The Oxidative Stress in Cigarette Smokers and Antioxidant Vitamins

  • The purpose of this study was to find the extent of lipid peroxidation of erythrocytes in cigarette smokers, and to determine the relative effectiveness of $\beta$-carotene, canthaxanthin , and $\alpha$-tocopherol as antioxidants. Thirty smokers and 30 nonsmokers participated in this study . No significant differences according to age, sex, and height were shown. Cigarette smokers in this study had higher hemoglobin concentrations and more oxidation of hemoglobin than non-smokers. In addition, the erythrocytes of cigarette smokers had significantly higher MDA concentrations than crythrocytes of nonsmokers, which suggests that smokers may have tocopherol were studied in vitro by measuring the concentration of malondialdehyde(MDA) and precent hemolysis of erythrocytes. The addition of any antioxidant to erythrocytes significantly decreased MDA concentrations(p<0.05) while antioxidants showed nonsignificant inhibition of hemolysis. Among the antioxidant used in this study, canthaxanthin showed the greatest inhibition of both lipid peroxidationand hemolysis. Meanwhile, $\alpha$-tocopherol showed potent inhibition of lipid peroxidation, but not of hemolysis.

Major Chemical Constituents of Supercritical Carbon Dioxide Extract of Pandanus odorus Leaves

  • Supercritical carbon dioxide was used as a solvent in the extraction of freeze-dried Pandanus odorus leaves. Analysis of the extract with GC-MS showed that Pandanus odorus leaves contain nutritional constituents such as ${\alpha}-tocopherol$ (Vitamin E) and squalene. The contents of ${\alpha}-tocopherol$ and squalene extracted from freeze-dried ground Pandanus odorus leaves at pressures ranging from 80 to $200\;kg\;cm^{-2}$ and temperatures between 40 to $80^{\circ}C$ were $134{\sim}300\;ppm$ and $750{\sim}1,200\;ppm$ respectively. The highest yield was obtained at $200\;kg\;cm^{-2}$ and $40^{\circ}C$. Other major components identified were hexadecanoic acid, 9,12,15-octadecatrien-1-ol, campesterol, stigmasterol and ${\beta}-sitosterol$.

Effects of Ethanol and Tocopherol on Hepatic Peroxidation and Mitochondrial Respiration in the Rat (에탄올과 토코페롤이 간조직의 지질산화와 미토콘트리아 산화능에 미치는 영향)

  • To investigate effects of chronic alcohol consumption and tocopherol on lipid peroxidation and mitochondrial respiration 48 male rats of Sprague-Dawley strain were divided into 4 groups. Each group received for 3 weeks one of 4 experimental diets: tocopherol deficient control (TDC), tocopherol deficient-ethanol (TDE), tocopherol-supplemented control (TSC) and tocopherol-supplemented-ethanol (TSE). Composition of the diets was based on the Lieber and Decarli liquid diet and $\alpha$-tocopherol was supplemented at the level of 30mg/liter of diet, and ethanol supplied 36kcal%. TDC and TSC were pair-fed to TDE and TSE, respectively. Increase of body weight of tocopherol deficient-ethanol group was the lowest and the effect was diminished with tocopherol supplementation. Respiration of liver mitochondria was depressed in ethanol-administered groups and the effect became larger with tocopherol deficiency. Hepatic lipid peroxide level was not influenced by ethanol, but hepatic tocopherol content decreased with ethanol treatment. The result indicated that, although lipid perroxide level was unchanged with chronic ethanol consumption, oxidative stress exists in tissues of rate administered ethanol and may be relieved by tocopherol supplementation.

  • ${\gamma}-Tocopherol$ methyltransferase (TMT) is an enzyme catalyzing ${\gamma}-tocopherol$ into ${\alpha}-tocopherol$ at the final step of ${\alpha}-tocopherol$ synthesis pathway. Putative TMT cDNA clone specific to Perilla frutescens immature seeds was isolated from cDNA library. The cDNA clone consisted of 1369 bp open reading frame encoding 369 amino acids with a relative Mw of 42 kDa. Results revealed the CDNA has 60% homology to Arabidopsis thaliana TMT, and possesses methyltransferase and S-adenosyl methionine-binding domains, suggesting that cDNA encodes a ${\gamma}-tocopherol$ methyltransferase To characterize the properties of the TMT gene, the cDNA sequences coding for mature TMT were expressed in E. coli and assayed to determine the enzyme activity in vitro.

Expression of γ-Tocopherol Methyltransferase Transgene Improves Tocopherol Composition in Lettuce (Latuca sativa L.)

  • A cDNA encoding ${\gamma}-tocopherol$ methyltransferase (${\gamma}-TMT$) from Arabidopsis thaliana was overexpressed in lettuce (Latuca sativa L.) to improve the tocopherol composition. Seven lines of lettuce ($T_0$) containing the ${\gamma}-TMT$ transgene were produced by Agrobacterium-mediated transformation. The inheritance and expression of the transgene were confirmed by DNA and RNA gel blot analyses as well as quantification of tocopherols and ${\gamma}-TMT$ activities. The ratio of ${\alpha}-/{\gamma}-tocopherol$ content (TR) varied from 0.6 to 1.2 in non-transformed plants, while the $T_0$ plants had ratios of 0.8 to 320. The ratio ranged from 0.4 to 544 in 41 $T_1$ progenies of the $T_0$ transgenic line gTM3, and the phenotypic segregation indicated monogenic inheritance of the transgene (i.e., 3:1 = dominant:wild-type classes). There was a tight relationship between the TR phenotype and ${\gamma}-TMT$ activity, and enzyme activities were affected by the copy number and transcript levels of the transgene. The TR phenotype was stably expressed in $T_2$ progenies of $T_1$ plants. The results from this study indicated that a stable inheritance and expression of Arabidopsis ${\gamma}-TMT$ transgene in lettuce results in a higher enzyme activity and the conversion of the ${\gamma}-tocopherol$ pool to ${\alpha}-tocopherol$ in transgenic lettuce.

Tocopherols and fatty acids in plant seeds from Korea (한국산 두류 종실중 토코페롤 함량 및 지방산 조성)

  • The concentrations of tocopherols and lipids, and compositions of fatty acids in the seeds of thirteen pulse plants, which are used for the raw materials of our traditional food as well as for the typical nutrients in Korea, were analyzed by HPLC and GC. The beans showed great differences of the tocopherol concentrations from 4.74 to 44.05 mg/100g, which were in the order of ${\gamma}->{\delta}->{\alpha}-tocopherol$ with the exception of the order of ${\delta}->{\gamma}->{\alpha}-tocopherol$ in jeock sodu and gal sodu of azuki bean. The contents of lipids were also remarkably different as 0.53 to 20.96%. Especially, the beans having from 15.78 to 20.96% of lipid contents showed high composition ratios of unsaturated fatty acid between 85.6 to 88.2%, of which the contents of linoleic acid were the highest as 56.0 to 58.6% of the total fatty acids, and also showed comparatively high concentrations of tocopherols as 23.2 to 44.05 mg/100g. There were significantly positive correlation between ${\gamma}-tocopherol$ and unsaturated fatty acids.

Retinol, α-tocopherol, and selected minerals in breast milk of lactating women with full-term infants in South Korea

  • BACKGROUND/OBJECTIVES: This study was performed to measure fat-soluble vitamins and minerals in breast milk of Korean lactating mothers who exclusively breastfed their babies. SUBJECTS/METHODS: Breast milk samples were collected from 334 mothers. Concentrations of retinol and ${\alpha}$-tocopherol were analyzed by high performance liquid chromatography ultraviolet spectrometry while concentrations of minerals were measured by inductively coupled plasma optical emission spectrometry. RESULTS: Retinol and ${\alpha}$-tocopherol contents of breast milk were $39.58{\pm}19.64{\mu}g/dL$ and $0.23{\pm}0.13mg/dL$, respectively. Average sodium, potassium, calcium, phosphorus, and magnesium levels in breast milk were $11.11{\pm}5.16$, $38.56{\pm}9.01$, $27.87{\pm}6.10$, $13.56{\pm}3.30$, and $3.05{\pm}0.65mg/dL$, respectively. Contents of trace elements such as iron, zinc, copper, and manganese were $40.26{\pm}46.21$, $98.40{\pm}62.47$, $24.09{\pm}9.03$, and $0.90{\pm}1.63{\mu}g/dL$, respectively. Fat-soluble vitamin concentration was positively correlated with total fat in milk samples, but no significant differences were observed in levels of retinol, ${\alpha}$-tocopherol, or minerals based on whether or not lactating women were taking dietary supplements. CONCLUSIONS: Micronutrient contents of breast milk samples from Korean lactating women were comparable to those of other nations. Retinol and ${\alpha}$-tocopherol levels were correlated and also with total fat in breast milk.

Enhanced Biosynthesis of ${\alpha}$-tocopherol in Transgenic Soybean by Introducing ${\gamma}$-TMT gene

  • This study was conducted to improve tocopherol (vitamin E) composition in soybean (Glycine max) by introducing a gamma-tocopherol methyl transferase (${\gamma}$-TMT) gene via Agrobacterium tumefaciens-mediated transformation. Immature cotyledon explants were cocultivated with Agrobacterium tumefaciens. Putative transgenic embryos were selected from immature cotyledons on MS medium supplemented with 40 mg/L 2,4-D containing 100 mg/L kanamycin, 500 mg/L carbenicillin and 250 mg/L cefotaxime. Plantlets were developed from somatic embryos, and then transferred to soil. Nineteen regenerated plantlets obtained on the selection medium from 1,460 cotyledons. However, only 9 plantlets were confirmed as transformed plants. Integration of the transgene into the soybean genomic DNA was confirmed by PCR and Southern blot analysis. HPLC analysis showed that the content of ${\alpha}$-tocopherol in transgenic soybean seeds (AT-1) was approximately 4-fold higher than that of non-transgenic plants. Conclusively, we obtained the transgenic soybean having increased ${\alpha}$-tocopherol content by the overexpression of ${\gamma}$-TMT transgene.

Effects of Antioxidants on Oxidation of Tallow by Gamma Irradiation (감마선 조사시 우지 산패억제를 위한 항산화제의 첨가효과)

  • The effects of antioxidants, ascorbyl palmitate (AP), ${\alpha}-tocopherol\;({\alpha}-Toc)$, BHA and ascorbyl $palmitate+{\alpha}-tocopherol\;(AP+{\alpha}-Toc)$ have been investigated on lipid peroxide formations in tallow immediately and during storage at $50^{\circ}C$ after gamma irradiation with the dose of $1{\sim}10kGy$. Immediately gamma irradiation greatly increased the initiative oxidation of tallow as was expected. But antioxidants were found to be greatly effective in minimizing the radiation-induced peroxidation of tallow and their antioxidative activities were $AP>BHA>AP+{\alpha}-Toc>{\alpha}-Toc$. Especially, AP showed greater antioxidative activity of initiate-oxidation than that of any other antioxidant. Oxidation of tallow during storage at $50^{\circ}C$ after irradiation induced the accelate of autoxidation. But the additions of antioxidants inhibited formation of peroxide. Their antioxidative activities were $BHA>AP+{\alpha}-Toc>{\alpha}-Toc>AP$. Especially $AP+{\alpha}-Toc$ mixture was not significantly different from BHA (p>0.05).

  • The main function of plastic materials in food packaging is to preserve a food for safe transportation and storage. The interactions between food and plastic materials in food packaging have become increasingly important for food quality and safety because monomer, low molecular weight components, or additives of plastic packaging materials can migrate into a food. The use of antioxidants in plastic materials can help protect the degradation of film itself and retard the oxidation of a packaged food containing lipid, through the migration of antioxidant from the packaging to a product via an evaporation / sorption mechanism. Nowadays, antioxidant (BHT) impregnated plastic materials are used for commercial food packaging application with the intention of achieving an extended shelf life of food in USA. Alpha tocopherol, as one of the most important free radical scavengers, has been well known in biological systems. Moreover, the potential use of alpha tocopherol as an additive for polymers used in the packaging industry may offer the most positive perception from both consumers and manufacturers. Alpha tocopherol has been used as an antioxidant for polyolefin resins fabricated to both bottles and film and has applications in the food packaging industry as a replacement for BHT. Today, alpha tocopherol offers an attractive choice for use as an antioxidant in polymers. This paper provides an overview of antioxidant effectiveness and applications for its use by the food packaging industry based on the evaporation-sorption mechanism of a packaging model product, where quality is associated with lipid oxidation. Important analytical techniques for predicting antioxidant interaction between the package system and product are discussed.
