brilliant blue, phenol red, congo red, and tryphan blue, were compared using chromagepic media. For the comparison, 19 fungal species belonging to three phyla, ascomycota, basidiomycota, and zygomycota were inoculated onto yeast nitrogen-based media containing different carbon substrates such as cellulose (carboxylmethyl and avicel types) and cellobiose labeled with each of the four dyes. Overall, the formation of clear zone on agar media resulting from the degradation of the substrates by the enzymes secreted from the test fungi was most apparent with media containing congo red. The detection frequency of cellulase activity was also most high on congo red-supplemented media. The results of this study showed that congo red is better dye than other three dyes in, a plate assay for, fungal enzyme detection.

Extracellular Enzyme Activities of the Monokaryotic Strains Generated from Basidiospores of Shiitake Mushroom

  • To obtain basic information on the biochemical property of basidiospores of shiitake mushroom (Lentinula edodes), the ability of producing extracellular enzyme was assessed using a chromogenic plate-based assay. For the aim, amylase, avicelase, $\beta$-glucosidase, CM-cellulase, pectinase, proteinase, and xylanase were tested against monokaryotic strains generated from forty basidiospores of two different parental dikaryotic strains of shiitake mushroom, Sanjo-101Ho and Sanjo-108Ho. These two parental strains showed different degree of extracellular enzyme activity. No identical patterns of the degree of enzyme activity were observed between monokaryotic strains and parental strains of the two shiitake cultivars. The degree of extracellular enzyme activity also varied among monokaryotic strains of the two shiitake cultivars. Our results showed that dikaryotic parental strains of shiitake mushroom produce monokaryotic basidiospores having very diverse biochemical properties.

  • Aucubin, an iridoid glucoside isolated from Aucuba japonica, has pharmacological effects such as antimicrobial effect, liver protective function and inhibition of liver RNA and protein biosynthesis, etc. This study was performed to observe the effect of aucubin on the pulp tissue after pulpotomy. Aucubin was converted to aucubinogenin as an active form by ${\beta}$-glucosidase. In 3 Mongrel dogs, the pulps were mechanically exposed with a sterile round bur and excised with sterile excavator. After bleeding control, aucubin was applied on remaining pulp tissues and then they were sealed with IRM in experimental group. In control group, $Ca(OH)_2$ powder was applied on remaining pulp tissues and then they were sealed with IRM. After interval of 1 and 12 weeks, the dogs were sacrificed. The teeth were prepared for histologic evaluation and examined by light microscope. Aucubin 1 week group showed that mild inflammation and vascular congestion in most of the specimen. More various degree of inflammation was found in experimental group than in control group. Premature calcified mass were found in the both 1 weeks groups. Continuous well-formed dentin bridge was found in both 12 weeks groups. Collectively, this study suggests that the possibility of aucubin as a medicament after pulpotomy.

Changes in Physiochemical Properties during the Fermentation of Doenjang Prepared with Black Soybeans

  • A physicochemical assessment of Doenjang (traditional fermented soyfood) prepared with Korean black soybeans (Glycine max) was carried out. The T-N rate increased slowly during storage up to 120 days and the A-N rate increased up to 80 days of ripening and then decreased slightly. The caseinolytic activity increased slowly during storage up to 80 days and then decreased after 80 days. In addition, the fibrinolytic and ${\beta}-glucosidase$ activities increased up to 80 and 30 days and then decreased. Genistin and daidzin concentrations gradually decreased with increased fermentation time. However, genistein and daidzein slowly increased with fermentation time. Genistein and daidzein reached maximum concentrations (316.8 and $305.2{\mu}g/g$, respectively) and plateaued thereafter. The anthocyanins increased greatly during fermentation up to 50 days and then remained constant between 50 and 90 days. Polyphenol contents showed a slight increase up to 80 days and then slowly decreased. The DPPH and ABTS radical scavenging activities increased linearly during storage up to 50 days, reached about 28.9% and 2.17 mg/g, respectively, and then slowly decreased. At 20 days of fermentation, macrophage-stimulating activity of the extract showed a maximum activity.

The Inhibitory Effect of Prunioside A Acyl Derivatives on NO Production in RAW 264.7 Cell

  • Prunioside A is a unique, highly oxidized monoterpene glycoside isolated from the methanol extract of Spiraea prunifolia var. Simpliciflora's root. The ester derivatives were synthesized from the hydrolyzed compounds of prunioside A by ${\beta}$-glucosidase. The derivatives showed suppressive effects on the generation of nitric oxide in murine machrophage-like RAW 264.7 cells stimulated by lipopolysaccharide and interferon- ${\gamma}$.

Antialgal Effect of a Novel Polysaccharolytic Sinorhizobium kostiense AFK-13 on Anabaena flos-aquae Causing Water Bloom

  • Isolation and identification of algal lytic bacteria were carried out. Nine strains of algal lytic bacteria were isolated by the double-layer method using Anabaena flos-aquae as a sole nutrient. The isolate, AFK-13, showing the highest algal lytic activity was identified as Sinorhizobium kostiense based on the l6S rDNA sequence. The algal lytic experiments of the culture supernatants of AFK-13 demonstrated that the bacterial cell growth reached a maximum at 36-h culture, but the supernatant of 72-h culture exhibited the highest activity. Components among the extracellular products in the crude enzyme of the supernatant from S. kostiense AFK-13 culture were responsible for degradation of cell walls of Anabaena flos-aquae. Algal lytic assay tests of the culture supernatants suggest that the main substances for algal lytic activity could be proteinaceous. The activity of glucosidase was observed highly by polysaccharolytic analysis using the crude enzyme from S. kostiense AFK-13, whereas activities of galactosidase, mannosidase, rhamnosidase, and arabinosidase were also detected in low levels. The molecular weights (MW) of ${\alpha}-\;and\;{\beta}$-glucosidases were estimated to be approximately 50-100 kDa by the ultrafiltration method.

Varying Inocula Permutations (Aspergillus oryzae and Bacillus amyloliquefaciens) affect Enzyme Activities and Metabolite Levels in Koji

  • In this study, we investigated the altered enzymatic activities and metabolite profiles of koji fermented using varying permutations of Aspergillus oryzae and/or Bacillus amyloliquefaciens. Notably, the protease and ${\beta}$-glucosidase activities were manifold increased in co-inoculated (CO) koji samples (co-inoculation of A. oryzae and B. amyloliquefaciens). Furthermore, gas chromatography-mass spectrometry (GC-MS)-based metabolite profiling indicates that levels of amino acids, organic acids, sugars, sugar alcohols, fatty acids, nucleosides, and vitamins were distinctly higher in CO, SA (sequential inoculation of A. oryzae, followed by B. amyloliquefaciens), and SB (sequential inoculation of B. amyloliquefaciens, followed by A. oryzae). The multivariate principal component analysis (PCA) plot based on GC-MS datasets indicated a clustered pattern for MA and MB (koji samples inoculated either with A. oryzae or B. amyloliquefaciens) across PC2 (20.0%). In contrast, the CO, SA, and SB metabolite profiles displayed segregated patterns across PLS1 (22.2%) and PLS2 (21.1%) in the partial least-square discriminant analysis (PLS-DA) model. Intriguingly, the observed disparity in the levels of primary metabolites was engendered largely by higher relative levels of sugars and sugar alcohols in MA, SA, and CO koji samples, which was commensurate with the relative amylase activities in respective samples. Collectively, the present study emphasizes the utility of integrated biochemical and metabolomic approaches for achieving the optimal permutation of fermentative inocula for industrial koji preparation.

The MAP Kinase Kinase Gene AbSte7 Regulates Multiple Aspects of Alternaria brassicicola Pathogenesis

  • Mitogen-activated protein kinase (MAPK) cascades in fungi are ubiquitously conserved signaling pathways that regulate stress responses, vegetative growth, pathogenicity, and many other developmental processes. Previously, we reported that the AbSte7 gene, which encodes a mitogen-activated protein kinase kinase (MAPKK) in Alternaria brassicicola, plays a central role in pathogenicity against host cabbage plants. In this research, we further characterized the role of AbSte7 in the pathogenicity of this fungus using ${\Delta}AbSte7$ mutants. Disruption of the AbSte7 gene of A. brassicicola reduced accumulation of metabolites toxic to the host plant in liquid culture media. The ${\Delta}AbSte7$ mutants could not efficiently detoxify cruciferous phytoalexin brassinin, possibly due to reduced expression of the brassinin hydrolase gene involved in detoxifying brassinin. Disruption of the AbSte7 gene also severely impaired fungal detoxification of reactive oxygen species. AbSte7 gene disruption reduced the enzymatic activity of cell walldegrading enzymes, including cellulase, ${\beta}$-glucosidase, pectin methylesterase, polymethyl-galacturonase, and polygalacturonic acid transeliminase, during host plant infection. Altogether, the data strongly suggest the MAPKK gene AbSte7 plays a pivotal role in A. brassicicola during host infection by regulating multiple steps, and thus increasing pathogenicity and inhibiting host defenses.
