Isolation and Identification of Tyrosinase Inhibitors from Loranthus tanakae

  • Various bioactive substances are found in Loranthus tanakae, including quercetin 3-rhamnoside (1), kaempferol 3-rhamnoside (2), rhamnetin 3-rhamnoside (3), and rhamnocitrin 3-rhamnoside (4), which inhibit tyrosinase. These compounds are mainly found in the EtOAc fraction of L. tanakae extract and demonstrate higher rates of tyrosinase inhibition than ascorbic acid, which was used as a control. Our results suggest that L. tanakae extracts can be utilized in skin whitening cosmetics.

A Study of Physiological Activities of the Thermal Treated Eggplant on the Skin (가지 열수추출물의 피부생리활성에 관한 연구)

  • The purpose of the present research was to investigate the physiological activities of the thermal treated eggplant on the skin. Five minute of thermal treatment at $100^{\circ}C$ had the highest polyphenol content of eggplant. However, below and over 5 min of thermal treatment time, they did not increase. When water and ethanol extracts were used, the maximum DPPH radical scavenging activities were obtained, 66.3 and 62.8%, respectively. Among various extracts, the acetone extract gave the highest cosmetic activity. Especially, when acetone extract (15.0 mg/mL) was used, the maximum inhibition activities of tyrosinase, elastase, and collagenase were obtained, 83.4, 78.2 and 62.5%, respectively. These results suggest the anti-wrinkle and whitening and effects of acetone extract were excellent. Therefore, it is should be considered as a promising candidate for novel cosmetic agents.

Inhibitory Effects of Crude Polysaccharide of Water Extract of Art on Melanin Biosynthesis (인진쑥 물추출물의 조다당제에 의한 Melanin 생성억제 효과)

  • Melanogenesis is a physiological process resulting in the synthesis of melanin pigment. We investigated the inhibitory effect of crude polysaccharide of water extract of Artemisia iwayomgi Kitamura (AI) on melanin biosystheies in B16/F10 melanoma cells. Crude polysaccharide of water extract of Artemisia iwayomgi Kitamura significantly inhibited tyrosinase activity and melanin contents with or without $\alpha$-MSH in vivo. Melanin contents and tyrosinase activity were decreased in a dose-dependent manner,These results show that crude polysaccharide of water extract of Artemisia iwayomgi Kitamura could be developed as skin whitening components of cosmetics.

  • Tyrosinase is known to accelerate the melanin polymer biosynthesis in melanocyte, so tyrosinase inhibitors hinder the melanin polymer biosynthesis and are useful not only for th e material used in cosmetics as skin-whitening agents but also as the remedy for disturbances in pigmentation. During our search for new melanin biosynthesis inhibitors from natural sources, 130 higher plants were tested for the inhibitory effect against tyrosinase activity by the mushroom tyrosinase assay. Among them, Carex humilis ($IC_{50}$, 10vg/ml), Sophora flavescence ($IC_{50},\;20{\sim}50{mu}g/ml$) and Styrax japonica ($IC_{50},\;10{\mu}g/ml$) inhibited the tyrosinase activity strongly.

  • To indentify inhibitors of melanogenesis. we compared the effect of some natural products on mushroom tyrosinase. human melanocytic tyrosinase activity and melanin content. The cytotoxicity of the component were also tested on cultured mouse melanoma cells, Each extract significantly inhibited tyrosinase activity and melanin synthesis in vitro and B 16 melanoma cell lines. In B 16 cell lines, watermelon's inner shell extract inhibited tyrosinase activity as strong as kojic acid at 150${\um}g$/${\mu}\ell$ concentration. And morning glory'seed extract inhibited melanin synthesis more than kojic acid at 150${\um}g$/${\mu}\ell$ concentration. Each extract were strong inhibitors of tyrosinase activity and total melanin synthesis in B 16 mouse melanoma cell lines at less than 100${\um}g$/${\mu}\ell$ concetration. These result show that extract of watermelon's inner shell. lettuce. morning glory's seed and licorice root could be developed as skin whitening component of cosmetics.

  • Dendropanax morbifera (D. morbifera) grows in the southern coastal areas and on Jeju Island in Korea. In this study, D. morbifera leaf extract was investigated to determine the mechanism of its whitening effect. The inhibitory activities of the extract on melanogenesis were tested in B16 melanoma cells treated with the ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH). D. morbifera leaf extracts remarkably decreased the melanin content at 25 and $50{\mu}g/mL$. The extracts significantly inhibited the intracellular tyrosinase activity and protein expression of tyrosinase and tyrosinase related protein-2 (TRP-2). In conclusion, D. morbifera leaf extracts would show a whitening effect by inhibiting intracellular tyrosinase activities and the expression of enzymes directly involved in the melanin biosynthesis. The results indicate that fractions of D. morbifera leaf extracts show potential for application as a whitening agent in the new whitening cosmetics.

Skin Whitening Effects of Angelica koreana and Cnidium monnieri Extracts (강활과 벌사상자 추출물의 피부 미백효과)

  • The objective of this study is to estimate the inhibition of skin melanin formation by extract of Angelica koreana and Cnidium monnieri and the possibility of functional cosmetic materials through anti-irritation and stability test. The extract used in this experiment is White-$AK^{TM}$ and the INCI name is Osthole. The main component of White-$AK^{TM}$ was identified as coumarin and EC50 value was 2.7ppm by mouse melanoma B 16 cell test. White-$AK^{TM}$ showed inhibitory effects 100 times lower concentration than arbutin. The main mechanism for skin whitening effect thought to be inhibition of tyrosinase-related gene expression. The basic essence formulation of White-$AK^{TM}$ 5% solution applied to the skin showed the effect of relieving skin irritation. White-$AK^{TM}$ in an opaque container, under UV conditions for 4 weeks, and showed close to 100% recovery and 97% recovery under $50^{\circ}C$ for 4 weeks. Therefore, it is thought that White-$AK^{TM}$ which helps skin whitening, relieving skin irritation and stable in UV condition is able to be used as the functional component in the cosmetic formulation.

In vitro Biological Activity Assay of Ethanol Extract of Radish (무 에탄올 추출물의 in vitro 생리활성 분석)

  • In vitro biological activities of ethanol extract of radish including whitening, hangover removal, antimicrobial and antioxidant activities were analyzed. For whitening activity assay, tyrosinase inhibition rate was measured as $IC_{50}$ (50% inhibitory concentration). The $IC_{50}$ values of radish trunk and root extracts were estimated as 0,9 mg/ml and 2.1 mg/ml, respectively. Radish trunk extract showed 2.5-fold tyrosinase inhibition activity of radish root extract, however, there was no significant difference according to radish species. By alcohol dehydrogenase (ADH) activity assay as a hangover removal activity assay, radish trunk extract (2.5 mg/ml) and root extracts (8 mg/ml) showed ]50% activation of ADH. TBA values of radish trunk and root extracts (1% of each) were 43-61 % level of ${\alpha}-tochoperol$ (2.2%). From the analysis of in vitro biological activities of radish, it was suggested that radish could be used in functional food or cosmetics containing hangover removal, whitening and antioxidant activities.

The Anti-Wrinkle and Whitening Effect of Extracts of Castanea crenata Inner Shell (밤부산물(율피)의 주름개선 및 미백 효과)

  • Aging in humans is inexorable and inescapable. The progressive decrease of physiological capacity and the reduction of the ability to cope with environmental stresses lead to increased susceptibility and vulnerability to human disease. Recently, in the cosmetic industry, many researchers have paid considerable attention to delaying or improving the symptoms of skin aging. Since the early 1990`s, there have been various challenges in developing cosmeceutical products which have strong anti-aging effects, and this has been an important issue in the cosmetic industry. Meanwhile, development of anti-aging cosmetics supported by biochemical activities in the skin has been researched. Castanea crenata inner Shell solvent extracts were investigated for anti-wrinkle and whitening effects, in order to apply it as a functional ingredient for cosmetic products. For anti-wrinkle effect, elastase inhibition activity of Castanea crenata inner shell acetone extract (CA) was 51.0% at a concentration of 100 ug/ml. The collagenase inhibition activity of CA and Castanea crenata inner shell ethanol extract (CE) was 96.4%, 94.3% at a concentration of 50 ug/ml. The tyrosinase inhibitory effect, which is related to skin-whitening, was 47.2% and 45.8% in CA and CE at a concentration of 500 ug/ml. All these results suggest that Castanea crenata inner shell can be effectively used as a cosmeceutical ingredient for the prevention of wrinkles.

Antioxidant, Anti-wrinkle and Whitening Effect of Fermented Extracts of Hwangryunhaedoktang (황련해독탕 발효물의 항산화, 항주름 및 미백 효과)

  • Hwangryunhaedoktang (HHT) has been traditionally used as a preventive and therapeutic medicine to treat enervation and diverse chronic diseases. This study was designed to compare the antioxidant, anti-wrinkle and whitening effects of HHT extract and its fermented extract by Leuconostoc mesenteroides (FHHT). FHHT was prepared by inoculation of L. mesenteroides after the extraction procedure with 70% ethanol. HHT and FHHT was investigated via high-performance liquid chromatography (HPLC). Simultaneous qualitative analysis of two bioacitive components, berberine and palmatine. was achieved by comparing their retention times ($t_R$) and UV spectra with those of the standard components. Cell viability test results indicated that both HHT and FHHT were non-toxic. In DPPH radical scavenging ability, $SC_{50}$ values of the FHHT was $68.85{\mu}g/mL$, which is more effective than HHT. Moreover, FHHT showed higher expression in production of procollagen type I than HHT. In nontoxic concentration range, FHHT showed strong melanin production inhibitory effect in ${\alpha}-melanocyte$ stimulating hormone (${\alpha}-MSH$)-stimulated B16F10 cell ($IC_{50}=9.82{\mu}g/mL$). These results suggested that fermented extracts of hwangryunhaedoktang had considerable potential as a cosmetics ingredient with an antioxidant and anti-wrinkle and whitening effects.
