Detection of Norovirus in Contaminated Ham by Reverse Transcriptase-PCR and Nested PCR

  • In order to enhance the efficacy of norovirus detection by reverse transcriptase-polymerase chain reaction (RT-PCR) and nested PCR, this study developed a norovirus mRNA concentration method using poly oligo dT-conjugated magnetic beads. An efficient norovirus detection protocol was performed on commercial ham using 2 viral elution buffers (glycine buffer and Tris beef extract buffer) and 2 concentration solutions [polyethylene glycol (PEG) and zirconium hydroxide]. The different approaches were verified by RT-PCR and nested PCR. This method was performed on ham in less than 8 hr by artificial inoculation of serial dilutions of the virus ranging from 1,000 to 1 RT-PCR unit/mL. The viral extraction and concentration method had 10-fold higher sensitivity using the combination of Tris beef extract buffer and PEG as compared to glycine buffer and zirconium hydroxide. This method proved that RT-PCR and nested PCR have the sensitive ability to detect norovirus in commercial ham, in that norovirus was successfully detected in artificially contaminated samples at a detection level as low as 1-10 RT-PCR unit/mL. Overall, such a detection limit suggests this protocol is both quick and efficient in terms of its potential use for detecting norovirus in meat products.

  • This research investigated the manufacturing process and characteristics analysis of ZPP(Zirconium Potassium Perchlorate) as pyrotechnic are commonly found in the aerospace, defense, and automotive industries. A solid pyrotechnic mixture is composed of an oxidizing agent, fuel, and binder. Precipitation process was used to uniformly mix the raw material. Through the analysis of the material characteristics and thermal response is designed optimum ratio by NASA CEA program. It was compared by performing the evaluation of these size, shape and calorimetry characteristics.

  • ALD와 CVD 공정을 진행 하는데 있어서 전구체의 평가 및 실시간 분해과정을 확인 하는 것은 매우 중요하다. 본 실험에서는 고유전 산화막에 쓰이는 Cyclopentadienyl Tris (dimethylamino) Zirconium, CpZr(NMe2)3 전구체의 증기압 특성과 FTIR, QMS를 활용하여 $250^{\circ}C$ 온도구간 에서의 분해과정을 실시간으로 측정 하였다. CpZr(NMe2)3의 Atomic mass 는 288 amu이며 증기압은 $60^{\circ}C$에서 0.075 Torr로 측정되었다. 온도가 증가 함에 따라 FTIR 에서 CH3 symmetric stretch (2776 cm-1), CH3 symmetric stretch (2865 cm-1) intensity가 줄어 들게 되었으며 QMS에서도 15 amu (Methyl)의 신호가 온도가 증가함에 따라 감소함을 확인 할수 있었다. QMS에서 Cp의 이온전류가 사라진 이유는 Cp가 모체인 CpZr에서 모두 다 분리되었고 신호가 없어 졌기 때문이다. 본 연구를 통해서 FTIR 단독으로는 얻을수 없는 온도에 따른 세부적인 분해과정을 QMS로 실시간 측정 함으로서 FTIR의 분석 결과와 산호보완 할수 있게 되었다.

Role of CaO in the Sintering of 12Ce-TZP Ceramics (12Ce-TZP 세라믹스의 소결에서의 CaO의 역할)

  • Role of CaO in the sintering of 12Ce-TZP ceramics was studied. The addition of small amounts of CaO increase the densification rate of 12Ce-TZP by altering lattice defect structure and the diffusion coefficient of the rate controlling species, namely cerium and zirconium cations. CaO also inhibits grain growth during sintering and allows the sintering process to proceed to theoretical density by maintaining a high diffusion flux of vacancies from the pores to the grain boundaries. The inhibition of grain growth is accomplished by the segregation of solute at the grain boundaries, causing a decrease in the grain boundary mobility. The segregation of calcium was revealed by AES study.

Li, Zr doped mesoporous silica: One pot synthesis and its application to $CO_2$ adsorption at low temperature (Li, Zr 담지 메조포러스 실리카 합성 : One pot 합성 및 저온 이산화탄소 흡착 응용)

  • Li, Zr doped mesoporous silica was synthesized in one pot and investigated for low temperature $CO_2$ adsorption. Herein CTAB and TEOS are used as structural directing agent and silica source respectively. The characteristics of the material was obtained from FT IR, XRD, SEM, TG and BET results. ICP AES results revealed the presence of lithium and zirconium. The material possesses high surface area ($962.22m^2g^{-1}$) with mono dispersed particles of about 2 nm. The maximum $CO_2$ adsorption capacity is 5 wt % (50 mg/g) of $CO_2$/g of sorbent at $25^{\circ}C$, which is regenerable at $200^{\circ}C$. This regeneration temperature of the adsorbent is lower than the reported lithium zirconium silicate powder. Until now, there is no report for the synthesis of Li, Zr doped mesoporous silica. The performance studies illustrate that Li, Zr doped mesoporous silica is tunable, regenerable, recyclable and selective sorbent and hence found to be a promising candidate for $CO_2$ adsorption.

A study on the transmittance due to thickness of zirconium core (지르코니아 코어의 두께에 따른 분광광도계 투과율에 관한 연구)

  • Purpose: The purpose of this study was to investigate the transmittance differences of zirconium core due to thickness within the visible light spectrum. Methods: 36 specimens were divided into 3 groups (0.6mm, 0.8mm, 1.0mm) which have each 12 specimens. The size of specimens was 10mm high and 10mm wide. The transmittance of specimens are measured by spectrophotometer Model Cary 500 that can measure infrared-ray, visible wave and ultraviolet-ray. Results: The results shows that there was no significant difference between specimen's thickness and transmittance. Conclusion: The individual's color perception is personal and there are numerous factors that influence on it. In general, human eye can perceive the color of thing only within visible light spectrum but in this experiment through spectrophotometer there was no big difference between specimen's thickness and transmittance. To sum up, The most important factors were a layed porcelain structure and its thickness rather than core thickness in the porcelain crown.

Study on the evaporation of high melting temperature metal by using the manufactured electron hem gun system (전자총 시스템 제작과 이를 이용한 고융점 금속 증발에 관한 연구)

  • An axial electron beam gun system, which emits the electron beam power of 50 kW, has been manufactured. The electron beam gun consists of two parts. One is the electron beam generation part. including the filament, cathode, and anode. The maximum beam current is 2 A and the acceleration voltage is 25 kV. The other part includes the focusing-, deflection-, and scanning coils. The beam diameter and ham trajectory can be controlled by these coils. The characteristic of each part is measured ior the optimum condition of evaporation process. Moreover, Helmholtz coil is installed inside the vacuum chamber to adjust the incident angel of the beam to the melting surface for the maximum evaporation. We report on the evaporation rates for zirconium(Zr) and gadolinium(Gd) metals which have the high melting temperatures.

  • Zirconium is used in nuclear reactors as a structural material due to its excellent corrosion resistance and to low neutron crosssection. Variation in the distribution and solubility of Zr(IV) with solution pH was obtained. Distribution of Zr(IV) containing species in HCl and $HNO_3$ solution was analyzed by considering the complex formation of Zr(IV) species with the anion of the inorganic acid. Bromley interaction parameter between $ZrO^{2+}$ and nitrate ion was estimated by using the reported data on the solvent extraction of Zr(IV) by Cyanex272 from $HNO_3$ solution. This Bromley parameter can be utilized in calculating extraction isotherm of Zr(IV) and in predicting the separation factor between Zr(IV) and Hf(IV).

PROPERTIES OF ZR ALLOY CLADDING AFTER SIMULATED LOCA OXIDATION AND WATER QUENCHING

  • In order to study the cladding properties of zirconium after a loss-of-coolant accident (LOCA)-simulation oxidation and water quenching test, commercial Zircaloy-4 and two kinds of HANA claddings were oxidized at temperatures ranging from $900^{\circ}C$ to $1250^{\circ}C$ and exposed for 300 s, and then cooled to $700^{\circ}C$ before quenching. Microstructural observations were made to evaluate the matrix characteristics with the chemical compositions after the LOCA-simulation test. Ring compression testing was then performed to compare the ductile behaviour of the HANA and Zircaloy-4 claddings. An X-ray diffraction (XRD) analysis was carried out for temperatures ranging from room temperature to $1250^{\circ}C$ for the oxide layer to verify the oxide crystal structure at each oxidation temperature.
