Changes of Gonadosomatic Index and Sex Steroid Hormone of Serum in Cultured Greenling (Hexagrammos otakii) (양식산 쥐노래미 (Hexagrammos otakii)의 생식소중량지수와 혈중 성 스테로이드호르몬의 변화)

  • Gonad and blood samples were taken from cultured greenling (Hexagrammos otakii), and changes of steroid hormone levels in serum were examined in relation to the gonadosomatic index (GSI). GSI of female and male were not changed significantly between March and August and then began to increase from September showing the highest value in January. Hepatosomatic index (HSI) of female was the highest in December,'then decreased gradually until May. $Estradiol-l7{\beta} (E_2)$ level in blood of female reached to the highest level in December, then decreased rapidly in January. The level was maintained low until August and started to increase In September. Testosterone (T) level in blood of female was similar to trend of $E_2$ in female. $17{\alpha}2{\beta}-dihydroxy-4-pregnen-3-one (17{\alpha}20{\brta}-OHP)$ level in blood of female reached to the maximum in January, and then decreased rapidly from Febuary and maintained low until July. T level in blood of male reached to the highest value in December, and it started to decrease from January and maintained low until July, after then increased again, while $17{\alpha}20{\beta}-OHP$ level of male was similar to trend of female's.

Quality Characteristics of Nochies Saccharified by Amylases from Various Sources (효소원을 달리한 노치의 품질 변화)

  • The present study was conducted to investigate quality characteristics of Nochi made with malted barley flour with (Cl) and without hull (C2), comparing with Nochi that was treated with different sources of commercial amylases. There was higher level of moisture content (18.4%) in Nochi treated with fungal ${\alpha}$-amylase (FU) comparing with the other Nochi samples. However, Nochi that was treated with bacterial ${\alpha}$-amylase and ${\beta}$-amylase (BA-${\beta}$) had the lowest level of moisture content (11.2%). Nochi samples which were treated with thermostable ${\alpha}$-amylase and fungal ${\alpha}$-amylase(TE-FU) were different from traditional Nochi samples in mechanical characteristics. According to the results of sensory evaluation, Cl was similar to C2 except in cohesiveness and malt flavor. TE-FU and Bh-${\beta}$ were not different from traditional Nochi in cohesiveness, sweetness and overall desirability.

Epidermal Growth Factor Decreases the Level of DNA Topoisomerase $II{\alpha}$ in Human Carcinoma A431 Cells

  • Human epidermoid carcinoma A431 cells have an extraordinarily large number of epidermal growth factor (EGF) receptors, and their growth is inhibited by EGF, which results in growth arrest at the Gl phase. In order to investigate the EGF-mediated inhibition mechanism, the expression level of DNA topoisomerase (topo) II was analyzed after EGF treatment. As a result, it was shown that EGF treatment lowered the amount of 170 kDa topo II (topo $II{\alpha}$) but not 180 kDa (topo $II{\beta}$). However, the A431 cell variant resistant to EGF was not sensitive to EGF treatment. These results suggest that EGF-induced growth arrest of A431 cells may be closely related to the depletion of topo $II{\alpha}$.

  • 동해의 SeaWiFS 클로로필-$\alpha$에서 나타나는 스펙클(speckle) 오차를 제거하고 Level-2에서 Level-3로의 자료합성을 실시하였다. 1998년부터 2007년까지 10년간 클로로필-$\alpha$의 시공간 변동성을 고려하여, 월 평균값으로 표준화된 공간적 표준 편차가 $7mg/m^3$ 이상 될 때를 경계 값으로 스펙클 제거기준을 설정하였다. 농도경사가 큰 전선지역과 연안으로부터 10-50 km 이내 지역은 제거 기준을 $12mg/m^3$ 이상으로 알고리즘을 구성하였다. 스펙클 제거 결과, 자료는 그대로 보존되고 스펙클이 존재하는 화소의 값만 줄어들었다. 특히 스펙클이 자주 발생하는 겨울철 클로로필 자료의 농도변화가 크게 나타났으며 스펙클이 많이 나타나는 시기에는 $5^{\circ}\times5^{\circ}$영역의 평균값이 10%이상 감소하는 결과를 보였다.

The Inhibitory Effect of Cornus walteri Extract Against ${\alpha}-amylase$ (말채나무 추출물의 ${\alpha}-amylase$ 저해 활성)

  • ${\alpha}-Amylase$ inhibitor is used to control blood glucose level by inhibiting starch digestion in the small intestine and delaying the absorption of glucose. In this study, we investigated the effect of the ethanol extracts from more than 1400 species of plants against ${\alpha}-amylase$ with the aim of developing a new ${\alpha}-amylase$ inhibitor. In the results, Cornus walteri extracts showed the highest inhibition activity. The inhibitory effect of Cornus walteri extract on the carbohydrate hydrolysis enzymes has different sensitivities against ${\alpha}-amylase$ from salivary and pancreatin and against ${\alpha}-glucosidase$ from yeast and porcine small intestine. In the study of inhibition kinetics of ${\alpha}-amylase$ and ${\alpha}-glucosidase$, Cornus walteri extract showed competitive inhibition against salivary and pancreatin while showing the combination of uncompetitive and noncompetitive inhibition against ${\alpha}-glucosidase$. The Cornus walteri extract was stable at acidic and thermal conditions. As for the blood glucose and body weight levels of Cornus walteri extract, we confirmed anti-hyperglycemic and anti-obesity effects. Also, in the investigation of the mRNA lever, Cornus walteri extract upregulated the level of GLUT4 mRNA in the quadriceps muscle.

Expression and Distribution of the Guanine Nucleotide-binding Protein Subunit Alpha-s in Mice Skin Tissues and Its Association with White and Black Coat Colors

  • Guanine nucleotide-binding protein subunit alpha-s ($Gn{\alpha}s$) is a small subunit of the G protein-couple signaling pathway, which is involved in the formation of coat color. The expression level and distribution of $Gn{\alpha}s$ were detected by quantitative real-time-polymerase chain reaction (qPCR), western blot, and immunohistochemistry to investigate the underlying mechanisms of coat color in white and black skin tissues of mice. qPCR and western blot results suggested that $Gn{\alpha}s$ was expressed at significantly higher levels in black mice compared with that of white mice, and transcripts and protein possessed the same expression in both colors. Immunohistochemistry demonstrated $Gn{\alpha}s$ staining in the root sheath and dermal papilla in hair follicle of mice skins. The results indicated that the $Gn{\alpha}s$ gene was expressed in both white and black skin tissues, and the expression level of $Gn{\alpha}s$ in the two types of color was different. Therefore, $Gn{\alpha}s$ may be involved in the coat color formation in mice.

Hypoglycemic Effect of Fermented Soymilk Extract in STZ-induced Diabetic Mice

  • This study investigated the hypoglycemic effect of fermented soymilk extract (FSE) in STZ-induced diabetic mice. FSE was prepared via fermentation of soymilk with Bacillus subtilis followed by methanol extraction. The hypoglycemic effect was determined by inhibitory activities against ${\alpha}$-glucosidase and ${\alpha}$-amylase as well as the alleviation of postprandial glucose level. The non-fermented soymilk extract (SE) was used as control in this experiment. FSE showed higher (p<0.05) inhibitory activities than SE against ${\alpha}$-glucosidase and ${\alpha}$-amylase. The $IC_{50}$ values of FSE for ${\alpha}$-glucosidase and ${\alpha}$-amylase were 0.77 ancd 0.94 mg/mL, respectively, which were comparable or even superior to those of acarbose (0.79 and 0.68 mg/mL, respectively). In addition, a further suppression on the postprandial blood glucose levels were observed in the FSE than SE group for both STZ-induced diabetic mice and normal mice. Furthermore, FSE significantly lowered the incremental area under the curve (AUC) in the diabetic mice and the AUC in normal mice corroborated the hypoglycemic effect of FSE (p<0.05). Results from this study suggest that FSE may help decrease the postprandial blood glucose level via inhibiting ${\alpha}$-glucosidase and ${\alpha}$-amylase and the usefulness of FSE was proven to be better than SE.

A Study on Teaching and Learning of the Limit Concept in High School (고등학교에서의 극한개념 교수.학습에 관한 연구)

  • The purpose of this study is to find out the problems which are caused when the limit concept of sequences is learned through an intuitive definition and to suggest a way of solving those problems. Students in Korea study the limit concept of sequences through an intuitive definition. They fail to apply the intuitive definition properly to the problems and they are apt to have misconception even though the Intuitive definition is applied properly. To solve these problems, this study examined the develop- mental process of the limit concept of sequences from the Intuitive definition to the formal definition, and looked into the way of students' internalization of the process through a field study. In this study, the levels of the limit concept of sequences possessed by the students at ZPD are as follows; level 0 : Students understand the limit concept of sequences through the intuitive definition. level 1 : Students understand the limit concept of sequences as 'The difference between $\alpha$$_{n}$ and $\alpha$ approaches 0' rather than 'The sequence approaches $\alpha$ infinitely.' level 2 : Students understand the limit concept of sequences through the formal definition. The levels of students' limit concept development were analysed by those criteria. Almost of the students who studied the limit concept of sequences through the intuitive defition stayed at level 0, whereas almost of the students who studied through the formal definition stayed at level 1. Through the study, I found that it was difficult for the students to develop the higher level of understanding for themselves but the teachers and peers could help the students to progress to the higher level. Students' learning ability was one of major factors that make the students progress to the higher level of understanding as the concept was developed hierarchically from Level 0 to Level 2. If you want to see your students get to the higher level of understanding in the limit concept, you need to facilitate them to fully develop understanding in lower levels through enough experiences so that they can be promoted to the highest level.

Studies on the Changes of Steroid Hormone Levels during the Reproductive Stage in Korean Native Goats II.   • The present study was conducted to find out the changes of progesterone and 20$\alpha$-dihydroprogesterone(20$\alpha$-OHP) levels during the gestation period in Korean native goats. 4 pregnant goats were offered for this experiment. Blood samples were taken from jugular vein on Day 5, 10, 20, 30, 40, 50, 60, 70, 80, 90, 100, 110, 120, 130, 140 and 145 of the pregnant goats. The serum levels of progesterone and 20$\alpha$-OHP were measured by radioimmunoassay. The progesterone level in goat serum during the gestaton was low as 2.94$\pm$0.21 at 5 days, and then increased to 4.51$\pm$0.25ng/ml at 10 days of gestation and increased greatly from Days 60 and reached a peak level of 7.21$\pm$0.58ng/ml at 100 days of gestation, and thereafter decreased to 6.01$\pm$0.57, 5.26$\pm$0.64ng/ml on Days 130, 140 of gestation, and decreased to 4.05$\pm$0.52ng/ml on Days 145 of pregnancy. The serum level of 20$\alpha$-OHP during the gestation was low as 0.3~0.4ng/ml in the early stage of pregnancy, and increased gradually and increased to 0.85$\pm$0.06ng/ml, 0.97$\pm$0.08ng/ml on Days 90 or 100, and then increased to 1.18$\pm$0.18, 1.25$\pm$0.21ng/ml on Days 140 or 145 of gestation. While the serum levels of progesterone during the luteal regression decreased, the 20$\alpha$-OHP increased continuously. From the above results, it was concluded that the enzyme 20$\alpha$-hydroxysteroid dehydrogenase (20$\alpha$-HSD) catalyzing theconversion of progesterone to a biologically inactive steroid, 20$\alpha$-OHP was active properly in the luteal cells what the levels of progesterone decreased and the levels of 20$\alpha$-OHP increased during the late pregnancy in Korean native goats.
