For the purpose of the development of a skin-whitening agent, Sophora flavescens was evaluated for tyrosinase inhibitory activity and its active principles were identified followed activity-guided isolation. The ethanol extract and dichloromethane fraction from S. flavescens showed significant inhibition of mushroom tyrosinase. From the dichloromethane fraction, three known prenylated flavonoids, sophoraflavanone G, kuraridin, and kurarinone, were isolated. Compared with kojic acid ($IC_50$=20.5 $\mu$M), these compounds possessed more potent tyrosinase inhibitory activity. (omitted)
The purpose of this study is to characterize the effects of KHG26792 (3-(naphthalen-2-yl(propoxy)methyl)azetidine hydrochloride), a potential skin whitening agent, on melanin synthesis and identify the underlying mechanism of action. Our data showed that KHG26792 significantly reduced melanin synthesis in a dose-dependent manner. Additionally, KHG26792 downregulated microphthalmia-associated transcription factor (MITF) and tyrosinase, the rate-limiting enzyme in melanogenesis, although tyrosinase was not inhibited directly. KHG26792 activated extracellular signal-regulated kinase (ERK), whereas an ERK pathway inhibitor, PD98059, rescued KHG26792-induced hypopigmentation. These results suggest that KHG26792 decreases melanin production via ERK activation. Moreover, the hypopigmentary effects of KHG26792 were confirmed in a pigmented skin equivalent model using Cervi cornus Colla (deer antler glue), in which the color of the pigmented artificial skin became lighter after treatment with KHG26792. In summary, our findings suggest that KHG26792 is a novel skin whitening agent.

RS White (resorcinol bis-ethylhexanoate) is used in cosmetics as a skin whitening agent. In this study, we studied the possibility of nano-emulsion formation containing whitening agent, RS White, with different types of emulsifying agents. With Tween 80, 60, HCO 60 and 40 as a hydrophilic surfactants and Span 80 as ahydrophilic surfactant, nano-emulsions were formed at appropriate concentrations, but they were not formed in the system using the Myrj 52, Montanov L, and Tegocare 450 with Span 80. The diameter of nano-emulsion sphere was smaller than 100 nm. The emulsion showed a translucent appearance and maintained stability in stability evaluation with time. In vitro skin permeation experiments showed that amounts of skin permeated nano-emulsion for 24 h were $70.84{\mu}g/cm^2$ and those of O/W emulsion were $28.97{\mu}g/cm^2$. In conclusion, a stable nano-emulsion containing the resorcinol bis-ethylhexanoate is effective for potential efficacy system as an efficient delivery system of the functional materials into skin.
Purpose: Barely sprout is a well-known oriental herbal medicine with a wide range of health benefits. Recent studies have provided scientific evidence of its therapeutic effects with expanded application. This study investigated anti-melanogenic effect of barley sprout water extract (BSE) in murine melanocyte B16F10. Methods: Various concentrations (0, 50, 125, and $250{\mu}g/mL$) of BSE and arbutin (150 ppm) were applied to B16F10 stimulated with or without alpha-melanocyte stimulating hormone (100 nM) for 72 hours. The whitening potency of BSE was determined altered cellular melanin contents. Activity and expression of tyrosinase and microphthalmia-associated transcription factor (MITF) were also assayed. Results: Experimental results revealed that treatment with BSE reduced cellular melanin production by approximately 40% compared to the control. Molecular findings supported that suppressed activity and expression of tyrosinase and MITF proteins by BSE were associated with declined cellular melanogenesis. Furthermore, anti-melanogenic effect of BSE ($250{\mu}g/mL$) was similar to that of arbutin, a commonly used whitening agent. Lastly, polyphenols including p-coumaric, ferulic, and vanillic acids were identified in BSE using HPLC analyses. They might be potential active ingredients showing such melanogenesis-reducing effect. Conclusion: BSE was evident to possess favorable anti-melanogenic potency in an in vitro model. As a natural food sourced material, BSE could be an effective depigmentation agent with potential application in pharmaceutical and cosmetic industries.

This paper describes a shake-flask fermentation of Acacia flower producing whitening- and antioxidant- agents. Tyrosinase activity was inhibited in the presence of traditional and flask-fermentation broths whereas no inhibitory effect of flower extract was observed. Tyrosinase inhibition was 40% in the presence of solution containing 10% of extract from fermentation broth and it increased by increase in the concentration of it. Arbutin ($20mg\;mL^{-1}$) and kojic acid ($80{\mu}g\;mL^{-1}$) gave 90% and 58% inhibition, respectively. The result indicates that whitening activity of 40% extract solution was comparable to that of kojic acid ($80{\mu}g\;mL^{-1}$). The comparable antioxidant activity was observed for 60% extract and $10mg\;mL^{-1}$ vitamin solution. No noticeable toxicity was observed with extract. The physicochemical stability of fermentation supernatant was observed at room temperature storage condition. The result clearly shows that shake-flask fermentation of Acacia flower produced whitening agent for functional cosmetics.

In this study, the antioxidative effect, cellular protective effect and inhibitory effect on tyrosinase of Cosmos bipinnatus extracts were investigated. The ethyl acetate fraction of Cosmos bipinnatus flower extract ($11.48\;{\mu}g$/mL) showed more excellent free radical (1,1-diphenyl-2-picrylhydrazyl radical, DPPH) scavenging activity (FSC50) than those of leaf and stem extracts ($17.45\;{\mu}g$/mL). Reactive oxygen species (ROS) scavenging activity (OSC50) of Cosmos bipinnatus extracts on ROS generated in $Fe^{3+}$-EDTA/H2O2 system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction of Cosmos bipinnatus flower extract ($0.56\;{\mu}g$/mL) showed 3 times more excellent ROS scavenging activity than L-ascorbic acid ($1.50\;{\mu}g$/mL). The protective effects of the ethyl acetate fractions of extracts from different parts of Cosmos bipinnatus on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fractions of leaf and stem extract and flower extracts suppressed photohemolysis in a concentration dependent manner ($10\sim50\;{\mu}g$/mL). The inhibitory effect of ethyl acetate fraction of Cosmos bipinnatus flower extract ($62.75\;{\mu}g$/mL) on tyrosinase was investigated to assess the whitening efficacy. The ethyl acetate fraction of Cosmos bipinnatus flower extract showed 3.5 times higher tyrosinase inhibitory effect than arbutin ($226.88\;{\mu}g$/mL) known as an effective whitening agent. These results indicate that fractions of Cosmos bipinnatus extracts can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O2$ and other ROS, and protect cellular membranes against ROS. Fractions of Cosmos bipinnatus extracts can be applicable to new functional cosmetics for antioxidant and whitening.

Objectives : In order to find out the whitening effects of Leonuri Herba, this study was designed to identify the effects and the action mechanism of LHE(Leonuri Herba extract) on Malignant melanoma cell lines. Methods : After treating LHE on the B16F10 cell-Malignant melanoma cell line-, the cell survival rate, melanin biosynthesis rate, intra&extracellular tyrosinase activity rate, SOD-like activity, tyrosinase mRNA gene expression were investigated. The results were compared with control group without LHE treatment or with positive control group treated with whitening efficacy substance. Results : B16F10 cell survival rate, melanin biosynthesis rate, and intra&extracellular tyrosinase activity were significantly inhibited depending on the concentration of treated LHE. Melanin biosynthesis rate and tyrosinase activity rate were also decreased when ${\alpha}-MSH$ was combined with LHE. In addition, the SOD-like activity was increased in a concentration-dependent manner in the treatment with the LHE, indicating signigicant activity at high concentrations, and the tyrosinase mRNA gene expression was decreased in both the LHE-treated group, the LHE and ${\alpha}-MSH-treated$ group. Conclusions : LHE seems to inhibit melanin synthesis through inhibition of tyrosinase activity and inhibition of tyrosinase mRNA gene expression. It also has the effect of promoting SOD-like activity and may be used clinically as a skin whitening agent in the future.

To investigate whitening Effects of Angelica dahurica Radix Ethanol Extract (ADEE), we used melan-a cell line, brown guinea pig, and HMB-45. We treated with ADEE of 6.25, 12.5, 25, and 50 ${\mu}g/m{\ell}$ concentration in order to evaluate the effect of ADEE on cell viability and on morphological observation of melan-a cells. Also we were induced the artificial tanning spots by 1,500 mJ/$cm^2$ of ultraviolet B radiation on the backs of brown guinea pigs (approximately 450~500g) and then the test agent of $30{\mu}{\ell}$ was applied on the spots twice a day, five days a week, for five weeks respectively. The visible whitening effect was evaluated once a week. At the end of the experiment, the animals were sacrificed under anesthetization. The artificial tanning spots were obtained by biopsy punch and stained with HMB-45 to observe the gp100 proteins which were melanosomes. Our results show that cell viability was not reduce at ADEE concentrations between 6.25 and 50 ${\mu}g/m{\ell}$, melanin synthesis and melanocyte dendricity were decreased in ADEE treated melan-a cells increasing ADEE concentration. In the gross observation, ADEE treated groups had lower pigmentation than the vehicle control groups. And in the histological observation, ADEE treated groups had lower melanocytes than the vehicle control groups. Also in the quantitative analysis of the gp100 proteins using image analysis software, ADEE treated groups had a significantly lower value (p<0.001) than the vehicle control group and this resultsagreed with the results of observation under microscope. From these results, weconcluded that ADEE had positive whitening effect.
Melanin is a natural pigment found in most plants and animals, and it is involved in determining the color of the skin and hair. Melanogenesis is a reactive occurrence in melanocytes aiming to protect the skin from external stimuli, such as ultraviolet rays. Tyrosine is involved in the biosynthesis of the substrate tyrosine into melanin. However, melanin overproduction can lead to skin diseases, such as melasma, blotching, hyperpigmentation, and skin cancer. Although many studies have been conducted on whitening substances, such as kojic acid and arbutin, some countries have banned or refrained from using them due to their side effects. Therefore, this study assessed the potential of horseradish (HR) as a new whitening agent in cosmetic products. For efficient extraction, subcritical water extraction was conducted. The results showed that the horseradish subcritical water 200℃ (SW 200) extract showed high DPPH radical scavenging ability, total phenolic contents (TPC), inhibiting tyrosinase activity and inhibiting melanin production of B16-F10 melanoma cell lines. To investigate its cytotoxicity to the B16-F10 melanoma cell lines, MTT reduction assay and morphological changes were observed. No cytotoxicity was found in horseradish methanol extract and SW 200. In conclusion, this research suggests the possibility of horseradish subcritical water may be useful as a natural whitening ingredient to be used in cosmetic products.
Objectives: Artemisiae Annuae Herba is the dried aerial part of Artemisia annua L. (AAL). In Oriental medicine, Artemisiae Annuae Herba (AAH) is traditionally used to treat fever. AAH clears summerheat or damp-Heat, clears deficiency fevers, cools the blood and stops bleeding, stops malarial disorders and relieves heat, clears liver heat and brightens the eyes. Recently, there were many studies about effects of AAH on anti-oxidative, anti-inflammatory, anti-cancer, hair growth and plasma lipid composition. So, we expected AAH has an availability that can effect on skin whitening and elasticity. Methods: The present study was designed to investigate the effects of AAH on skin whitening and elasticity in SK-MEL-2 cells. In this experiment, the effects of AAH on proliferation rates, melanin synthesis, tyrosinase activities and production levels of tyrosinase, MMP-1 and MMP-9 in vitro were examined. Results: AAH did not affect viability of SK-MEL-2 cells and inhibited melanin synthesis induced by ${\alpha}$-Melanocyte-stimulating hormone (${\alpha}$-MSH) significantly. In addition, AAH also inhibited tyrosinase activity and lowered tyrosinase level in SK-MEL-2 cells. Finally, AAH inhibited productions of Matrix metalloproteinase-1 (MMP-1) and Matrix metalloproteinase-9 (MMP-9). Conclusions: These data suggest that AAH can be used to treat patients with skin diseases such as freckled face and also used as skin whitening agent.
