The effect of ginseng on the reactivity of penile corpus cavernosal smooth muscle strips of rabbits was investigated to support the clinical application of ginseng for the treatment of erectile dysfunction. New Zealand white rabbits were randomly divided into two groups, normal diet group (n=9) and ginseng saponin diet group (n=5). Then, each group was fed normal diet and Korean red ginseng saponin diet (50 mg/kg/day) for 10 weeks. We measured concentration dependent relaxation of corporal smooth muscle to acetylcholine (10-8 M to 10-4M) in organ chamber. The degree of relaxation was expressed as percentage of maximal relaxation obtained by papaverine (10-4M). Dose dependent relaxation of corpus cavernosal smooth muscle to acetylcholine, at the concentration of 10-8 M to 10-4M by half log increment, was 4.06$\pm$0.00, 4.30$\pm$1.30, 5.32$\pm$0.68, 11.64$\pm$1.74, 16.24$\pm$1.61, 23.33$\pm$ 2.29, 26.45$\pm$2.25, 30.43$\pm$2.40 and 33.41 $\pm$2.48 (%), respectively in normal diet group and 9. 83$\pm$4.15, 20.60$\pm$4.62, 24.1815.12, 35.75$\pm$5.71, 43.35$\pm$6.11, 51.30$\pm$6.22, 56.33$\pm$6.22, 54.30$\pm$4.17 and 51.98$\pm$3.92 (Vc), respectively in ginseng group. These data suggest that ginseng enhances ondothelium-dependent acetylcholine-induced relaxation of penile corpus cal.ernosal smooth muscle in rabbits.
In this study, quality in terms of the surface color and constituents of white ginseng prepared with different peeling time by using barker were investigated. The color of the white ginseng become better according to the increasing of peeling time. The components, such as contents of crude fat, crude protein, fatty acids, amino acids were slightly increased by the peeling, but carbohydrate and sugars were decreased. The contents of crude saponin and ginsenosides were markedly influenced. Compared with intact ginseng roots, peeling of ginseng roots resulted in a substantial decrease (approximately 20-30%) in total ginsenoside contents. The results suggest that peeling for white ginseng preparation leads to improve the surfacecolor formation of roots, while lose the contents of ginsenosides as the major active ingredients of ginseng.
This study was carried out to investigate a point of difference in chemical characteristics between normal and inferior Korean red ginseng (Naeback red ginseng=red ginseng with white part of clear boundary in phloem and/or xylem of ginseng body, saengnaeback red ginseng=red ginseng with white part of indistinct boundary). content of total sugar as chemical components of naeback and saengnaeback part from Korean red ginseng were less than that of normal part, and content of reducing sugar in normal and saengnaebakc part showed higher than that of naeback part. But differences in content of total phenolic compounds was not found. The content of crude saponin in normal part was highest, amounts of ginsneoside were about same. The content of constituent amino acid in normal part was about 2 times as compared with those in naeback and saengnaeback part in red ginseng. Among the various amino acids, the contents of arginine, glutamic acid, aspartic acid, leucine and alanine of normal and naeback part were higher than others, but in saengnaeback proline, glutamic acid, aspartic acid, threonin and glycine etc. were higher. In the total amount of free amino acids, those in normal, naeback and saengnaeback part were about the same one another, and arginine, glutamic acid and aspartic acid were major free amino acids.

A bacterium isolated from contaminated white ginseng was identified using API kit and electron microscope. This isolate was determined as rod shaped bacterium having about 1.0 ${\mu}m$ in diameter and 2.0 to 6.0 ${\mu}m$ in length. It had motility by peritrichous flagellum. The isolate had ${\beta}-galactosidase$, arginine dihydrolase and ornithin decarboxylase. It did not have ability not only to use citrate as sole carbon source and but also to produce $H_2S$. However, it could ferment glucose, manitol, sorbitol, rhamnose, arabinose and amygdalin. From these obserbations, the isolate was identified as Citrobacter sp. Ginseng saponin was added to culture of Citrobacter sp. in order to investigate saponin's influence on its growth. The strain was incubated at $38^{\circ}C$ for 3 days after addition of 0.05, 0.5, 2.0 and 4.0% (w/v) of saponin, respectively and the growth rates was investigated. The relative bacterial growth inhibition rates showed 28.6, 66.7, 92.4 and 97.7%, respectively, when compared with saponin non-treated group. These results suggest that the growth of Citrobacter sp. is inhibited by saponin in a concentration-dependent manner.
This study was carried out to determine changes in general chemical composition, free sugars, physicochemical properties of extract, and ginsenoside contents depending upon processing methods. Ginseng roots harvested from the same field were employed for the processing into white ginseng (WG), taegeuk ginseng (TG), red ginseng A (RGA, steamed one time), and red ginseng B (RGB, steamed three times). The fat content decreased by increasing duration of treatment and number of steaming treatment. On the other hand, there was no significant variation in contents of ash and carbohydrate depending on processing methods. Contents of sucrose and maltose was higher in Taegeuk and red ginseng than those of white ginseng. Steamed ginseng root (taegeuk and red ginseng root) showed higher amount of water extractable solid than the unsteamed white ginseng, but the variation of crude saponin content was not distinctive depending on processing methods. The contents of total ginsenosides increased by the order of white, taegeuk, red A, and red B root. In summary, chemical composition and total ginsenoside content were different according to part of root and processing methods, thus implies the importance of quality control as well as pharmacological activity of ginseng root.
In this study, white ginseng water extract (WGWE) solutions were analyzed to set up the functional saponin content and quality optimization condition. The highest saponin content among the total white ginseng extracts was 8.32 mg/10 ml which was extracted at $75^{\circ}C$ for 18 hours. In addition, the saponin content decreased according to the increased extraction temperature and time. The highest content of $Rb_2$ and Re was 0.89 mg/10 ml, 0.82 mg/10 ml at $75^{\circ}C$ for 18 hours which decreased according to the increased extracted temperature and time. The highest content of $Rg_3$ was 1.67 mg/10 ml at $95^{\circ}C$ for 24 hours which decreased according to the increased time. The turbidity, sweetness and reducing sugar content were increased according to the increased extracted time at $75^{\circ}C$, $85^{\circ}C$, $95^{\circ}C$, but pH were decreased according to the increased extracted time. Therefore, the most appropriate white ginseng extracting method have to extracted the proper temperature for saponin content at first time in combination with raise the temperature for taste at second time.
Ginseng Radix, the root of Panax ginseng C. A. Meyer has been used in Eastern Asia for 2000 years as a tonic and restorative, promoting health and longevity. Two varieties are commercially available: white ginseng(Ginseng Radix Alba) is produced by air-drying the root, while red ginseng(Ginseng Radix Rubra) is produced by steaming the root followed by drying. These two varieties of different processing have somewhat differences by heat processing between them. During the heat processing for preparing red ginseng, it has been found to exhibit inactivation of catabolic enzymes, thereby preventing deterioration of ginseng quality and the increased antioxidant-like substances which inhibit lipid peroxide formation, and also good gastro-intestinal absorption by gelatinization of starch. Moreover, studies of changes in ginsenosides composition due to different processing of ginseng roots have been undertaken. The results obtained showed that red ginseng differ from white ginseng due to the lack of acidic malonyl-ginsenosides. The heating procedure in red ginseng was proved to degrade the thermally unstable malonyl-ginsenoside into corresponding netural ginsenosides. Also the steaming process of red ginseng causes degradation or transformation of neutral ginsenosides. Ginsenosides $Rh_2,\;Rh_4,\;Rs_3,\;Rs_4\;and\;Rg_5$, found only in red ginseng, have been known to be hydrolyzed products derived from original saponin by heat processing, responsible for inhibitory effects on the growth of cancer cells through the induction of apoptosis. 20(S)-ginsenoside $Rg_3$ was also formed in red ginseng and was shown to exhibit vasorelaxation properties, antimetastatic activities, and anti-platelet aggregation activity. Recently, steamed red ginseng at high temperature was shown to provide enhance the yield of ginsenosides $Rg_3\;and\;Rg_5$ characteristic of red ginseng Additionally, one of non-saponin constituents, panaxytriol, was found to be structually transformed from polyacetylenic alcohol(panaxydol) showing cytotoxicity during the preparation of red ginseng and also maltol, antioxidant maillard product, from maltose and arginyl-fructosyl-glucose, amino acid derivative, from arginine and maltose. In regard to the in vitro and in vivo comparative biological activities, red ginseng was reported to show more potent activities on the antioxidant effect, anticarcinogenic effect and ameliorative effect on blood circulation than those of white ginseng. In oriental medicine, the ability of red ginseng to supplement the vacancy(허) was known to be relatively stronger than that of white ginseng, but very few are known on its comparative clinical studies. Further investigation on the preclinical and clinical experiments are needed to show the differences of indications and efficacies between red and white ginsengs on the basis of oriental medicines.
The patterns of saponins of lateral gingengs cultivated different areas and various ginseng products were investigated by quantitative thin-layer chromatography. In the case of ginseng cultivated in the Kum San and Gang Hwa area, some parts of the panaxatriol series of the saponins (peak 6 and 7.8.9) were higher in concentration than in ginseng grown in other areas while the other ingredients were almost the same. In the process of heat treatment the quantity of peak 2 was generally decreased. However, in the case of red and white ginseng, one part of the panaxatriol saponins, peak 6 was increased. This tendency was also found in honeyed ginseng and ginseng tea which were not exposed to sunlight, but the increase was much less. The change in the red and white ginseng which were exposed to sunlight was very substantial. Therefore we can assume that the increase of peak 6 comes about due to the combination of heat treatment and exposure to sunlight, especially due to exposure to sunlight.

Ginseng saponin was added to culture of Enterobacter sp. in order to investigate saponin's influence on its growth. The strain was incubated at 38$^{\circ}C$ for 3 days after addition of 0.05, 0.5, 2.0 and 4.0% (w/v) of saponin, respectively and the growth rates were investigated. The relative bacterial growth rates showed 75.0, 37.5, 7.5 and 0.5%, respectively, when compared with 100% of saponin non-added group. These results suggest that the growth of Enterobacter sp. is inhibited by saponin with the concentration dependency.

This investigation was carried out to study the effect of heating treatment on the saponin fractions in ginseng extract and crude saponin. The changes of saponin patterns and amounts were investigated using HPLC and compared with peak area of each fraction. Shape of crude saponin was changed more easy to compare with in ginseng extract. The more extracting temperature risen and treated time longer, the more changes of saponin amounts and patterns were shown, expecially at $100^{\circ}C$ over. All of the saponin fraction except ginsenoside-Rd were relatively unstable in heating treatment. Suitable extracting condition was extracted at $80^{\circ}C$ for 40 hours with $H_2O$.
