Zygotorulaspora cornina sp. nov. and Zygotorulaspora smilacis sp. nov., Two Novel Ascomycetous Yeast Species Isolated from Plant Flowers and Fruits

  • Three isolates belonging to the ascomycetous genus Zygotorulaspora were obtained from the fruits of Cornus officinalis and Smilax china, and flowers of Dendranthema zawadskii var. latilobum in Gongju-si, Korea. Phylogenetic Analyses of the LSU D1/D2 domain and ITS region sequences supported the recognition of two new species: Zygotorulaspora cornina sp. nov. (type strain NIBRFGC000500475 = KACC93346PPP) and Zygotorulaspora smilacis sp. nov. (type strain NIBRFGC000500476 = KACC93347PPP). The two novel species revealed no growth on D-Galactose, unlike the other six species in the genus Zygotorulaspora. They are distinguished from each other by their phylogenetic differences and phenotypic characteristics such as assimilation of xylitol, 5-keto-D-gluconate, and ethanol. All species in the genus Zygotorulaspora including the two novel species have phenotypic traits of genus Zygotorulaspora: asci are persistent, sucrose and raffinose are assimilated, and m-inositol is not required for growth, and they are mainly associated with plants.

Effect of Oral Healthcare Program of Exemplary Kindergartens for Prevent the Dental Caries in Incheon City, Korea (치아우식증 예방을 위한 모범 유치원 구강건강관리프로그램 효과 - 인천 일부지역 유치원을 중심으로)

  • The purpose of the study is to confirm the effect of the oral health management program in exemplary kindergartens for prevent the dental caries. The study performs the survey on the parents about the changes in the oral health behaviors after the oral health education in April 2012 and measures the patient hygiene performance (PHP) index before the education, after the education and 3 weeks after the education to assess the removal capability to plaque on the teeth surface for the children. The analysis is performed on 130 people who join the program and respond the survey. There is no change in the behavior related to the tooth brushing of children after the oral health education. No change is found from the interest in the oral health education (p>0.05) but the recognition in the importance of the tooth positively change, as well as recognition of the tooth brushing method and the reaction to recommending to tooth brushing. The PHP index of children is significantly improved from 3.87 before the education through 2.71 after the education and 3.27 3 weeks after the education (p<0.001). The observation from the patents shows that their children have interest in preventing the cavities including 33.1% after having xylitol and 60.7% after the oral health education. In conclusion, the study confirms the effect of the oral health management program in exemplary kindergartens to prevent the cavities, meaning that it is required to activate the programs and keep implementing the programs including providing xylitol and education for teachers, parents and children to habituate the oral health behavior of the children and completely learn toothbrushing method under continuous management and reeducation.

Influences of Sugar Substitutes on the Physicochemical and Sensory Properties and Hardness of Baksulgi during Storage (대체감미료 사용이 백설기의 이화학적·관능적 특성 및 저장 중 경도변화에 미치는 영향)

  • Baksulgi with various sugar substitutes was prepared and characterized for physicochemical and sensory qualities. The 10% sugar in a control baksulgi was replaced with 16.1% glucose, 13.3% erythritol, 10.2% xylitol, 0.2% stevioside (enzymatically modified stevia 100%), 0.05% aspartame, or 1.2% sucralose. Instrumental and sensory analyses indicated that the baksulgi with glucose was significantly different in color from the others, which was attributed to high susceptibility to browning reaction of the glucose. Baksulgi with aspartame showed significantly lower pH than that of control baksulgi, suggesting that the aspartame might have been hydrolyzed into its constituent amino acids under elevated steaming temperature. A sensory evaluation revealed that baksulgi with sucralose or xylitol was comparable to the control baksulgi in all sensory characteristics. However, the baksulgi with sucralose showed appreciably increased hardness with increased storage time at $4^{\circ}C$, partially due to the anhygroscopic property of sucralose.

Characterization of Streptococcus mutans Ingbritt Sucrose-glucan Glucosyltransferase and the Inhibition Effect of Chitin Derivatives on its Activity (Streptococcus mutans Ingbritt sucrose-glucan glucosyltransferase 특성과 그 활성에 미치는 키틴 유도체들의 효과)

  • Sucrose-glucan glucosyltransferase (Gtf) is an important enzyme involved in the cavity formation process where insoluble glucan is synthesized. In this study, we purified Gtf from Streptcoccus mutans Ingbritt through ammonium sulfate precipitation, Sephadex G-150, CM-Sephadex, and DEAE-Sephadex column chromatographies. A 13-fold of purification was achieved with a total yield of 6.3%. The apparent molecular mass of the enzyme was determined to be 66 kDa on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The optimal pH and temperature were established to be 6.0 and $40^{\circ}C$, respectively. The enzyme activity could be inhibited to 22-59% by 1 mM $Hg^{2+}$, $Cu^{2+}$ and $Al^{3+}$, and to 68% by 1 mM EDTA. It was also inhibited 40% by 2 mM xylitol and 35-45% by 0.05% soluble chitosan, glycol chitosan, and glycol chitin. This is the first report to reveal the inhibition effect of chitin derivatives on Gtf activity, which may be further applicable to develop gargles to overcome cavity.

The Relationship between Oral Health Knowledge and Its Education Recognizance of Childcare Teacher (보육교사의 구강보건교육경험 인지도에 따른 구강보건지식의 관련성)

  • This study was conducted to examine the influence of the experience of childcare teachers about oral health education on oral health knowledge. Questionnaires to be self-filled up on the experience of oral health education were applied to 267 childcare teachers who are working in a nursery schools in Kyoungnam province. The data were analysed using the SPSS 12.0 program. The obtained result were as follows: The oral health knowledge was higher in the case with oral health education than without it. A public health center would provide the highest oral health knowledge to nursery teachers. The educations about peridontal disease, oral health effect of fluorine and Xylitol were the most demanding among the some items. When the education topics are related with peridontal disease and Xylitol, the most respondents are willing to participate in the education course. The high correlation (r=0.874) was shown in the necessity of the education and the necessity of attending the course. Also there was a low correlation (r=0.345) between the knowledge items of peridontal disease and the participation in the oral health education. The relationship of oral health knowledge with experience and recognizance of oral health education appeared. Therefore systematic and regular program for the oral health education would be required.

Mycelial Culture Conditions of Lepista nuda and Extracellular Enzyme Activity (민자주방망이버섯(Lepista nuda) 균사체 배양조건 및 효소활성)

  • The culture condition and medium composition for the enhanced mycelial growth of Lepista nuda DGUM 26501 were investigated. The optimal temperature and pH for the mycelial growth were $24^{\circ}C$ and $7.0\~8.0$, respectively. The partial pressure of oxygen for the enhanced mycelial growth was more than $10\%\;O_2$. When Czapek-Dox medium was used as a minimal medium, manitol and xylitol were very good carbon sources. Organic nitrogen sources were better than inorganic ones for mycelial growth. As the nitrogen source tested, com steep liquor, soytone and protease peptone were the best as a source of organic nitrogen sources. When ammonium phosphate as phosphorus sources was used, the enhanced mycelial growth was shown. Nicotinic acid was proved to be the most appropriate source of vitamin. After the mycelia of L. nuda DGUM 26501 was cultivated at $24^{\circ}C$ for 10 days in LNM broth (pH 7.0), the activities of extracellular enzyme were determined. The specific activity of $\alpha-amylase$ was much higher than those of other enzymes. However, little or no enzyme activities of $\beta-glucosidase$, CMCase, laccase and lipase were found.

Quality Characteristics of Vanilla Sauce with Various Sweeteners (감미료의 종류를 달리한 바닐라 소스의 품질특성)

  • The study examined the kinds of sweeteners(xylitol, sorbitol, acesulfame K, aspartame, stevioside) instead of sugar for vanilla sauce to satisfy customers' health needs. According to the results, the sauce with sugar had lowest salinity, highest sugar content, and sauce with aspartame had lowest sugar content. pH were highest in sauce with sorbitol, with aspartame were lowest. L-value, a-value was the highest in sauce with aspartame, while b-value was the lowest. The moisture content was the lowest, while viscosity was the highest in the sauce with xylitol. In a sensory evaluation, texture and overall preference was the highest in vanilla sauces with aspartame which have proper color intensity and strong flavor in mouth feel. The result indicated that health-oriented consumers and satisfy the health of modern people's needs when using the vanilla sauce with aspartame instead of sugar. It is also advised to vanilla sauce with aspartame for preventing adult disease and using the diets for patient.

Isolation, Identification, and Characterization of Bacillus spp. from the Traditionally fermented Cheonggukjangs in the Gyeonggi and the Gangwon Provinces (경기.강원 지역의 전통발효 청국장으로부터 Bacillus균주의 분리 동정 및 특성 분석)

  • Traditionally fermented Cheonggukjangs were collected from Gyeonggi and Gangwon provinces and 22 strains were isolated and identified by using 165 rDNA sequences. Most of the identified strains were Bacillus subtilis and B. licheniformis, B. subtilis and B. licheniformis are dominant in the Gyeonggi area and B. licheniformis in the Gangwon area. In the growth pattern of the isolated strains, the duration of lag phase was generally 5 to 7 hours and stationary phase was reached after 23 to 40 hours of incubation. Total cell populations at the stationary phase were between $1{\times}10^6\;CFU/ml$ and $5{\times}10^7\;CFU/ml$. The fermenting ability of carbohydrates of isolates showed some differences among the regions. The isolated strains from Yong-In, Gyeonggi showed higher fermenting abilities with D-xylose, xylitol, D-tagatose and Methyl-$\alpha$-D-mannopyranoside. D-lactose, D-tagatose, D-xylose, Methyl-$\alpha$-D-mannopyranoside, amygdalin, arbutin, esculin and 2-keto-gluconate were well fermented with the An-Seong's strains; L-rhamnose, inositol, D-mannitol, D-sorbitol, celibiose and gluconate with the Kawang-Ju's stains; and D-lactose with the Odaesan's strains.

Effect of Carbon sources and Vitamins on Mycelial Growth of Tricholoma matsutake DGUM 26001 (송이균사(Tricholoma matsutake DGUM 26001)의 생육에 미치는 탄소원 및 비타민의 영향)

  • The mycelium was isolated from the fruiting body of Tricholoma matsutake collected from Mt. Namsan, Kyongju and it was named as Tricholoma matsutake DGUM 26001. For the mycelial growth of T. matsutake DGUM 26001, the complex media, yeast-malt extract medium and Czapek-Dox medium supplemented with yeast extract, were excellent. The media such as nutrient glucose medium, mushroom complex medium, and Tricholoma matsutake medium (TMM), were effective. However, There was no a mycelial growth in the media used for bacterial cultivation such as colombia medium, brain heart infusion medium, Luria-Bertani medium supplemented with glucose, and brucella medium. When carbohydrate as a carbon and energy source was supplemented in the TMM medium for the mycelial growth, starch as a polysaccharide was best. As a disaccharide, trehalose and maltose were excellent. Sorbitol, xylitol and glucose were excellent carbon sources of monosaccharose. When the mycelia were cultivated for 30 days at $24^{\circ}C$ in the TMM supplemented with 2.0% starch, the dry weight of the mycelia harvested was 8.85 g/L. When organic acid was given as a carbon source, only succinic acid was utilized. As a vitamin source, coconut water and pyridoxine were excellent. After 30 day-cultivation in the TMM medium, the dry weights with coconut water and pyridoxine were 8.65 and 8.32 g/L, respectively.

Studies on the Mixture Wine Processing using Omija and Pear (오미자와 배를 이용한 혼합 발효주 제조 방법에 대한 연구)

  • Omija contains high organic acid content (5~7%) that must be reduced in order to produce high quality wine for consumption. In this study, we used pear, which has low total acid but plenty of sugar, to relieve the sourness of Omija material. Our group mixed Omija and pear at a ratio of 1:9 to 1:15 in order to attain a 0.5~0.7% total acid level, similar to commercial wine. As Omija was mixed with pear, the red color of Omija changed to a lighter red color. Alcohol fermentation solution of Bokbunja and Gaeryangmeoru were used to make up for the light red color of Omija pear wine. The red color of Bokbunja was confirmed to be similar to the original red color of Omija. It was proven that addition of 3~5% Bokbunja to Omija pear mixture wine produced replicated the red color image of Omija. Our results show that addition of 2~5% xylitol to Omija pear mixture wine was good by sensory test.
