Optimization study of the hot water extraction for enhancing antioxidative activity from Auricularia auricula was performed by Taguchi approach using orthogonal matrix $L_9(3^4)$ method. The correlation between DPPH radical scavenging activity and the components of samples extracted from different extraction conditions were also analyzed. The correlation coefficient between DPPH radical scavenging activity and melanin content of A. auricula were 0.93, indicating 'good correlation'. The optimum extraction conditions was obtained at the extraction time of 1 hr. temperature of $85^{\circ}C$, solid: water ratio of 1: 40(w/v) and frequency of 2 times. Under these conditions, values of maximum DPPH free radical scavenging activity and melanin contents of A. auricula were $67.21{\pm}2.17$ and $52.94{\pm}2.10 mg/g$, respectively. Melanin content of 1.6 times and DPPH free radical scavenging effect of 130% were enhanced by optimization.
In this work, the anti-aging skin effects of bacteriochlorophyll a isolated from Rhodobacter sphaeroides are first reported, with notably low cytotoxicity in the range of 1% to 14% in adding 0.00078 (% (w/w)) of the extracts, compared with the normal growth of both human dermal fibroblast and keratinocyte cells without any treatment as a control. The highest production of procollagen from human fibroblast cells (CCD-986sk) was observed as 221.7 ng/ml with 0.001 (% (w/w)) of bacteriochlorophyll a, whereas 150 and 200 ng/ml of procollagen production resulted from addition of 0.001 (% (w/w)) of the photosynthetic bacteria. The bacteriochlorophyll-a-induced TNF-α production increased to 63.8%, which was lower secretion from HaCaT cells than that from addition of 0.00005 (% (w/w)) of bacteriochlorophyll a. Additionally, bacteriochlorophyll a upregulated the expression of genes related to skin anti-aging (i.e., keratin 10, involucrin, transglutaminase-1, and MMPs), by up to 4-15 times those of the control. However, crude extracts from R. sphaeroides did not enhance the expression level of these genes. Bacteriochlorophyll a showed higher antioxidant activity of 63.8% in DPPH free radical scavenging than those of water, ethanol, and 70% ethanol extracts (14.0%, 57.2%, and 12.6%, respectively). It was also shown that the high antioxidant activity could be attributed to the skin anti-aging effect of bacteriochlorophyll a, although R. sphaeroides itself would not exhibit significant anti-aging activities.
The fringe tree, Chionanthus retusus (Oleaceae) has been used as a natural remedy that helps a paralytic stroke, dementia, phlegm and malaria. This study was conducted to investigate the antioxidant and anti-aging activities of water extracts obtained from the flesh of C. retusus by reflux extraction at room temperature and $80^{\circ}C$, and pressure heating extraction at $110^{\circ}C$, for an increment of their practical use. The highest extraction yield from the flesh of C. retusus was 67.94% extract by pressure heating extraction at $110^{\circ}C$. The content of total polyphenol compounds was the highest by 72.71 mg/g in $80^{\circ}C$ extract by reflux extraction. The highest content of total flavonoid compounds was 7.60 mg/g at $110^{\circ}C$ extract. The results of soluble protein and reducing sugar contents showed the highest content of 4.93 mg/g and 46.77 mg/g in $110^{\circ}C$ extract, respectively. In the analysis of DPPH radical scavenging activity, all extracts showed an excellent effect of 92% in 1.0 mg/mL concentration. The highest effect of ABTS radical scavenging activity was 88.67% at $110^{\circ}C$ extraction in 1.0 mg/mL concentration. Nitrite scavenging ability was the highest as 66.16% at $110^{\circ}C$ extract at pH 1.2 and 1.0 mg/mL concentration. The results of SOD-like activity and xanthine oxidase inhibition activities showed the highest effect of 14.95% and 59.45% at $110^{\circ}C$ extract in 1.0 mg/mL, respectively. Tyrosinase inhibition activity for whitening effect was the highest of 35.31% at $85^{\circ}C$ in 1.0 mg/mL. In the analysis of anti-aging effect, collagenase and elastase inhibition activity showed the highest effects of 37.78% at $110^{\circ}C$ and 20.39% at $80^{\circ}C$, respectively. All results of antioxidant activities and anti-aging effects increased along with the concentration increases. These results indicated that the extracts extracted from the flesh of C. retusus at $80^{\circ}C$ and $110^{\circ}C$ have a large amount of useful ingredients, an excellent antioxidant activity, like as DPPH and ABTS radical scavenging activity, and anti-aging effects to develop functional products than those of $25^{\circ}C$.
Cereal grains are the most important food source for humans. As the global population continues to grow exponentially, the need for the enhanced yield and minimal loss of agricultural crops, mainly cereal grains, is increasing. In general, harvested grains are stored for specific time periods to guarantee their continuous supply throughout the year. During storage, economic losses due to reduction in quality and quantity of grains can become very significant. Grain loss is usually the result of its deterioration due to fungal contamination that can occur from preharvest to postharvest stages. The deleterious fungi can be classified based on predominance at different stages of crop growth and harvest that are affected by environmental factors such as water activity ($a_w$) and eco-physiological requirements. These fungi include species such as those belonging to the genera Aspergillus and Penicillium that can produce mycotoxins harmful to animals and humans. The grain type and condition, environment, and biological factors can also influence the occurrence and predominance of mycotoxigenic fungi in stored grains. The main environmental factors influencing grain fungi and mycotoxins are temperature and $a_w$. This review discusses the effects of temperature and $a_w$ on fungal growth and mycotoxin production in stored grains. The focus is on the occurrence and optimum and minimum growth requirements for grain fungi and mycotoxin production. The environmental influence on aflatoxin production and hypothesized mechanisms of its molecular suppression in response to environmental changes are also discussed. In addition, the use of controlled or modified atmosphere as an environmentally safe alternative to harmful agricultural chemicals is discussed and recommended future research issues are highlighted.

Chitosan, a cationic polysaccharide, has been widely used as a dietary supplement and in a variety of pharmacological and biomedical applications. The antifungal activity and mechanism of action of low molecular weight water-soluble chitosan (LMWS-chitosan) were studied in fungal cells and vesicles containing various compositions of fungal lipids. LMWS-chitosan showed strong antifungal activity against various pathogenic yeasts and hyphae-forming fungi but no hemolytic activity or cytotoxicity against mammalian cells. The degree of calcein leakage was assessed on the basis of lipid composition (PC/CH; 10:1, w/w). Our result showing that LMWS-chitosan interacts with liposomes demonstrated that chitosan induces leakage from zwitterionic lipid vesicles. Confocal microscopy revealed that LMWS-chitosan was located in the plasma membrane. Finally, scanning electron microscopy revealed that LMWS-chitosan causes significant morphological changes on fungal surfaces. Its potent antibiotic activity suggests that LMWS-chitosan is an excellent candidate as a lead compound for the development of novel anti-infective agents.
Carbon tetrachloride ($CCl_4$) has been used to treat a variety of parasitic infection in both large and small animals, including Fasciola hepatica. Recently, we can easily contest with carbon tetrachloride in air, food, water, rain and Industry area etc. and it is using in order to induce liver injury in laboratory. In this study. we investigated activitis of LDH, ALP, AST and ALT and amount of cholesterol, triacylglycerol, glucose and BUN in mice serum exposed to $CCl_4.$ The mice divided Into a Naive control(A), corn oil control(B) and experimental group(C, D). Naive control group(A) was given feed and water only. Corn oil control group(B) was given corn oil 1ml /100g of body weight(B.W). Experimental group(C) was given carbon tetrachloride 1ml /100g B.W. ($CCl_4$: corn oil=1:20(V /V) ), Experimental group(D) was given carbon teterachloride 1ml /100g B.W. ($CCl_4$: corn oil=1:10(V /V) ). The results obtained were summarized as follows : 1) The body weight was declined after the l0th day In mice exposed to carbon tetra-chloride. 2) The total protein level in serum was significantly in mice exposed to carbon tetra-chloride($P{\leq}0.05$). The albumin and A /G ratio was decresed significantly in mice exposed to carbon tetrachloride($P{\leq}0.01$) 3) All of the activity of LDH, AST, ALT and ALP in mice serum exposed to carbon tetra chloride inclosed significantly activity of LDH ($P{\leq}0.05$), inclosed significantly activity of AST($P{\leq}0.05, \;p {\leq}0.01$), inclosed significantly activityof ALT 3nd ALP($P {\leq}0.05,\;p {\leq}0.01$). 4) The amount of cholesterol and triacylglycerol, lipid metabolite products in serum was inclosed in case of cholesterol but did not change in case of triacylglycerol.

This study is to evaluate moisturizing effect and durability of UV A/B blocking activity with multiple (W/O/W) emulsion system. Most of the sun protective products come to be hot issue having both high SPF and long-lasting activity as using special products when is going out, mountain climbing and sports. Also, many consumers prefer the products which have the excellent waterproofing activity of sun care cosmetics as well as the non-sticky feeling that carried out the study of the sensorial science and texture preference. Therefore, development of the specific formulation using this multiple (W/O/W) emulsion technology, it has O/W type hydro skin feel having soft and moist texture when it is treated on the skin. Finally, this formulation is instantly changed to W/O type feel after adsorbed into the skin. The purpose of this study is to get high SPF lasting effect having high water resistance tactivity with high functional multiple (W/O/W) emulsion cream. We used major ingredients, UV-B absorbers were selected with ethylhexyl methoxycinnamate, isoamyl-p-methoxycinnamate, ethylhexylsalicylate, and octocrylene, UV-A absorbers were selected with butylmethoxydibenzoylmethane, bis-ethylhexyloxyphenol methoxy phenyltriazine. SPF effect of O/W type cream was 34.1. SPF effect of W/O/W type cream was 40.6 (increased about 19%). Water resistance effect after 4 hours, SPF effect of O/W type cream was 3.6 (quickly drop down). SPF effect of W/O/W type cream having 81.0 % waterproofing effect was 32.7 (decreased about SPF 7.9). Moisturizing effect of O/W cream at first was superior comparing multiple emulsion. But after 3 hours quickly was drop-down. Moisturizing effect of multiple emulsion was high comparing O/W type and other sun block creams after 4 hours was constantly maintaining water-content.
This study was conducted to evaluate the estrogen activity of silkworm (Bombyx mori) pupa extracts and their fractions. Powdered samples of freeze-dried silkworm pupa were extracted at room temperature (RT), $40^{\circ}C$, $60^{\circ}C$, $80^{\circ}C$, and $100^{\circ}C$ in water (D.W), chloroform, ethyl acetate, and methanol for 6h and then filtered (0.45 um). The extracts were then freeze-dried. The estrogenic activity of these extracts was then investigated by competition binding assays using estrogen receptor ${\alpha}\;(ER{\alpha})$ and $ER{\beta}$, and by evaluating their effects on the proliferation of the human breast cancer cell line, MCF-7. Among the extracts evaluated, water extracts prepared at RT showed the highest binding affinity to $ER{\alpha}$ ($IC_{50}$, 1.76 ug/ml) and $ER{\beta}$ ($IC_{50}$, 0.07 ug/ml). In addition, MCF-7 cells that were treated with 62.5 ug/ml of the RT extract showed the greatest increase in proliferation (2-fold; 1291.79%) when compared to control cells (659.82%). Next, the water extract that was prepared at RT (sample 1) was dissolved in D.W. and further fractionated using a Dowex 50W - 8X ($H^+$) column. The flow-through and wash were then pooled together and freeze-dried (sample 2). The bound materials were then eluted with 20 mM NaCl, after which they were applied to a Dowex 1X2 - 200 ($Cl^-$) column and washed with D.W. to remove the sodium ions. The eluants were then freeze-dried (sample 3). Of these fractions, sample 2 showed the highest binding affinity to ER{\alpha} ($IC_{50}$, 1.44 ug/ml) and $ER{\beta}$ ($IC_{50}$, 1.18 ug/ml). In addition, MCF-7 cells that were treated with sample 2 (15.6 ug/ml) showed the largest increase in growth (1159.39%) when compared to control cells (525.26%). Taken together, these results suggest that the fraction of the RT water extract of silkworm pupa referred to as sample 2 may be useful as a phytoestrogen.
This research was carried out to determine the differences of physiological activites between Grifola frondosa of log cultivation(LC) and Grifola frondosa of bottle cultivation(BC). Total flavonoids content, total phenolics content, electron donating ability(EDA), nitrite-scavenging ability(NSA), SOD-like activity and inhibitory effect of Xanthine oxidase were examined. The highest value of total flavonoid content is $5.96{\pm}0.81mg/g$ in water extract from Grifola frondosa of log cultivation at $40^{\circ}C$ (LC-W40) but, one of total phenolics compound is $44.53{\pm}0.89mg/g$ in water extract from Grifola frondosa of bottle cultivation at $40^{\circ}{\cdots}$ (BC-W40). The EDA using DPPH of BC-W40 extract showed the highest value of $97.14{\pm}0.71%$. Nitrite-scavenging ability was $62.55^{\circ}{\ae}0.36%$ in extract from Grifola frondosa of BC-W40 at pH 1.2. The value was SOD-like activity showed the highest value of $18.95^{\circ}{\ae}1.39%$ in extract from LC-W40. Xanthine oxidase inhibitory activity was the highest value of $54.31{\pm}0.40%$ in extract from Grifola frondosa BC-W40, and dependent on concentrations. These results showed that a the antioxidant effects of Grifola frondosa is excellent. However, physiological activities of Grifola frondosa were not depend on caltivation method regulary, and were different according to kind of solvents, concentraitions and physialogical factors examined such as EDA, SOD-like activity and NSA.
The effect of water activity ($a_w,\;0.9{\sim}0.995$) and temperature ($18{\sim}30^{\circ}$C) on in vitro growth and interactions between ochratoxin-producing Aspergillus ochraceus and six other fungi (Alternaria alternata, Aspergillus candidus, A. flavus, A. niger, Eurotium amstelodami, E. rubrum) isolated from maize grain were investigated. A. ochraceus and each six other species were paired and their interactions given a numerical score to obtain an index of dominance ($I_D$) for each species. Generally A. ochraceus was very competitive and dominant against other fungi. It was, however, dominanted by Alternaria alternata and A. niger at high $a_w\;(0.995\;a_w)$, and mutually antagonistic when paired with E. amstelodami and E. rubrum at low $a_w\;(0.9\;a_w)$. The growth rates of each species were also calculated under the same range of environmental conditions. They were markedly influenced by aw and temperature. At high temperature ($30^{\circ}C$), A. ochraceus grew most rapidly under slightly drier conditions ($0.95\;a_w$), while A. alternata, A. flavus and A. niger did at high water availability level ($0.995\;a_w$). At $18^{\circ}C\;and\;25^{\circ}C$, and high $a_w$ level ($0.995\;a_w$), A. alternata grew fastest, while A. candidus, E. amstelodami and E. rubrum grew very slowly. Using Biolog plates the effect of $a_w$ and temperature on utilization patterns of carbon sources in maize was evaluated. The niche overlap index (NOI) relative to A. ochraceus was determined and compared with that of each interacting species. Under high water available condition ($0.995\;a_w$). the NOI of A. ochraceus was often >0.9, indicative of the coexistence with other interacting species. However, against E. amstelodami and E. rubrum at $18^{\circ}C$, the species had NOI <0.8, indicative of occupation of different niches. At low $a_w\;(0.95\;a_w)$, NOI for A. ochraceus was <0.8 when paired with A. alternata and A. niger also suggested the occupation of different niches.
