Skin Permeation Characteristics of Antihyperlipoproteinemic Agent using Natural Polymer Bases in Rats (천연고분자 기재에 의한 수용성 항고지단백혈증제의 흰쥐 피부투과 특성)

  • Transdermal therapeutic system(TTS) is often used as the method of drug dosage into the epidermic skin. Natural polymer were selected as ointment material of TTS. We investigated the permeation of natural polymer ointment containing drug in rat skin using horizontal membrane cell model. Permeation properties of materials were investigated for water-soluble drug such as oxiniacic acid in vitro. These results showed that skin permeation rate of drug across the composite was mainly dependent on the property of ointment base and drug. Proper selection of the polymeric materials which resemble and enhance properties of the delivering drug was found to be important in controlling the skin permeation rate. This result suggests a possible use of natural polymer ointment base as TTS of antihyperlipoproteinemic agent.

Mechanical Properties of Cement Mortar with Polymers (폴리머 모르타르의 기계적 특성)

  • A polymer reinforced grouts using ordinary portland cement mortar and water soluble polymer{polyvinyl alcohol(PVA), styrene-butadiene rubbre(SBR), etylene-vinyl acetate copolymer(EVA)} were made. The mechanical properties of the hardened specimens were investigated through the observation of the microstructure and application of fracture mechanics. When the PVA, SBR and EVA was added with 1.5 wt% to the grouts, the compressive strength were about 54 MPa, 63 MPa and 68 MPa respectively, and the flexural strength was about 11 MPa, 12.8 MPa, and 13.6 MPa respectively, and Young's modulus was about 3.8 GPa, 4.4 GPa and 4.6 GPa respectively, and critical stress intensity was about 0.73 MNm-1.5, 0.85 MNm-1.5 and 0.9 MNm-1.5 respectively. It can be considered that the strength improvement of polymer mortar grouts may be due to the removal of macropores and the increase of various fracture toughness effects, such as grain bridging, frictional interlocking and polymer bridging.

Mechanical degradation kinetics of poly(ethylene oxide) in a turbulent flow

  • Turbulent drag reduction (DR) efficiency of water soluble poly(ethylene oxide) (PEO) with two different molecular weights was studied as a function of polymer concentration and temperature in a turbulent flow produced via a rotating disk system. Its mechanical degradation behavior as a function of time in a turbulent flow was also analyzed using both a simple exponential decay function and a fractional exponential decay equation. The fractional exponential decay equation was found to fit the experimental data better than the simple exponential decay function. Its thermal degradation further exhibited that the susceptibility of PEO to degradation increases dramatically with increasing temperature.

  • Polyethylene oxide (PEO) surfaces are recognized as having an effective steric stabilization character. A theoretical scaling analysis involves the osmotic and elastic coefficients of the polymer as a function of molecular weight, in a good solvent. The calculated results show that PEO in water may exhibit the greatest flexibility among water soluble polymers, probably due to its lowest elastic contribution.

  • Submicron-sized polymeric colloidal particles can self assemble into 3-dimensional (3D) opal structure which is a useful template for photonic crystal. Narrowly dispersed polymer microspheres can be synthesized by emulsion polymerization in water using water-soluble radical initiator. In this report, we demonstrate a facile and reproducible emulsion polymerization method to prepare various polymeric microspheres within 200 - 400 nm size ranges which can be utilized as colloidal photonic crystal template. By controlling the amount of monomer and surfactant, monodisperse polymer colloids of polystyrene (PS) and acrylates with various sizes were successfully prepared without complicated synthetic procedures. Such polymer colloids self-assembled into 3D opal structure exhibiting bright colors by reflection of visible light. The colloidal particles and the resulting opal structures were rigorously characterized, and the wavelength of the structural color from the colloidal crystal was confirmed to have quantitative relationship with the size of constituting colloidal particles as predicted by Bragg equation. The tunability of the structural color was achieved not only by varying the particle size but also by infiltration of the colloidal crystal with liquids having different refractive indices.

  • Acyclovir(ACV) is an important antiviral drug used extensively against infections caused by herpes viruses, especially herpes simplex and varicella zoster. Because of high crystallinity and large particle size, solubility of intact ACV is very low in water(1.3 mg/ml). The goal of this work is to enhance the solubility of ACV. To make solid dispersion, Polyethyleneglycol, Hydroxyprophylmethylcelluose and Polyvinylpyrrolidone were used as polymer carriers in this work. Polymer carriers and drug were dissolved in acetic acid. And then spray drying method and freeze drying method were used as solvent extraction. Morphology, crystallization and functional group were characterized using SEM, XRD and FT-IR. The result of in vitro test showed the sample using PVP as polymer carrier had higher dissolution rate(up to 466%) than intact ACV.

Water Soluble Cyclosporine Monomethoxy Poly(ethyleneglycol) Conjugates as Potential Prod rugs

  • The highly water-soluble monomethoxypoly(ethyleneglycol) (mPEG) prod rugs of cyciosporin A(CsA) were synthesized. These prod rugs were prepared by initially preparing intermediate in the form of carbonate at the 3'-positions of CsA with chloromethyl chloroformate, in the pres-ence of a base to provide a 3'-carbonated CsA intermediate. Reaction of the CsA intermediate with mPEG derivative in the presence of a base provides the desired water-soluble prod rugs. As a model, we chose molecular weight 5 kDa mPEG in the reaction with CsA to give water soluble prodrugs. To prove that the prod rug is decomposed in the body to produce CsA, the enzymatic hydrolysis test was conducted using human liver homogenate at $37^{\circ}C$. The prodrug was decomposed in human liver homogenate to produce the active material, CsA, and the hydrolysis half-life ($t_{1/2}$) of the prodrug, KI-306 was 2.2 minutes at $37^{\circ}C$. However, a demon-stration of non-enzymatic conversion in pH 7.4 phosphate buffer was provided by the fact that the half-life ($t_{1/2}$) is 21 hours at 37$^{\circ}C$. The hydrolysis test in rat whole blood was also conducted. The hydrolysis was seen with half-life ($t_{1/2}$) of about 9.9, 65.0, 14.2, 3.4, 2.1 9.5, and 1.6 minutes for KI-306, 309, 312, 313, 315, 316, and 317, respectively. This is the ideal for CsA prodrug. The pharmacokinetic study of the prodrug, KI-306, in comparison to the commer-cial product (Sandimmune Neoral Solution) was also carried out after single oral dose. Each rat received 7 mg/kg of CsA equivalent dose. Especially, the prodrug KI-306 exhibits higher AUC and $C_{max}$ than the conventional Neoral. The AUC and $C_{max}$ were increased nearly 1.5 fold. The kinetic value was also seen with $T_{max}$ of about 1.43 and 2.44 hours for KI-306 and Neoral, respectively.

  • $\omega$-Anhydride-functionalized poly(ethylene oxide) (PEO) obtained from chain-end functionalization and anionic ring-opening polymerization of ethylene oxide using n-butyllithium with potassium t-butoxide in the presence of dimethylsulfoxide (DMSO) was found to be an efficient material for the preparation of water-soluble, polymeric pro-drugs. The reaction of $\omega$-anhydride-functionalized PEO with sulfonamide or with vancomycin provided an efficient method to produce corresponding, water-soluble, PEO-conjugated sulfonamide or PEO-conjugated, vancomycin pro-drugs. These were used successfully to prepare water-dispersible, silver nanoparticles. In this study, the particle sizes were in the range of $5{\sim}40$ nm. The resulting products were characterized by $^1H$ NMR spectroscopy, transmission electron microscopy, electron and X-ray diffraction, size exclusion chromatography, and UV/Visible spectroscopy.

  • The synthesis of poly(2-ethynylpyridine) having glycidyl functionality was performed by the direct polymerization of 2-ethynylpyridine and epibromohydrin under mild reaction conditions without any initiator and catalysts. The polymerization proceeded well to give the resulting poly(2-ethynylpyridinium bromide) with a glycidyl functionality having relativity high molecular weight in high yields. The polymer structure was characterized by various instrumental methods to have the conjugated polymer backbone structure having glycidyl functionality. This ionic polymer was completely soluble in water, methanol, DMF, DMSO, and N,N-dimethylacetamide, but insoluble in THF, toluene, acetone, nitrobenzene, and n-hexane. This polymer system exhibited the UV-visible absorption around 300 and 520 nm and red photoluminescence spectrum around 725 nm.
