  • $^{93}Nb(n,\alpha)^{90}Y$, $^{90}Zr(n,p)^{90}Y$,$^{93}Nb(n,\alpha)^{90m}Y$$^{90}Zr(n,p)^{90m}Y$반응의 평균 핵분열 반응단면적을 결정하였다. $\alpha-$히드록시 이소부티르산을 용출제로한 양이온 교환 수지통을 사용하여 생성된 핵종을 정량적으로 분리하였다. $^{90m}Y$$^{90}Y$의 절대측정은 $\gamma-$선 분광법 및 보정된 $2\pi$계측기로 각각 $0.14\pm0.01mb$, $0.83\pm0.02mb$, $0.018\pm0.02mb$$0.33\pm0.02mb$이었다. $^{90}Y$ 대신 $^{90m}Y$ 를 사용하는 편이 니오브를 정량하기 위한 보다 좋은 분석방법이 될 수 있음을 알았으며 그 가능성에 대하여 검토하였다.

Theanine이 남자 성인의 대뇌 $\alpha$파 발현에 미치는 영향 (Effects of Theanine on the Release of Brain Alpha Wave in Adult Males)

  • L-theanine Is an amino acid in green tea and has been known to decrease serotonin and increase norepinephrine in rat brains, and also reported to produce mental relaxation, lower blood pressure and improve learning ability in human beings. But, few studies on these effects for human beings have been conducted so far. This study was conducted to evaluate the effect of L-theanine on the release of brain alpha waves known to be related with mental relaxation and concentration. Twenty healthy male volunteers aged 18 to 30 years without any Physical and Psychological diseases were recruited through written advertisement. Alpha power values of EEG as a surrogate marker of mental relaxation and concentration were measured in frontal and occipital regions for 40 minutes after administration of four placebo or test tablets and 20 minute resting period. The same procedure crossed over at 7-day intervals. We analyzed average alpha power values in frontal and occipital regions at 10 minute intervals. Repeated ANOVA revealed that there were significant differences of occipital alpha power values between placebo and test groups with high anxiety (p < 0.05). The mean values at 20,30,40,50 and 60 minute intervals were 0.23, 024, 0.28, 0.25 and 0.34 in placebo, respectively and 0.23, 0.29, 0.40, 0.34, and 0.45 in test, respectively. But there were no significant differences of frontal and occipital alpha power values between placebo and test groups with low anxiety (p > 0.05) . The results of this study suggest that L-theanine containing tablets promote the release of alpha waves related to mental relaxation and concentration in young adult males.

정액 내 Tumor Necrosis Factor-alpha 농도와 정자 DNA 손상과의 관련성 (Seminal Tumor Necrosis Factor-alpha Level and Sperm Nuclear DNA Integrity in Healthy Donors)

  • 목 적: 정액 내 tumor necrosis factor-alpha (TNF-${\alpha}$) 농도와 정자 DNA 손상 및 정액 검사 소견과의 관련성을 평가하고자 하였다. 연구방법: 정액 표본은 45명의 건강한 남성에서 자위에 의하여 획득하였다. 정자의 상태는 컴퓨터 정액 분석기를 이용하여 판정하였으며, 두부의 DNA 손상은 TUNEL 분석방법에 의해 측정하였다. TNF-${\alpha}$ 농도는 동결-융해된 정장액에서 ELISA법으로 측정하였다. 결 과: 정자 DNA 손상율은 1.9%에서 53% (mean ${\pm}$ SD, 12.4${\pm}$9.6%)로 매우 광범위하게 나타났다. 단변량분석에 의하면 DNA 손상 정도와 정자의 농도, 운동성과는 관련이 없었으나, 직진운동성 (linearity)과는 음의 상관 관계를 나타내었으며 (r=-0.325, p=0.03) 연구 대상 남성의 연령과는 양의 상관 관계를 나타내었다 (r=0.484, p=0.001). 정액내에 존재하는 TNF-${\alpha}$ (>1 pg/mL)는 연구 대상 남성의 73.3% (33/45)에서 검출되었으며 평균 농도는 4.9 pg/mL, 범위는 1.1에서 22.6 pg/mL이었다. 정액 검사 상의 정자 상태와 정자 DNA 손상과는 유의한 관련성이 나타나지 않았다. 결 론: 본 연구에서는 정자 DNA의 손상이 남성의 연령과 관련성이 있음을 확인하였으나, TNF-${\alpha}$와의 관련성은 확인할 수 없었다.

Prostaglandin $F2{\alpha}$ 투여가 수퇘지의 성행동과 정액 채취 훈련에 미치는 영향 (The Effects of $PGF_2{\alpha}$ on Sexual Activity and Semen Collection Training)

  • Prostaglandin $F_2{\alpha}$ ($PGF_2{\alpha}$) can facilitate release of epinephrine from the adrenal gland. The objective was to extend these findings and determine the effects of $PGF_2{\alpha}$ on sexual activity and semen collection training in sexually inexperienced boars. Boars (n=32; $281{\pm}18$ days of age) were moved individually once weekly to a semen collection room equipped with an artificial sow. Before entering the semen collection room, boar received i.m. treatments of $PGF_2{\alpha}$ at doses of 5 (n=8), 10 (n=8), or 20 (n=8), and control boar (n=8) were not treated. Reaction time (elapsed time after entering collection pen until the start of mounting) for boars receiving 5mg ($3.3{\pm}0.9\;min$), 10mg ($3.3{\pm}0.8\;min$) $PGF_2{\alpha}$ was shorter (p<0.05) than for controls ($6.7{\pm}0.9min$). Duration of ejaculation (min) per session was longer (p<0.05) for $PGF_2{\alpha}$ (10 mg, 20 mg)-treated boars ($7.3{\pm}0.7\;min$, $6.9{\pm}0.7\;min$), compared to control ($3.4{\pm}0.8\;min$). The number of training session per boars was less (p=0.056) for $PGF_2{\alpha}$ 10mg-treated boars ($1.0{\pm}0.4$), compared to control ($2.0{\pm}0.4$). Semen characteristic such as volume, concentration, the number of total ejaculated sperm, were similar for $PGF_2{\alpha}$-treated and controls. There was no apparent difference on sperm movement characteristics (Mot: motility, VCL : curve linear velocity, VSL : straight line velocity, VAP : average path velocity, LIN : linearity) after semen preservation by collected with or without $PGF_2{\alpha}$ treatment. In summary, administration of $PGF_2{\alpha}$ in boars increased the sexual activity and facilitated the training boars to mount an artificial sow for semen collection, but did not affect semen characteristic.

Bacillus sp. DSNC 101이 생산하는 $\alpha$-L-Arabinofuranosidase의 정제 및 특성 (Purification and Characterization of an $\alpha$ -L-Arabinofuranosidase from Bacillus sp. DSNC 101)

  • Bacillus sp. DSNC 101의 배양 상징액으로부터 $\alpha$-L-arabinofuranosidase를 정제하여 그 특성을 조사하였다. 정제된 효소의 분자량을 SDS-PAGE를 이용하여 측정한 결과 56kDa로 나타났으며 효소 활성도에 대한 최적 온도와 최적 pH는 각각 55$^{\circ}C$와 7.0이었다. p-nitrophenyl-alpha-L-arabinofuranoside에 대한 Km 값과 Vmax는 1.0mM과 113.6U/mg이었으며 Hg$^{2+}$과 Cu$^{2+}$의 첨가는 효소 반응을 완전히 저해하였다. Ba-cillus sp. DSNC 101은 탄소원으로 볏짚을 사용할 때 $\alpha$-L-arabinofuranosidase를 생산하였으나 xylan이나 glucose, cellulose를 탄소원으로 배양할 때는 생산되지 않았다.

  • 본연구(本硏究)에서는 한우유(韓牛乳)와 Holstein cow milk의 단백질(蛋白質)을 Zone electrophoresis에 의하여 분석(分析)하여 그 결과(結果)를 다음과 같이 얻었다. (1) 한우유(韓牛乳)와 Holstein cow milk의 이화학적(理化學的) 성질(性質)이 Table 1, Table 2에 나타나 있다. (2) 한우유(韓牛乳)의 유청단백질(乳淸蛋白質)의 단백질(蛋白質) 분포(分布)는 Lactoalbumin 1.74mg %, Alpha-1 globulin 1.19mg%, Alpha-2 globulin 0.68mg %, Beta globulin 2.57mg %, Gamma globulin 0.93mg %이었다. (3) Holstein cow milk protein의 단백질(蛋白質) 분포(分布)는 Lactoalbumin 1.23mg %, Alpha-1 globulin 1.93mg %, Alpha-2 globulin 0.86mg %, Beta globulin 2.56mg %, Gamma globulin 1.74mg %를 얻었다. (4) 한우(韓牛)(female)와 Holstein cow(male)와의 교잡종인 F-1인 유청단백질(乳淸蛋白質) 분포(分布)는 Lactoalbumin 1.30mg %, Alpha-1 globulin 1.58mg %, Alpha-2 globulin 2.46mg %, Beta globulin 1.39mg %이었고 F-2의 그것은 Lactoalbumin 1.57mg %, Alpha-1 globulin 1.74mg %, Alpha-2 globulin 2.19mg %, Beta globulin 1.17mg %, F-4에서는 Lactoalbumin 2.46mg %, Alpha-1 globulin 1.24mg %, Alpha-2 globulin 1.57mg %, Beta globulin 1.74mg %를 각각(各各) 얻었다.

Starch-iodine Complex의 변색반응을 이용한 ${\alpha}-Cyclodextrin$의 정량 (Colorimetric Determination of ${\alpha}-Cyclodextrin$ by Using the Decoloration of Starch-iodine Complex)

  • ${\alpha}-Cyclodextrin(CD)$이 starch-iodine 복합체를 변색시킬 수 있다는 성질을 이용하여 ${\alpha}CD$의 새로운 비색정량법을 확립하였다. 이 새로운 ${\alpha}CD$의 정량법은 다음과 같다. ${\alpha}CD$의 표준용액을 2 mg/ml까지 농도별로 준비하였다. 그리고 동량으로 혼합된 starch-iodine 복합체 용액 1ml에 ${\alpha}CD$ 표준용액 $200\;{\mu}l$와 증류수 3 ml를 함께 가하고, 570 nm에서 흡광도를 측정하였다. 이 방법으로 ${\alpha}CD$의 농도범위는 $0{\sim}2\;mg/ml$까지 측정할 수 있었고, 570 nm에서의 흡광도도 가장 좋은 직선성을 나타내었다. 한편, ${\beta}CD$와 glucose는 ${\alpha}CD$ 정량을 위한 본 방법에 아무런 방해효과가 없었다.

Effects of $\alpha$-lipoic acid on cell proliferation and apoptosis in MDA-MB-231 human breast cells

  • The role that antioxidants play in the process of carcinogenesis has recently gained considerable attention. $\alpha$-Lipoic acid, a naturally occurring disulfide molecule, is a powerful antioxidant that reportedly exerts beneficial effects in patients with advanced cancer by reducing the level of reactive oxygen species and increasing glutathione peroxidase activity. In this study, we examined changes in the protein and mRNA expression associated with cell proliferation and apoptosis in MDA-MB-231 breast cancer cultured in the presence of various concentrations (0, 250, 500, and 1000 ${\mu}mol/L$) of $\alpha$-lipoic acid. The results revealed that $\alpha$-lipoic acid inhibited the growth of breast cancer cells in a dose-independent manner (P < 0.05). Additionally, $ErbB_2$ and $ErbB_3$ protein and mRNA expressions were significantly decreased in a dose-dependent manner in response to $\alpha$-lipoic acid (P < 0.05). Furthermore, the protein expression of phosphorylated Akt (p-Akt) levels and total Akt, and the mRNA expression of Akt were decreased dose-dependently in cells that were treated with $\alpha$-lipoic acid (P < 0.05). Bcl-2 protein and mRNA expressions were also decreased in cells that were treated with $\alpha$-lipoic acid (P < 0.05). However, Bax protein and mRNA expressions were increased in cells treated with $\alpha$-lipoic acid (P < 0.05). Finally, caspase-3 activity was significantly increased in a dose-dependent manner in cells treated with $\alpha$-lipoic acid (P < 0.05). In conclusion, we demonstrated that $\alpha$-lipoic acid inhibits cell proliferation and induces apoptosis in MDA-MB-231 breast cancer cell lines.

Interval oscillation criteria of the El-Sayed type and the Kong type are obtained. These criteria are further extended to equations with deviating arguments. As special cases, our work generalizes, unifies, and improves many existing results in the literature.
