The Growth and Yield Differences in Kenaf (Hibiscus cannabinus L.) in Reclaimed Land Based on the Physical Types of Organic Materials (간척지에서 유기물 투입 형태에 따른 케나프의 생육반응)

  • To improve the soil of reclaimed land, we added organic materials at a level of 3,000 kg/10 a. As a result, the electrical conductivity (EC) value of reclaimed soil decreased by 58%, the organic material content increased from 6.7 to 16.0 g/kg, the porosity increased from 1.57 to 1.31%, the soil hardness decreased from 20.2 to 17.9 mm and the plow layer was deepened from 19.8 to 26.8 cm. After these physiochemical improvements to the reclaimed soil, the growth phase of crops was improved compared to that of non-treatment crops. The height of kenaf (Hibiscus cannabinus L.) cultivated in the reclaimed land containing organic materials was increased by 18.8%. Especially, the improvement effects of pellet type manure compost and rice straw on kenaf were more preferable than those of other organic materials. When the kenaf was cultivated in the reclaimed land containing organic materials, the yield increased. The average yield of the treatment crops was 9,218 kg/10 a, 2.1 times higher than that of non-treatment crops. The most effective treatments to increase the yields were pellet type manure compost (10,848 kg/10 a, 148% increase), rice straw (120% increase) and chopped kenaf (95% increase). To increase the physicochemical enhancements to the reclaimed land soil and most improve yields, the most effective type of organic materials was the pellet. The organic material types that maintained a better growth phase and most increased the yield were the liquid and pellet types. When we used pellet type organic material, the plant height of kenaf was increased by 41% in comparison with that of the non-treatment crops and yield was increased by more than 122%. Additionally liquid type organic material improved the yield (by 127%).

Reduction of Perchlorate and Nitrate by Citrobacter Amalonaticus Strain JB101 : Kinetics and the Applicability of MBR (Citrobacter Amalonaticus Strain JB101에 의한 과염소산염과 질산염의 환원 : Kinetics 및 MBR을 이용한 처리 가능성)

  • This study was performed to evaluate the characteristics of the competition between two electron acceptors, perchlorate and nitrate, with Citrobacter Amalonaticus strain JB101. In addition, the applicability of membrane bioreactor(MBR) for perchlorate removal was evaluated. The maximum growth rate of strain JB101 on perchlorate and nitrate are 0.27 and 0.58 $hr^{-1}$, and maximum substrate utilization rates were 35.1 mg $ClO_4^-/g$ protein-day and 45.6 mg $NO_3^-/g$ protein-day, respectively. Nitrate was a competitive inhibitor for perchlorate, and strain JB101 prefer nitrate to perchlorate as electron acceptor. Complete removal of perchlorate could be achieved up to the surface leading rate of 4.6 g $ClO_4^-/m^2-day$ with the MBR fed with 20 mg $ClO_4^-/L$(HCMBR). When 5 mg/L of nitrate was added to the same influent, perchlorate removal efficiency decreased to 96.5%, while nitrate was completely removed. For the MBR fed with 0.7 mg/L of perchlorate (LCMBR), the maximum perchlorate removal efficiency was 100% up to the loading rate of 0.23 g $ClO_4^-/m^2-day$. Membrane fouling was found to be a problem at high leading rate for both MBRs. The acetate consumption ratio per perchlorate was $13.7{\sim}51.7\;e^-eq./e^-eq.$ in LCMBR, while the value was $2.5{\sim}3.6\;e^-eq./e^-eq.$ in HCMBR. This difference could be related to the acetate consumption with oxygen as electron acceptor. Therefore, the amount of acetate addition must be determined considering the concentrations of other electron acceptors in the influent.

Proliferation Assay of Splenocyte and PBMC by the Evaluation of Alamar Blue Dye Reduction Value in Broiler Chicks (Alamar Blue 색소의 환원량 평가에 의한 급성기 반응중 육계병아리의 비장세포와 PBMC 증식도 측정)

  • In this study, hatched male broiler chicks(Ross) were fed on a basal diet and LPS was administered via intraperitoneal injection three times every other day, on the 9th, 11th and 13th days of the experiment, and then PBMC and splenocytes were isolated on day 14. The degree of alama blue reduction was evaluated at 4, 24, 48, 96 and 120 h in the splenocytes, and at 4, 8, 12, 24 and 48 h for PBMC of incubation after the addition of alama blue solution to the media. The cell numbers used in this experiment were 103, 104 and 105 cells per well, and the con A levels were 0.0, 1.0, 5.0, and 10.0 ㎍ per ml of medium. 1. The degree of alama blue reduction was found to increase in a linear fashion with increasing incubation time and cell numbers, for both splenocytes and PBMC. 2. During acute phase response, the degree to which alama blue was reduced was significantly elevated (p<0.05) at an incubation time of 24 hr for the splenocytes, 4 hr for PBMC, and a cell number of 105 cells per well, respectively. 3. The raised reduction of alama blue to control was linear with Con A levels in medium, and higher reduction in Con A 10.0 ㎍ relative to 1.0 or 5.0 ㎍ in ml medium was shown 4. The medium with autologous serum evidenced a significantly (p<0.05) higher reduction of alama blue relative to FBS. 5. Splenocytes and PBMC from the LPS-injected birds evidenced significantly higher levels of alama blue reduction regardless of incubation time, number of cells, level of Con A added, or serum type, as compared with what was observed in normal birds. The results indicated that the assay conditions for proliferative activity using the alama blue method in birds in which the acute phase response had been activated via intraperitoneal LPS injection requires 4 hrs of incubation for PBMC, 24 hrs of incubation for splenocytes, and 10㎍ of Con A per ml of medium.

Effect of Processing Additives on Vulcanization and Properties of EPDM Rubber (EPDM 고무의 첨가제에 따른 가류 및 물성에 미치는 영향 연구)

  • Effects of three different types of dispersions and flow improving additives composed with fatty acid esters, fatty acid metal salts and amide compound on the vulcanization and the mechanical properties properties of rubber compounds of EPDM and carbon black as fillers. were investigated using Mooney viscometer, moving die rheometer, hardness tester, and universal test machine. The aging characteristics of vulcanized EPDM compounds were also investigated. The Mooney viscosity measured at $125^{\circ}C$ showed a tendency to decrease in the order of amide type> metal salt type > ester type additive. Scorch time showed little or no difference with the addition of ester or metal salt type additives, but the amide type additive shortened a scorch time more than one minute. Rheological measurement data obtained at $160^{\circ}C$ showed that the vulcanization time was faster for metal salt type and amide type additive systems. Delta torque values of EPDM compound increased with metal salt type and amide type additives, but slightly decreased with ester type additive. The tensile strength of the EPDM compound was greatly improved when an ester type additive was added, but the amide type or metal salt type additive had no significant effect. The elongation was significantly improved for metal salt type additive, while the rest were not significantly affected. The tear strength of the EPDM compounds increased with the addition of all kinds of additives, and it increased remarkably in the case of metal salt type additive. Hardness of the EPDM compounds was nearly same value regardless of additive types. The thermal aging of the EPDM blend at $100^{\circ}C$ for 24 h showed little change in the case of metal salt type or amide type additive, but the elongation tends to decrease by 10-20% for all EPDM compounds containing additives.

Effect of phenobarbital sodium and 3-methylcholanthrene on metabolism in vitro and toxicity of $^{14}C$-carbofuran in rat (쥐에서 phenobarbital sodium 및 3-methylcholanthrene이 $^{14}C$-carbofuran의 독성과 in vitro 대사에 미치는 영향)

  • In order to elucidate the effect of phenobarbital sodium(PB) and 3-methylcholanthrene(3-MC) on metabolism in vitro and toxicity of $^{14}C$-carbofuran in rat, they were administered by the chemicals, alone or in combination, and their survival ratios and metabolites were investigated. The $LD_{50}$(96 hrs) value of carbofuran to rats was 6.9 mg/kg. The toxicities of the major metabolites were in the decreasing order of 3-hydroxycarbofuran, 3-ketocarbofuran, 3-hydroxycarbofuran phenol and were much lower than that of the parent compound. When the rats were orally administered by the dose of carbofuran alone, 8.4 mg/kg, the survival ratio was 0%, whereas that was raised up to $60{\sim}80%$ with 20 mg/kg of PB or 3-MC, and 100% with 60 mg/kg of PB or 3-MC. Their metabolism in vitro occurred in the microsomal fraction. In case of carbofuran alone, the major metabolite was 3-hydroxycarbofuran. When carbofuran with PB or 3-MC, on the other hand, was treated, it was 3-ketocarbofuran. In addition, when the co-factor(NADP+G-6-P+G-6-P-DG) was added to the microsomal fraction(phase I system), and a mixture of NADPH+GSH to the 105,000g supernatant(phase II system) taken by carbofuran alone, each metabolites were produced by the maximum levels, respectively. In case of the carbofuran treatment with PB or 3-MC, the microsomal fraction of phase I system produced the maximum levels of metabolites, as in the treatment of carbofuran alone, whereas the 105,000g supernatant supplemented with the co-factor NADPH+FAD(phase II system) was brought about the maximum production of metabolites. The ratio of the formation of metabolites was 2 to 3 times higher in the combined treatment of carbofuran with PB or 3-MC than in the treatment of carbofuran alone.

The research for the yachting development of Korean Marina operation plans (요트 발전을 위한 한국형 마리나 운영방안에 관한 연구)

  • The rise of income and introduction of 5 day a week working system give korean people opportunities to enjoy their leisure time. And many korean people have much interest in oceanic sports such as yachting and also oceanic leisure equipments. With the popularization and development of the equipments, the scope of oceanic activities has been expanding in Korea just as in the advanced oceanic countries. However, The current conditions for the sports in Korea are not advanced and even worse than underdeveloped countries. In order to develop the underdeveloped resources of Korean marina, we need to customize the marina models of advanced nations to serve the specific needs and circumstances of Korea As such we have carried out a comparative analysis of how Austrailia, Newzealand, Singapore, japan and Malaysia operate their marina, reaching the following conclusions. Firstly, in marina operations, in order to protect personal property rights and to preserve the environment, we must operate membership and non-membership, profit and non-profit schemes separately, yet without regulating the dress code entering or leaving the club house. Secondly, in order to accumulate greater value added, new sporting events should be hosted each year. There is also the need for an active use of volunteers, the generation of greater interest in yacht tourism, and the simplification of CIQ procedures for foreign yachts as well as the provision of language services. Thirdly, a permanent yacht school should be established, and classes should be taught by qualified instructors. Beginners, intermediary, and advanced learner classes should be managed separately with special emphasis on the dinghy yacht program for children. Fourthly, arrival and departure at the moorings must be regulated autonomically, and there must be systematic measures for the marina to be able, in part, to compensate for loss and damages to equipment, security and surveillance after usage fees have been paid for. Fifthly, marine safety personnel must be formed in accordance with Korea's current circumstances from civilian organizations in order to be used actively in benchmarking, rescue operations, and oceanic searches at times of disaster at sea.

Enzymatic characterization of Paenibacillus amylolyticus xylanases GH10 and GH30 for xylan hydrolysis (Paenibacillus amylolyticus 유래 xylanase GH10 및 GH30의 xylan 가수분해 특성)

  • The enzymatic degradation of xylans is the most versatile way to obtain the high value-added functional compounds or the fermentable sugars for renewable energy. The endo-${\beta}$-xylanases are the major enzymes which hydrolyze the internal ${\beta}$-1,4-linkages of xylan backbones to produce the mixtures of xylooligosaccharides including xylobiose and xylotriose. Among them, glucuronoxylanase GH30 can exclusively hydrolyze the internal ${\beta}$-1,4-linkages of xylans decorated with methylglucuronic acid branches. In the present study, two xylanolytic enzyme (PaXN_10 and PaGuXN_30) genes were cloned from Paenibacillus amylolyticus KCTC 3005, and expressed in Escherichia coli, respectively. PaXN_10 (38.7 kDa) belongs to the endo-${\beta}$-xylanases GH10 family, while PaGuXN_30 (58.5 kDa) is a member of glucuronoxylanase GH30. They share the same optimal reaction conditions at $50^{\circ}C$ and pH 7.0. Enzymatic characterization proposed that P. amylolyticus can utilize the hardwood glucuronoarabinoxylans via the cooperative actions of xylanases GH10 and GH30. The extracellular PaGuXN_30 is secreted into the medium and hydrolyzes glucuronoarabinoxylans to release a series of aldouronic acid mixtures with a methylglucuronic acid branch. The resultant products being transported into the microbial cell are successively degraded into the smaller xylooligosaccharides by the intracellular PaXN_10, which will be utilized for the cellular metabolism.

Properties of Baechu Kimchi treated with Black Rice Water Extract (흑미를 첨가하여 항산화성이 강화된 배추김치의 개발 및 품질 특성)

  • To develop a new functional kimchi with antioxidative properties, salted baechu was soaked in black rice water extract for 6 h at room temperature. The antioxidative property of the water extract was $78.75{\pm}1.18%$ that of the control (0.1% [w/v] alpha-tocopherol). The black rice gel was added to the baechu kimchi preparation. The color of baechu kimchi treated with black rice water extract changed to dark violet and/or black. Control kimchi and black rice water-treated kimchi were stored at $4^{\circ}C$ for 30 days. No significant differences were detected between the control and the black rice water-treated group in the early stages of fermentation. As fermentation time increased, pH decreased and titratable acidity increased rapidly in control kimchi. However, such marked changes were not evident in test kimchi. The hardness value of black rice water-treated kimchi was higher than that of control kimchi after the midpoint of the fermentation period. The storage life of baechu kimchi treated with black rice water extract was prolonged by up to 5 days compared with control samples, owing to a decline in lactic acid bacteria and yeast levels during the final fermentation period in black rice water-treated kimchi. The total phenolic levels and the antioxidative capacity of black rice water-treated kimchi (83%) were approximately 1.5-fold higher than in control kimchi (57%). In sensory evaluation, black rice water-treated kimchi scored higher than did control kimchi using a blind test protocol.

Effects of Vitamins and Nucleic acids on the mycelial growth and the sclerotial production of Sclerotium rolfsii (Vitamin과 핵산이 Sclerotium rolfsii의 균사생장 및 균핵형성에 미치는 영향)

  • The study was performed to clear the effects of thiamine, biotin, nicotinic acid, pyridoxine, inositol, deoxyribonucleic acid (DNA) and ribonucleic acid (RNA) on the mycelial growth and the sclerotial production of Sclerotium rolfsii Sacc. isolated from Magnolia kobus. The results are abstracted as follows: 1. Tested fungus was thiamine- deficient and required thiamine 20r/l for maximum growth of mycelia. At higher concentrations than thiamine 20r/l, however, mycelial growth was decreased with increasing the concentrations and was inhibited little less than that of thiamine-free control at 150r/l. 2. The effecfivenesses of the nitrogen sources on the mycelial growth under the thiamine presence were recognized in order of $NH_4NO_3>(NH_4)_2SO_4 >asparagine> KNO_3$, and on the sclerotial production were $KNO_3>NH_4NO_3>asparagine>(NH_4)_2SO_4$. The optimum concentrations of thiamine were about 12r/1 in $KNO_3$, about 16r/1 in asparagine on the growth of mycelia, and were about 8r/l in $KNO_3\;and\; NH_4NO_3,\; 16r/1$ in asparagine on the production of sclerotia. 3. After the organism began to grow, the pH value of cultral filtrate was rapidly dropped down to about 3.5. Hereafter it was slowly fallen down as the growth amount was increased, but was not depreciated below pH 2.2. 4. Nicotinic acid was not effective individually on the mycelial growth and the sclerotial formation of tested fungus without thiamine, but slight effect of it was recognized with thiamine 10r/l, even though maximum growth was shown at 7-10mg/1. Beyond that concentration, however, mycelial growth was rather depressed. 5. When ammonium sulphate or asparagine as the nitrogen sources was used, pyridoxine, biotin and inositol had not any effectivenesses on the mycelial growth and the sclerotial production of examined fungus. 6. In the concentrations of thiamine, biotin, pyridoxine and inositol, as long as thiamine was not added in those, their correlating effects on the growth of the organism were not observed at all. Equivalent or more effects on the mycelial growth were recognized in combinations of thiamin + pyridoxine, thiamine + inositol, thiamine + biotin + pyridoxine, and thiamine + biotin + pyridoxine + inositol compared with thiamino alone, and in combinations of thiamine + biotin and thiamine + biotin + inositol, mycelial growth was inhibited rather than that of thiamine alone. Sclerotial production of those combinations was increased more than that of thiamine alone in dry weight. 7, The little effects of DNA and RNA on the mycelial growth of the organism were recognized compared with the control(DNA-and RNA-free), and RNA was more effective than DNA. Maximum growth of mycelia was observed at RNA 2-6mg/1 and DNA 6mg/l. No effectivenesses on the sclerotial production were recognized in the RNA and DNA. 8. Mycelial growth of the organism was increased with increasing the concentrations of the RNA and the thiamine, that is, the effectiveness of RNA was revealed apparently under presence of thiamine, but was not shown in the sclerotial formation.

Technical Efficiency in Korea: Interindustry Determinants and Dynamic Stability (기술적(技術的) 효율성(效率性)의 결정요인(決定要因)과 동태적(動態的) 변화(變化))

  • This paper, a sequel to Yoo and Lee (1990), attempts to investigate the interindustry determinants of technical efficiency in Korea's manufacturing industries, and also to conduct an exploratory analysis on the stability of technical efficiency over time. The hypotheses set forth in this paper are most found in the existing literature on technical efficiency. They are, however, revised and shed a new light upon, whenever possible, to accommodate any Korea-specific conditions. The set of regressors used in the cross-sectional analysis are chosen and the hypotheses are posed in such a way that our result can be made comparable to those of similar studies conducted for the U.S. and Japan by Caves and Barton (1990) and Uekusa and Torii (1987), respectively. It is interesting to observe a certain degree of similarity as well as differentiation between the cross-section evidence on Korea's manufacturing industries and that on the U.S. and Japanese industries. As for the similarities, we can find positive and significant effects on technical efficiency of relative size of production and the extent of specialization in production, and negative and significant effect of the variations in capital-labor ratio within industries. The curvature influence of concentration ratio on technical efficiency is also confirmed in the Korean case. There are differences, too. We cannot find any significant effects of capital vintage, R&D and foreign competition on technical efficiency, all of which were shown to be robust determinants of technical efficiency in the U.S. case. We note, however, that the variables measuring capital vintage effect, R&D and the degree of foreign competition in Korean markets are suspected to suffer from serious measurement errors incurred in data collection and/or conversion of industrial classification system into the KSIC (Korea Standard Industrial Classification) system. Thus, we are reluctant to accept the findings on the effects of these variables as definitive conclusions on Korea's industrial organization. Another finding that interests us is that the cross-industry evidence becomes consistently strong when we use the efficiency estimates based on gross output instead of value added, which provides us with an ex post empirical criterion to choose an output measure between the two in estimating the production frontier. We also conduct exploratory analyses on the stability of the estimates of technical efficiency in Korea's manufacturing industries. Though the method of testing stability employed in this paper is never a complete one, we cannot find strong evidence that our efficiency estimates are stable over time. The outcome is both surprising and disappointing. We can also show that the instability of technical efficiency over time is partly explained by the way we constructed our measures of technical efficiency. To the extent that our efficiency estimates depend on the shape of the empirical distribution of plants in the input-output space, any movements of the production frontier over time are not reflected in the estimates, and possibilities exist of associating a higher level of technical efficiency with a downward movement of the production frontier over time, and so on. Thus, we find that efficiency measures that take into account not only the distributional changes, but also the shifts of the production frontier over time, increase the extent of stability, and are more appropriate for use in a dynamic context. The remaining portion of the instability of technical efficiency over time is not explained satisfactorily in this paper, and future research should address this question.
