Study of plasma β-carotene concentration in dairy cows (젖소의 혈장 베타카로틴 농도 조사)

  • This study was carried out to determine the effects of ${\beta}-carotene$ on the control of mastitis in dairy cows during the dry period. The relationship between the levels of plasma ${\beta}-carotene$ and the status of udder health in Holstein dairy cows were investigated. Blood samples were collected from 117 cows to compare the levels of plasma ${\beta}-carotene$ in lactating cows. The levels of plasma ${\beta}-carotene$ were $1.82{\mu}g/ml$ in healthy cows(n = 65) and $1.12{\mu}g/ml$ in mastitic cows(n = 52), respectively(p < 0.01). In the experiment to compare the level of plasma ${\beta}-carotene$ in the cows at different stages of lactation, the plasma ${\beta}-carotene$ levels were $1.73{\mu}g/ml$ in lactating cows(n = 22), $1.29{\mu}g/ml$ in nonlactating cows(n = 35) and $0.43{\mu}g/ml$ in cows after calving(n = 16)(p < 0.05).

Optimization for Extraction of ${\beta}-Carotene$ from Carrot by Supercritical Carbon Dioxide (초임계 유체에 의한 당근의 ${\beta}-Carotene$ 추출의 최적화)

  • Supercritical fluid extraction of ${\beta}$-carotene from carrot was optimized to maximize ${\beta}$-carotene (Y) extraction yield. A central composite design involving extraction pressure ($X_1$ 200-,100 bar), temperature ($X_2,\;35-51^{\circ}C$) and time ($X_1$$ 60-200min) was used. Three independent factors ($X_1,\;X_2,\;X_3$) were chosen to determine their effects on the various responses and the function was expressed in terms of a quadratic polynomial equation,$Y={\beta}_0+{\beta}_1X_1+{\beta}_2X_2+{\beta}_3X_3+{\beta}_11X_12+{\beta}_22X_3^2+{\beta}_-12X_1X_2+{\beta}_12X_1X_2+{\beta}_13X_1X_3+{\beta}_23X_2X_3,$ which measures the linear, quadratic and interaction effects. Extraction yields of ${\beta}$-carotene were affected by pressure, time and temperature in the decreasing order, and linear effect of tenter point (${\beta}_11$) and pressure (${\beta}_1$) were significant at a level of 0.001(${\alpha}$). Based on the analysis of variance, the model fitted for ${\beta}_11$-carotene (Y) was significant at 5% confidence level and the coefficient of determination was 0.938. According to the response surface of ${\beta}$-carotene by cannoical analysis, the stationary point for quantitatively dependent variable (Y) was found to be the maximum point for extraction yield. Response area for ${\beta}$-carotene (Y) in terms of interesting region was estimated over $10,611{\mu}g$ Per 100 g raw carrot under extraction.

  • The micronucleus test using peripheral blood reticulocytes (RETs) was evaluated in ICR mice treated with N-methyI-N-nitrosourea (MNU) and benzo(a)pyrene [B(a)P] as model clastogens. The frequency of micronucleated reticulocytes (MNRETs) in both positive compounds was similar to other results which were reported previously. On the other hand, an anticlastogenic effect of the natural antioxidant, $\beta$-carotene and one of taroholds, galangin as model anticlastogens were investigated using simultaneous treatment. Mice were treated with a model clastogen alone, or with a model clastogen and a model anficlastogen simultaneously. Both $\beta$-carotene and galangin showed anticlastogenic effects against MNU- or B(a)P-induced micronuclei in mice. However, galangin has stronger activity than $\beta$-carotene. Results from our experiment suggest that the in vivo supravital staining micronucleus test using peripheral blood is useful in the evaluation of clastogenic and anticlastogenic effects.

Levels and Factors Affecting Serum Carotenoids Concentrations of Adults Living in Rural Area of Korea (한국농촌 성인의 혈청 Carotenoids 농도와 이에 영향을 미치는 요인에 관한 연구)

  • Recent researches suggest that carotenoids are important not only as provitamin A but also for prevention of chronic diseases. This study was conduction to determine levels and factors affecting serum levels of lutein + zeaxanthin, $\beta$-cryptoxanthin, and $\beta$-carotene in 93 adults living in rural area of Korea. Fasting blood samples were collected and serum carotenoid levels were measured by HPLC. Dietary intake was estimated by 24 hour recall method and frequency questionnare of major food groups. Mean serum concentration of lutein + zeaxanthin was 616.32 nmol/L, $\beta$-cryptoxanthin was 856.95nmol/L, and $\beta$-carotene was 242.90nmol/L. Serum $\beta$-carotene levels in study subjects were very low. Both $\beta$-cryptxanthin and $\beta$-carotene were negatively correlated with serum triglyceride and positively correlated with total-choesterol and LDL-cholesterol. Serum levels of female subjects were significantly higher than males in all carotenoids. For age groups, subjects in their 30's were shown to have the highest concentration of all carotenoids. Lutein + zeaxanthin were lowest in subjects in theri 40's while $\beta$-crytoxanthin and $\beta$-carotene levels were lowest in subjects in their 60's. The $\beta$-carotene levels in non-smokers were significantly higher than in drinkers. Lutein+zeaxanthin levels were significantly higher among subjects consuming more green and yellow vegetables by frequency questionnarie. In conclusion, serum carotenoids were affected by sex, age, serum lipids, smoking, and alcohol intake. Intake of vegetables and fruits could affect by sex, serum lipids, smoking, and alchol intake. Intake of vegetables and fruits could affect serum lutein+zeaxanthin level. This data indicated that compared to other studies, Korean adults in rural areas have high lutein+zeaxanthin concentratins and low $\beta$-carotene concentrations in serum. High lutein+zeaxanthin levels may be related to high consumption of vegetables in these subjects.

  • the Chinese cabbage kimchi was fermented with the various submaterials such as hot pep-per garlic ginger leek green onion fermented anchovy juice and sugar according to the average contents of each submaterial described in the 39 kinds of references. And then the effects of each submaterial and lactic acid bacteria such as L. brevis. Leu. mesenteroides. P cerevisiae and L. plantarum on the content of carotenes were investigated, The major carotene in kimchi was $\beta$-carotene. And also $\delta$-carotene and $\alpha$-carotene were detected. Contents of $\beta$-carotene and total carotene were high in the kimchi containing leek red pepper powder green onion and fermented anchovy juice as a submaterial. But the kimchi containing or omitting the other submaterials were litter affected to the contents of carotene. Contents of $\beta$-carotene and total carotene were high in kimchi fermented with Leu. msenteroides L. brevis and P. cerevi-siae as a starter but was low with L plasntarum.

  • Carotenoids in commercial green-yellow vegetables(carrot, mugwort, perilla leaf, leek and water dropwort) were analyzed by HPLC. Carotenoids detected were lutein, ${\alpha}$-carotene, and ${\beta}$-carotene. ${\beta}$-Carotene and lutein were detected in every sample analyzed, but ${\alpha}$-carotene could only be detected in carrot. Blanching vegetables in 3% saline increased the content of carotenoids, however, the components of carotenoids were not changed. This result suggests that blanching increases the amount of available carotenoids.

  • Effects of beta-carotene on the immunobiological responses were studied in ICR mice. ICR male mice were divided into 8 groups (10 mice/group), and beta-carotene at doses of 4, 20 and 100 mg/kg were orally administered to ICR mice once daily for 28 consecutive days. Cyclophosphamide (CY) was injected intraperitoneally (i.p.) to ICR mice with a single dose of 5 mg/kg body weight at 2 days before secondary immunization. Mice were sensitized and challenged with sheep red blood cells (5-RBC). Immune responses were evaluated by humoral immunity, cellular immunity and non-specific immunity. The results of this study were summarized as follows: (1) Beta-carotene significantly increased the weight ratios of liver, spleen and thymus to body weight depending on dose, and significantly increased the increasing rate of body weight and the number of circulating leukocyte. (2) Beta-carotene dose-dependently increased hemagglutination titer, Arthus reaction and hemolytic plaque forming cell related to humoral immunity. (3) Beta-carotene significantly increased delayed-type hypersensitivity reaction and rosette forming cell related to cellular immunity. (4) Beta-carotene dose-dependently increased phagocytic activity, and significantly increased natural killer (NK) cell activity. (5) Beta-carotene dose-dependently inhibited reductions in humoral immunity, cellular immunity, NK cell activity and phagocytic activity by treatment with CY.

Production of $\beta$-Carotene-Enriched Rice Bran Using Solid-State Fermentation of Rhodotorula glutinis

  • This work was aimed at utilizing rice bran as a substrate for $\beta$-carotene production by Rhodotorula glutinis DM 28 under optimized conditions of solid-state fermentation. The biomass and $\beta$-carotene content of Rhodotorula glutinis DM 28 grown on rice bran as a sole substrate under solid-state fermentation were 54 g/kg rice bran and 1.65 mg/kg rice bran, respectively. Its biomass and $\beta$-carotene content, however, could be improved by 60% and 30%, respectively, using the Central Composite Design for the optimization of its cultivation conditions. The optimized conditions obtained were a pH of 5, a moisture content of 70% (w/w), and a carbon-to-nitrogen ratio of 4. Under these conditions, rice bran containing R. glutinis DM 28 had nutritional values of $\beta$-carotene, protein, and fat higher than those of rice bran alone. Yeast-grown rice bran could be suitable, therefore, to use as a $\beta$-carotene-enriched supplement in animal feeds.

Effect of Tocopherols and $\beta$-Carotene on the Oxidation of Linoleic Acid Mixture in the Solid Model System (리놀레산 함유 고형 모델시스템의 산화에 미치는 토코페롤 및 베타 카로틴의 영향)

  • Effects of tocopherols and $\beta$-carotene on the oxidation of the solid model system of a free fatty acid mixture (64.5% of linolic acid ; 26.4% of oleic acid ; 5.0% of palmitic acid) with tocopherols and $\beta$-carotene were studied. $\alpha$-tocopherol revealed an antioxidant activity at the concentration below 0.05%, however, it showed a prooxidant activity when the concentration was higher than 0.05%. The antioxidant activity of ${\gamma}$ -tocopherol was not affected by the concentrations in the range of 0.01~0.10% in the model and ${\gamma}$-tocopherol showed higher antioxidant activity than that of $\alpha$-tocopherol. It seemed that $\alpha$-tocopherol was unstable compared to ${\gamma}$-tocopherol during oxidation. $\beta$-carotene showed a weak antioxidative activity at the initial stage of this system while $\beta$-carotene showed a prooxidant activity in the presence of tocopherol. $\beta$ -carotene was highly susceptible to autoxidative degradation during oxidation.

Effects of $\beta-Carotene$ on the Thermal Oxidation Stability of Deep Fried Lard (튀김용 돈지의 열산화 안정성에 미치는 $\beta-Carotene$의 효과)

  • This study was designed to investigate the effect of $\beta-carotene$ on the heat oxidation stability of deep fried laid with different healing temperature. Commercially purchased $\beta-carotene$ was used to this experiment $\beta-carotene$ was added to the deep fried lard at final concentration of $0\%\;0.001\%\;and\;0.004\%$ (wt/wt fat basis). Total plate count peroxide value, acid value, and TBA(thiobarbituric acid) values of the samples were determined regulary at $50^{\circ}C$ during 15 days. The result of the study are as follows: The number of microorganism was decreased by $\beta-carotene$ added to the deep fried lard Deep fried lard with $0.004\%$ $\beta-carotene$ have higher antioxidative effect than those of control and $0.001\%$ treated group. Peroxide and TBA values were significantly (P<0.05) decreased in all samples by added $\beta-carotene$ at $0.004\%$ Antioxidative effect increased with increasing concentration of $\beta-carotene$. These results suggest that $\beta-carotene$ might inhibit the growth of microganisms and retard lipid oxidation in the deep fried lard.
