Biological activity of supercritical extraction residue 60% ethanolic extracts from Ulmus davidiana (느릅나무 초임계 추출박 60% 주정추출물의 생리활성)

  • Ulmus davidiana supercritical fluid residue EtOH extracts(USCFR) and ethyl acetate solvent fraction (USCFREA) of supercritical extraction foil were investigated in order to examine the recycling of supercritical extraction foil in the process of studying Ulmus davidiana branch supercritical extract. Experiments were performed for the determination of total phenol content. The $IC_{50}$ value(ppm) of DPPH radical scavenging activity and ABTS radical scavenging activity was $7.42{\pm}0.09$, $7.50{\pm}0.05$, $22.94{\pm}0.09$, $6.43{\pm}0.10$, and USCFREA, respectively, as compared with the positive control (vitamin C) with values $17.80{\pm}0.14$ and $5.34{\pm}0.06$, respectively. The antioxidative activities of USCFR and USCFREA were confirmed to be superior to the positive control group. In anti-allergic activity studies, both USCFR and USCFREA showed concentration-dependentanti-allergic activity, and USCFREA showed strong anti-allergic activity even at very low concentrations. Thetotal phenolic contents (ugEG, ugGA; ppm) of USCFR were $134.17{\pm}0.13$, $132.02{\pm}0.24$ and USCFREA were $154.77{\pm}1.05$ and $153.18{\pm}1.10$, respectively. Based on the above results and strong antioxidant activity, USCFR and USCFREA hold the potential to be considered as basic research materials for the development of therapeutic supplements based medicines or functional cosmetics related to chronic inflammatory skin immunity diseases.

Antimicrobial Activity and Antioxidant Effect of Curcuma longa, Curcuma aromatica and Curcuma zedoaria (국내산 울금, 강황 및 보라울금의 항균활성과 항산화 효과)

  • We investigated the antimicrobial and antioxidant activities. and total polyphenol contents of Curcuma longa. Curcuma aromatica. and Curcuma zedoaria. In antimicrobial activity assays of solvent extracts, ethanol extract showed the highest activity compared with other extracts. The antimicrobial activity of ethanol extract of C. longa showed higher than those of C. aromatica, and C. zedoaria. Antimicrobial substance in ethanol extract of C. longa was maintained after heating at $100^{\circ}C$ for 30 min and not affected by changes in pH. DPPH free radical scavenging activity of solvent extracts were high in the order of ethanol, ethylacetate, ether, water, and hexane fractions, C. longa showed greater DPPH free radical scavenging activity than those of the C. aromatica, and C. zedoaria. Total polyphenol content of C. longa and C. aromatica exhibited were similar levels, whereas C. zedoaria was slightly lower concentration.

Antioxidant Activity and Inhibitory Effect on Oxidative DNA Damage of Ethyl Acetate Fractions Extracted from Cone of Red Pine (Pinus densiflora) (솔방울 에틸아세테이트 분획물의 항산화 및 산화적 DNA 손상 억제 활성)

  • Antioxidant activity and inhibitory effect on oxidative DNA damage of ethyl acetate fractions extracted from Cone of Red Pine (Pinus densiflora) were investigated to find utilization of Cone, by-product of Red Pine, thrown out after berry shatter, as a new natural plant resource. Cone from P. densiflora was extracted with methanol (MeOH) and separated to petroleum ether, ethyl acetate and water fraction. Among them, ethyl acetate fraction was used. The antioxidant activity was conducted by the 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical, 2, 2'-Azino-bis (3-ethylbenzothiazoline-6 sulfonic acid) diammonium salt (ABTS) radical scavenging assay, Fe2+ chelating assay and reducing power assay. The inhibitory effect on oxidative DNA damage was determined by DNA cleavage assay using φX-174 RF I plasmid. The results of DPPH and ABTS radical scavenging activity at 200 ㎍/㎖ of extracts were 86.50% and 95.80% respectively, which were similar figures compared with L-ascorbic acid as control. Fe2+ chelating activity was 77.96% and reducing power was 0.77 at 200 ㎍/㎖. Total phenolic component was 27.29±0.3 ㎎/g and Vitamin C content was 1.84±0.1 ㎎/g. Also ethyl acetate fraction from Cone has inhibitory effect, using φX-174 RF I plasmid on DNA cleavage assay. In conclusion, Cone, by-product of P. densiflora, showed high antioxidant activity and inhibitory effect on oxidative DNA damage. Therefore this study suggests Cone, useless by-product, can be developed as a new natural plant resource with lots of utilization such as an effective antioxidant, natural medicine, food, cosmetics and so on.

Effect of the Concentration of Nutrient Solution on Early Yield and Fruit Quality of Tomato(Lycopersicon esculentum Mill.) in Substrate Culture (고형배지경에서 배양액농도가 토마토의 초기수량 및 품질에 미치는 영향)

  • This study was carried out to investigate the effect of the concentration of nutrient solution on growth of tomato(Lrcopersicon esculentum Mill. cv. seokwang) in substrate culture. The substrates used in the experiment were perlite, vermiculite, and peatmoss. Tomato plants were treated with different concentrations of nutrient solution, viz. 0.5, 1.0, 2.0, 3.0, and 5.0mS/cm at seedling stage and transferred to different treatments, 1.0, 2.0, and 3.0mS/cm after transplanting in each substrate. Total fruit number, total yield, marketable fruit number, and marketable yield were much higher at 2.0-3.0mS/cm than at 1.0mS/cm in all three substrate culture. The percentage of malformed fruit was in order of peatmoss>perlite> vermiculite and Vitamin C content was vermiculite> perlite> peatmoss. In all three substrate culture, high marketable yield was shown when tomato plants grew with concentration of 2.0-5.0mS/cm at seedling stage and 2.0-3.0mS/cm after transplanting. The concentrations of nutrient solution after transplanting as well as at seedling stage had a great influence on total yield, marketable yield, and soluble solids. However total and marketable fruit number were considerably affected by the concentrations of nutrient solution after transplanting.

The Study on the Quality of Pre-Processed Vegetables in School and Institutional Food-Service (단체급식에서 사용되는 전처리 농산물의 품질 특성 분석)

  • This study was performed to investigate the quality of pre-processed vegetables used in school and institutional food-services. Pre-processed food materials (carrot, potato, and cabbage) frequently used in food-service were collected from 14 various processing company sources. The sensory and physico-chemical qualities of the pre-processed food materials were determined using sensory and instrumental analysis. For the physico-chemical analysis of the food materials, pH, total acidity, hardness, Hunter colorimeter value, reducing sugar and vitamin C content were determined. For the sensory quality evaluation, 15 panelist were trained and consensus was reached on the quality standards of the preprocessed materials (carrot, potato, and cabbage). Finally, appearance, color, texture, off-odor/taste, and overall quality were determined. In the physico-chemical analysis, there were no significant differences among samples collected from various processing companies. In sensory quality evaluations, the color quality of pre-processed potato was lower than that of other materials. From the coefficient correlations and partial least squares regression analysis between sensory and instrumental data, pH, total acidity, colorimeter values, and hardness were considered important components in assessing the quality of pre-processed vegetables.

Characteriatics of Single Cell Suspension of Garlic, Red Pepper and Cucumber Prepared by Protopectin Hydrolytic Enzymes (펙틴분해효소를 이용한 마늘, 고추와 오이의 단세포화물의 제조)

  • Protopectinase (PPase) from Bacillus subtilis was used to investigate enzymatic maceration of vegetable tissues. Optimum concentration and pH of PPase were 0.75, 0.75, and 0.5%, and 5.0, 8.0, 7.0 for red pepper, garlic, and cucumber, respectively. Optimum shaking-rate, reaction time, and temperature of PPase were 250 rpm, 150 min, and $37^{\circ}C$, respectively. Yields of mechanically macerated red pepper, garlic, and cucumber were 45.8, 47.5, and 82.1%, whereas those treated with PPase were 81.8, 84, and 98%. Over 40% Vitamin C, the most unstable component during mechanical maceration, remained intact for 12 days after enzymatic treatment. Color differences $({\Delta}E)$ of mechanically macerated red pepper, garlic, and cucumber were 1.16, 2.86, and 3.27, whereas those of PPase-treated ones were 2.87, 7.68, and 5.22 after heat treatment at $100^{\circ}C$ for 20 min. Capsaicin content of mechanically macerated red pepper was 0.4 mg/100 g, whereas that treated with PPase was 1.32 mg/100 g. Viscosity of PPase-treated vegetable decreased slowly with increasing storage period, whereas that of mechanically macerated vegetable sharply decreased. These results indicate PPase treatment of vegetable could be better choice for preparation of high-values and functionally processed food and for extending preservation period.

Pasteurization Efficiency and Physico-chemical Changes of Soymilk HTST Pasteurized Using Microwaves (두유의 마이크로파 고온단시간 살균시 살균효과 및 이화학적 성분 변화)

  • This work was to determine the microbial and physico-chemical changes of HTST-pasteurized soymilk using microwave energy. Soymilk was HTST pasteurized$(at\;90^{\circ}C\;for\;20\;sec)$ by three methods: by heating in a stainless steel tube immersed in a hot water bath(MP0), by heating in a microwave cavity to a defiled temperature and then holding in a hot water bath(MP1), and by both heating and holding in a microwave cavity(MP2). The microbial quality based on the total plate count was in the order of MP0, MP2 and MP1. The three samples pasteurized by different methods showed the similar microbial quality with respect to the coliform count, psychrotrophic bacterial count and phosphatase activity. The destruction of trypsin inhibitor was in the order of MP0, MP1 and MP2. There were no significant differences in pH, titratable acidity, viscosity and vitamin $B_2$ content before and after pasteurization and among the different pasteurization methods. The similar or higher quality retention of the MP1 or MP2 supports the possibility of using microwave energy for the HTST pasteurization of soymilk and other fluid food products.

Development of Functional Modified Atmosphere Film for Winter Date (중국산 대추(Winter date)의 신선도 유지를 위한 기능성 MA필름 개발)

  • In order to study the storage stability of winter date (Zizyphus jujuba Miller) with functional MA film storage, linear low density polyethylene $(LLDPE,\;30{\mu}m\;and\;60{\mu}m)$ film packaging and non-packaging were compared $30{\mu}m$ LLDPE packaging, $60{\mu}m$ LLDPE packaging and non-packaging samples were stored at $0^{\circ}C,\;8^{\circ}C$ (low temperature) and $25^{\circ}C$(room temperature) for 10 weeks. Weight loss of $30{\mu}m$ LLDPE packaging, $60{\mu}m$ LLDPE packaging and non-packaging in room temperature were decreased by 3.6% 0.4% and 36.1% respectively. Titratable acidity showed the trend of similarity in all treatment during the storage conditions, and especially LLDPE film packaging showed higher than non-packaging in the last storage. Soluble solids showed an increase in non-packaging, a decrease in LDPE film packaging highly. Vitamin C content showed the trend of decrease in all treatments, but non-packed jujubes sustained higher than LLDPE film packaging.

Pink Pigmented Facultative Methylotrophic Bacteria(PPFMs): Introduction to Current Concepts (분홍색 색소를 형성하는 methylotrophic acteria(PPFMs): 최근 경향소개)

  • The non infecting, plant associated bacteria have attracted increased attention for stimulating plant growth and as environmental friendly plant protecting agents. Pink-pigmented facultatively methylotrophic bacteria (PPFMs), classified as Methylobacterium spp., are persistent colonizers of plant leaf surfaces. As the leaves of most or all plants harbor PPFMs that utilize leaf methanol as their sole source of carbon and energy, which is a specific attribute of the genus Methylobacterium. Although they are not well known, these bacteria are co-evolved, interacting partners in plant metabolism. This claim is supported, for example, by the following observations: (1) PPFMs are seed-transmitted, (2) PPFMs are frequently found in putatively axenic cell cultures, (3) Low numbers of seed-borne PPFMs correlate with low germinability, (4) Plants with reduced numbers of PPFM show elevated shoot/root ratios, (5) Foliar application of PPFMs to soybean during pod fill enhances seed set and yield, (6) Liverwort tissue in culture requires PPFM-produced vitamin B12 for growth, (7) treated plants to suppress or decrease disease incidence of sheath blight caused by Rhizoctonia solani in rice, and (8) the PPFM inoculation induced number of stomata, chlorophyll concentration and malic acid content, they led to increased photosynthetic activity. Methylobacterium spp. are bacterial symbionts of plants, shown previously to participate in plant metabolism by consuming plant waste products and producing metabolites useful to the plant. There are reports that inform about the beneficial interactions between this group of bacteria and plants. Screening of such kind of bacteria having immense plant growth promoting activities like nitrogen fixation, phytohormone production, alleviating water stress to the plants can be successfully isolated and characterized and integration of such kind of organism in crop production will lead to increased productivity.

Effect of Semi-dry Red Pepper Powder on Quality of Kimchi (김치의 품질에 미치는 반건조 고춧가루의 영향)

  • We compared two kinds of red peppers, semi-dry red popper and hot air dry red pepper, and made two kinds of Kimchies with them for this study. The semi-dry red pepper is better to maintain natural color of raw red pepper, to keep hygenity and to protect its loss of the nutrition, than the hot air dry red pepper. The results are as follows: The content of vitamin C and ASTA color value in semi-dry red pepper powder were much higher than those in hot air dry red pepper The cell numbers of total bacteria in semi-dry red pepper powder were much lower than those in hot air dry red pepper. The changes in quality of the two Kimchies were evaluated by pH, acidity, total microbes, lactic acid bacteria cells count and sensory. The patterns of changes in pH, acidity, total microbes and lactic acid bacteria cells count of two kinds of samples were the same. But in case of sensory evaluation, Kimchi made with semi-dry red powder showed best sensory scores than of all in overall acceptability.
