Synthesis of Trifluoromethylated Dihydro-1,4-oxathiin Carboxanilides and Their Fungicidal Activity (삼불화메틸기가 포함된 디히드로-1,4-옥사티인 카르복스아닐리드 유도체의 합성과 살균 활성)

  • ${\alpha},{\beta}$-Unsaturated carboxanilides 5 with trifluromethylated dihydro-1,4-oxathiins were synthesized for the development of new agrochemical fungicide. Chlorination of trifluoromethylated ${\beta}-keto$ ester 6 followed by the reaction with 1,2-mercaptoethanol gave intermediate 1,4-oxathiane 11. Without purification of 11, substitution of hydroxy group by chlorine, followed by dehydrochlorination of 10 in the presence of triethylamine afforded trifluoromethylated dihydro-1,4-oxathiin ethyl ester 9. Acylation of the hydroxy group of the carboxylic acid 12 followed by treatment of various amines gave the corresponding trifluoromethylated dihydro-1,4-oxathiin carboxamides 5. Antifungal screening (in vivo) of the synthesized compounds against typical plant diseases, which include rice blast, rice sheath blight, cucumber gray mold, tomato late blight, wheat leaf rust, and barley powdery mildew, was carried out. Where meta position of the phenyl group was substituted with isopropoxy or isopropyl group, excellent antifungal activities against rice sheath blight and wheat leaf rust were detected.

Design of new 1,3-thiazoline derivatives by isosterism and antifungal activity of new 2,4-diimino-1,3-thiazolidines (Isosterism을 이용한 새로운 1,3-thiazoline 유도체의 디자인 및 신규 2,4-diimino-1,3-thiazolidine 유도체의 살균 활성)

  • For the purpose of a development of new agrochemical fungicides, new compound 4 in which 1,3-thiazoline scaffold as well as urea moiety in the structure was designed through molecular modification of lead compound, 2-imino-1,3-thiazone based on isosterism. The reaction of N-alkylthiourea 5 and bromoacetonitrile in ethanol gave 2,4-diimino-1,3-thiazolidine hydrobromide 6 regioselectively, which was treated with isocyanates gave the corresponding 8 which is tautomer of 4. Antifungal screening (in vivo) of the synthesized compound 8 against typical plant diseases, which include rice blast, rice sheath blight, cucumber gray mold, tomato late blight, wheat leaf rust, and barley powdery mildew, was carried out. Antifungal activities against rice blast of the compound 8 were weaker than those of 2-phenylimino-1,3-thiazoline 1. Some compounds showed weak antifungal activities against wheat leaf rust.

Control of Late Blight of Tomato and Potato by Oilgochitosan (올리고키토산에 의한 토마토 역병과 감자 역병의 방제)

  • Chitosan is a linear polysaccharide composed of randomly distributed ${\ss}$-(1-4)-linked D-glucosamine and Nacetyl-D-glucosamine. There have been many reports on the induced systemic resistance and in vivo antifungal activities of higher molecular weight chitosans with molecular weights over 3,000 amu (atomatic mass unit), but there are few papers on in vivo antifungal activities of low molecular weight chitosans (oligochitosans) with molecular weights less than 3,000 amu. In our study, an oligochitosan sample (320.3,000 amu) showed a potent 1-day protective activity with control values more than 94% at concentrations of 500 and 1,000 ${\mu}g$/ml especially against tomato late blight caused by Phytophthora infestans under growth chamber conditions. It also displayed a moderate 1-day protective activity with control values of 67.89% at concentrations of 500 and 1,000 ${\mu}g$/ml against wheat leaf rust and red pepper anthracnose. On the other hand, it showed a 16-hr curative activity against red pepper anthracnose, but not against tomato late blight and wheat leaf rust. In field experiments, oligochitosan effectively suppressed the development of late blight on potato and tomato plants with control values of 72% and 48%, respectively. The results strongly indicate that oligochitosan can be used as an eco-friendly organic material for the control of late blight on tomato and potato plants.

  • [ ${\alpha},{\beta}$ ]-Unsaturated carboxamides 12 with trifluromethylated dihydro-1,4-dithiins were synthesized for the purpose of development of new agrochemical fungicide. Chlorination of trifluoromethylated ${\beta}$-ketoester 4 followed by tile reaction with 1,2-ethanedithiol gave intermediate 1,4-dithiane 9. Without purification of 9 substitution of hydroxy by chlorine followed by dehydrochlorination in the presence of triethylamine afforded trifluoromethylated dihydro-1,4-dithiin ethyl ester 7. Activation of the hydroxy of the carboxylic acid 10 obtained from the hydrolysis of 7 and then reacted with various amines gave the corresponding trifluoromethylated dihydro-1,4-dithiin carboxamides. Antifungal screening (in vivo) against typical plant diseases, Rice Blast, Rice Sheath Blight, Cucumber Gray Mold, Tomato Late Blight, Wheat Leaf Rust, and Barley Powdery Mildew of the synthesized compounds was carried out. As a result, most of the compounds shlowed weak antifungal activities and some compounds in which isopropyl group was substituted in meta of the phenyl showed antifungal activity (99%) at 250 ppm against the disease Wheat Leaf Rust.

  • A Streptomyces sp. isolated from a soil sample collected in Taejeon, Korea has previously been found to produce two new polyene antibiotics. The two new antibiotics were named "16-methyloxazolomycin (antibacterial)" and "tetrin C (antifungal)", and their chemical structures are presented elsewhere [10, 11]. In the current study, chemotaxonomy, numerical taxonomy, and ISP methods were all employed for the taxonomic study. The spore chains were spirales and the spore surface was smooth. The spore mass was a gray series and no melanin pigment was produced. On the basis of the morphological and physiological properties, the microorganism was identified to be Streptomyces erumpens, belonging to the gray series of category IV, as defined by Bergey′s Manual. Tetrin C at the concentration of 20 ${\mu}g$/ml demonstrated a potent in vivo (pot test) preventive effect against rice blast, rice sheath blight, cucumber gray mold, wheat powdery mildew, and barley leaf rust.

  • New ${\beta}$-methoxyacrylate derivatives 1-4 having thio-enol side chain were prepared and subjected to in vivo screening for fungicidal activity against phytopathogenic fungi and many of them showed good fungicidal activities against especially rice blast and wheat leaf rust at 100 ppm.

  • $({\pm})-2-(4-Chlorophenyl)-2-cyano-2-phenyl-1-hexanol$ (2) and acetate ester (3) were resolved by various lipases. (R) and (S)-systhane were synthesized by the resolved compound 2. The antifungal screening of (R), (S)-systhane and $({\pm})-systhane$ against wheat leaf rust and barley powdery mildew gave activity over 92% in concentration of 2 ppm, but (R)- and (S)-systhane were not more active than $({\pm})-systhane$.

  • Two dimeric sesquiterpenes were separated from Chloranthus japonicus Sieb. and identified as shizukaols C and F. They exhibited potent antifungal activities ($MICs=4-16{\mu}g/ml$) in vitro against various plant pathogenic fungi (Pythium ultimum, Phytophthora infestans, Botrytis cinerea, Colletotrichum lagenarium, Alternaria kikuchiana, and Magnaporthe grisea). Shizukaol C showed 88% and 91% protective activities in the greenhouse against Puccinia recondita (wheat leaf rust) and Phytophthora infestans (tomato late blight), respectively, at $100{\mu}g/ml$; shizukaol F exhibited 93% antifungal activity against Puccinia recondita at the same concentration. Therefore, these compounds might serve as interesting candidates for effective antifungal agents.

  • A synthesis of new $\beta$-ketoacetoanilide chloride derivatives and anti fungal activity of these compounds library against 6 typical plant pathogens were described. Reaction of ketene dimer with chlorine followed by treatment of aniline derivatives gave 89 kinds of the corresponding $\beta$-ketoacetoanilide chlorides through combinatorial synthetic technology using Carousel Reaction Stations. Evaluation of antifungal activity (in vivo) of this chemical library against rice blast, rice sheath blight, tomato aray mold, tomato late blight, wheat leaf rust and barley powdery mildew was carried out. In general, $\beta$-ketoacetoanilide chlorides which present a substituent at 4 in phenyl group(para) of the compounds showed selective control activity against tomato late blight caused by Phytophthora infestans.

In vivo Antifungal Activity Against Various Plant Pathogenic Fungi of Curcuminoids Isolated from the Rhizomes of Curcuma longa

  • In a search for plant extracts with potent in vivo antifungal activity against various plant pathogenic fungi, the methanol extract of the Curcuma longa rhizomes effectively controlled the development of rice blast catised by Magnaporthe grisea and tomato late blight caused by Phytophthora infestans. Three curcuminoids such as curcumin, demethoxycurcumin, and bisdemethoxycurcumin were purified from the methanol extract of C. longa rhizomes as antifungal principles. Among the three curcuminoids, demethoxycurcumin was the most active to both rice blast and tomato late blight, followed in order by curcumin and bisdemethoxycurcumin. However, they all exhibited no or little in vivo antifungal activity against other fungal pathogens causing rice sheath blight (Corticium sasaki), tomato gray mold (Botrytis cinerea), wheat leaf rust (Puccinia recondita), or barley powdery mildew (Blumeria graminis f. sp. hordel).
