α-Glucosidase Inhibitory Activity of the Ethanol Extract of Peanut (Arachis hypogaea L.) Skin (땅콩 속껍질 에탄올 추출물의 알파-글루코시데이즈 억제활성)

  • Background: Owing to its high efficiency in lipid and protein production, peanut (Arachis hypogaea L.) is considered one of most important crops world-wide. The kernels of peanuts are undoubtedly the most important product this plant, whereas the skin is almost completely neglected in nutraceutical terms. However, peanut skin contains potentially health-promoting phenolics and dietary fiber, and there is considerable potential for commercial exploitation. In this study, we evaluated the α-glucosidase inhibitory activity of an extract of peanut skin (PS). Methods and Results: The α-glucosidase inhibitory effects of 80% ethanol extracts of peanut (A. hypogaea L. 'Sinpalkwang') skin were evaluated and found to have a half-maximal inhibitory concentration (IC50) value of 1.2 ㎍/㎖. Progress curves for enzyme reactions were recorded spectrophotometrically, and the inhibition kinetics revealed time-dependent inhibition with enzyme isomerization. Furthermore, using ultra-high performance liquid chromatography combined with quadrupole-orbitrap mass spectrometry, we identified 26 compounds in the peanut skin extract, namely, catechin, epicatechin, and 24 proanthocyanidins. Conclusions: The results suggest that peanut skin can be utilized as an effective source of α-glucosidase inhibition in functional foods and nutraceuticals.

  • Methylelaiophylin isolated from Streptomyces melanosporofaciens was selected as an ${\alpha}$-glucosidase inhibitor with an $IC_{50}$ value of 10 ${\mu}M$. It showed mixed-type inhibition of ${\alpha}$-glucosidase with a $K_i$ value of 5.94 ${\mu}M$. In addition, methylelaiophylin inhibited the intracellular trafficking of hemagglutinin-neuramidase (HN), a glycoprotein of Newcastle disease virus (NDV), in baby hamster kidney (BHK) cells. Methylelaiophylin inhibited the cell surface expression of NDV-HN glycoprotein without significantly affecting HN glycoprotein synthesis in NDV-infected BHK cells.

Antioxidant Activity and Alpha-Glucosidase Inhibitory Activity of Stings of Gleditsia sinensis Extracts (조각자 추출물의 항산화 활성 및 alpha-glucosidase 저해 활성)

  • This study was performed to investigate the physiological activities of stings of Gleditsia sinensis extracts. Antioxidant activity was evaluated by measuring total phenolic contents (TPC), comet assay, and 2.2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity (RSA). Anti-diabetic activity was measured by inhibition activities on $\alpha$-glucosidase. Stings of Gleditsia sinensis extracts were prepared by extracting them with methanol and ethanol. The methanolic extracts showed the highest phenol content (1.12 g/100 g gallic acid equivalents). The $\alpha$-glucosidase inhibitory activity of methanol extracts were 17.9% higher, and that of ethanol extracts were 10.3% higher at a concentration of 1 mg/ml. These results indicate that stings of Gleditsia sinensis might be potential candidates as antioxidant and anti-diabetic agents.

Gynura procumbens Extract Alleviates Postprandial Hyperglycemia in Diabetic Mice

  • This study was designed to investigate the inhibitory effect of Gynura procumbens extract against carbohydrate digesting enzymes and its ability to ameliorate postprandial hyperglycemia in streptozotocin (STZ)-induced diabetic mice. G. procumbens extract showed prominent ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory effects. The half-maximal inhibitory concentration ($IC_{50}$) of G. procumbens extract against ${\alpha}$-glucosidase and ${\alpha}$-amylase was $0.092{\pm}0.018$ and $0.084{\pm}0.027mg/mL$, respectively, suggesting that the ${\alpha}$-amylase inhibition activity of the G. procumbens extract was more effective than that of the positive control, acarbose ($IC_{50}=0.164mg/mL$). The increase in postprandial blood glucose levels was more significantly alleviated in the G. procumbens extract group than in the control group of STZ-induced diabetic mice. Moreover, the area under the curve significantly decreased with G. procumbens extract administration in STZ-induced diabetic mice. These results suggest that G. procumbens extract may help alleviate postprandial hyperglycemia by inhibiting carbohydrate digesting enzymes.

Inhibitory Effect of Endarachne binghamiae Extract on Melanin Synthesis (미역쇠(Endarachne binghamiae)추출물의 멜라닌 생성 억제 효과)

  • In this study, we investigated the effect of 88 marine algae extracts on melanin synthesis to develop new whitening agents. Among varieties of marine algae tested, the ethyl acetate extracts from Endarachne binghamiae (EB), Scytosiphon lomentaria, Sargassum yezoense, Ecklonia cava and Sargassum fusiforme inhibited melanin synthesis in melan-a cells. EB treatment showed the strongest inhibitory activity in melanin synthesis, compared with that of other extracts. EB-mediated inhibition of melanin synthesis appeared to be associated with inhibition of ${\alpha}$-glucosidase-dependent glycosylation of tyrosinase in melan-a cells. In addition, EB treatment did not affect mushroom tyrosinase or cell-extracted tyrosinase activity in vitro. Taken together, our findings suggest that anti-browning effect of EB on skin is mediated through regulation of ${\alpha}$-glucosidase activity and subsequent inhibition of tyrosinase activity and melanin synthesis, and further development of EB as a potential agent for skin whitening.

강원도산 상엽과 누에의 혈당강하 활성물질의 특성

  • It was proved that both ethanol extracts from Mori Folium from Kangwon do and silk worm had higher inhibition acitivity on $\alpha$-glucohydrolase than the water extracts. In adding above 8.5 (mg/L) of silkworm extracts, the inhibition rate on $\alpha$-1,4 glucosidase was saturated while the inhibition rate was continuously increased in adding the extracts from Moli Folium. It was also found that the diethyl ether fraction showed much better inhibition activity than water fraction from ethanol extracts, yielding ca. 85% of inhibition rate for the extract of Moli Folium, compared to 91% for a commercially available hypoglycemic drug, Chloropropamide. In separating the diethyl ether fractions by Consecutive Sephadex gel filtration and Thin layer chromatography, three and four active spots were obtained from Moli Folium and silkworm, respectively. It is interesting that the similar Rf spots from both species among several spots in TLC have the highest inhibition acitivity on a target enzyme, which can imply that the active substances from both species are same or similar molecular weight and structure. Glucose-lowering activities of both speciese were also examined in vivo, showing that the fraction from Moli Folium had better activity than that from silkworm, and its activity was similar to that of a commercial drug.

Biological activity of crude polyphenol fractions of Cedrela sinensis isolated using different extraction methods (참죽의 추출방법에 따른 폴리페놀 분획의 생리기능성)

  • The biological activity of crude polyphenol fractions (WphF, EphE, VphF, and SphF) extracted from Cedrela sinensis using hot water, ethanol, and enzymes such as Viscozyme and Shearzyme was examined in this study. The yield of VphF was the highest (43.44%) among all fractions. The total polyphenol and flavonoid content of the fractions were highest after ethanol extraction (447.98 and 337.49 mg/g, respectively). Fractions obtained after hot water and ethanol treatment showed high antioxidant activity. All fractions, except for WphF, showed a significantly higher ${\alpha}$-glucosidase inhibitory activity than the acarbose. EphF and WphF showed the high acetylcholinesterase inhibition activity. All fractions showed more than 50% tyrosinase inhibition activity at 2 mg/mL concentration. According to these results, the crude polyphenol fractions from C. sinensis showed high antioxidative, ${\alpha}$-glucosidase inhibitory, and tyrosinase inhibitory activities. This study suggests that crude polyphenol fractions from C. sinensis, especially the WphF and EphF fractions, are good sources of functional food.

Study on antioxidative, antidiabetic and antiobesity activity of solvent fractions of smilax china L. leaf extract (청미래덩굴잎 추출물 용매분획의 항산화, 항당뇨 및 항비만 활성연구)

  • Smilax china L., a native plant found in Asian countries, has several medicinal properties including antioxidant, anti-inflammatory, and anti-cancer effects. Although the root of the plant is commonly used as traditional herbal medicine in Korea and China, the medicinal properties of the leaves have not gained the same attention. In this study, we analyzed the antioxidant activity, ${\alpha}$-glucosidase inhibitory effect and lipid accumulation inhibition effect of Smilax china L. leaf water extract (SCLE) and its solvent fractions. SCLE was fractionated by using a series of organic solvents, including ethylacetate (EA) and n-butanol (BuOH). The EA fraction had the highest total polyphenol content ($440.20{\pm}12.67$ mg GAE/g) and total flavonoid content ($215.14{\pm}24.83$ mg QE/g). The radical scavenging activity $IC_{50}$ values of the EA fraction for 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2-azino-bis-(3-ethylbenzthiazoline)-6-sulfonic acid (ABTS) were 0.022 mg/mL and 0.13 mg/mL, respectively. Further, SOD-like activity and reducing power values of the EA fraction were higher than those of the other fractions. However, both the ${\alpha}$-glucosidase and lipid accumulation inhibition assays showed that the BuOH fraction ($83.35{\pm}4.18%$ at 1 mg/mL) and water extract ($11.27{\pm}2.67%$) were more effective than the EA fraction ($64.13{\pm}6.35%$, and $45.66{\pm}7.20%$). These results provide new insights into the potential anti-diabetic and anti-obesity effects of Smilax china L. leaf.

Biological Activities of Phellinus linteus Mycelium Culture with Cassiae Semen Extract on β-Glucuronidase Inhibitory Activity (β-Glucuronidase 저해 활성이 우수한 결명자를 첨가한 상황 균사체 배양액의 생리활성)

  • We examined the effects of biological activity Phellinus linteus mycelium culture with cassiae semen extract. Firstly, the optimal temperature, initial pH and culture period for mycelial growth in a liquid culture of P. linteus were determined, and they were $30^{\circ}C$, pH 5.0 and 8 days respectively. The five herbal materials were examined against several health functional efficacies, and, as a result, Cassiae semen was chosen, with its superior inhibitory effects in ${\beta}$-glucuronidase inhibitory activity, electron donating activity, ACE inhibitory, and ${\alpha}$-glucosidase inhibitory activities(95.3%, 80.9%, 96.1 and 24.2%, respectively). P. linteus fruit body was investigated on ${\beta}$-glucuronidase inhibitory activity, electron donating activity, ACE inhibitory, and ${\alpha}$-glucosidase inhibitory activities, and they were 54.7%, 81.9%, 30.0% and 20.1%, respectively. Accordingly, C. semen was used in the following experiment, to give an additive functional effect on the P. linteus. As the amount of C. semen in the cultural media increased, mycelial weight and ${\beta}$-glucan contents also increased, but final pH was not influenced. In addition, the ${\beta}$-glucuronidase inhibitory activity, electron donating activity, and ${\alpha}$-glucosidase inhibitory activity increased. P. linteus mycelium culture showed higher activities in the other three tests above, except for electron donating activity, when C. semen was added to the medium before cultivation.

Effects of novel chalcone derivatives on α-glucosidase, dipeptidyl peptidase-4, and adipocyte differentiation in vitro

  • Chana series are new chalcone derivatives. To evaluate the possibility of Chana series as therapeutic agents of type 2 diabetes, the inhibitory effects of Chana series on the activities of ${\alpha}$-glucosidase and DPP-4 were investigated using in vitro enzyme assays, and their effects on adipocyte differentiation were investigated in C3H10T1/2 cells. Chana 1 and Chana 7 among the Chana series showed significant inhibition of ${\alpha}$-glucosidase activity. In DPP-4 enzyme assay, Chana 1 exhibited the highest inhibitory activity while Chana 7 did not. In MTT assay, Chana 1 did not show significant cytotoxicity up to a concentration of $250{\mu}M$, whereas cytotoxicity was observed with Chana 7 at a concentration of $300{\mu}M$. In addition, Chana 1 induced adipocyte differentiation. Therefore, Chana 1 showed inhibitory effects on ${\alpha}$-glucosidase and DPP-4 as well as a stimulatory effect on adipocyte differentiation, suggesting that Chana 1 may be a potential beneficial agent for the treatment of type 2 diabetes.
