A Novel Synthesized Tyrosinase Inhibitor, (E)-3-(4-hydroxybenzylidene) chroman-4-one (MHY1294) Inhibits α-MSH-induced Melanogenesis in B16F10 Melanoma Cells (신규 합성물질 (E)-3-(4-하이드록시벤질리딘)크로마논 유도체의 티로시나아제 효소활성 저해 및 멜라닌 생성 억제 효과)

  • Melanin pigments are abundantly distributed in mammalian skin, hair, eyes, and nervous system. Under normal physiological conditions, melanin protects the skin against various environmental stresses and acts as a physiological redox buffer to maintain homeostasis. However, abnormal melanin accumulation results in various hyperpigmentation conditions, such as chloasma, freckles, senile lentigo, and inflammatory pigmentation. Tyrosinase, a copper-containing enzyme, plays an important role in the regulation of the melanin pigment biosynthetic pathway. Although several whitening agents based on tyrosinase inhibition have been developed, their side effects, such as allergies, DNA damage, mutagenesis, and cytotoxicity of melanocytes, limit their applications. In this study, we synthesized 4-chromanone derivatives (MHY compounds) and investigated their ability to inhibit tyrosinase activity. Of these compounds, (E)-3-(4-hydroxybenzylidene)chroman-4-one (MHY1294) more potently inhibited the enzymatic activity of tyrosinase (IC50 = 5.1±0.86 μM) than kojic acid (14.3±1.43 μM), a representative tyrosinase inhibitor. In addition, MHY1294 showed competitive inhibitory action at the catalytic site of tyrosinase and had greater binding affinity at this site than kojic acid. Furthermore, MHY1294 effectively inhibited α-melanocyte stimulating hormone (α-MSH)-induced melanin synthesis and intracellular tyrosinase activity in B16F10 melanoma cells. The results of the present study indicate that MHY1294 may be considered as a candidate pharmacological agent and cosmetic whitening ingredient.

Melanin Inhibitory Effect and Anti-inflammatory Effects of Dietyota coriacea Extracts Derived from Adjacent Sea of the Jeju Island (제주도 근해에 자생하는 참가죽그물바탕말 [Dictyota coriacea] 추출물의 멜라닌 억제 효과 및 항염증 효과)

  • We investigated several biological activities using the ethanol extract and its fractions from Dictyota coriacea to evaluate the usefulness of its extract as a functional biomaterial. The ethanol extract and n-hexane and ethyl acetate fractions showed dependently inhibitory effect on tyrosinase activity and melanin content in B16F10 cells. The ethanol extract and its fractions showed inhibitory effect on Tyrosinase and TRP-1 gene transcription but didn't showed inhibitory effect on TRP-2 gene transcription. Also, the n-hexane and ethyl acetate fractions dose-dependently inhibited the NO production in a RAW 264.7 cells. These results suggest that extract of Dictyota coriacea could be used as functional biomaterial in developing a skin whitening agent having the anti-inflammatory activity.

Inhibitory Effects on Melanin Production in B16 Melanoma Cells of Fallen Pear (B16F10 Melanoma 세포에서 낙과 배 물 추출물의 멜라닌 생성 저해 효과)

  • This study investigated the water extracts of fallen pear (FPWE) on tyrosinase activity and melanogenesis. In the present study, we examined the effects of FPWE on mushroom tyrosinase activity in vitro, B16F10 melanoma cell tyrosinase activity, melanin contents, and expression of melanogenic enzyme proteins such as tyrosinase. An apparent down-regulatory effect on tyrosinase activity was observed when B16F10 cells were incubated with FPWE. Results of melanin assay using B16F10 cells treated with different concentrations (50, 125, and $250{\mu}g/mL$) of FPWE showed a dose-dependent decrease in melanin content. To determine whether or not FPWE indirectly affects tyrosinase activity, we assessed mushroom tyrosinase activity upon treatment with various concentrations (125, 250, 500, and $1,000{\mu}g/mL$) of FPWE. In addition, we investigated changes in the protein level of tyrosinase by using Western blotting. Tyrosinase and microphthalmia-associated transcription factor expression levels in B16F10 melanoma cells were reduced in a dose-dependent manner by FPWE. These results suggest that FPWE reduced melanin formation by inhibition of tyrosinase activity. Therefore, we suggest that FPWE could be used an effective whitening agent for skin.

  • In this study, a new compound, 1-(2-hydroxy-3,4,5,6-tetramethoxyphenyl)-1-methoxy-3-phenylpropane; (Jeju-Erythrane) was isolated and identified from the bark of Lindera erythrocarpa Makino. Also, we investigated the effects of Jeju-Erythrane on alpha melanocyte-stimulating hormone (MSH)-induced melanogenesis in mouse B16F10 melanoma cells. The new compound dose dependently inhibited the tyrosinase activity and melanin synthesis in B16F10 cells. The new compound showed inhibitory effect on the Tyrosinase and TRP-1 gene transcription but not on the TRP-2 gene. These results suggest that the new compound of L. erythrocarpa could be used as a functional biomaterial in developing skin whitening agent.

Evaluating Safety of Recycled and Non-recycled Paper-based Box for Food Packaging (식품 포장재로서 재활용 및 비재활용 종이 상자의 안전성 분석)

  • There are many possibilities that recycled papers adulterate contaminants during the process. The objective of this research was to evaluate safety of paper-based packaging materials for food, especially commercial pizza boxes in Korea. Ultimately, we compared recycled and non-recycled commercial paper packaging materials. This study was based on Korean Food and Drug Administration (KFDA) regulation. PCBs were measured by GC-ECD. Arsenic and lead were analyzed using an ICP-MS. Formaldehyde was determined by LC-MS. Fluorescent materials were detected by UV lamp. The result of this study shows that non-recycled paper packaging materials meets all the tested hygienic specifications for food packaging materials, whereas recycled paper-based paperboard box may be contaminated by fluorescent whitening agent. Though these results need to be further studied, it is recommended to avoid using recycled paper in direct contact with food when used in food packaging.

Tyrosinase Inhibitory Compounds Isolated from Persicaria tinctoria Flower (쪽 꽃에서 분리한 타이로시네이즈 저해 화합물)

  • To develop a new natural whitening agent, we investigated the tyrosinase inhibitory effects of Persicaria tinctoria Flower extracts (PTFE). PTFE showed inhibitory activity on mushroom tyrosinase with the $IC_{50}$ values of $70.8{\pm}2.2{\mu}g/mL$. We purified two active compounds from PTFE by LH-20 column chromatography and prep-high performance liquid chromatography (HPLC) and identified as quercetin-3-O-rhamnoside (Q3R) and myricetin-3-O-rhamnoside (M3R) by $^1H$nuclear magnetic resonance (NMR) and liquid chromatography-mass spectrometry (LC-MS) analysis. Q3R and M3R showed tyrosinase inhibitory activities with the $IC_{50}$ values of $47.0{\pm}0.1{\mu}g/mL$ and $150.5{\pm}1.6{\mu}g/mL$, respectively. These results suggest that PTFE and its active compounds reduced melanin formation by the inhibition of tyrosinase activity. Thus, P. tinctoria flower extracts may be a candidate for cosmetic use.

  • Ascorbic acid (vitamin C, AsA) has been known as a strong reducing agent and is supposed to retard the synthesis of melanin pigment. A main problem that arose in using vitamin C in cosmetic formulation was its poor stability and low skin permeability, which result in low lightening efficacy in clinical trials. In this study, iontophoretic gel patch with flexible thin layer battery was employed in order to enhance skin permeation of vitamin c derivative (ascorbyl glucoside, AsAG) and to increase its lightening efficacy. in vitro iontophoretic skin permeation and stability of AsAG, safety and clinical lightening efficacy of iontophoretic patch containing 2% AsAG solution were examined. A optimun current of ionthophoretic patch for korean women was 0.1 mA, considering the skin permeability and skin irritation of consumers. We suggest that iontophoretic gel patch could be a safe system for enhancing the skin permeation of AsAG and lightning efficacy.

Antioxidant Effects and Melanin Inhibitory Effect of Natural Pimpinella komarovii Extracts in Jeju Island (제주도 자생 노루참나물 (Pimpinella komarovii) 추출물의 항산화 효과 및 멜라닌 억제 효과)

  • We investigated several biological activities using the ethanol extract and its fractions from Pimpinella komarovii leaves to evaluate the usefulness of its extract as a functional biomaterial. The ethanol extract showed antioxidant activities, such as DPPH scavenging activity $(IC_{50}=231.8{\mu}g/m{\ell})$. superoxide scavenging activity $(IC_{50}=23.6{\mu}g/m{\ell})$, and xanthine oxidase inhibitory activity $(IC_{50}=587.8{\mu}g/m{\ell})$. Its EtOAc fraction showed the strongest antioxidant activities among several fractions. The inhibitory effect of ethanol extract on tyrosinase activity was higher than water fraction. When $50{\mu}g/m{\ell}$ of EtOAc fraction was applied, the inhibition ratio of tyrosinase activity was much higher (42%) than that of melasolv. The EtOAc fraction also showed higher inhibitory effect on melanogenesis in Melan-a cells. The n-hexane and EtOAc fractions dose-dependently inhibited the NO production in a RAW 264.7 cells. These results suggest that extract of Pimpinella komarovii could be used as functional biomaterial in developing a skin whitening agent having the antioxidant activity.

Antioxidant and Whitening Activity of Essential Oils (Essential Oils의 항산화 활성 및 미백작용)

  • This study is performed to investigate the effects of citrus essential oils on melanin production in B16 melanoma cells and reactive oxygen species (ROS) generation in RBL 2H3 cells. Five kinds of citrus essential oil (bergamot, grapefruit, lemmon, mandarin, petigrain) did not have any influence on DPPH radical scavenger activity, cell growth and cytotoxicity in B16 melanoma cells. In purified tyrosinase assay, both mandarin and petigrain essential oils dose-dependently inhibited its activity, but bergamot did not. In $1{\mu}M\;{\alpha}-MSH-stimulated$ B16 melanoma cells, all of 5 citrus essential oils inhibited melanin production in $\underline{a}$ dose dependent manner. On the other hand, four kinds of citrus essential oil dose-dependently increased ROS generation in RBL 2H3 mast cells, but mandarin did not. From the above results, it is possible that citrus essential oils nay be developed to be anti-melanogenic agent on the basis of their inhibitory effect on MSH-induced melanin production. Hut we can not rule out the possibility of the induction of allergy and inflammation since citrus essential oils caused ROS generation in RBL 2H3 mast cells.

  • The purposes of this study were to examine the effect of 35% hydrogen peroxide (HP) bleaching agent on the changes in physical and chemical characteristics of tooth. The bleached teeth showed an apparent color changes. The whiteness increased linearly within the tested period as the period of bleaching increased. The microhardness between bleached groups after bleaching showed any statistically significant difference according to the paried t-test. The bleached enamel surface showed any apparent morphological changes compared to the enamel which was stored in distilled water only. The difference of the total mineral contents for the distilled water and hydrogen peroxide did not show statistical significance. These results demonstrated that bleaching using 35% hydrogen peroxide were adversely affects application time of experimental group and may confirm the safety of using these agents for a short time in dentist-monitored bleaching.
