Characteristics of Morphological and Physiological Changes during the Autolysis Process of Saccharomyces cerevisiae FX-2

  • In this paper, the autolysis process of Saccharomyces cerevisiae FX-2 (S. cerevisiae FX-2) via, a variety of endogenous enzyme, was investigated systematically by analyzing changes in physicochemical parameters in autolysate, surface morphology and the internal structure of the yeast cells. As an explicit conclusion, the arisen autolysis depended on the pH and the optimal pH was found to be 5.5. Based on the experimental data and the characteristics of mycelia morphology, a hypothesis is put forward that simple proteins in yeast vacuolar are firstly degraded for utilization, and then more membrane-bound proteins are hydrolyzed to release hydrolytic enzymes, which arouse an enzymatic reaction to induce the collapse of the cell wall into the cytoplasm.

  • Metal ions are essential to life. However, some metals such as mercury are harmful, even when present at trace amounts. Toxicity of mercury arises mainly from its oxidizing properties. Ionizing radiation (IR) is an active tool for destruction of cancer cells and diagnosis of diseases, etc. IR induces DNA double strand breaks in the nucleus, In addition, it causes lipid peroxidation, ceramide generation, and protein oxidation in the membrane, cytoplasm and nucleus. Yeasts have been a commonly used material in biological research. In yeasts, the physiological response to changing environmental conditions is controlled by the cell types. Growth rate, mutation and environmental conditions affect cell size and shape distributions. In this work, the effect of IR and mercury chloride (II) on the morphology of yeast cells were investigated. Saccharomyces cerevisiae cells were treated with IR, mercury chloride (II) and IR combined with mercury chloride (II). Non-treated cells were used as a control group. Morphological changes were observed by a scanning electron microscope (SEM). The half-lethal condition from the previous experimental results was used to the IR combined with mercury. Yeast cells were exposed to 400 and 800 Gy at dose rates of 400Gy $hr^{-1}$ or 800 Gy $hr^{-1}$, respectively. Yeast cells were treated with 0.05 to 0.15 mM mercury chloride (II). Oxidative stress can damage cellular membranes through a lipidic peroxidation. This effect was detected in this work, after treatment of IR and mercury chloride (II). The cell morphology was modified more at high doses of IR and high concentrations of mercury chloride(II). IR and mercury chloride (II) were of the oxidative stress. Cell morphology was modified differently according to the way of oxidative stress treatment. Moreover, morphological changes in the cell membrane were more observable in the frequently stress treated cells than the temporarily stress treated cells.

배양액의 pH가 Aureobasidium pullulans의 풀루란 생성과 형태에 미치는 영향 (Effects of pH on the Elaboration of Pullulan and the Morphology of Aureobasidium pullulans)

  • The effects of pH on the cell growth, the elaboration of pullulan, and the morphology of Aureobasidium pullulans IFO 4464 were examined. A. pulluans grew in yeast-like form at constant pH7.5 and in mycelial form at constant pH2.5. At the both pH conditions, the elaboration of pullulan was very low, about 6.0~6.5g/l. The mixture of yeast-like form and mycelial form of cells was found at the constant pH4.5, at which condition, the elaboration of pullulan was high, about 24.5g/l. The pH shift experimemts showed that the specific production rates of pullulan were 0.048($hr^{-1}$)for the mycelial form and 0.058($hr^{-1}$)for the yeast like form, which indicated that the yeast-like form has the similar, only slightly higher, biosynthetic activity of pullulan to the mycelial form at pH4.5 and the pH of culture broth is more important factor for the elaboration of pullulan than the morphology of A. pullulans.

Nanoscopic Morphological Changes in Yeast Cell Surfaces Caused by Oxidative Stress: An Atomic Force Microscopic Study

  • Nanoscopic changes in the cell surface morphology of the yeasts Saccharomyces cerevisiae (strain NCYC 1681) and Schizosaccharomyces pombe (strain DVPB 1354), due to their exposure to varying concentrations of hydrogen peroxide (oxidative stress), were investigated using an atomic force microscope (AFM). Increasing hydrogen peroxide concentration led to a decrease in cell viabilities and mean cell volumes, and an increase in the surface roughness of the yeasts. In addition, AFM studies revealed that oxidative stress caused cell compression in both S. cerevisiae and Schiz. pombe cells and an increase in the number of aged yeasts. These results confirmed the importance and usefulness of AFM in investigating the morphology of stressed microbial cells at the nanoscale. The results also provided novel information on the relative oxidative stress tolerance of S. cerevisiae and Schizo pombe.

Effect of Environmental Stress on Morphological Change of an Extremely Cadmium-Tolerant Yeast, Hansenula anomala B-7

  • An extremely cadmium-tolerant budding yeast, Hansenula anomala B-7 underwent a morphological switch in response to either heat shock treatment or cadmium stress, respectively. It exhibited a morphological transition from a unicellular yeast form to a pseudohyphae-like coagulation when subjected to prolonged heat shock treatment. In contrast, the yeast cells showed an irregularity in surface morphology when given thermal stress for a short time. Patterns of proteins expressed in the pseudohyphae-like cells demonstrated that several proteins were overexpressed while others were underexpressed in comparison with those prepared from the cells in the yeast form. It was a striking feature, however, that nearly 40% of the proteins extracted from the cells in the pseudohyphae form appeared to be composed of a single polypeptide. This polypeptide was apparently overexpressed during the pseudohyphae phase and its molecular weight was estimated to be 58 kDa according to SDS-PAGE analysis. However, a significant level of the protein was not observed in the cells before transition to pseudohyphae. The architecture of the cell shape was also damaged when incubated in a medium containing more than 1,000 ppm (8.9mM) of cadmium ions, although able to proliferate at a slow rate. However, the irregularity in the cell morphology exerted either by the brief heat shock treatment or by the cadmium stress with the high concentrations of the metal ions was not repaired, even though the damaged cells were allowed to grow for sufficient time in fresh, cadmium-free medium.

Effects of yeast hydrolysate supplementation on intestinal morphology, barrier, and anti-inflammatory functions of broilers

  • Objective: This study was conducted to evaluate the effects of dietary yeast hydrolysate (YH) supplementation on intestinal morphology, barrier, and anti-inflammatory functions of broilers. Methods: A total of 320 one day old male broilers were randomly allocated into four groups with eight replicates of ten broilers each. The broilers were supplemented with a basal diet (the control group) or basal diets adding 50, 100, 150 mg/kg YH, respectively. This trial lasted for 42 days. The orthogonal polynomial contrasts were used to determine the linear and quadratic effects of increasing levels of YH. Results: In our previous research, supplementing YH improved growth performance by enhancing body weight gain but decreased feed-to-gain ratio. In this study, compared with the control group, dietary YH addition linearly and quadratically decreased serum diamine oxidase activity (p<0.05). Additionally, supplementing YH linearly and/or quadratically decreased jejunal crypt depth (CD), tumor necrosis factor-alpha (TNF-α) concentration as well as mucin 2, interleukin-6 (IL-6), IL-1β, TNF-α, nuclear factor kappa B, and myeloid differentiation factor 88 gene expression levels (p<0.05). Whereas the jejunal villus height (VH), VH/CD, IL-10 concentration as well as zonula occludens-1 and IL-10 gene expression levels were linearly and/or quadratically increased by YH supplementation (p<0.05). Conclusion: Dietary YH supplementation improved intestinal morphology, barrier and anti-inflammatory functions while decreased intestinal permeability of broilers, which might be related with altering pertinent genes expression. This study provides evidence of YH as a promising feed additive for broilers.

Isolation and Identification of Yeasts from Wild Flowers Collected around Jangseong Lake in Jeollanam-do, Republic of Korea, and Characterization of the Unrecorded Yeast Bullera coprosmaensis

  • Several types of yeasts were isolated from wild flowers around Jangseong Lake in Jeollanam-do, Republic of Korea and identified by comparing the nucleotide sequences of the PCR amplicons for the D1/D2 variable domain of the 26S ribosomal DNA using Basic Local Alignment Search Tool (BLAST) analysis. In total, 60 strains from 18 species were isolated, and Pseudozyma spp. (27 strains), which included Pseudozyma rugulosa (7 strains) and Pseudozyma aphidis (6 strains), was dominant species. Among the 60 strains, Bullera coprosmaensis JS00600 represented a newly recorded yeast strain in Korea, and its microbiological characteristics were investigated. The yeast cell has an oval-shaped morphology measuring $1.4{\times}1.7{\mu}m$ in size. Bullera coprosmaensis JS00600 is an asporous yeast that exhibits no pseudomycelium formation. It grew well in vitamin-free medium as well as in yeast extract-malt extract broth and yeast extract-peptone-dextrose (YPD) broth, and it is halotolerant growing in 10% NaCl-containing YPD broth.

  • 효모(Saccharomyces cerevisiae, SC)의 구성성분인 yeast cell-extract(YE)과 yeast cell-wall(CW)이 육계의 생산성, 장내융모 발달 및 혈청 콜레스테롤에 어떠한 영향을 미치는지 알아보고자 사양실험을 실시하였다. 육용 수평아리(Ross) 240수를 4처리 6반복, 반복당 10수를 공시하였다. 옥수수 및 대두박 위주 기본사료에 SC, YE 그리고 CW를 각각 0.5, 0.25 그리고 $0.25\%$로 첨가한 실험사료를 제조하여 육계에 5간 급여하였다. 효모의 급여는 육계의 생산성에 어떠한 영향을 미치지 못하였으며, villus height, crypt depth 및 villus:crypt ratio 역시 처리간 유의성은 발견되지 않았다. SC 급여 받은 육계의 혈청 콜레스테롤은 대조구에 비하여 $19.7\%$ 감소(P<0.05)를 나타내었다. 특히, 효모성분 중 YE가 혈청 콜레스테롤을 유의적으로 낮추었다(P<0.05). 결론적으로 사료내 효모의 첨가는 육계의 혈청 콜레스테롤을 유의적으로 감소시켰으며(P<0.05), 콜레스테롤을 감소시키는 성분은 효모의 세포내용물에 포함되어 있는 것으로 본 연구결과 알 수 있었다.

Rahnella aquatilis Strain AY2000 Produces an Anti-Yeast Substance

  • To screen for an anti-yeast substance (AYS), many bacteria were isolated from soil and a strain AY2000 was selected. The strain AY2000 was identified as Rahnella aquatilis by morphology, biochemical properties, and 16S r-RNA nucleotide sequence analyses. The strain AY2000 showed anti-yeast activity against Candida albicans and Saccharomyces cerevisiae, whereas R. aquatilis ATCC33071 as a type strain did not show the activity against the yeasts under the same condition. The growth of yeast cell was significantly inhibited by AYS produced by the strain AY2000, as shown by optical density and MTT assay. The minimum inhibitory concentration (MIC) of the AYS against S. cerevisiae and C. albicans at $28^{\circ}C\;was\;20{\mu}g/ml\;and\;60{\mu}g/ml$, respectively. The MIC of AYS against hyphae of C. albicans at $37^{\circ}C\;was\;600{\mu}g/ml$. Scanning electron microscopic analysis revealed that yeast cells treated with AYS had an irregular form with a wrinkled and rough surface.

Isolation and characterization of unrecorded yeasts species in the family Metschnikowiaceae and Bulleribasidiaceae in Korea

  • The goal of this study was to isolate and identify wild yeasts from soil samples. The 15 wild yeast strains were isolated from the soil samples collected in Pocheon city, Gyeonggi Province, Korea. Among them, four yeast stains were unrecorded, and 11 yeast stains were previously recorded in Korea. To identify wild yeasts, microbiological characteristics were observed by API 20C AUX kit. Pairwise sequence comparisons of the D1/D2 domain of the 26S rRNA were performed using Basic Local Alignment Search Tool(BLAST). Cell morphology of yeast strains was examined by phase contrast microscope. All strains were oval-shaped and polar budding and positive for assimilation of glucose, 2-keto-ᴅ-gluconate, N-acetyl-ᴅ-glucosamine, ᴅ-maltose and ᴅ-saccharose (sucrose). There is no official report that describes these four yeast species: one strain of the genus Kodamaea in the family Metschnikowiaceae and three strains of the Hannaella in the family Bulleribasidiaceae. Kodamaea ohmeri YI7, Hannaella kunmingensis YP355, Hannaella luteola YP230 and Hannaella oryzae YP366 were recorded in Korea, for the first time.
