Recently new soybean saponins with D DMP (2,5-dihydroxy-6-methyl-2,3,- dihydro-4H-pyran-4-one) moiety have been isolated from legumes. The purpose of this study is to characterize ROS scavenging activities of DDMP saponins ($\alpha$g, $\beta$g saponin) isolated from Glycine max (L.) Merrill. The scavenging activity on OH was examined in terms of lipid peroxidation in the rat liver homogenates and the same activity on $O_2$ was also determined in the xanthine-xanthine oxidase system, respectively. Up to 0.25 mg DDMP saponins ($\alpha$g and $\beta$g saponins) did not cause any significant effects on the prevention of lipid peroxidation as compared with the control group. In terms of superoxide scavenging activities, 0.25 and 0.5 mg $\alpha$g saponin inhibits only 2.6% and 5.5% (p<0.05) of the control group, respectively. However, $\alpha$g saponin dose-dependently (p<0.01, r=0.955) inhibits the formation of superoxide radical unto 21.3% of the control group with a maximal dose of 0.5 mg (p<0.01), equivalent to 0.17 units of superoxide dismutase activity.
There is a potential role of collagenase-3 in alveolar bone loss and periodontal disease progression, we need to develope or find chemotherapeutic drugs or herbal agents which may regulate the expression of MMP-13. Ginseng saponin, one of the major components of Korea ginseng(panax ginseng) root, has many various biologic effects, such as cytotoxic effect, tumoricidal effects, cytokine regulations, and protein biosynthesis effect. The purpose of this study was to determine the effects of Korea red ginseng saponin on MMP-13 gene expression in osteoblasts. The experimental groups were cultured with ginseng saponin in concentration of 1.0, 10, 25, 50, 100, 250 and $500{\mu}g/ml$ for MTT assay. Primary rat calvarial cells were pre-treated for 1 hour with ginseng saponin(100 ${\mu}g/ml$) and then stimulated with $IL-1{\beta}(1.0ng/ml)$ and PTH(10 nM). MMP-13 gene expression was evaluated by RT-PCR. The results were as follows: Ginseng saponin was cytotoxic to osteoblast at concentration exceeding $250{\mu}g/ml$ for longer than 24 hours in tissue culture(p<0.01). In RT-PCR analysis, steady state MMP-13 mRNA levels were increased approximately 350% by $IL-1{\beta}$, and 400% by PTH when normalized to untreated control. $IL-1{\beta}-indued$ MMP-13 mRNA expression was reduced 50% by pretreatment with ginseng saponin. But ginseng saponin didn't inhibit MMP-13 expression from PTH stimulated cells. This results suggest that ginseng saponin Inhibit $IL-1{\beta}-indued$ MMP-13 mRNA expression.
항균 활성이 대체로 상승되었으나 Sarcina maginata의 경우는 약간 감소되었다. $\beta$-lactam 항생 물질인 penicillin G.Na와 ampicillin.Na도 인삼 saponin의 첨가로 항균 활성이 상승되었으나 Serratia 속 세균은 penicillin G.Na의 항균활성에 일정한 효과를 나타내지 않았다. 인삼 saponin 의 첨가에 의한 항생 물질들의 항균 활성의 변화는 Gram양성 세균 및 Gram음성 세균에 대해 일정하지 않은 상승 작용 혹은 길항작용과 같은 비특이성을 나타내었다. 세균의 약제 내성 현상도 인삼 saponin자 항생물질의 투여로 배제될 수 있을 것으로 사료되었다.
There is much evidence suggesting that compounds present in soybean can prevent cancer in many different organ systems. Especially, soybean is one of the most important source of dietary saponins, which have been considered as possible anticarcinogens to inhibit tumor development and major active components contributing to the cholesterol-towering effect. Also they were reported to inhibit of the infectivity of the AIDS virus (HIV) and the Epstein-Barr virus. The biological activity of saponins depend on their specific chemical structures. Various types of triterpenoid saponins are present in soy-bean seeds. Among them, group B soyasaponis were found as the primary soyasaponins present in soybean, and th e 2, 3-dihydro-2, 5-dihydroxy-6- methyl-4H-pyran-4-one(DDMP)-conjugated soyasaponin $\alpha\textrm{g}$, $\beta\textrm{g}$, and $\beta$ a were the genuine group B saponins, which have health benefits. On the other hand, group A saponins are responsible for the undesirable bitter and astringent taste in soybean. The variation of saponin composition in soybean seeds is explained by different combinations of 9 alleles of 4 gene loci that control the utilization of soyasapogenol glycosides as substrates. The mode of inheritance of saponin types is explained by a combination of co-dominant, dominant and recessive acting genes. The funtion of theses genes is variety-specific and organ specific. Therefore distribution of various saponins types was different according to seed tissues. Soyasaponin $\beta\textrm{g}$ was detected in both parts whereas $\alpha\textrm{g}$ and $\beta$ a was detected only in hypocotyls and cotyledons, respectively. Soyasaponins ${\gamma}$g and $\gamma\textrm{g}$ were minor saponin constituents in soybean. In case group A saponins were mostly detected in hypocotyls. Also, the total soyasaponin contents varied among different soy-bean varieties and concentrations in the cultivated soy-beans were 2-fold lower than in the wild soybeans. But the contents of soyasaponin were not so influenced by environmental effects. The composition and concentration of soyasaponins were different among the soy products (soybean flour, soycurd, tempeh, soymilk, etc.) depending on the processing conditions.
The effects of ginseng saponins on the sarcolemmal $Na^{+}$, $K^{+}$-ATPase were compared to gypsophila saponin, sodium dodecylsulfate (SDS), and Triton X-100 to elucidate whether the effects are due to the membrane distruption, using a highly enriched preparation of cardiac sarcolemma prepared from dog ventricular myocardium. About 26% and 29% of vesicles in the preparation, enriched in ouabain-sensitive $Na^{+}$, $K^{+}$-ATP ase, $\beta$-adrenergic and muscarinic receptors are rightside-out and inside-out orientation, respectively. Ginseng saponins (triol>total> diol) inhibited $Na^{+}$, $K^{+}$-ATP ase activity, $Na^{+}$, $K^{+}$-ATPase activity and [$^{3}$H]ouabain binding of sarcolemmal vesicles. However, gypsophila saponin, SDS (0.4$\mu$g/$\mu$g protein) and Triton X-100 (0.6 $\mu$g/$\mu$g protein) caused about 1.35 and 1.40-fold increase in $Na^{+}$, $K^{+}$-ATPase activity and [$^{3}$H] oubain binding, respectively. Especially, the activating effect of gypsophila saponin on membrane Na+, K+ ATPase was detected at gypsophila saponin to sarcolemmal protein ratios as high as 100. Low dose of ginseng saponin (3$\mu$g/$\mu$g protein) decreased the phosphorylation sites and the concentration of ouabain binding sites (Bmax) without affecting the turnover number and affinity for ouabain binding, while gypsophila saponin, SDS(0.4 ug/ug protein), ahd Triton X-100 (0.6$\mu$g/$\mu$g protein) increased the Bmax. The results suggest that ginseng saponins cause a decrease in the number of active sites by interacting directly with $Na^{+}$, $K^{+}$-ATPase before disruption of membrane barriers of sarcolemmal vesicles.
수천 년 동안 전통적 약제로 사용되어온 고려인삼은 중추신경계의 항상성을 유지하고 면역기능을 강화하는 효능을 나타내는 것으로 알려져 있다. 신경계질환의 진행이 대부분 염증 또는 면역반응을 수반하며, 이로 인하여 손상된 신경세포의 수복과정에 교세포 기원의 매개물질들이 기여하므로, 교세포에서의 cytokine 및 NO생성에 대한 연구는 신경기능과 신경면역기능의 조절 뿐 아니라 신경계 질환에 대한 연구의 초석이라고 할 수 있다. 따라서 본 연구에서는 고려인삼의 신경면역 및 염증반응 조절효과에 대하여 연구하고자 하였으며, 이를 위하여 흰쥐 대뇌피질의 교세포를 일차배양하며 고려인삼사포닌 분획을 처치하여 TNF-$\alpha$, IL-1$\beta$, 및 NO의 생성변동을 연구하였다. 백삼 사포닌은 50-500$\mu\textrm{g}$/ml 용량에서 TNF-$\alpha$와 IL-1$\beta$ 생성을 증가시켰으며, 미세교세포에서 iNOS 발현 및 NO생성을 유도하였고 성상세포의 stellation을 초래하였다. 특히 백삼사포닌 50-100 $\mu\textrm{g}$/ml는 세포독성을 거의 나타내지 않았으므로 이들 용량에 의한 교세포의 적절한 활성화가 중추신경계 면역기능 증가 및 염증반응 조절에 기여할 것으로 생각된다.
건강 기능성 농산물로 주목을 받고 있는 검정콩의 isoflavone과 soyasaponin ${\alpha}g$와 ${\beta}g$의 축적 추이를 검토하였다. 또한 이들 항산화 성분을 고함유한 고 기능성 검정콩 생산을 위한 재배기술 확립에 기여하고자 하였다. 얻어진 결과는 다음과 같다. 1. 검정콩의 등숙기간 중 항산화 성분인 isoflavone과 soyasaponin ${\alpha}g$와 ${\beta}g$의 함량은 대체로 수확기$(R_8)\;6{\sim}7$일전에 가장 높았다. 이는 고항산화 콩을 생산하려면 수량 중심의 수확시기보다 $3{\sim}7$일 정도 빨라야 함을 의미한다. 2. 검정콩 종실의 등숙기간 중 총 isoflavone 함량은 일품 검정콩은 개화 후 61일, 흑청콩은 개화 후 77일에 가장 높았다. 검정콩 종실의 soyasaponin ${\alpha}g$와 ${\beta}g$의 함량은 일품 검정콩은 개화 후 61일에, 흑청콩은 개화 후 71일에 가장 높았다. 3. 검정콩 잎의 등숙기간 중 총 isoflavone함량은 일품검정콩과 흑청콩 모두 개화 후 55일에 가장 높았다. 검정콩 잎의 soyasaponin ${\alpha}g$의 함량은 일품검정콩은 개화 후 $18{\sim}41$일, 흑청콩은 개화 후 $55{\sim}71$일에 높았다. 4. 검정콩 잎의 생리활성 성분 함량이 높은 것으로 보아 이를 신소재 기능성 농산물로 활용 및 개발할 수 있을 것으로 사료되었다. 검정콩 잎을 신소재 기능성 농산물로 활용하기 위한 수확적기는 종실비대가 완료되는 생리적성숙기($R_7$)이었다.
방울비짜루의 MeOH 추출물 및 수포화 BuOH추출물로부터 항균활성을 확인하고 그 원인물질을 구명하던 중 두 종의 steroid saponin을 분리하였다. 이들의 구조확인을 위해 $^1H,\;^{13}C$ NMR 및 2D NMR 등 분광학적 방법을 사용하였으며 최종 이들의 구조가 25S-spirostane계의 steroid saponin인$3-O-[{\beta}-D-glucopyranosyl-(1{\rightarrow}2)-{\beta}-D-glucopyranosyl]-(25S)-spirostan-3{\beta}-ol$ 및 $3-O-{{\beta}-D-glucopyranosyl-(1{\rightarrow}2)-[{\beta}-D-xylopyranosyl-(1{\rightarrow}4)]-{\beta}-D-glucopyranosyl}-(25S)-spirostan-3{\beta}-ol$인 것으로 동정되었다. 이 두 화합물은 방울비짜루로부터는 처음으로 분리되었다. 이들 saponin의 항균활성 및 항균 스펙트럼을 검사하기 위하여 20개 균주에 대한 MIC를 실시하였다. 이들은 그중 10개 균주에 대해 $100\;{\mu}g/ml$의 농도에서 세균의 성장을 저지하는 항균력이 있었으며 광범위한 항균 스펙트럼을 나타내었다.
인삼을 배지로 사용하여 목질진흙버섯(Phellinus linteus), 영지버섯(Ganoderma lucidum) 및 노루궁뎅이버섯(Hericium erinaceum) 균사체를 접종 배양하여 얻어진 균사체 인삼배양물과 원료 인삼에 대한 사포닌과 $\beta$-glucan의 변화를 분석하였다. 원료인삼의 구성사포닌 함량은 22.47mg/g이었으나 균사체로 배양된 인삼의 사포닌 함량은 목질 진흙버섯이 39.78 mg/g, 영지버섯이 55.88 mg/g 그리고 노루궁뎅이버섯이 32.69 mg/g으로 원료인삼에 비해 증가하였다. 균사체 인삼배양물의 사포닌은 PPT계는 감소하였고 PPD계는 증가하였다. 구성사포닌 중 ginsenoside Rd는 원료인삼에 비해 균사체 인삼배양물에서 각각 17, 30 및 12배 증가하였다. 3종의 균사체와 균사체 인삼배양물들의 총 glucan 함량은 각각 46.24, 34.74 및 34.02% 그리고 28.77, 17.78 및 21.77%이었다. $\beta$-glucan 함량은 균사체에서는 각각 29.14, 19.44 및 23.39%이었고 인삼배양물에서는 각각 8.85, 5.46 및 5.51%로 나타나 목질진흙버섯이 균사체와 인삼배양물에서 가장 높은 함량을 나타내었다.
오염된 인삼분말로부터 1종의 세균을 순수분리하여 API kit 및 전자현미경을 이용하여 형태적, 생리적 특성을 조사하였다. 분리균은 직경 약 1.0 ${\mu}m$이었고 길이는 $2.0{\sim}6.0{\mu}m$로 세포표면에 규칙적으로 분포되어 있는 편모로 이동하는 간균이었다. 분리균은 ${\beta}-galactosdase$, arginine dihydrolase, ornithin decarboxylase를 가지고 있으며 탄소원으로 citrate를 이용할 수 없었고, $H_2S$는 생성하지 못하였다. 그러나 glucose, manitol, sorbitol, rhamnose, arabinose 등의 당 및 amygdalin을 탄소원으로 이용할 수 있었다. 이상의 API kit를 이용한 생리적 특성분석 및 전자현미경을 이용한 미세 형태적 특성 관찰결과에 의해 본 균주는 장내세균과 (Enterobacteriaceae)에 속하는 Citrobacter sp.로 동정되었다. 한편 분리균주 Citrobacter sp.의 생육에 미치는 인삼사포닌의 영향을 조사한 결과 사포닌은 농도의존적으로 균의 생육을 억제하는 경향을 나타내었다. 즉, 사포닌을 0.05, 0.5, 2.0, 4.0% 첨가하고 $38^{\circ}C$에서 3 일 동안 배양한 후 사포닌 비첨가군과 비교한 결과 균의 상대적 생육저해율은 각각 28.6, 66.7, 92.4, 97.7%로 나타났다. 사포닌의 미생물 생육 억제작용은 비교적 고농도인 4.0% 사포닌 첨가시에도 초기의 $6.2{\times}10^6$에서 $1.0{\timex}10^8$ CFU/g으로 완만한 가경향을 나타내어 사멸작용보다는 정균작용일 것으로 추측된다.
