The Preparation of Mask-pack Sheet Blended with Styela clava tunics and Natural Polymer (미더덕껍질과 천연고분자 혼합물을 이용한 마스크팩시트의 제조방법)

  • Ultraviolet radiation have much influenced with a deep wrinkles, roughness, laxity of skin damage and pigmentation through oxidative stress and oxidative photo-damage. This study investigates the functional properties of hydrogel facial mask sheets made from agar, Styela clava tunics and Broussonetia papyrifera tunics. The skin of S. clava is covered with a hard cellulose containing glycoprotein, glycosaminoglycan and chondroitin sulfate. B. papyrifera is better known as Paper mulberry. It contains kazinol which serves as a tyrosinase inhibitor and skin whitening agent. The tensile strength of facial mask sheet was measured by universal testing machine, and the water absorption and moisture permeability of facial mask sheet were measured by dryer. Additionally, the DPPH assay and MTT assay were conducted for anti-oxidative activity and cytotoxicity of facial mask sheet. The whitening effect of the facial mask sheet was measured by tyrosinase inhibitor assay. These tests showed that the three ingredients are suitable cosmetic materials. The results reveal that they produce a high quality hydrogel facial mask sheet when the membrane contains 1%(W/V) of agar, 0.1%(W/V) of B. papyrifera tunics and 0.05%(W/V) of S. clava tunics.

Melanogenesis Inhibition Effects of Nemopilema nomurai Hydrolized Extracts (노무라입깃해파리 가수분해 추출물의 멜라닌합성 저해효과)

  • This study was aimed to investigate skin whitening effects of Nomura's jellyfish (Nemopilema nomurai) hydrolyzed extracts (NHE). The extracts were prepared through the hydrolysis of N. nomurai using commercial proteolytic enzymes such as Protamex, Alcalase, Flavourzyme and Neutrase with optimum pHs (pH 5-8) at $55^{\circ}C$. Their whitening activities were examined from the inhibition of melanin synthesis using B16-F1 cell lines. Among the examined samples, Neutrase-treated extract (N-NHE) showed the most significant inhibition effect on melanin synthesis by 89.9% at a concentration of $100{\mu}g/mL$. This sample decreased the expression of tyrosinase and TRP-1 (tyrosinase related protein-1) proteins as well. These results suggested the potential application of NHE as whitening ingredients in cosmetic preparation.

Verification of anti-oxidative activity of Aruncus dioicus, a native plant of Ulleungdo (울릉도 자생식물 삼나물(Aruncusdioicus)의 항산화 활성 검증)

  • The extracts of Aruncus dioicus were investigated for anti-oxidant and biological activities in order to verify its potential as functional ingredients of cosmetic products. The ethyl acetate (EtOAc) extract of A. dioicus showed excellent scavenging activity using DPPH and ABTS anti-oxidant analysis at the $1,000{\mu}g/mL$ concentration. While tyrosinase inhibitory effect was measure for whitening assay and showed 59.2%, the suppression levels of elastase and collagenase were 56% and 90% respectively for anti-wrinkle activity. Therefore, it is plausible to conclude that A. dioicus extract, especially EtOAc fraction which has outstanding whitening, anti-oxidant, and anti-wrinkle activities, could be used as a new functional materials for cosmetics.

Skin Whitening Effects of Caragana sinica Rehder Extract Fermented by Saccharomyces cerevisiae KCTC 7913 (골담초 효모 발효 추출물의 피부 미백 효능에 관한 연구)

  • In this study, we evaluated antioxidative effects and skin whitening effects of extract of Caragana sinica fermented by S. cerevisiae KCTC 7913. At first, Caragana sinica was fermented via inoculation of Saccharomyces cerevisiae KCTC 7913 and then extracted for fermented C. sinica. It has shown that more increased of melanogenesis inhibition activity in a dose-dependent manner on B16F10 melanoma cells and elevated the amount of resveratrol contents by HPLC analysis than non-fermented. Furthermore, the extract of fermented C. sinica was inhibitory effects against tyrosinase, a key enzyme of melanogenesis pathway, more than non-fermented C. sinica extract. And it did not show the skin irritation. Therefore, fermented C. sinica extracts might be used as safe cosmetic ingredients.

  • This study was conducted to effect the cell activity and cytotoxicity of dentifrice. For the study, 6 kinds of general dentifrice, 3 kind of whitening dentifrice, 2 kinds of natural dentifrice and SLS(sodium lauryl sulfate) of positive control group. Immortalized human gingiva fibroblast cell was used for the study, WST test for cell activity and Agar diffusion test for cytotoxicity. Agar diffusion test showed high cytotoxicity in general dentifrice test group and whitening dentifrice test group, but low cytotoxicity in natural dentifrice test group. As a result of cell nucleus staining, cell shape and nuclear activity showed that the highest activity in natural dentifrice group, followed by whitening dentifrice group and general dentifrice group. As a result of this study, the cytotoxicity of different ingredient and according to the use to dentifrice. As a result of this study, we confirm cytotoxicity of kind and components according to the purpose of using dentifrice. Therefore, it is necessary to indicate the detailed ingredients of dentifrice for the smart choice of consumers.

  • Curcuma longa L. of Zingiberaceae family, has been reported to have radical scavenging activity and antiinflammatory effect. On these facts, biological activity and whitening effect were examined to evaluate a bioactivity of the extracts of Curcumma longa L. by extraction conditions as a possible pharmaceutical material. When compared extraction methods, supercritical extraction was the best showing various biological activities and whitening effect. The highest electron donating ability $(89%\;at\;1,000{\mu}g/ml)$ were detected and SOD-like activity was inhibited up to $95%\;at\;500{\mu}g/ml$ in supercritical extraction. To evaluate the whitening effect, tyrosinase inhibitory activity was conducted 89% of tyrosinase inhibitory effect examined at $1,000\;{\mu}g/ml$. From these results, we suggest that the supercritical extracts from Curcuma longa L. is useful for cosmetic ingredients.

Preliminary Screening of Some Jeju Island Native Plants for Whitening and Antioxidant Activity (제주도 자생식물들에 대한 미백 및 항산화 효능 탐색)

  • In this study, we investigated the whitening and anti-oxidant activity of 37 Jeju island native plants. The active ingredients of the plants were prepared by methanol extraction. Whitening activity of plant extracts was examined from the inhibitory effect of tyrosinase and the inhibition of melanin synthesis of the B16-F1 cell line. Their anti-oxidant activity was measured by electron donating ability of DPPH (1,1-diphenyl-2-picrylhydrazyl) and ROS (reactive oxygen species) scavenging activity in V79-4 lung fibroblast cells using DCF-DA (dichlorofluorescin diacetate). Cytotoxicity of the extracts on cell s based experiments was investigated by MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide) assay. Also, the toxicity test using a rabbit and the human skin patch test were carried out for examining the safety of the extracts which showed the high whitening activity. It is interesting that the extracts of Lespedeza cuneata, Ligustrum lucidum (stem), Morus bombycis (stem) and Prunella vulgaris var. lilacina showed both potent whitening and anti-oxidant activities.

Whitening and Anti-inflammatory Constituents from the Extract of Citrullus lanatus Vines (수박 덩굴 추출물 유래 미백 및 항염 활성 성분)

  • In this study, we investigated whitening and anti-inflammatory constituents from a watermelon (Citrullus lanatus, C. lanatus) vines (leaves and stems). As anti-melanogenesis and anti-inflammatory activities were screened for the ethanol extract and solvent fractions, n-hexane (n-Hex) and ethyl acetate (EtOAc) fractions showed the most potent activities. Three constituents were isolated from the n-Hex and EtOAc fractions of C. lanatus; ${\alpha}-linolenic$ acid (1), sigmast-7-en-O-${\beta}$-D-glucopyranoside (2), 1-feruloyl-${\beta}$-D-glucopyrinoside (3). The chemical structures of the isolated compounds were elucidated based on the spectroscopic data including $^1H$ and $^{13}C$ NMR spectra, as well as comparison of the data to the literature values. Whitening and anti-inflammatory effects were studied for the isolated compounds. Upon the anti-melanogenesis tests using ${\alpha}-MSH$ stimulated B16F10 melanoma cells, the compounds 1 and 3 inhibited the cellular melanogenesis and intracellular tyrosinase activities effectively. For the anti-inflammation tests using lipopolysaccharide (LPS)-induced RAW 264.7 cells, the isolates 1 and 3 were determined to decrease the production of nitric oxide (NO) and pro-inflammatory cytokines ($TNF-{\alpha}$, IL-6). Based on these results, C. lanatus vines extract could be potentially applicable as whitening and anti-inflammatory ingredients in cosmetic formulations.

A Study on the Application of New Cosmetic Materials of Whitening Effect and the Physiological Activities of Chestnut Inner Shell (율피의 생리활성 몇 미백효과를 이용한 화장품신소재에 관한 연구)

  • Objectives : This is the study of the application as the ingredients of cosmetics through the examination of the function for physiological activity of Chestnut inner shell. Methods : Chestnut inner shell, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect, nitric oxide inhibition have been investigated in the physiological activity measurement of function experiment. Results : We used BHA and kogic acid for the comparative. As a result of testing electron donating ability, at over 100ppm of water extract and ethanol extract, BHA showed relatively high donating ability by more than 90%. And as a result of measuring SOD like activity, 1000ppm of water extract showed an effect of 30% and ethanol extract showed an effect of 40%, BHA showed an effect of 30%. In the xanthine oxidase inhibition test, 1000ppm of water extract showed an effect of 70% and ethanol extract showed an effect of 63%, BHA showed an effect of 100%. In the tyrosinase inhibition test, 1000ppm of water extract showed an effect of 55% and ethanol extract showed an effect of 87%, Kogic acid showed an effect of 98%. In the anti-inflammatory test, the water extract and ethanol extract inhibited the generation of nitric oxide. Conclusions : The results indicated that extract of Chestnut inner shell can be used as a natural ingredients with biological function in cosmetics ingredients.

Multifuctional Activities of Cultured extracts from Lactobacillus plantarum Ml as cosmeceutical ingredients.

  • The effects of Lactic acid bacteria have been investigated on anti-tumor. cholesterol reduction in blood. promotion of immune and skin-beauty. We are focused on cosmeceutical activity of Lactic acid bacteria (LAB). Ml, which is found in Korean traditional food. Kimchi The LAB.Ml has been identified as Lactobacillus plantarum Ml and individually cultured with Soybean soup and Soybean-Curd whey, until the total acidity has been reached the highest. After then, cell-free extracts from Ml have been used for the following studies. We assessed the effect of Lactobacillus plantarum Ml on the depigmentation of B16FlO melanoma cell. The melanin content of cells was decreased with 1-3% of cultured extracts. The tyrosinase activity was reduced by cell-free extracts of Lactobacillus plantarum Ml. Anti-aging and anti-oxidative activity of Ml cultured extract was also studied in NIH-3T3 human fibroblast cells. It showed that induction of cell proliferation. collagen synthesis and free radical scavenging activity. Additional studies for anti-fungal and anti-acne activity were also detected on Staphylococcus aureus and Propionibacterium acnes, respectively. These results suggest that cultured extract of Lactobacillun plantarum Ml would be used for cosmeceutical ingredients through multifunctional reaction on skin such as whitening, anti-wrinkle. anti-oxidation and anti-acnes.
