A Comparison of Natural (D-α-tocopherol) and Synthetic (DL-α-tocopherol Acetate) Vitamin E Supplementation on the Growth Performance, Meat Quality and Oxidative Status of Broilers

  • The present study was conducted to compare the supplementation of natural (D-${\alpha}$-tocopherol) and synthetic (DL-${\alpha}$-tocopherol acetate) vitamin E on the growth performance, meat quality, muscular antioxidant capacity and genes expression related to oxidative status of broilers. A total of 144 1 day-old Arbor Acres broiler chicks were randomly allocated into 3 groups with 6 replicates of 8 birds each. Birds were given a basal diet (control group), and basal diet supplemented with either 20 IU D-${\alpha}$-tocopherol or DL-${\alpha}$-tocopherol acetate for 42 days, respectively. The results indicated that treatments did not alter growth performance of broilers (p>0.05). Compared with the control group, concentration of ${\alpha}$-tocopherol in the breast muscle was increased by the supplementation of vitamin E (p<0.05). In the thigh, ${\alpha}$-tocopherol content was also enhanced by vitamin E inclusion, and this effect was more pronounced in the natural vitamin E group (p<0.05). Vitamin E supplementation increased the redness of breast (p<0.05). In the contrast, the inclusion of synthetic vitamin E decreased lightness of thigh (p<0.05). Dietary vitamin E inclusion reduced drip loss at 24 h of thigh muscle (p<0.05), and this effect was maintained for drip loss at 48 h in the natural vitamin E group (p<0.05). Broilers given diet supplemented with vitamin E showed decreased malondialdehyde (MDA) content in the breast (p<0.05). Additionally, natural rather than synthetic vitamin E reduced MDA accumulation in the thigh (p<0.05). Neither natural nor synthetic vitamin E supplementation altered muscular mRNA abundance of genes related to oxidative stress (p>0.05). It was concluded that vitamin E supplementation, especially the natural vitamin E, can enhance the retention of muscular ${\alpha}$-tocopherol, improve meat quality and muscular antioxidant capacity of broilers.

Simultaneous Determination of Vitamin A and E in Infant Formula by HPLC with Photodiode Array Detection

  • The objective of this study was to develop a method to simultaneously quantify vitamins A and E in infant formula. To determine the vitamin A and E content, vitamin A and four different vitamin E isomers (${\alpha}$-, ${\beta}$-, ${\gamma}$-, and ${\delta}$-tocopherol) were separated by high performance liquid chromatography with a photodiode array detector using a Develosil RPAQUEOUS RP-$C_{30}$ column ($4.6{\times}250$ mm, 5 ${\mu}M$). The vitamin A and E contents in the certified reference material determined using this method were within the certified range of standard values. The limits of detection (LODs) and limits of quantitation (LOQs) for vitamin A were 0.02 and 0.06 ${\mu}g/L$, respectively. LODs and LOQs for the vitamin E isomers ranged from 0.20 to 0.55 and from 0.67 to 1.81 ${\mu}g/L$, respectively. Linear analyses indicated that the square of the correlation coefficient for the vitamin A and E isomers was 0.9997-0.9999. The recovery of vitamins ranged from 96.69 to 97.79%. The results demonstrate that this novel method could be used to reliably analyze vitamin A and E content in infant formula.

DFT/B3LYP Study to Investigate the Possible Ways for the Synthesize of Antioxidants with High Efficiency Based on Vitamin E

  • The possible ways for increasing the antioxidant properties of vitamin E have been investigated with density function theory. The effect of replacing three methyl groups of vitamin E with various substituents such as electron donating and electron withdrawing groups on the antioxidant properties of vitamin E were investigated. Also the effects of the reducing the number of atoms in the heterocyclic ring and replacing the oxygen heteroatom with other heteroatoms on the antioxidant properties of vitamin E were investigated. The novel structures that obtained from replacing methyl groups with substituents such as $NH_2$, OH, COOH and NHMe have greater antioxidant activity than vitamin E. Obtained results reveal that novel structure that obtained with replacing O with NH hetroatom would be a better antioxidant than vitamin E. The results reveal that reducing the number of atoms in the heterocyclic ring is a better way to synthesize novel antioxidants.

Effect of Supplementation of Vitamin E on In Vitro Maturation and Activation of Bovine Oocytes

  • This study was carried out to assess the effect of vitamin E against the reactive oxygen species (ROS) on chemical activation of in vitro matured oocytes. Bovine oocytes were aspirated from slaughtered ovaries and transferred to maturation medium with or without vitamin E ($100\;{\mu}M$). After 22 hours of culture, oocytes with polar bodies were selected and submitted to activation treatments with or without vitamin E. After activation, oocytes were cultured in mSOF medium and rate of development was monitored. For ROS ($H_2O_2$) detection, in vitro matured and activated oocytes were selected and stained with DCFDA and observed under fluorescence microscope. The ROS contents were not significant differences in IVM rate, activation process and embryonic development to blastocysts with or without vitamin E. The cell number of blastocyst showed significant difference (p<0.05) in embryos matured and activated with vitamin E. The results of the present study demonstrated that the exposure of vitamin E in IVM and activation process improved the quality of embryos evaluated by the cell number of blastocysts.

Effect of Vitamin E and Selenium Administration on The Reproductive Performance in Dairy Cows

  • Incidence of retained placenta in dairy cows was investigated in 120 parturitions. Prior to calving, cows were allotted into four groups; 1) control, 2) the infection of Vitamin E (500 IU), 3) the injection of selenium (40 mg), and 4) the infection of Vitamin E (500 IU) and selenium (40 mg). Selenium (Se) and Vitamin E were injected intramusculary 20 d prior to the estimated calving date. No effects of Se or Vitamin E administration alone was observed for number of service per conception, conception rate, and estrus rate (p > 0.05). But the Vitamin E administration with Se significantly (p < 0.05) reduced the incidence of retained placenta and the days required for calving the first service. Incidence of retained placenta was not significantly (p > 0.05) influenced by parity, but it was significantly (p < 0.01) influenced by season, especially, increased during July and August. Concentration of Se in plasma was not significantly (p > 0.05) changed during peripartum period whether the treatment. The results of this study on retained placenta suggest that this disorder will be reduced by the administration of Se and Vitamin E prior to calving in dairy cows.

The Effect of Dietary Selenium Source and Vitamin E Levels on Performance of Male Broilers

  • Selenium and vitamin E are micronutrients essential for normal health and maintenance in poultry. They are necessary in preventing free radical damage to phospholipid membranes, enzymes and other important molecules. Two experiments were conducted in a semi-commercial environment to examine the effect of Se source and vitamin E level in diet on broiler performance and meat quality. Increasing vitamin E from 50 IU to 100 IU did not affect growth performance of broilers although the 24 h drip-loss was tended to be reduced (p=0.06). There was an interaction between vitamin E and the source of Se in glutathione peroxidase activity (GSH-Px) and Se concentration in excreta. Increasing vitamin E from 50 IU to 100 IU elevated GSH-Px and Se concentration in excreta by 42 IU/g Hb and 0.9 ppm for the organic Se group, respectively, but reduced GSH-Px and Se concentration in excreta by 16 IU/g Hb and 1.3 ppm for inorganic group, respectively. Vitamin E played no role in the feather coverage of the birds when scored on day 37. Organic Se is more effective in improving feather score and 24 h drip-loss, with a markedly higher deposition rate in breast muscle and a lower excretion rate in the excreta (p<0.05) compared to the inorganic Se source. Both vitamin E and the source of Se did not affect (p>0.05) the energy utilisation by birds.

  • It has been known that quercetin, a bioflavonoid widely distributed in fruits and vegetables as dietary-derived flavonoid and exert significant multiple biological effects such as antioxidant and anti-inflammatory, anti-tumor effects. In addition, it has been shown to have a chemoprotective role in cancer, though complex effects on signal transduction involved in cell proliferation and angiogenesis. The present study investigated whether quercetin can enhance antioxidant enzyme activity (glutathione peroxidase: GPx, superoxide dismutase: SOD, catalase: CAT) and regulate the reactive oxygen species (ROS) generation in the presence of vitamin E, L-ascorbic acid, reduced glutathione (GSH) on B16F10 murine melanoma cells. After 48h treatment of cells with quercetin in the presence of vitamin E, L-ascorbic acid, GSH, we measured the cytotoxicities by MTT assay. The cells exhibited a dose-dependent inhibition in their proliferation in the presence of vitamin E, L-ascorbic acid, GSH respectively. We also investigated the effects of antioxidant enzyme activity and ROS generation. The antioxidant enzyme activity of quercetin in the presence of vitamin E was stronger than GSH, L-ascorbic acid, the same treatments decreased ROS generation in B16F10 murine melanoma cells. Taken together, these result demonstrate that the antioxidant effect of quercetin can enhanced in the presence of vitamin E and it might plays an important role in anti-oxidative effects.

Streptozotocin 유발 당뇨쥐 신장조직의 항산화계에 미치는 Vitamin E의 영향 (effects of Vitamin E on the Antioxidative Defense System of Kidney in Streptozotocin-Induced Diabetic Rats)

  • The purpose of this study was to investigate the effects of vitamin E on the antioxidative defense system of kidney in streptozotocin induced diabetic rats. Sprague Dawley male rats weighing 100$\pm$10g were randomly assigned to one normal and three STZ induced diabetic groups, which were subdivided into vitamin E free diet(DM 0E group), 40mg vitamin E per kg diet(DM 40E group) and 400mg vitamin E per kg diet(DM 400E group). Vitamin E level of normal group was 40 mg per kg diet. Diabetes was experimentally induced by intravenous injection of 55mg/kg of body weight of streptozotocin(STZ) in citrate buffer(pH 4.3) after 4 weeks feeding of experimental diets. Animals were sacrificed at the 6th day of diabetic states. There were no significant on body weights, food intakes, and food efficiency ratio before the diabetic occurrence. But after the injection of STZ, body weights and food efficiency ratios were significantly decreased and the food intakes was increased. Kidney weights were significantly increased in diabetic groups compared to normal group. However, there were no significant differences among the diabetic groups. Plasma insulin levels of diabetic groups were significantly decreased, whereas, blood sugar levels were increased compared to that of normal group. There were no significant differences among diabetic groups in plasma insulin and glucose levels. Activities of superoxide dismutase(SOD) in DM 0E and DM 40E groups were signi ficantly decreased by 33% and 27%, respectively, compared to normal group. But that of DM 400E group was increased by 35% compared to DM 0E group. Activity of glutathione peroxidase(GSHpx) in DM 0E group was decreased by 20% compared with normal group. GSHpx activity in DM 400E group was increased by 29% compared to normal group. The contents of vitamin E in kidney were 58% and 49% lower in DM 0E and DM 40E group, respectively, than normal group. There was no significant difference in renal vitamin E contents between DM-400E group and normal group. The contents of superoxide radical(O2 ) in kidney were 150% and 98%, respectively, higher in DM 0E and DM 40E groups than normalgroup. DM 400E and normal groups were similar levels in their superoxide radical contents of kidneys. These results indicate that vitamin E functioned as chain breaking antioxidant in kidney such as in other tissues.

식이 Vitamin E가 Streptozotocin 유발 당뇨쥐의 간 및 폐조직에서의 Microsomal Mixed Function Oxidase System에 미치는 영향 (Effect of Dietary Vitamin E on the Microsomal Mixed Function Oxidase System of Liver and Lung in Streptozotocin-induced Diabetic Rats)

  • 본 연구는 streptozotocin 유발 당뇨쥐의 간과 폐조직에서의 MFO계 활성 변화 및 지질과산화에 미치는 vitamin E의 영향을 관찰하고자 본 실험을 행하였다. 체중 140g 정도의 Sparague-Dawley종 숫컷을 대조군(40mg vitamin E/kg diet group)과 당뇨유발 실험군으로 나눈 후 실험군은 다시 식이내 vitamin E 공급수준에 따라 DM-0E군(0mg vitamin E/kg diet group), DM-40E군(40mg vitamin E/kg diet group), DM-400E군(400mg vitamin E/kg diet group) 나누어 4주간 사육한 후 대조군을 제외한 실험군을 STZ로 당뇨를 유발시켰다. 당뇨유발 6일 후 쥐를 희생시켜 간조직 및 폐조직 microsome 중의 cytochrome P$_{450}$ 및 cytochrome b$_{5}$함량과 NADPH-cytochrome P$_{450}$ reductase 활성도를 측정하고 아울러 microsome내의 지질과산화물을 측정하여 다음과 같은 결과를 얻었다. 1. 간조직 중의 cytochrome P$_{450}$ 함량은 대조군에 비해 DM-0E군, DM-40E 군 및 DM-400E군이 각각 162%, 150%, 56% 증가하였으며 또 DM-400E군은 DM-0E, DM-40E군에 비해 낮았다. 폐조직 중의 cytochrome P$_{450}$ 함량도 간조직에 비해 함량은 낮았지만 경향은 비슷하였다. 2. 간조직 중의 cytochrome b$_{5}$은 대조군에 비해 DM-0E군, DM-40E군은 각각 70%, 53%씩 증가하였으나 DM-400E군은 대조군 수준이었다. 폐조직 중의 cytochrome b$_{5}$함량도 대조군에 비 해 DM-0E, DM-40E 및 DM-400E군에서 각각 149%, 116%, 50%씩 증가하였다. 3. 간조직에서의 cytochrome P$_{450}$ reductase 활성은 대조군에 비해 DM-0E군은 58%, DM-40E군에서는 47% 증가하였으며 DM-400E는 대조군 수준이었다. 폐조직에서도 간조직에서와 비슷한 경향이었다. 4. 간조직의 지질과산화물가는 대조군에 비해 DM-0E군, DM-40E군, DM-400E군이 각각 143%, 95%, 31%씩 높았다. 또 DM-400E군은 DM-0E, DM-40E군에 비해 약 50% 정도 낮았다. 폐조직에서도 경향은 비슷했는데 간조직 보다 과산화적 손상이 다소 높았다. 이상의 결과에서 STZ 유발 당뇨쥐에서는 MFO system의 활성이 증가되고 지질과산화가 촉진되었다. 그러나 식이 중의 vitamin E의 적절한 공급은 MFO system 활성이 저하되고 과산화적 손상이 현저하게 완화됨을 볼 수 있었다.

습사료에 비타민 C와 E 첨가가 넙치 치어의 성장과 체성분에 미치는 영향 (Effects of Supplementary Vitamin C and E to Moist Pellet Diet on Growth and Body Composition of Juvenile Olive Flounder, Paralichthys olivaceus)

  • This study was conducted to investigate the supplementary effects of vitamin C and E to moist pellet on growth and body composition in juvenile olive flounder (Paralichthys olivaceus). Four groups of diet made : moist pellet (MP) diet group was used as the control group where the ratio of raw feed and commercial compound meal is 5:5 $(CP\;30\%,\;CL\;17\%),$ vitamin C added group (VC), vitamin I added group (VE) and vitamin C and I added group (VCE). The supplementary amount of vitamin C and E corresponded to 1000 mg/kg (dry wt.) and 220mg/kg (dry wt.), respectively. Vitamin C was destroyed $50\%$ and vitamin E was destroyed $20\%$ for manufacturing process. After 8 weeks feeding trial, weight gain was $121.9\%$ in MP group, while it ranged from 180.5 to $184.9\%$ in the VC, VE and VCE group. Feed efficiency was $71.\6%$ in MP group, whereas it ranged from 78.7 to $80.6\%$ in the VC, VE and VCE group, weight gain and feed efficiency of fish fed vitamin supplemented diets were significantly higher than control group and no significant difference among the vitamin added groups was observed (p>0.05). There were no significant differences in moisture, crude protein, crude lipid, crude ash, condition factor (CF), hepatosomatic index (HSI), and visceraweight index (VWI) of body composition. Hemoglobin (Hb) was $3.79\%$ in MP group, whereas $4.99\%$ in VC group, $4.55\%$ in VE group and $5.07\%$ in VCE group; Hb was significantly higher in the vitamin added groups than the control group (p<0.05). Vitamin C retention in liver was 18.1, 20.7, 46.6, 54.0 mg/kg in MP, VE, VC and VCE groups, respectively. Vitamin E retention in liver was 25.9, 53.8, 88.2, 124.5 mg/kg in MP, VC, VE and VCE groups, respectively. These results indicate that supplementation of vitamin C and E to moist pellet diet significantly improved growth and feed efficiency of olive flounder, and the mixed addition of vitamin C (1,000 mg/kg dry wt.) and vitamin E (220 mg/kg dry wt.) did not lead to the synergy effect.
