  • We have investigated for amino acid composition, molecular weight and terminal amino acid of the main gliadin protein which was purified by Sephadex column. The results were obtained as follows: 1. The highest content of amino acid was glutamic acid for the main gliadin protein. 2. The molecular weight was estimated 60,200 ${\pm}$200 for the main gliadin protein. 3. Identified N-terminal and C-terminal amino acid of the main gliadin protein were phenylalanine and methionine, respectively.

An XML Tag Indexing Method Using on Lexical Similarity (XML 태그를 분류에 따른 가중치 결정)

  • For more effective index extraction and index weight determination, studies of extracting indices are carried out by using document content as well as structure. However, most of studies are concentrating in calculating the importance of context rather than that of XML tag. These conventional studies determine its importance from the aspect of common sense rather than verifying that through an objective experiment. This paper, for the automatic indexing by using the tag information of XML document that has taken its place as the standard for web document management, classifies major tags of constructing a paper according to its importance and calculates the term weight extracted from the tag of low weight. By using the weight obtained, this paper proposes a method of calculating the final weight while updating the term weight extracted from the tag of high weight. In order to determine more objective weight, this paper tests the tag that user considers as important and reflects it in calculating the weight by classifying its importance according to the result. Then by comparing with the search performance while using the index weight calculated by applying a method of determining existing tag importance, it verifies effectiveness of the index weight calculated by applying the method proposed in this paper.

Single Oral Dose Toxicity Test and Four Weeks Repeated Oral Dose Determination Test of Crude Antifungal Compounds Produced by Bacillus subtilis SN7 in Rats (Bacillus subtilis SN7이 생성한 조항균 물질의 단회 경구투여 독성 시험 및 4주 반복 경구투여 용량 결정 시험)

  • To provide information on the safety of crude antifungal compounds produced by Bacillus subtilus SN7 isolated from Meju, we carried out an acute (single) oral dose toxicity test and 4 week repeated oral dose determination test on crude antifungal compounds in male and female Sprague Dawley rats. In the acute toxicity test, rats were treated with crude antifungal compounds produced by Bacillus subtilus SN7 orally at increasing dose levels (500, 1,000, and 2,000 mg/kg) and observed for 2 weeks. In the repeated-dose 28-day oral dose determination study, rats were orally administered doses of 500, 1,000, and 2,000 mg/kg daily for 4 weeks. There were no test article-related deaths or abnormal clinical signs in the two studies. In the 4 week repeated oral dose determination test, there were also no significant differences in clinical signs, body and organ weight changes, or any other hematological and biochemical parameters between the control and treated groups. The results suggest that the crude antifungal compounds produced by Bacillus subtilus SN7 up to a dosage level of 2,000 mg/kg are not toxic in male and female rats.

Image Processing Methods for Measurement of Lettuce Fresh Weight

  • Purpose: Machine vision-based image processing methods can be useful for estimating the fresh weight of plants. This study analyzes the ability of two different image processing methods, i.e., morphological and pixel-value analysis methods, to measure the fresh weight of lettuce grown in a closed hydroponic system. Methods: Polynomial calibration models are developed to relate the number of pixels in images of leaf areas determined by the image processing methods to actual fresh weights of lettuce measured with a digital scale. The study analyzes the ability of the machine vision- based calibration models to predict the fresh weights of lettuce. Results: The coefficients of determination (> 0.93) and standard error of prediction (SEP) values (< 5 g) generated by the two developed models imply that the image processing methods could accurately estimate the fresh weight of each lettuce plant during its growing stage. Conclusions: The results demonstrate that the growing status of a lettuce plant can be estimated using leaf images and regression equations. This shows that a machine vision system installed on a plant growing bed can potentially be used to determine optimal harvest timings for efficient plant growth management.

Protein Profiles of Major Korean Rice Cultivars

  • The protein profiles among Korean rice cultivars were assessed by total protein determination, solubility fractionation, SDS-PAGE analysis and scanning densitometry. In the extraction of protein, the SDS/urea system at a neutral pH was more efficient than that at alkaline pH. The determination of total protein showed that the protein content was similar among cultivars, ranging from 87.9 to 92.7 mg/g dry weight. Additionally, the water/NaCl-soluble protein fraction, containing 14${\sim}$16 kDa albumin and 22 kDa globulin ${\alpha}$-globulin, was also similar among cultivars, with a range of 9.94 to 11.98 mg/g dry weight. The SDS-PAGE/densitometry of total protein showed that there was no discernable difference in proteins of higher molecular weights among various cultivars, whereas the amount of lower molecular weight proteins (14${\sim}$16 kDa) is somewhat variable among cultivars. Furthermore, SDS-PAGE analysis of water/NaCl-soluble and propanol-soluble fractions indicates that there is a discernible change in the content of albumin, globulin or prolamin among cultivars. Thus, the PAGE/densitometry method, preceded by solubility fractionation, is useful for examining differences in protein profiles of rice cultivars.

A Simplified Method for the Local Blood Flow Determination in Rats and Rabbits by means of $^{86}Rb$ ($^{86}Rb$을 이용한 국소 혈류량 측정편법)

  • A simplified method for the local blood flow determination by means of $^{86}Rb$ was developed in rats and rabbits. $^{86}Rb$ in the form of chloride mixed with physiological saline was intravenously injected. The doses were $10{\mu}Ci$ for rats and $100{\mu}Ci$ for rabbits, which were injected in less than 5 seconds. The rats were sacrificed after 30 seconds, and the rabbits at the intervals of 10, 20, 40 and 60 seconds, by decapitation or rapid intravenous injection of 3 to 5ml of saturated KCI. After bleeding, the organ and tissue samples, e.g. lungs, renal cortex, jejunum and skeletal muscle were quickly removed. The $^{86}Rb$ uptake in 1 gram of the organs and tissues were measured. On the basis of uptake value, administered dose and body weight, the local blood flow was calculated. Following were the results: 1. The uptake values of $^{86}Rb$ in the above organs and tissues of rats were different from other previous reports, in which the large rats were used. It appears, therefore, that the correction on the basis of body weight is necessary. 2. The uptakes of $^{86}Rb$ in the above organs and tissues of rabbits remained rather stationary within 20 to 40 seconds. 3. The local blood flow in the above organs and tissues were calculated from $^{86}Rb$ uptake in per cent dose per 1 gram tissue for 200 gram body weight. The formula could be applied not only to the rabbits but to the rats. 4. The present method could be applied to the comparison of the local blood flow between the various organs and tissues of the control and experimental animals.

A Turbidimetric Determination of Protein by Trichloroacetic Acid

  • Based on the turbidimetric response of protein with 50% trichloroacetic acid (TCA), this study aims to introduce an assay method for protein in solution. The standard procedure consists of mixing equal volume of sample solution (standard or unknown) with 50%-TCA solution and measuring the absorbance at 450 nm after 20 min. The absorbances of the solutions were almost stable over 120 min at room temperature. This assy method is simple, reproducible, and tolerant to many interfering substances. It can detect less amount than $10\mu$g/ml of bovin serum albumin. The assay method has low protein-to-protein variability over wide range of molecular weight.

Spectrochemical Determination of Impurities in Rutile (金紅石內에 포함된 不純物의 分光化學的 測定)

  • A spectrochemical method for the determination of the major impurities, such as aluminum, iron, magnesium and silicon, in rutile single crystals and variously doped rutile is presented. By applying higher current (12 amp) and a 1:2 sample-to-graphite dilution by weight, the elaborate sample preparation needed for conventional fusion technique was avoided, and relatively higher detection limits were established. Average deviations are approximately ${\pm}8%$ for iron and magnesium in the concentration ranges of 0.007 to 0.7% and 0.006 to 0.6% respectively, and ${\pm}5%$ for aluminum and silicon in the range of 0.005 to 0.5%.

Characteristics of egg coagulates cooked conventionally or by microwaves (Microwave 처리가 알찜의 품질 특성에 미치는 영향)

  • Comparisions were made for cooking times, internal temperatures, thiamin contents and textural properties of various portion weights of egg mixture cooked in a microwave oven at high and/or low power levels and in a conventional double boiler. The mean internal temperatures of conventionally cooked egg mixtures were 76.4~80.7$^{\circ}C$. When cooking was made by steaming, the mean internal temperature were comparable among samples. With the microwave cooking, the range of mean temperature was 83.8~96.4$^{\circ}C$, and they were significantly higher than the conventionally cooked egg mixtures. The hardness determination was conducted using universal testing machine and a taste panel. Samples cooked with steam or with larger portion weight had softer texture than samples cooked by microwaves or smaller amount, respectively. Mean internal temperature was significantly(r=0.99, p<0.05) correlated with mechanical hardness determination. Statistical analyses indicated no significant difference in the thiamin content among various treatments.

Treatment of Nitrogen Oxides in Ambient Air using a Ion-Selective Electrode (대기중 질산화물의 이온 선택성 전극에 의한 처리)

  • For the determination of polluant NOx in ambient air, nitrate ion-selective electrode(ISE) was made. To comparison of NOx in each method, the nitrate-ISE, NEBA, Orion electrode were used to determinee NOx in ambient air. In this work, the concentration of NOx in ambient air was average 0.06ppm. The results were good agreement with those obtained by each method within a relative error of 3%, Absorbing efficiency of nitrogen oxides in ambient air was good for Alkali solution. The determination of nitrogen oxides in ambient air using the Aliquat 336N-PVC membrane electrode was one of the useful method. The best characteristics of the Aliquat 336N-PVC me,mbrane electrode were obtained with the ion-exchanger concentration level of 6.5-9.1 percent by weight. The optimal membrane composition, was 9.09wt.% of ion-exchanger, 30.95wt.% of PVC, 60.6wt.% of plasticizer (DBP), and 0.5mm of thickness. Under the above condition, the electrode approached the Nernstian slope most closely, and the linear response ranges produced the best results.
