We examined the antimicrobial activity of electrolyzed alkaline water against spoilage microorganisms in rice warehouses. Dominant microbial species were isolated from paddy, dust and air samples of three different warehouses located in Gyeongnam. We used electrolyzed alkaline water (EW) manufactured over various treatment times. The acidities (pH) of EW treated for 30 sec, 1, 2, 3, and 4 min were 8.89, 8.91, 9.20, 9.35, and 9.22, respectively. HClO contents were 150.7, 314.2, 191.8, 104.1, and 255.3 ppm, respectively. EW inhibited bacteria, yeasts, and molds; also, it inhibited three yeast species strongly. The antimicrobial effects of EW increased as pH values and HClO contents increased. EW obtained after 30 sec treatment retained the antimicrobial activities after 14 days whereas EW obtained after 1 and 2 min treatments retained the antimicrobial activities after 21 and 24 days, respectively.
BACKGROUND: Contaminated water was a major source of food-borne pathogens in various recent fresh produce-related outbreaks. This study was conducted to investigate the microbial contamination level and correlations between the level of sanitary indicator bacteria and the detection ratio of pathogens in agricultural water by logistic regression analysis. METHODS AND RESULTS: Agricultural water was collected from 457 sites including surface water (n=300 sites) and groundwater (n=157 sites) in South Korea from 2018 to 2020. Sanitary indicator bacteria (total coliform, fecal coliform, and Escherichia coli) and food-borne pathogens (pathogenic E. coli, E. coli O157:H7, Salmonella spp., and Listeria monocytogenes) were analyzed. In surface water, the coliform, fecal coliform, and E. coli were 3.27±0.89 log CFU/100 mL, 1.90±1.19 log CFU/100 mL, and 1.39±1.26 log CFU/100 mL, respectively. For groundwater, three kinds of sanitary indicators ranged in the level from 0.09 - 0.57 log CFU/100 mL. Pathogenic E. coli, Salmonella and Listeria monocytogenes were detected from 3%-site, 1.5%- site, and 0.6%-site water samples, respectively. According to the results of correlations between the level of sanitary indicator bacteria and the detection ratio of pathogens by logistic regression analysis, the probability of pathogen detection increased individually by 1.45 and 1.34 times as each total coliform and E. coli concentration increased by 1 log CFU/100mL. The accuracy of the model was 70.4%, and sensitivity and specificity were 81.5% and 51.7%, respectively. CONCLUSION(S): The results indicate the need to manage the microbial risk of agricultural water to enhance the safety of fresh produce. In addition, logistic regression analysis is useful to analyze the correlation between the level of sanitary indicator bacteria and the detection ratio of pathogens in agricultural water.
After preparation of acetone, ethanol, methanol, and water extracts (10 g/300 mL) of dried persimmon (Diospyros kaki cv. Fuyu) flower-buds, total phenolic contents (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (RSA), 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) RSA, reducing power (RP), and tyrosinase inhibitory activity of the extracts were evaluated. The methanol extracts produced the highest TPC (113.39 mg gallic acid equivalents/g), DPPH RSA ($IC_{50}=40.25\;{\mu}g/mL$), ABTS RSA ($IC_{50}=58.17\;{\mu}g/mL$) and RP ($IC_{50}=69.43\;{\mu}g/mL$) activities while the water extracts generated the lowest values. The ethanol extract showed the highest tyrosinase inhibitor activity (88.90%) at a concentration of 1 mg/mL. These results indicated that persimmon flower-buds may be a useful source of natural antioxidants.
Platycodon grandiflorum have been used as a traditional remedy and food source. This study was performed to investigate the immunomodulating effects of Platycodon grandiflorum in mouse, using ex vivo experiments. Six to seven-week old mice were fed ad libitum on a chow diet, and water extract of Platycodon grandiflorum was orally administrated at two different concentractions (50 and 500 mg/kg B.W./day) every other day for four weeks. In ex vivo experiments, the highest proliferation of splenocytes and levels of cytokine (IL-$1{\beta}$, IL-6, TNF-${\alpha}$) production were observed in 500 mg/kg BW/day supplementation group for all three cytokines stimulated by LPS. In conclusion, this study suggests that Platycodon grandiflorum extracts may enhance the immune function by regulating the splenocytes proliferation and cytokine production capacity by activating macrophages in mice.
This study was carried out to make the new products of color moot as a functional food by variation of the color and flavor through addition of gamgyul, Daccus Carota Var. Sativa, Vitis vinifera, soy sauce, and Pimpinella brachycarpa N., respectively. For all the products, the color was observed by Colorimeter and the sensory evaluation was performed to evaluate their effectiveness and usefulness. The A$_w$ (water activity) test of color mooks has performed to examine their water activity individually. The A$_w$ of color moot was higher than that of the white one. The characters of texture of color mook as a function of the amount of the color and the flavor added were measured by a texture analyzer (XT-RA, Texturometer). By the TPA test, the rainbow and orange color mook examined were high in the springiness.
Rhodiola spp. and red ginseng have been used for food and medicinal applications in disease chemoprevention in many Asian countries. Increased oxidative stress by reactive oxygen species (ROS) has been proposed to be a major cause of muscle fatigue. The present study was designed to investigate the protective effects of a fermented hot-water extract mixture from Rhodiola sachalinensis and red ginseng (MFR) on cell damage and the antioxidant enzyme system in $H_2O_2$-induced oxidative stress in skeletal muscle cells. C2C12 myoblasts were treated with various concentrations of NFR (non-fermented Rhodiola sachalinensis extract), FR (fermented hot-water extract from Rhodiola sachalinensis) and MFR for up to 5 days after the standard induction of differentiation, followed by semi-quantitative RT-PCR. MFR treatment dose-dependently protected oxidative damage of C2C12 cells. The treatment with MFR also enhanced mRNA expressions of MyoD, Cu/Zn SOD, Mn-SOD and GPX up to 16%. These results indicate that MFR exerts an anti-oxidative effect through a mechanism (s) that may involve the up-regulation of antioxidant enzymes, which may be important for the cellular redox environment in muscle cells.

The purpose of the present study was to examine the effect of water extracts from Phellinus linteus on lipid composition and antioxidative system in rats fed high fat high cholesterol diet. Rats were divided into four experimental groups which are composed of normal diet group, high fat high cholesterol diet group, high fat high cholesterol diet with 50 mg/kg b.w water extracts from Phellinus linteus supplemented group (PA group), high fat high cholesterol diet with 100 mg/kg b.w water extracts from Phellinus linteus supplemented group (PB group). The serum TG and cholesterol contents of the water extracts from Phellinus linteus supplemented groups (PA and PB groups) were decreased compared to the high fat high cholesterol diet group. The serum HDL-cholesterol contents of the water extracts from Phellinus linteus supplemented groups were increased compared to the high fat high cholesterol diet group. The serum LDL-cholesterol and AI of the water extracts from Phellinus linteus supplemented groups were significantly decreased compared to the high fat high cholesterol diet group. Supplementation of water extracts from Phellinus linteus groups (PA and PB groups) resulted in increased activities of superoxide dismutase. Hepatic glutathione peroxidase (GSH-px) were increased compared to the high fat high cholesterol diet group. TBARS values in liver and plasma were reduced in water extracts from Phellinus linteus supplemented groups. These result suggest that supplementation of water extracts from Phellinus linteus may have a pronounced impact on lipid composition and antioxidative system in the rats fed high fat high cholesterol diet.
Water is an important component in the production of fresh produce. It is mainly used for irrigation and application of pesticides and fertilizers. Several outbreaks cases related to fresh produce have been reported and water has been identified as the most likely source. On the other hand, wildlife has been identified as a possible source of the waterborne pathogens. The purpose of this study was to investigate the effect of wildlife access on irrigation water used in the cultivation of Chinese cabbage in Goesan. The frequency of wild animals access to upstream water source and the contamination level of bacteria such as Escherichia coli and Enterococci of irrigation water used in Chinese cabbage farm was examined. A total of 37 wildlife including the wild bear and water deer were observed in upstream of water source during the cultivation of Chinese cabbage. The result indicated the presence of hygienic indicator bacteria from the upstream where there is no human access. The contamination range of coliforms, E. coli, and Enterococcus spp. Detected in the irrigation water were 2.13~4.32 log MPN / 100 mL, 0.26~2.03 log MPN / 100 mL, and 1.43~3.49 log MPN / 100 mL, respectively. Due to low water temperatures, the contamination levels of coliform bacteria and E. coli in the irrigation water during harvesting time was lower compared to those recorded during transplanting of Chinese cabbage. However, no significant difference was detected in the number of Enterococci during the cultivation of Chinese cabbage. The results indicated the need to manage the microbial risk in irrigation water to enhance safety in cultivation of Chinese cabbage.

The objectives of this study were to determine inhibitory effect on tyrosinase activity and antioxidant activity of the methanolic extract from grape seeds and to investigate relationships between tyrosinase inhibitory activity and antioxidant activity of the extract. The 80% methanol extracts of grape seeds were fractionated subsequently with hexane, chloroform, ethyl acetate and water. The methanolic extract and fractions from grape seeds inhibited tyrosinase activity in a concentration dependent manner. The methanolic extracts showed the highest inhibitory effects on tyrosinase activity. The inhibitory effects of the ethyl acetate fraction from the methanolic extract on tyrosinase activity was higher than other fractions. The ethyl acetate fraction from methanolic extracts showed higher antioxidant activity and contained higher polyphenolic and flavonoid contents compared to other fractions. The correlation coefficients among the polyphenoilc content of methanolic extracts, ABTS radical cation scavenging activity and inhibitory effect of tyrosinase were relatively high. These results suggest that grape seeds may have potential as a depigmentation agent for cosmetics and functional food products.
