Composition Analysis and Antioxidative Activity from Different Organs of Aster ciliosus Kitamura (부위별 개쑥부쟁이 (Aster ciliosus Kitamura)의 이화학적 성상 및 항산화 활성)

  • In the present study, we investigated the mineral and metabolite composition and antioxidant activities in the flower, stem and root of Aster ciliosus Kitamura in order to detect biologically active substances and develop novel functional resources. The mineral and metabolite compositions differ significantly among the organ of Aster ciliosus Kitamura. The flower is composed of K as 827 mg/100g. while the stem and root are composed of Na as 654.64mg/100g and 771.70mg/100g, respectively. Contents of unsaturated fatty acids, such as oleic, linoleic, and linolenic acid, were higher than those of saturated fatty acid, such as palmitic acid in flower. The contests of phenolic compounds were measured at 285 nm using different solvents: 2.53 and 2.03 optical density (OD) in chloroform and 1.864 and 1.715 in ethanol extracts for the stem and root, respectively. Antioxidant activity was measured in 75% ethanol extracts: 41.3, 42.0 and $45.2\;{\mu}g/ml$ in stem, root and flower at $IC_{50}$ value, respectively. These results suggest that extracts from Aster ciliosus Kitamura can be potentially used as novel resources for antioxidant and biological active substances.

Optimizing Conditions for Streptomyces chibaensis J-59 Glucose Isomerase Production Using Response Surface Methodology (반응표면분석에 의한 방선균 Streptomyces chibaensis J-59 포도당 이성화효소의 생산 최적화)

  • Using response surface methodology(RSM), the various conditions(agitation speed, air flow, glucose concentration) in jar fermentor culture were investigated to find the optimum conditions for maximum enzyme production. Central-composite-design was used to control the variable constant in the experiment. The glucose isomerase production of Steptomyces chibaensis J-59 was mostly affected by the air flow rate and glucose concentration. The estimated optimum conditions were as follows: 1% birchwood xylan, 1.5% CSL, 0.1% $MgSO_4{\cdot}7H_2O$, 0.012% $CoCl_2{\cdot}6H_2O$, pH 7.0; air flow, 2.2vvm; agitation speed, 587rpm; glucose concentration, 0.586%. Experimental values(7.43GIU/ml) for the enzyme production obtained from the given optimum conditions had a almost resemblane to response values(7.67GIU/ml) predicted by the RSM. The jar fermentor culture by the RSM produced xylose isomerase about 2.7 times as much as the baffled flask culture.

  • 본 논문에서는 무선인터넷 환경에 적합한 개인화된 상품추천에이전트를 제안한다. 기존에 유선인터넷상의 많은 개인화 추천시스템에서는 초기 사용자 모델링을 위해 사용자에게 수많은 질의를 하고 응답을 요구하였다. 그러나 이러한 방식은 무선인터넷 환경에서 정보 전송량에 따른 높은 사용요금을 고려할 때 적용하기 힘든 방식이다. 본 제안 시스템은 사용자의 Social data률 이용하여 사용자를 비슷한 연령과 성별 그룹으로 나누고, 해당 그룹에서 구매율이 높은 상품을 우선 제시한 후, 사용자 행동을 모니터링 하여 암시적(Implicit)피드백을 통해 프로파일을 생성함으로써, 번거로운 질의-응답 과정 없이도 초기 사용자 모델링을 수행할 수 있다. 프로파일 생성 이후에는 이를 기반으로 하여 사용자몰 유사한 취향을 가진 그룹으로 다시 군집화한 후 협력적 추천을 하게 되며, 프로파일에는 해당 상품의 최종 카테고리명과 키워드를 수집함으로써, 상품의 브랜드와 규격정보를 반영한 추천이 가능하다. 또한 추천 상품과 사용자의 구매데이터와의 비교를 수행하여 사용자가 해당상품을 구매하였을 경우, 상품에 대한 취향정보는 그대로 유지하고 관련 상품을 추천하되, 구매한 상품이 중복 추천되지 않도록 하였다. 시스템 평가를 위해 프로토타입을 구현하여, 다수의 사용자에게 시스템을 이용하며 관심품목을 체크하도록 하였고. 추천횟수가 반복되며 히트율이 증가하는 결과를 통해 시스템의 학습속도와 성능을 평가하였다. 그리고 쇼핌몰에서 구매경험이 있는 사용자의 기존 구매데이터와 Social data를 이용한 초기 제시상품을 역으로 비교하여 오랜 시간과 비용 발생 없이도 초기 프로파일 생성의 유효성을 증명하였다. 포함하는 XML 질의에 대해서도 웹에서 캐쉬를 이용한 처리가 효율적임을 확인하였다.키는데 목적이 있다.RED에 비해 향상된 성능을 보여주었다.웍스 네트워크상의 다양한 디바이스들간의 네트워크 다양화와 분산화 기능을 얻을 수 있었고, 기존의 고가의 해외 솔루션인 Echelon사의 LonMaker 소프트웨어를 사용하지 않고도 국내의 순수 솔루션인 리눅스 기반의 LonWare 3.0 다중 바인딩 기능을 통해 저 비용으로 홈 네트워크 구성 관리 서버 시스템 개발에 대한 비용을 줄일 수 있다. 기대된다.e 함량이 대체로 높게 나타났다. 점미가 수가용성분에서 goucose대비 용출함량이 고르게 나타나는 경향을 보였고 흑미는 알칼리가용분에서 glucose가 상당량(0.68%) 포함되고 있음을 보여주었고 arabinose(0.68%), xylose(0.05%)도 다른 종류에 비해서 다량 함유한 것으로 나타났다. 흑미는 총식이섬유 함량이 높고 pectic substances, hemicellulose, uronic acid 함량이 높아서 콜레스테롤 저하 등의 효과가 기대되며 고섬유식품으로서 조리 특성 연구가 필요한 것으로 사료된다.리하였다. 얻어진 소견(所見)은 다음과 같았다. 1. 모년령(母年齡), 임신회수(姙娠回數), 임신기간(姙娠其間), 출산시체중등(出産時體重等)의 제요인(諸要因)은 주산기사망(周産基死亡)에 대(對)하여 통계적(統計的)으로 유의(有意)한 영향을 미치고 있어 $25{\sim}29$세(歲)의 연령군에서, 2번째 임신과 2번째의 출산에서 그리고 만삭의 임신 기간에, 출산시체중(出産時體重) $3.50{\sim}3.99kg$사이의 아

  • 웹의 확산과 더불어 웹 페이지 검색의 성능 즉, 빠른 응답시간과 확장성(scalability)은 각 웹 사이트의 절대적 평가 기준이 되었다. 웹 옹용은 일반적으로 불특정 다수를 대상으로 하기 때문에 확장성 또한 주요 성능의 척도가 된다. 이와 같은 웹 사이트 성능을 담보하기 위한 대표적 요소기술이 웹 캐슁이다. 본 논문은 웹 상에서 XML 데이터베이스 기반의 웹 응용(XML database-backed web application)을 위한 응용서버의 XML 캐쉬를 이용하여 주어진 XML 질의를 변환, 처리하는 기법과 구현에 관한 것으로 XPath의 경로표현식 중 가장 중요한 세 가지 기능인 조건을 명시하는 필터 연산자, 부모-자식 관계를 나타내는 경로 연산자(/), 그리고 조상-후손 관계를 나타내는 경로 연산자(//)를 연구 범위로 하였다. Effect of storage time and the level of formic acid on fermentation characteristics, epiphytic microflora, carbohydrate components and in vitro digestibility of rice straw silage

  • Objective: The study aimed to evaluate the effect of storage time and formic acid (FA) on fermentation characteristics, epiphytic microflora, carbohydrate components and in vitro digestibility of rice straw silage. Methods: Fresh rice straw was ensiled with four levels of FA (0%, 0.2%, 0.4%, and 0.6% of fresh weight) for 3, 6, 9, 15, 30, and 60 d. At each time point, the silos were opened and sampled for chemical and microbial analyses. Meanwhile, the fresh and 60-d ensiled rice straw were further subjected to in vitro analyses. Results: The results showed that 0.2% and 0.6% FA both produced well-preserved silages with low pH value and undetected butyric acid, whereas it was converse for 0.4% FA. The populations of enterobacteria, yeasts, moulds and aerobic bacteria were suppressed by 0.2% and 0.6% FA, resulting in lower dry matter loss, ammonia nitrogen and ethanol content (p<0.05). The increase of FA linearly (p<0.001) decreased neutral detergent fibre and hemicellulose, linearly (p<0.001) increased residual water soluble carbohydrate, glucose, fructose and xylose. The in vitro gas production of rice straw was decreased by ensilage but the initial gas production rate was increased, and further improved by FA application (p<0.05). No obvious difference of FA application on in vitro digestibility of dry matter, neutral detergent fibre, and acid detergent fibre was observed (p>0.05). Conclusion: The 0.2% FA application level promoted lactic acid fermentation while 0.6% FA restricted all microbial fermentation of rice straw silages. Rice straw ensiled with 0.2% FA or 0.6% FA improved its nutrient preservation without affecting digestion, with the 0.6% FA level best.

Characteristics of Fibrinolytic Enzymes of Bacillus licheniformis CY-24 Isolated from Button Mushroom Compost (양송이 배지로부터 분리한 Bacillus licheniformis CY-24의 섬유소분해 효소의 특성)

  • The present study was performed to improve the technique used for fermenting the mushroom growth medium. Taxonomic analysis of 16S rDNA sequence from the predominant Bacillus strain CY-24 isolated during the fermentation phase of the rice straw medium identified it as Bacillus licheniformis. In addition, the growth environment of B. licheniformis was also examined in this study, which revealed the optimal growth temperature and pH to be 30 ℃ and 6.0, respectively. This study also revealed that carboxymethyl cellulase (CMCase) and polygalacturonase (PGase) enzymes isolated from B. licheniformis achieved their maximal activities at 50 ℃ and 60 ℃ respectively. Furthermore, the study confirmed that the two enzymes, i.e., CMCase and PGase in B. licheniformis are stable at temperatures above 60 ℃. The present study thus demonstrates that B. licheniformis CY-24 possesses excellent enzymatic properties. It also reveals that the action of enzymes during the production of growth mediums used for the cultivation of mushrooms is closely associated with the promotion of fermentation and softening of the rice straw. Overall, this study provides elementary information regarding the role of B. licheniformis enzymes during growth medium fermentation for Agaricus bisporus cultivation.

Hydrolysis of Arabinoxylo-oligosaccharides by α-ʟ-Arabinofuranosidases and β-ᴅ-Xylosidase from Bifidobacterium dentium

  • Two α-ʟ-arabinofuranosidases (BfdABF1 and BfdABF3) and a β-ᴅ-xylosidase (BfdXYL2) genes were cloned from Bifidobacterium dentium ATCC 27679, and functionally expressed in E. coli BL21(DE3). BfdABF1 showed the highest activity in 50 mM sodium acetate buffer at pH 5.0 and 25℃. This exo-enzyme could hydrolyze p-nitrophenyl arabinofuranoside, arabino-oligosaccharides (AOS), arabinoxylo-oligosaccharides (AXOS) such as 32-α-ʟ-arabinofuranosyl-xylobiose (A3X), and 23-α-ʟ-arabinofuranosyl-xylotriose (A2XX), whereas hardly hydrolyzed polymeric substrates such as debranched arabinan and arabinoxylans. BfdABF1 is a typical exo-ABF with the higher specific activity on the oligomeric substrates than the polymers. It prefers to α-(1,2)-ʟ-arabinofuranosidic linkages compared to α-(1,3)-linkages. Especially, BfdABF1 could slowly hydrolyze 23,33-di-α-ʟ-arabinofuranosyl-xylotriose (A2+3XX). Meanwhile, BfdABF3 showed the highest activity in sodium acetate at pH 6.0 and 50℃, and it has the exclusively high activities on AXOS such as A3X and A2XX. BfdABF3 mainly catalyzes the removal of ʟ-arabinose side chains from various AXOS. BfdXYL2 exhibited the highest activity in sodium citrate at pH 5.0 and 55℃, and it specifically hydrolyzed p-nitrophenyl xylopyranoside and xylo-oligosaccharides (XOS). Also, BfdXYL2 could slowly hydrolyze AOS and AXOS such as A3X. Based on the detailed hydrolytic modes of action of three exo-hydrolases (BfdABF1, BfdABF3, and BfdXYL2) from Bf. dentium, their probable roles in the hemiceullose-utilization system of Bf. dentium are proposed in the present study. These intracellular exo-hydrolases can synergistically produce ʟ-arabinose and ᴅ-xylose from various AOS, XOS, and AXOS.

Isolation and Determination of Microbiological Characteristics of Unrecorded Wild Yeasts from Waters and Soils of Haegeumgang in the Southern Sea, and from Namdaecheon and Geumsancheon Upstream of Geumgang, Korea (해금강과 금강상류 남대천 및 금산천 주변으로부터 야생효모의 분리 및 국내 미기록 효모들의 균학적 특성)

  • The goal of this study was to investigate the diversity of wild yeasts from the waters and soils of Haegeumgang in Gyungsangnam-do, and Namdaecheon and Geumsancheon in upstream of Geumgang, Korea and to characterize any previously unrecorded wild yeast strains. In total, 52 strains comprising 22 different species of wild yeasts were isolated from 35 samples obtained from Haegeumgang. Forty three and sevent nine wild yeast strains were isolated from 90 samples taken from Namdaecheon and Geumsancheon, respectively. Among the total 174 isolated wild yeast strains, 4 strains, i.e., Exobasidium rhododendri HGG10-5 (NNIBR2022633FG1), Udeniomyces pyricola NDC29-1 (NNIBR2022633FG2), Diddensiella caesifluorescens GSC2-2 (NNIBR2022633FG5) and Pichia scaptomyzae BAC2-3 (NNIBR2022633FG4) were previously unrecorded yeasts were oval or spherical in shape, only Pichia scaptomyzae BAC 2-3 formed ascospores. Three strains with the exception of Udeniomyces pyricola NDC 29-1 grew well in vitamin-free medium and Exobasidium rhododendri HGG 10-5 grew well in YPD medium containing 10% NaCl. All four novel strains assimilated fructose, lactose, raffinose, starch and xylose.

Studies on the Production of Alcohol from Woods (목재(木材)를 이용(利用)한 Alcohol 생산(生産)에 관(關)한 연구(硏究))

  • In order to examine the alcohol production from softwoods (Pinus densiflora Sieb. et Zucc., Pinus rigida Miller, Larix leptolepis Gordon) and hardwoods (Alnus japonica Steud., Castanea crenata Sieb. et Zucc. Populus euramericana CV 214), chemical compositions were analyzed and conditions of acid hydrolysis with wood meals were established. Also strains which could remarkably decompose the cellulose were identified, and conditions of cellulase production of strains, characteristics of cellulase, and alcohol fermentation were examined. The results were summarized as follows. 1) In acid hydrolysis of wood, the high yield of reducing sugars was shown from 1.0% to 2.0% of hydrochloric acid and 2.0% of sulfuric acid. The highest yield was produced 23.4% at wood meals of Alnus japonica treated with 1.0% of hydrochloric acid. 2) The effect of raising the hydrolysis was good at $1.5kg/cm^2$, 30 times (acid/wood meal), and 45 min in treating hydrochloric acid and 30 min in treating sulfuric acid. 3) The pretreatments with concentrated sulfuric acid were more effective concentration ranged from 50% to 60% than that with hydrochloric acid and its concentration ranged from 50% to 60%. 4) The quantative analysis of sugar composition of acid hydrolysates revealed that glucose and arabinose were assayed 137.78mg and 68.24mg with Pinus densiflora, and 102.22mg and 65.89mg with Alnus janonica, respectively. Also xylose and galactose were derived. 5) The two strains of yeast which showed remarkably high alcohol productivity were Saccharomyces cerevisiae JAFM 101 and Sacch. cerevisiae var. ellipsoldeus JAFM 125. 6) The production of alcohol and the growth of yeasts were effective with the neutralization of acid hydrolysates by $CaCO_3$ and NaOH. Production of alcohol was excellent in being fermented between pH 4.5-5.5 at $30^{\circ}C$ and growth of yeasts between pH 5.0-6.0 at $24^{\circ}C$. 7) The production of alcohol was effective with the addition of 0.02% $(NH_2)_2CO$ and $(NH_4)_2SO_4$, 0.1% $KH_2PO_4$, 0.05% $MgSO_4$, 0.025% $CaCl_2$, 0.02% $MnCl_2$. Growth of yeasts was effective with 0.04-0.06% $(NH_2)_2CO$ and $(NH_4)_2SO_4$, 0.2% $K_2HPO_4$ and $K_3PO_4$, 0.05% $MgSO_4$, 0.025% $CaCl_2$, and 0.002% NaCl. 8) Among various vitamins, the production of alcohol was effective with the addition to pyridoxine and riboflavin, and the growth of yeasts with the addition to thiamin, Ca-pantothenate, and biotin. The production of aocohol was increased in 0.1% concentration of tannin and furfural, but mas decreased in above concentration. 9) In 100ml of fermented solution, alcohol and yeast were produced 2.201-2.275ml and 84-114mg for wood meals of Pinus densiflora, and 2.075-2.125ml and 104-128mg for that of Alnus japonica. Residual sugars were 0.55-0.60g and 0.60-0.65g for wood meals of Pinus densiflora and Alnus japonica, respectively, and pH varied from 3.3 to 3.6. 10) A strain of Trichoderma viride JJK. 107 was selected and identified as its having the highest activity of decomposing cellulose. 11) The highest cellulase production was good when CMCase incubated for 5 days at pH 6.0, $30^{\circ}C$ and xylanase at pH 5.0, $35^{\circ}C$. The optimum conditions of cellulase activity were proper in case of CMCase at pH 4.5, $50^{\circ}C$ and xylanase at pH 4.5, $40^{\circ}C$. 12) In fermentation with enzymatic hydrolysates, the peracetic acid treatment for delignification showed the best yields of alcohol and its ratio was effective with the addition of about 10 times. 13) The production of alcohol was excellent when wood meals and Koji of wheat bran was mixed with 10 to 8 and the 10g of wood meals of Pinus densiflora produced 2.01-2.14ml of alcohol and Alnus japonica 2.11-2.20ml.

Study on Quality Characteristics of Korean Traditional Kukhwaju by Addition of Dried Chrysanthemum indicum L. Extract into Mash (건조된 감국 열수추출물의 첨가 방법에 따른 국화주의 특성 연구)

  • This study investigated the effects of addition of Chrysanthemum indicum L. extracts on quality characteristics of Kukhwju during fermentation. Fermented liquors without C. indicum L. were used as the control group. For experimental groups, we added 1, 2, 5, and 10% C. indicum L. extracts and brewed according to the addition method written in Yorok. The pH level during fermentation phase, alcohol concentration, level of reducing sugars, and acidity were measured in each of the experimental groups. After fermentation, we measured total contents of phenols, antioxidant effects, free sugars, organic acids, and chromaticity. During fermentation, no significant difference was observed between the C. indicum L. group and control group. Regarding total phenol contents and antioxidant effects, only the 5% and 10% C. indicum L. groups showed higher contents of phenols than the non-addition group. In general, 5% and 10% C. indicum L. addition groups were positively evaluated. In conclusion, Kukhwaju with 10% C. indicum L. extract showed the best antioxidant effects. In the present study, we obtained different characteristics and determined the optimum addition amount of C. indicum L.
