Preparation and Quality Characteristics of Yogurt with Black Garlic Extracts (흑마늘 농축액을 첨가한 요구르트의 제조와 품질특성)

  • Yogurt base was prepared from skim milk added with 0.5, 1.0 and 1.5% (w/v) black garlic extracts(BGE, 60 brix), fellowed by fermentation with Lactic acid bacteria (the mixed strain of Streptococcus thermophilus, Lactobacillus delbrueckii subsp. bulgaricus) at $42^{\circ}C$ for 24 hr. The yogurt products were evaluated for acid production (pH, titrable acidity), number of viable cells, viscosity, color value, and sensory properties. The pH of yogurt with BGE was below 6.0 at 6 hr fermentation, after which it was rapidly acidifies. Aftert 24 hr, the titratable acidity of yogurt with 1.5% BGE was 0.74%, which was 5.7 times higher than that before fermentation. There was no significant difference in viable cell count between the samples after 3 hr fermentation. The viscosity of yogurt was decreased by the addition of BGE. As the percentage of BGE increased, the L value (lightness) decreased while the a value (redness) and b value (yellowness) increased. The overall sensory score of yogurt with BGE was lower than that of yogurt with only skim milk. Therefore, moderate addition of BGE was below 1% for the preparation of yogurt.

Production of Curd Yogurt from Platycodon grandiflorum(Jacq.) A.DC. (길경(桔梗)을 이용한 호상 요구르트 제조(製造))

  • The curd yogurt was prepared from skim milk powder added with the juice, puree and powder, respectively, of the roots of Platycodon grandiflorum (Jacq.) A.DC. Twelve hours were proved to be best for fermentation of the curd yogurt, which showed 4.1 in pH and 1.15% in titratable acidity. Quality of the curd yogurt in sensory evaluation was best when 2 % of juice, 1 % of puree and 1 % of powder were added to the skim milk powder, respectively. When curd yogurt was kept at $5^{\circ}C$ for 9 days, pH decreased, while titratable acidity increased. The viscosity was highest with addition of puree 1%. After fermentation, number of viable cell was $6.2{\times}10^8/ml$ in control. $4.4{\times}10^8/ml$ in juice 2%, $3.7{\times}10^8/ml$ in puree 1% and $4.2{\times}10^8/ml$ in powder 1%. When curd yogurt was kept at $5^{\circ}C$ for 9 days, its keeping quality (pH, titratable acidity, viscosity and number of viable cells) were good.

THE CYTOTOXIC EFFECTS OF GLASS-IONOMER CEMENT LINERS ON FIBROBLASTS IN HUMAN PULP (Glass-ionomer Cement 이장재의 세포독성에 관한 연구)

  • The purpose of this study was to evaluate for the cytotoxicity of glass-ionomer cement liners(GC liningcement, Ketac-bond, Vitrebond and Fuji lining LC) on the fibroblasts cultured from human pulp. The fibroblasts were cultured in DMEM-10% FBS medium. The measurement of pH, succinate dehydrogenase (SDH) activity test and $^{51}Chromium$ release test were performed. Viable cell count and $^{14}C$-leucine incorporation rate were evaluated following culture time of 2, 4 and 6 days. The results of this study were as follows : 1. The pH in all cements was to be neutralized as time elapsed, and Fuji lining LC was the lowest pH value among them. 2. SDH activity was more inhibited in GC lining cement and Vitrebond than Ketac-bond and Fuji lining LC with the setting process, and GC lining cement and Ketac-bond were reduced after 5 minute's setting and then elevated as time elapsed. 3. In SDH activity test following exposure time, the activity in Vitrebond, GC lining cement and Fuji lining LC was inhibited with increased exposure time, but it was fairly constant in Ketac-bond. 4. Overall the liquid component was more inhibited than the powder component of glass-ionomer cement in SDH activity test. 5. In $^{51}Cr$-release test, Fuji lining LC was the most released of all the cements tested and followed by : Vitrebond, Ketac-bond, GC lining cement. 6. In viable cell count, the number of cells increased as the culture day proceeded in Ketac-bond, but they decreased in GC lining cement. Fuji lining LC was only observed after 2 days culture and there was not observed the whole culture days in Vitrebond. 7. In $^{14}C$-leucine incorporation rate test, protein synthesis was decreased with the number of culture days in GC lining cement, Vitrebond and Fuji lining LC, but it was followed that of control in Ketacbond.

Anti-inflammatory effects and GABA production of old antler and Auricularia auricula-judae extract fermented by Lactobacillus plantarum (녹각 및 목이버섯의 젖산발효를 통한 GABA 생산 및 항염증활성 효과)

  • The optimization of lactic acid fermentation was conducted to produce an old antler fortified with functional ingredients. For the over-production of gamma aminobutyric acid (GABA), the extract of old antlers (OA) was fermented by Lactobacillus plantarum EJ2015 with 0.5% YE, 1.5% glucose, and 3.5% MSG at $30^{\circ}C$ for 7 days. The lactic acid fermented OA showed high viable cell counts of $2.0{\times}10^8CFU/mL$, pH 6.56 and 0.77% acidity after 7 days. Addition of Auricularia auricula-judae (AAJ) enhanced the cell growth of L. plantarum EJ2014, resulting in higher viable cell counts of $2.0{\times}10^9CFU/mL$ and acid production after fermentation for 1 day. In particular, acidity was greatly decreased after fermentation for 3 days and 1.4% GABA was produced by converting efficiently mono sodium glutamate as a substrate. Fermented OA/AAJ mixture indicated the reduced cytotoxicity compared with that of unfermented OA. The fermented OA/AAJ mixture indicated anti-inflammatory effect with less production of NO in microphage cells. The production of NO dropped to $17.75{\mu}M$ at 4 mg/mL, and to $5.58{\mu}M$ at 6 mg/mL old antler after fermentation. Thus, lactic acid fermented OA with AAJ could fortify GABA, probiotics and dietary fiber.

Quality Characteristics of Traditional Kochujang Adding Pear Juices during Fermentation (배즙을 첨가한 전통고추장의 숙성과정 중 품질특성)

  • In order to improve qualities of traditional kochujang, pear juice was added to kochujang, and the physicoche-mical and microbial characteristics of pear-added kochujang were investigated for 2 months of fermentation at 30$^{\circ}C$. Moisture contents of pear-added kochujangs increased except of 9$\%$ pear-added kochujang and crude protein contents of them decreased as the fermentation proceeded. Total sugar contents of pear-added kochujangs did not change during the fermentation period, and reducing sugar contents of them increased remarkably from early stage of fermentation. The pH of pear-added kochujangs decreased and titratable acidity of them increased during fermentation. Amino-nitrogen contents of pear-added kochujangs increased remarkable after 45 days of fermentation. Viable cells counts of yeasts in pear-added kochujangs increased to 2.2$\∼$2.9$\times$$10^{3}$CFU/g, and bacterial cell counts were in relatively constant range of 2.1$\∼$2.9$\times$$10^{7}$CFU/g at the late stage of fermentation. Color of pear-added kochujangs had no difference and 'a' value decreased slightly during fermentation. The result of sensory evaluation showed that 6$\%$ pear added kochujang were more acceptable than others.

Phylogenetic diversity of bacterial communities in a gray solar saltern and isolation of extremely halophilic bacteria using culturomics (토판염전 결정지 내 세균군집의 계통학적 다양성 및 Culturomics법을 이용한 고도 호염균의 분리)

  • In this study, we investigated the phylogenetic diversity of the bacterial community and isolation of extremely halophilic bacteria using culturomics in a gray solar saltern. The number of bacterial living cells, enumerated in a gray solar saltern by direct fluorescence microscopy was three to four orders of magnitude greater than those enumerated by plate counts, suggesting the distribution of 'viable but non-culturable bacteria'. The biodiversity of bacterial communities in a gray solar saltern was investigated by pyrosequencing, 1,778 OTUs of bacteria were comprised of 18 phyla 46 classes 85 orders 140 families 243 genera with 6.16 diversity index. Archaea communities were composed of 3 phyla 6 classes 7 orders 7 families 38 genera with 4.95 diversity index from 643 OTUs. Totally 137 isolates were isolated by 59 different cultural methods based on culturomics considering culture media and conditions suitable for the growth of extremely halophilic bacteria. Phylogenetic analyses of extremely halophilic isolates based on 16S rRNA gene sequences, extremely halophilic isolates were composed of 4 phyla and 11 genera. Haloterrigena and Haloferax can be successfully isolated from culturomics. These culturomics were effective methods for collection of diversity of extremely halophilic bacteria.

Mode of Action of the Bacteriocin from Lactobacillus sp. GM7311 against Gram Positive Bacteria (Lactobacillus sp. GM7311이 생산하는 박테리오신의 Gram 양성균에 대한 작용형태)

  • The bacteriocin produced by Lactobacillus sp. GM7311 showed strong inhibitory activity against the growth of three Gram positive bacteria, Listeria monocytogenes, Bacillus subtilis, and Staphylococcus aureus. When the bacteriocin was added to the culture at different phases, viable cells of all of the tested strains were decreased, although the most inhibited phase was different. Thereby, when the bacteriocin $(100\;Bu/m{\ell})$ was added to exponential and stationary phase of L. monocytogenes, the rapid reduction of viable cell counts occured. And, in the case of B. subtilis, the highest inhibitory effect occured at lag phase and mid-exponential phase by the addition of the bacteriocin under same condition as mentioned above. Also, we can observe the accelerated reduction of survivors counts for the all of the phase except stationary phase in the S. auresus. Transmission electron microscopic observation of L. monocrogenes and B. subtilis treated with bacteriocin revealed apparent Iysis of the cell wall and excretion of the cell contents, indicating bacteriolysis. Also, the amino acids and fatty acids compositions were different from controls. However, the Iysis of cell wall didn't occur in S. aureus, though the cytoplasmic materials were reduced. This result indicates that the bacteriocin inhibits the synthesis of nuclear materials such as DNA, RNA and proteins.

Quality Characteristics of Korean Traditional Rice Wine with Glutinous Rice (찹쌀 첨가에 따른 전통발효주의 품질 특성)

  • This study is carried out to investigate the physicochemical characteristics, microbial population, and sensory characteristics during fermentation of Korean traditional rice wine with addition of glutinous rice. The fed-batch fermentation of rice was performed by Nuruk and yeast for 10 days at $28^{\circ}C$ in a water bath. The four fermentation batches included 0, 10, 15 and 20% of glutinous rice based on the total rice contents. The growth of total viable cells, lactic acid bacteria (LAB), and yeasts were similar among the four batches during the fermentation period. The population for total viable cells and LAB were increased for the first 3 days, and decreased slowly until 10 days. The number of yeast cells was rapidly decreased after day 6, when the alcohol content reached about 15% for all the fermentation batches. Physicochemical characteristics, such as pH, total acidity, and reducing sugars, were not different with the increase of additional glutinous rice contents. The ethanol production was higher in Korean traditional rice wine from non-glutinous rice (17.1%) than ones from glutinous rice (15.8~16.7%). For the sensory evaluations, Korean traditional rice wine with 15% glutinous rice was highly preferred due to the highest sweetness.

High Glucose Induces Apoptosis through Caspase-3 Dependent Pathway in Human Retinal Endothelial Cell Line (인간망막 내피세포주에서 고농도 포도당이 caspase-3 경로를 통해 세포자연사 유도)

  • Diabetic Retinopathy (DR) is a leading cause of blindness among adults in the western countries. Hyperglycemia is a condition, that induces apoptotic cell death in a variety of cell types in diabetes, but the mechanism remains unclear. The aim of the study is to understand the effects of high Glucose on Human Retinal Endothelial Cells. Retinal endothelial cells were cultured in Iscove's Modified Dulbecco's Medium (IMDM) containing 5, 25 and 50 mM Glucose, incubated for 24, 36 and 48 hours in humidified 5 % CO$_2$ incubator at 37$^{\circ}C$. Human Retinal Endothelial Cell Line (HREC) were characterized for morphology with different treatment by phase contrast microscopic analysis. Number of dead and viable cells was counted by trypan blue exclusion and supported by MTT assay. The intracellular Hydrogen peroxide (H$_2$O$_2$), a Reactive Oxygen Species (ROS) generation in high glucose conditions was assessed by FOX II assay and apoptosis by caspase-3 assay. The high glucose treated cells undergoing DNA fragmentation was witnessed by Agarose gel electrophoresis. We found that the cells incubated with 25 and 50 mM glucose containing medium for 48 hours altered the morphology of the cell, induced apoptosis and DNA fragmentation. The dead cell number were high in 25 and 50 mM when compared to the cells incubated with 5 mM glucose for 24, 36, and 48 hours. Also, the H$_2$O$_2$ levels and the activity of caspase-3 were increased in high glucose treated cells. Conclusions/interpretation: Our results demonstrated that elevated glucose induces apoptosis in cultured HREC. The hyperglycemia-induced increase in apoptosis may be dependent on caspase activation. The association between ROS generation and caspase-3 activation on high glucose treated cells is yet to be investigated.

Effect of ultrasonification on immuno-stimulatory activities of the extracts from Korean medicinal herbs

  • The effect of ultrasonification on immune-stimulatory activities of extracts Acanthopanax senticosus Harms, Ephedra sinica Stapf, Rubus coreanus Miq and Artemisia capillaris Thunb was observed by the growth of human immune cells. The extracts yield make a comparative study of water extracts and water extract with ultrasonification at $40^{\circ}C,$ $60^{\circ}C$ and $100^{\circ}C$. The yield is highest in the water extracts with ultrasonification at $60^{\circ}C$ The growth of human T-cell was increased up to $13.8{\times}10^{-4}$ viable cells/ml in adding ultrasonification extracts. The treated extracts increased up to $10.5{\times}10^{-4}$ pg/cells that TNF-${\alpha}$ specific secretion of human T-cell. These results suggested that biological activities of water extracts with ultrasonification have more strong than water extracts in human immune system.
