Characterization of Extracellular \alpha-galactosidase Produced by Streptomyces sp. YB-4. (균체외 \alpha-galactosidase를 생산하는 Streptomyces sp. YB-4의 분리 및 효소 특성)

  • A strain YB-4 producing the extracellular $\alpha$-galactosidase was isolated from soil, and has been identified as Streptomyces sp. on the basis of its cultural, morphological and physiological properties. The partially purified $\alpha$-galactosidase was most active on paranitrophenyl-$\alpha$-D-galactopyranoside at pH 6.0 and 6$0^{\circ}C$. The enzyme retained 90% of its maximum activity between pH 4.0 and pH 10.0 after pre-incubation for 1 h. The enzyme was able to hydrolyze oligomeric substrates such as melibiose, raffinose and stachyose to liberate galactose residue, indicating that the $\alpha$-galactosidase of Steptomyces sp. YB-4 hydrolyzed $\alpha$-1,6 linkage.

  • It is well known that the continued fraction expansion of ${\sqrt{d}}$ has the form $[{\alpha}_0,\;{\bar{{\alpha}_1,\;{\ldots},\;{\alpha}_{l-1},\;2_{{\alpha}_0}}]$ and ${\alpha}_1,\;{\ldots},\;{\alpha}_{l-1}$ is a palindromic sequence of positive integers. For a given positive integer l and a palindromic sequence of positive integers ${\alpha}_1,\;{\ldots},\;{\alpha}_{l-1},$ we define the set $S(l;{\alpha}_1,\;{\ldots},\;{\alpha}_{l-1})\;:=\;\{d{\in}{\mathbb{Z}}{\mid}d>0,\;{\sqrt{d}}=[{\alpha}_0,\;{\bar{{\alpha}_1,\;{\ldots},\;{\alpha}_{l-1},\;2_{{\alpha}_0}}],\;where\;{\alpha}_0={\lfloor}{\sqrt{d}}{\rfloor}\}.$ In this paper, we completely determine when $S(l;{\alpha}_1,\;{\ldots},\;{\alpha}_{l-1})$ is not empty in the case that l is 4, 5, 6, or 7. We also give similar results for $(1+{\sqrt{d}})/2.$ For the case that l is 4, 5, or 6, we explicitly describe the fundamental units of the real quadratic field ${\mathbb{Q}}({\sqrt{d}}).$ Finally, we apply our results to the Mordell conjecture for the fundamental units of ${\mathbb{Q}}({\sqrt{d}}).$

The Effects of Fe and V on the Characteristics of $\beta$to$\alpha$ Transformation for Zr-0.8Sn Alloys (Fe와 V이 Zr-0.8Sn 합금의 $\beta{\rightarrow}\alpha$ 상변태 특성에 미치는 영향)

  • The effects of Fe and V content on the characteristics of $\beta$ to $\alpha$ phase transformation for Zr-0.8Sn alloys were studied by optical and transmission electron microscopy. With increasing V content, $\beta{\rightarrow}\alpha+\beta$ transformation temperature decreased, thus allowing the width of $\alpha$-lath to be fine air-cooled Zr-0.8Sn-xV alloys. The width of $\alpha$-lath, however, was slightly increased with Fe content. While the $\beta$ to $\alpha$ transformed microstructures of water-quenched Zr-0.8Sn, Zr-0.8Sn-0.1Fe, Zr-0.8Sn-0.2Fe, Zr-0.8Sn-0.4Fe, Zr-0.8Sn-0.1V and Zr-0.8Sn-0.2V were mainly slipped martensite, that of water-quenched Zr-0.8Sn-0.4V was predominantly twinned martensite. The transition of slipped martensite to twinned martensite in Zr-0.8Sn-Xv was thought to be due to the decrease of $M_S$ temperature.

  • In order to study the physiological properties of the intestinal bacteria, we isolated the intestinal bacteria of Koreans and tested the enzymatic patterns. Isolated Bifidobacterium sp. Int-57 had the higher activity of $\alpha$-glucosidase, $\beta$-glucosidase, $\alpha$-galactosidase, $\beta$-galactosidase. $\beta$-xylosidase and $\alpha$-arabinofuranosidase than other intestinal microorganisms. The effect of the carbon sources on the production of each enzymes of Bijidobacterium sp. Int-57 was investigated. The most suitable carbon source for the production of $\beta$-glucosidase was maltose, for a-glucosidase cellobiose, for $\alpha$-galactosidase raffinose, for $\beta$-galactosidase lactose, and for $\beta$-xylosidase and $\alpha$-arabinofuranosidase xylose, respectively. In addition, we investigated the optimal conditions and pH stability of each crude enzymes. The optimal condition of a-glucosidase was pH 6.0 and $40^{\circ}C$. that of Jj-glucosidase pH 7.0 and 50oe, that of $\beta$-galactosidase pH 7.0 and $50^{\circ}C$, that of $\beta$-xylosidase pH 6.0 and $40^{\circ}C$ , and that of $\alpha$-arabinofuranosidase pH 5.0 and $50^{\circ}C$. respectively. a-Glucosidase was stable at pH 4.0-9.0. Jj-glucosidase at pH 4.0-7.0. $\beta$-galactosidase at pH 4.0-9.0, $\beta$-xylosidase at pH 4.0-6.0, and /3-arabinofuranosidase at pH 7.0-9.0, respectively.

  • Let $0 < \alpha < 2$ and let $T_\alpha (\theta) = \alpha tan(\theta/2)$. $T_\alpha$ has an attractive fixed point at $\theta = 0$. We denote by $C(\alpha)$ the set of points in $I = [-\pi, \pi]$ which are not attracted to $\theta = 0$ by the succesive iterations of $T_\alpha$. That is, $C(\alpha)$ is the set of points in I where the dynamics of $T_\alpha$ is chaotic.

The Study on Antioxidation of Retinal (Retinol에 대한 항산화 연구)

  • In an attempt to compare the antioxidation effects of constrain the oxidation and improve the structural stability, retinol and various antioxidants were together encapsulated by liposome. Four water soluble and four oil soluble antioxidants were tested for performance. The influence of tertiary butylhydroquinone(TBHQ), ${\alpha}$-glycosyl rutin(${\alpha}$-G rutin), licorece, pycnogenol as water soluble antioxidants and butylated hydroxytoluene(BHT), ${\alpha}$-lipoic acid, ferulic acid, natural concentrated tooopherol(no-tocopherol) as oil soluble antioxidants on the constraint of oxidation of retinol were investigated. Additional study was conducted to compare the synergic effect of antioxidation for retinol with licorice, pycnogenol, ${\alpha}$-lipoic acid and BHT. All the antioxidant used at the study constrained oxidation of retinol. The effect of antioxidation for retinol increased in order of licorice, pycnogenol, TBHQ, ${\alpha}$-G rutin as water soluble antioxidants and ${\alpha}$-lipoic acid, BHT, no-tocopherol, ferulic acid as oil soluble antioxidants. In conclusion, ${\alpha}$-lipoic acid is more effective retinol antioxidants than BHT. And the combination of ${\alpha}$-lipoic acid and BHT gave best synergic among six combinations.

Mechanical Properties of Partially Stabilized $\alpha$-Sialon Synthesized from Kimcheon Quartzite (김천규석으로부터 제조한 부분안정화 $\alpha$-Sialon의 기계적 물성)

  • In order to synthesize the partially stabilized $\alpha$-Sialon, A1N and Y2O3 were added to synthesized $\alpha$-Si3N4. The phase composition, mechanical properties, micro structure, etc, of the synthesized $\alpha$-Sialon were investigated. Partially stabilized $\alpha$-Sialon ceramics could be synthesized from the composition which was a little deviated from x=0.4, x=0.6 composition along the Si3N4.0.1Y2O3:0.9AlN tie line at 1750-180$0^{\circ}C$ for 2 hrs in N2 atmosphere. It is assumed that A1N is more closely related than Y2O3 to the formation of $\alpha$-Sialon, and that A1N is more easily dissolved into $\alpha$-structure than into $\beta$-structure. In Ya2O3-rich phase mechanical properties were observed to be poor because of formation of mellilite, grain growth, and thermal decomposition of $\alpha$-Sialon. The maximum values of M.O.R, KIC and hardness are 723 MPa, 4.5MN/㎥/2 and 19.3 GPa, respectively, and they were observed for the $\alpha$-Sialon ceramics sintered at 178$0^{\circ}C$.

Synthesis of Glycosides by Transglycosylation of α-Amylase from Starch (전분으로부터 α-amylase에 의한 배당체의 합성)

  • Glycosides were synthesized using transglycosylation reaction of amylase in water system. The glycosides synthesized in water phase by a-amylase with starch as a glycosyl donor and benzylalcohol as an acceptor were identified as benzylalcohol-${\alpha}$-glucoside (BG) and benzylalcohol-${\alpha}$-maltoside (BM) of which one molecule of benzylalcohol was bound to 1-OH of glucose. The final products were BG in reaction system of pH 5.0, and BM in that of pH 8.0. The transglycosylation reaction by ${\alpha}$-amylase were carried out in water system containing 50 mg starch, 50 mg benzylalcohol, and 10 units enzyme at $30-35^{\circ}C$ for 3 days. The synthesized BG was hydrolyzed to glucose and benzylalcohol by ${\alpha}$-glucosidase, while ${\alpha}$-amylase hydrolyzed BM to glucose and benzylalcohol-${\alpha}$-glucoside in pH 5.0. Maltotriose resemble structurally to BM was rapidly hydrolyzed to glucose and maltose by ${\alpha}$-amylase at pH 5.0, being slightly hydrolyzed at pH 8.0, but not transglycosylated in present of benzylalcohol.

Kinetics and Mechanism of the Hydrolysis of ${\alpha}$-Cyano-${\beta}$-piperonylacrylic Acid (${\alpha}$-Cyano-${\beta}$-Piperonylacrylic Acid의 가수분해 메카니즘과 그의 반응속도론적 연구)

  • The rate constants of the hydrolysis of ${\alpha}$-Cyano-${\beta}$-piperonylacrylic acid were determined by Ultraviolet spectrophotometry at various pH and a rate equation which can be applied over wide pH range was obtained. The reaction mechanism of hydrolysis of ${\alpha}$-Cyano-${\beta}$-piperonylic acid and especially the catalytic contribution of hydroxide ion which not studied carefully before in acidic media, can be fully explained by the rate equation obtained. The rate equation reveals that; below pH 4.0, the reaction is initiated by the addition of water molecule to ${\alpha}$-Cyano-${\beta}$-piperonyl acrylic acid. At pH $5.0{\sim}7.5$, ${\alpha}$-Cyano-${\beta}$-piperonylacrylic acid compete with ${\alpha}$-Cyano-${\beta}$-piperonyl acrylate ion in adding of water. At pH 8.0, water is the only nucleophile for ${\alpha}$-Cyano-${\beta}$-piperonylacrylate ion, however, above pH 12.0, hydroxide ion is an addendum and the accepter is ${\alpha}$-Cyano-${\beta}$-piperonylacrylate ion.
