Optimization of Extraction Condition of Methyl Jasmonate-treated Wild Ginseng Adventitious Root Cultures using Response Surface Methodology

  • The usage of wild ginseng (Panax ginseng C.A. Meyer) has been limited due to short supply and high price. Therefore, sufficient production as well as efficient extraction of mountain ginseng are required for the development as products. In this study, wild ginseng adventitious root cultures were prepared for efficient production with advantages of fast growth and stable production. Treatment of methyl jasmonate (MJ) to wild ginseng adventitious root cultures increased the extraction yield and antioxidative activity. Further investigation on effect of extraction conditions suggested the importance of ethanol concentration on antioxidative activity and extraction yield of MJ-treated wild ginseng adventitious root cultures. Optimized extraction condition of MJ-treated wild ginseng adventitious root cultures for maximum extraction yield and antioxidative activity was determined using response surface methodology with three-level-three-factor Box-Behnken design (BBD). Extraction of 1 g MJ-treated wild ginseng adventitious root culture with 30 ml of 9% ethanol at $30^{\circ}C$ produced 310.2 mg extract with 71.0% antioxidative activity at $100{\mu}g/ml$. Taken together, MJ-treated wild ginseng adventitious root culture is valuable source for wild ginseng usage and optimized extraction condition can be used for the development of functional products or folk remedies.

산삼 부정배양근의 진세노사이드 함량 증진과 성분 변환 (Enhancement and Conversion of Ginsenoside Contents in Cultured Wild Ginseng Adventitious Root)

  • Background: Culturing wild ginseng adventitious root using plant factory technology provides genetic safety and high productivity. This production technology is drawing attention in the fields of functional raw materials and product development. The cultivation method using elicitors is key technology for controlling biomass and increasing secondary metabolites. Methods and Results: Elicitor treatments using methyl jasmonate, pyruvic acid, squalene, β-sistosterol were performed to amplify total ginsenosides (Rb1, Rc, Rb2, Rb3, and Rd) of cultured wild ginseng adventitious root. Thereafter, fermentation and steaming processes were performed to convert total ginsenosides into minor molecular ginsenosides (Rg3, Rk1, and Rg5). The result indicated that methyl jasmonate minimizes the reduction in fresh weight of cultured wild ginseng adventitious root and maximizes total ginsenosides (sum of Rb1, Rc, Rb2, Rb3, and Rd). Ginsenoside conversion results showed a maximum degree of conversion of 131 mg/g. Conclusions: In this study, we demonstrated that the optimal elicitor treatment method increased the content of total ginsenosides, while the steaming and fermentation processing method increased the content of minor ginsenosides.

Adventitious Root Development and Ginsenoside Production in Panax ginseng, Panax quinquefolium and Panax japonicum

  • This work was carried out to establish adventitious root culture system in three Panax species (wild-grown P. ginseng, P. quinquefolium, and P. japonicum) to analyze their ginsenoside productivity. Adventitious roots were induced directly from segments of seedlings after cultured on MS(Murashige andSkoog 1962) solid medium containing 3.0 mg/l IBA. Omission of $NH_4NO_3$ from the medium greatly enhanced both the frequency of adventitious root formation and number of roots per explants in all the three Panax species. However, elongation of post-induced adventitious roots was enhanced on medium with $NH_4NO_3$. Two-step culture protocol: $NH_4NO_3$-free medium for first two weeks of culture, followed by $NH_4NO_3$ containing medium for further 4 weeks, greatly enhanced the fresh weight increase of adventitious roots in all the three ginseng species. The fresh weight of adventitious roots was high in P. quinquefolium and low in P. ginseng, followed by P. japonioum regardless of the composition of medium. Pattern and content of ginsenosides in adventitious roots differed among the three Panax species. Total ginsenoside content of adventitious roots in P. quinquefolium, P. ginseng, and p. japonicum was 8.03, 15.7 and 1.2 mg/g dry weight, respectively. Among the three speices, adventitious roots in P. quinquefolium produced hig-hamount of ginsenosides. The pattern of ginsenoside fractions between P. ginseng and P. quinquefolium was similar but the amount of ginsenoside differed between the two, While, in P japonicum, total ginsenoside content was very low and some ginsenosides such as ginsenoside Rb2 and Rf were not detected. Conclusively, we demonstrate that same culture condition was required for induction and elongation of adventitious roots of three ginseng species but growth of adventitious roots and their ginsenoside production were different among them.

  • 산삼은 고유의 생약으로 민간 또는 한방에서 효능을 인정받아 왔으나 산삼의 희귀성으로 인하여 산삼에 대한 연구가 활발하지 못하였다. 따라서 본 실험은 식물 조직 배양 기술을 이용하여 산삼 부정근을 대량으로 배양하고 추출하여 화장품 원료로서의 적용 가능성을 연구하였다.

산삼 배양근 추출물의 혈압강화 및 혈관이완 효과 (The Antihypertensive and Vasodilating Effects of Adventitious Root Extracts of Wild Ginseng)

  • 본 연구에서는 산삼 배양근이 NO 생성과 NO와 연관된 생리활성에 미치는 효과에 대해 조사하였다. ECV304 세포에 산삼 배양근 열수 추출물(WE) 혹은 부탄올 추출물의 수용액 분획물(ABE)를 처리하게 되면 상당량의 NO가 발생하는 것을 확인하였다. 추출물에 의한 ECV304 세포 내 endothelial nitric oxide synthase(eNOS)의 발현 양 변화는 거의 없었으며 100${\mu}g$의 ABE에 의해 약 6%의 ACE 억제 효과가 관찰되었다. 동맥 혈관에서의 혈관이완 효과는 WE는 2.5 mg/ml일 때 44.8%의 이완율을 나타낸 것에 비해 ABE는 0.1 mg/ml일때 91.3%의 혈관 이완율을 보였다. 선청성 고혈압 쥐인 SHR에서의 단 회 경구투여 시 혈장강화 효과는, 8시간 경과 후 최저혈압(154.5${\pm}$8.6 mmHg)을 보였고, 24시간이 지나면 초기 수준으로 회복되는 것을 확인할 수 있었다.

산삼 부정근박 급여가 육성-비육돈의 생산성, 혈액성상 및 육질특성에 미치는 영향 (Effects of Dietary Wild-Ginseng Adventitious Root Meal on Growth Performance, Blood Characteristics and Meat Quality in Growing-Finishing Pigs)

  • 본 시험은 산삼 부정근박 급여에 따른 육성-비육돈의 생산성, 혈액성상 및 육질특성에 미치는 영향을 평가하였다. 시험 동물은 3원 교잡종(Landrace×Yorkshire×Duroc) 육성-비육돈 96두를 공시하였다. 시험 개시시의 체중은 68.29±0.31kg이었으며, 사양시험은 70일간 실시하였다. 시험설계는 1) CON(Basal diet), 2) WGR1(Basal diet+0.5% cultured wild ginseng roots), 3) WGR2(Basal diet+1.0% cultured wild ginseng roots) 및 4) WGR3(Basal diet+1.5% cultured wild ginseng roots)로 4개 처리를 하여 처리당 6반복, 반복당 4두씩 완전임의 배치하였다. 성장률에서는 종료 체중과 일당 증체량에서는 WGR3 처리구가 CON 처리구구에 비해 유의적으로 낮은 결과를 나타내었다(Linear effect, P=0.005). 혈액 특성에서 red blood cell 함량은 종료시 CON과 WGR2 처리구가 WGR1 처리구에 비해 유의적으로 높은 함량을 나타내었다(Quadratic effect, P=0.019). White blood cell 함량은 종료시 WGR2 처리구가 CON와 WGR1 처리구에 비해 유의적으로 높은 함량을 나타내었다(Linear effect, P=0.041). 변화량에서도 WGR2 처리구가 다른 처리구들에 비해 유의적으로 높게 나타내었다(Linear effect, P=0.042). Total protein 함량은 종료시 WGR2 처리구가 CON처리구에 비해 유의적으로 높은 함량을 나타내었다(Quadratic effect, P=0.011). 혈액 내 콜레스테롤 함량에서는 Total cholesterol, high-density lipoprotein-cholesterol, low-density lipoprotein-cholesterol 및 triglyceride의 함량은 처리구간 유의적인 차이를 나타내지 않았다. 육질 특성에서 pH는 WGR1 처리구가 WGR3 처리구에 비해 유의적으로 높게 나타내었다 (Quadratic effect, P=0.022). 보수력에서도 WGR2 처리구가 WGR3 처리구에 비해 유의적으로 높게 나타내었다(Quadratic effect, P=0.050).

초임계 유체로 추출된 산삼 부정 배양근 오일의 효모균 처리에 의한 비사포닌계 지방산 함량 변화 및 Collagenase 및 Elastase 저해 활성 증대 (Changes in Non-saponin Fatty Acid Content and Increases in Inhibitory Activities of Collagenase and Elastase by Treatment with Saccharomyces cerevisiae of the Supercritical Fluid Extracted Oil of the Adventitious Roots Culture of Wild Mountain Ginseng)

  • Background: To obtain useful cosmetic resources, this study aimed to determine the non-saponin fatty acid and inhibitory activities of collagenase and elastase by treatment of Saccharomyces cerevisiae in supercritical fluid extracted oil of the adventitious root culture of wild mountain ginseng. Methods and Results: We performed supercritical fluid extraction at various conditions such as pressure, temperature, time, and use of co-solvents, unlike the n-hexane extraction for the adventitious roots culture of wild mountain ginseng. The non-saponin-fatty acid obtained from the oil of the adventitious roots culture was incresed by treatment with S. cerevisiae. The supercritical fluid extraction was conducted using gas chromatography. Non-saponin-fatty acid content, in the oil of adventitious roots culture of wild mountain ginseng treated with S. cerevisiae for 2 days were three times higher than that in the control. In addition, the oil of the adventitious roots culture treated with S. cerevisiae was investigated for the anti-wrinkle effect by using collagenase and elastase. The oil of adventitious roots culture treated with S. cerevisiae exhibited higher collagenase and elastase inhibitory activities than those in the control. Conclusions: Supercritical fluid extracted oil of the adventitious roots culture of wild mountain ginseng treated with S. cerevisiae was found to have decreased ratio of saturated fatty acids and incresed ratio and content of unsaturated fatty acids increased. Furthermore, it showed anti-wrinkle effects in vitro.

THE STUDY ON TISSUE CULTURED WILD MOUNTAIN GINSENG(Panax Ginseng C.A. Meyer) ADVENTITIOUS ROOTS EXTRACT AS A COSMETIC INGREDIENT

  • Korean ginseng(Panax Ginseng C.A. Meyer) known as a oriental miracle drug is an important medicinal plant. Ginseng has been used for geriatric, tonic, stomachic, and aphrodisiac treatments for thousands years. Also, it is an antibiotic and has therapeutic properties against stress and cancer. Ginseng is widely distributed all over the world. Among them, Korean mountain ginseng has the most valuable effect on pharmaceuticals. The roots of mountain ginseng contained several kinds of ginsenosides that have many active functions for the human body. However, the study of mountain ginseng has a limit because the mountain ginseng is very expensive and rare. So, we artificially cultured mountain ginseng adventitious roots using the bioreactor culture system. We induced callus from original mountain ginseng, directly dug up in mountain and aged about one hundred ten years. Separated adventitious roots were precultured in 500ml conical flasks and then, transferred in 20L bioreactors. The adventitious roots of mountain ginseng were harvested after culturing for 40days, dried and then, extracted with several solvents. In this study, we investigated the whitening effect, anti-wrinkle effect and the safety of tissue cultured adventitious roots extract of mountain ginseng in order to identify the merit as a cosmetic ingredient. Particularly, extract of mountain ginseng adventitious roots showed whitening and anti-wrinkle effects. The inhibitory effect of this extract on the melanogenesis was examined using B-16 melanoma cell. When B-16 melanoma cells were cultured with adventitious root extract, there was a dramatically decrease in melanin contents of 8-16 melanoma cell. And we identified this extract inhibited Dopa auto-oxidation significantly. Also, when transformed mouse fibroblast L929 cells were treated with this extract, there was a significant increase in collagen synthesis. The results show significant inhibited melanization and wrinkle without inhibiting cell viability.

Ginsenoside Production and Morphological Characterization of Wild Ginseng (Panax ginseng Meyer) Mutant Lines Induced by γ-irradiation (60Co) of Adventitious Roots

  • With the purpose of improving ginsenoside content in adventitious root cultures of Korean wild ginseng (Panax ginseng Meyer), the roots were treated with different dosages of ${\gamma}$-ray (5, 10, 25, 50, 75, 100, and 200 Gy). The growth of adventitious roots was inhibited at over 100 Gy. The irradiated adventitious roots showed significant variation in the morphological parameters and crude saponin content at 50 to100 Gy. Therefore, four mutant cell lines out of the propagation of 35 cell lines treated with 50 Gy and 100 Gy were selected on the basis of phenotypic morphology and crude saponin contents relative to the wild type control. The contents of 7 major ginsenosides ($Rg_1$, Re, $Rb_1$, $Rb_2$, Rc, Rf, and Rd) were determined for cell lines 1 and 3 from 100 Gy and lines 2 and 4 from 50 Gy treatments. Cell line 2 showed more secondary roots, longer length and superior growth rate than the root controls in flasks and bioreactors. Cell line 1 showed larger average diameter and the growth rate in the bioreactor was comparable with that of the control but greater in the flask cultured roots. Cell lines 1 and 2, especially the former, showed much more ginsenoside contents than the control in flasks and bioreactors. Therefore, we chose cell line 1 for further study of ginsenoside contents. The crude saponin content of line 1 in flask and bioreactor cultures increased by 1.4 and 1.8-fold, respectively, compared to the control. Total contents of 7 ginsenoside types ($Rg_1$, Re, $Rb_1$, $Rb_2$, Rc, Rf, and Rd) increased by 1.8 and 2.3-fold, respectively compared to the control. Crude saponin and ginsenoside contents in the bioreactor culture increased by about 1.4-fold compared to that the flask culture.

Effective Purification of Ginsenosides from Cultured Wild Ginseng Roots, Red Ginseng, and White Ginseng with Macroporous Resins

  • This study was aimed (i) to develop an effective method for the purification of ginsenosides for industrial use and (ii) to compare the distribution of ginsenosides in cultured wild ginseng roots (adventitious root culture of Panax ginseng) with those of red ginseng (steamed ginseng) and white ginseng (air-dried ginseng). The crude extracts of cultured wild ginseng roots, red ginseng, and white ginseng were obtained by using a 75% ethanol extraction combined with ultrasonication. This was followed sequentially by AB-8 macroporous adsorption chromatography, Amberlite IRA 900 Cl anion-exchange chromatography, and Amberlite XAD16 adsorption chromatography for further purification. The contents of total ginsenosides were increased from 4.1%, 12.1%, and 11.3% in the crude extracts of cultured wild ginseng roots, red ginseng, and white ginseng to 79.4%, 71.7%, and 72.5% in the final products, respectively. HPLC analysis demonstrated that ginsenosides in cultured wild ginseng roots were distributed in a different ratio compared with red ginseng and white ginseng.
