Optimal Extraction Conditions of Anti-obesity Lipase Inhibitor from Phellinus linteus and Nutritional Characteristics of the Extracts

  • In an effort to develop novel mushroom-derived anti-obesity nutraceuticals, water and ethanol extracts containing the lipaseinhibitory compound from Phellinus linteus were prepared, and their nutritional components were determined. The optimal conditions for the extraction of P. linteus lipase inhibitor involved the treatment of the fruiting bodies with distilled water at $80^{\circ}C$ for 72 hr and 80% ethanol at $100^{\circ}C$ for 60 hr, respectively. The distilled water extract and ethanol extract contained 10.9% and 6.11% of crude protein, and 0.96% and 15.86% of crude fat, respectively. Additionally, the distilled water extract contained a large quantity of minerals, including 239.5 mg of K, 39.3 mg of Mg, and 39.3 mg of Na. The free amino acid content of the distilled water extracts was also higher than that of the ethanol extracts, and in particular, the distilled water extracts contained 5,139 mg of asparagine, 3,891 mg of tryptophan, 2,598 mg of alanine, and 2,066 mg of serine in 100 g of the distilled water extracts. 100 g of the distilled water and ethanol extracts were found to contain 12.31 g and 8.16 g of malic acid, respectively.

  • In order to establish effective extracting method of ginseng extracts from fresh ginseng, the yield, chemical composition, physical properties and organoleptic quality of the extracts, which are extracted with various concentrations of ethanol, were investigated. The results are as follows : 1. The yield of the extracts was increased with decreasing the concentration of ethanol as solvent. As in case of water as a solvents, the highest yield was achieved when 23.64% of water was used. The yield were 12.3% and 9.05%, when 70% and 90% of ethanol were used, respectively lively. 2. Crude protein content is the highest level and nitrogen·free extracts content is the lowest at the concentration of 50% ethanol. Lipid was increased linerly while ash was decreased as increment of ethanol concentration. 3. Viscosity and residue of the extracts also decreased in accordance with the increament of ethanol concentration and the transmittance value and pH of extract solutions were almost similar except transmittance value of the water extracts. 4. The extracts extracted with 70% ethanol gave the best result of sensory test. The total sensory test score of each extracts (70%, 90%, 50%, 0% and 30%) were 70, 65, 50, 46 and 41, respectively.

결명자 추출물의 추출특성 및 항산화 효과 (Extraction Characteristics and Antioxidative Activity of Cassia tora L. Extracts)

  • 결명자 추출액의 추출 특성과 추출액의 항산화성을 검토하여 결명자의 추출 조건을 결정함으로써 결명자 유효성분의 식품 소재화를 위한 기초자료를 얻고자 하였다. 용매와의 비율을 결정하기 위해 8배, 10배, 12배, 14배의 water를 가하여 추출 수율을 비교한 결과 water와의 비율이 10배일 때 추출 수율이 가장 좋은 것으로 나타났다. 탁도는 water 추출액에서 가장 높았으며, 추출용매의 ethanol 공노가 증가할수록 낮아졌다. 적정산도는 추출용매의, 50% ethanol에서 가장 높았으나, 전체적으로 산도가 매우 낮았다. 유리당 함량은 fructose $0.08{\sim}0.11mg%$, glucose $0.09{\sim}0.32%$, sucrose $0.08{\sim}1.26mg%$로 나타났다. 총 폴리페놀 함량을 나타낸 것으로 50% ethanol 추출액에서 2.03%로 가장 높은 함량을 나타내었으며. 75% ethanol 추출액은 0.02%로 다른 용매에 비해 현저하게 낮은 함량을 나타내었다. 전자공여능은 50% ethanol에서 66.,39%로 나타나 다른 용매에 비해 높은 전자공여 효과를 보여 주었으며, 폴리페놀 함량과 비슷한 경향을 나타내었다. SOD 유사활성 측정 결과는, 50% ethanol 추출액에서 50.56%로 가장 높은 활성을 나타냈었으며, 총 폴리페놀 함량과 전자공여능 측정 결과와 비슷한 경향으로, 50% ethanol 추출액이 다른 두 용매 보다 우수한 것으로 나타났다.

흑삼의 열수 및 에탄올 추출물의 항비만 효과 (Anti-obesity Effects of Water and Ethanol Extracts of Black Ginseng)

  • 본 연구는 구중구포법 과정의 흑삼 1~9포를 대상으로 열수 및 70% 에탄올 추출물의 ginsenosides을 분석한 결과 생리 활성이 높다고 판단한 흑삼 5포와 9포 열수 및 에탄올 추출물을 각각 3T3-L1 지방세포, RAW264.7 대식세포, 지방유래 줄기세포에 처치하여 세포독성, 지질축적(adipogenesis) 및 염증인자 변화를 관찰하였다. 본 연구에서는 열수 추출물보다 에탄올 추출물 흑삼에서 효율적으로 ginsenosides가 추출되었다. 또한 흑삼 에탄올 추출물이 $PPAR{\gamma}$ 및 C/$EBP{\alpha}$ 감소, AMPK의 인산화 증가 등 adipogenesis 관련인자를 억제하였으며, 아울러 염증성 사이토카인의 분비를 억제하였다. 따라서 비만 및 대사성 질환의 예방에도 잠정적으로 이용될 수 있을 것으로 기대되지만 ginsenosides와 비만과의 조절기전 연구 및 항염증성 기전 등에 대한 정확한 기전 규명이 필요하다. 현재 인삼에 함유된 ginsenosides를 이용하여 체지방 감소 등의 기능성식품으로 많이 개발되고 있는 현실을 감안해 볼 때 홍삼 및 흑삼의 제조 기술인 구증구포법의 효율적인 표준화 공정기술, 유효 생리활성 성분의 대량 생산 및 안전성 기술 등이 개발된다면 항비만에 대한 고부가 가치 상품개발이 가능할 것으로 기대된다.

함초의 항산화 및 산화 촉진 효과 (Antioxidant and Pro-oxidant Activities of Hamcho (Salicornia herbacea L.))

  • Hamcho (Salicornia herbacea) extracts were evaluated for total polyphenol content, antioxidant and pro-oxidant activities. The total polyphenol content was 1.81 g and 0.72 g per 100 g of dried sample in water and ethanol extracts respectively. Both water and ethanol extracts of Hamcho significantly exhibited antioxidant activity. The scavenging activity of hydroxyl radical was 13.8~26.2% and 14.2~16.0% in water and ethanol extracts respectively. The inhibitory effect of conjugated diene formation was 24.6~39.1% and 28.4~39.6% in water and ethanol extracts respectively. However, pro-oxidative effect was also observed in the Hamcho extracts. The Hamcho water extract showed the pro-oxidant effect by enhancing the formation of hydroxyl radical and conjugated diene. The ethanol extract of Hamcho induced conjugated diene formation at 0.5 mg/mL but not at 1 mg/mL. The hydroxyl radical formation was not induced by the Hamcho ethanol extract. Taken together, these results show that Hamcho extracts can act as pro-oxidants by generating hydroxyl radical or conjugated diene in addition to their antioxidant properties. Therefore, this study suggests that the physiological properties of Hamcho and its use as food materials should be considered with caution because antioxidant dietary plants such as Hamcho possess possible adverse effects induced by pro-oxidant activity.

추출 조건을 달리한 감잎과 녹차의 물 및 에탄올 추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향 (Effect of Water and Ethanol Extracts of Persimmon Leaf and Green Tea Different Conditions on Lipid Metabolism and Antioxidative Capacity in 12-month-old Rats)

  • This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism. 딱총나무(Sambucus williamsii var. coreana) 추출물의 항산화효과 (Antioxidative Activity of Extracts from Sambucus williamsii var. coreana)

  • 본 연구에서는 딱총나무의 항산화 활성을 조사해 보았다. 딱총나무 잎과 줄기의 물추출물에는 6.6 mg/g과 2.0 mg/g의 phenolic compound가 함유되어 있었다. 추출물의 항산화 활성 실험에서 전자공여능은 딱총나무 잎의 물과 에탄올 추출물 phenolic 200 ${\mu}g/mL$의 함량에서는 99.5%, 89.7%, 줄기의 물과 에탄올 추출물 phenolic 200 ${\mu}g/mL$의 함량에서는 92.2%, 94.3%의 높은 항산화 효과를 나타내었다. 또한 ABTS radical cation decolorization 측정에서도 딱총나무 잎의 물과 에탄올 추출물 phenolic 200 ${\mu}g/mL$의 함량에서는 79.8%, 99.1%, 줄기의 물과 에탄올 추출물 phenolic 200 ${\mu}g/mL$의 함량에서 90.8%, 97.2%의 매우 높은 항산화 효과를 나타내었다. Antioxidant protection factor에서는 딱총나무 잎의 물과 에탄올 추출물 phenolic 200 ${\mu}g/mL$의 함량에서는 1.1과 1.1 PF, 줄기의 물과 에탄올 추출물 phenolic 200 ${\mu}g/mL$의 함량에서는 1.4와 1.0 PF의 항산화효과를 나타내었다. Thiobarbituric acid reaction substance 측정에서도 딱총나무 잎의 물과 에탄올 추출물 phenolic 200 ${\mu}g/mL$의 함량에서는 88.7%, 98.1%, 줄기의 물과 에탄올 추출물 phenolic 200 ${\mu}g/mL$의 함량에서는 93.6%, 90.6%로 positive control인 BHT 및 vitamin C보다 우수한 항산화 효과를 나타내었다. 본 연구의 결과로 딱총나무의 항산화 활성을 이용한 천연 기능성 식품소재로서의 활용이 가능할 것으로 판단되었다.

층꽃나무(Caryopteris incana Miq.) 추출물의 생리활성 (Biological activities of extracts from Caryopteris incana Miq.)

  • 본 연구에서 층꽃나무(Caryopteris incana Miq.)는 항산화, 피부미백 및 주름개선 활성을 측정하는 데에 사용되었다. 층꽃나무의 생리활성은 고형분 함량보다 첨가된 total phenolic compounds의 농도에 의해 영향을 받는 것으로 확인되어 이후 모든 실험은 층꽃나무로부터 추출한 물과 80 % ethanol 추출물의 total phenolic compounds 함량을 조절하여 실시하였다. 층꽃나무 추출물의 항산화 활성은 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azinobis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS), antioxidant protection factor (PF), Thiobarbituric acid reactive substances (TBARs)를 측정하였다. $200{\mu}g/mL$의 농도에서 물과 ethanol 추출물로 DPPH free radical 소거능을 측정한 결과 각각 84, 92 %의 활성을 나타내었고, ABTS radical 소거능은 물과 ethanol 추출물 둘 다 약 99 %의 활성을 나타내었다. PF를 측정한 결과 물과 ethanol 추출물이 각각 1.56, 1.67 PF 값을 나타내었고, TBARs는 각각 62, 82 %의 활성을 나타내었다. 주름개선 및 피부미백 활성을 측정하였을 때, $200{\mu}g/mL$의 농도에서 80 % ethanol 추출물이 물 추출물보다 눈에 띄게 높은 효과를 나타내었다. 주름개선과 연관된 elastase와 collagenase 저해활성은 ethanol 추출물에서 각각 58, 89 %로 측정되었다. 피부미백과 연관된 tyrosinase의 저해활성은 ethanol 추출물에서 13 %로 측정되었고, 수렴효과는 50 %로 측정되었다. 이러한 결과로 보아, 층꽃나무 추출물은 항산화 활성, 주름개선 및 미백효과가 우수하므로 기능성 화장품 원료의 새로운 자원이 될 것으로 기대되었다.

화피, 석류피, 염부수백피 추출물의 항산화 및 항균 효과 (Antioxidative and Antimicrobial Activities of Water- and Ethanol-Extracts from Betula platyphylla var. japonica, Punica granatum and Rhus javanica)

  • Objectives : This study was conducted to measure the antioxidative and antimicrobial activities of water- and ethanol-extracts from Betula platyphylla var. japonica, Punica granatum and Rhus javanica against various species of anaerobic bacteria. Methods : In order to evaluate the antioxidative and antimicrobial activities of water- and ethanol-extracts, DPPH radical scavenging and superoxide dismutase (SOD)-like activities were measured in various species such as Staphylococcus epidermidis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans and Propionibacterium acnes that induce skin inflammation. Also the total amount of phenol in each water- and ethanol-extract was measured to identify its role in the antioxidative and antimicrobial activities of water- and ethanol-extracts from Betula platyphylla var. japonica, Punica granatum and Rhus javanica. The antibacterial activity of the extracts was measured by confirming the lowest concentrations in disk diffusion and minimum inhibitory concentration (MIC) tests. Results : It was observed that RJE (Rhus javanica-EtOH extracts) show the highest content of total phenol. In addition, for RJE the total phenol content was higher from samples taken from domestic sources than from samples taken from foreign sources. DPPH radical scavenging activity was increased by treatment with PGE (Punica granatum-EtOH extracts), RJE and RJW (Rhus javanica-water extracts). It was observed that SOD-like activity was highest in the treatment with PGE. All of the extracts showed antimicrobial activity on S. epidermidis, S. aureus, P. acnes and E. coli, including those from Punica granatum, and it was noted that the activity was higher with RJE than with RJW. Conclusions : These results provide evidence that ethanol extracts of Punica granatum and Rhus javanica may have a beneficial role as antioxidants and antibiotics. Extracts from domestic samples of Betula platyphylla var japonica appeared to have a greater efficacy than extracts from foreign samples.

산수유, 황기, 감초 추출물의 생리활성 (Biological Activities of Extracts from Corni fructus, Astragalus membranaceus and Glycyrrhiza uralensis)

  • Objectives : This study was conducted to evaluate the antioxidative and anticancer activity of the water and ethanol extracts from medicinal herbs. Methods : Three kinds of medicinal herbs(Corni fructus, Astragalus membranaceus and Glycyrrhiza uralensis) were extracted with distilled water and 70% ethanol, and the extracts were tested for their antioxidative and anticancer activities. Results : The highest polyphenol contents of the water and ethanol extracts from medicinal herbs were 342.14 mg and 435.62 mg per 100 g of Cornus officinalis, respectively. The highest electron donating abilities (EDA) of the water and ethanol extracts from Glycyrrhiza uralensis were 88% and 91% at 1,000 ppm, respectively. The water and ethanol extracts from Astragalus membranaceus had the highest nitrite scavenging abilities (NSA) at 1,000 ppm. The highest anticancer activity of the extracts were from Glycyrrhiza uralensis against both of MDA and A549 cells. Conclusions : These results suggest that the medicinal herbs can be used as natural antioxidant to prevent oxidative damage in normal cells probably because of their antioxidant characteristics.
