One bacterium, which showed strong antagonistic activity against H. pylori KCCM 41756, was isolated from kimchi. The strain NO1 was designated as Lactobacillus plantarum NO1 based on Gram staining, biochemical properties, and 16S rRNA gene sequencing. The culture medium $(2{\sim}4{\mu}g/ml)$ of Lb. plantarum NO1 reduced $(40{\sim}60%)$ the urease activity of H. pylori KCCM 41756. Lb. plantarum NO1 inhibited the binding of H. pylori to human gastric cancer cell line, AGS cells, by more than 33%. Lb. plantarum NO1 exhibited high viability (maintained initial viable cell count of $10^9CFU/ml$) in 0.05 M sodium phosphate buffer (pH 3.0) for 2 h, in artificial gastricjuice for 2 h and in 0.3%, 0.5% oxgall for 24 h. Hemolysis phenomena did not observed when Lb. plantarum NO1 was incubated in the blood agar media. We concluded that Lb. plantarum NO1 can be a good candidate as a probiotic, harboring anti-H. pylori activity.

This study was conducted to find out the effect of herbicides on soil micro-organism and soil enzyme in loam soil, and on pathogenic microorganism in continuous pepper cropping soil. The result was summarized as follows: When herbicides were treated, the number of soil microorganism generally decreased at the early stage of incubation, and gradually increased at 30 days incubation. The number of fungi was significantly decreased seven times comparison with control plot. The number of actinomycetes was increased at 20 days incubation and that of pytium was increased for all days incubation in Linuron treatment plot. Although the activity of soil enzyme tended to decrease when herbicides were treated, the activities of urease and phosphatase were higher than that of control plot at the early stage of incubation in MO, Linuron and Simetryne treatment plot. In Simetryne treatment plot, the activity of protease was increased at the early stage, and suddenly decreased after 30 days incubation. There was no significant effect of herbicides on the activities of ${\beta}$-glucosidase and polygalacturonase. The activity of cellulase was inhibited at the early stage of incubation, but that of cellulase was higher than that of control plot after 20 days incubation. High significance was showed the correlation coefficient between soil microorganisms except fungi and soil respiration as herbicides were treated, and the total microorganism and soil respiration in Linuron plot. when Dicamba and Simetryne were treated, the correlation coefficient between the total microorganism and the activities of protease and urease were considerably significant.
The effects of some soil conditions on the degradation rate and decomposing pattern of thiolix were investigated and the obtained results are summarized as follows: Thiolix degraded more rapidly in flood soils than in noon-flooded, and in wet soils than in dry soils under non-flooded soils. The degradation rates in non-flooded soils increased with higher pesticide concentration. Thiolix was more persistent in non-flooded soils under soil sterelization than under non-sterilization and degraded rapidly in glucose application. The metabolites identified from the soils by TLC and GLC include Thiolix alcohol, Thiolix sulfate, Thiolix ether and a unknown metabolite. Soil enzyme, acid phosphatase activity decreased at higher pesticide concentration, lower moisture contents of soil and the activity in glucose application was increased. Soil enzyme, urease and dehydrogenase activity decreased at higher pesticide concentrations.
The novel compound KH 3218 was synthesized and evaluated for its ability to inhibit the gastric H$^{+}$+ $K^{+}$ ATPase activity in vitro as well as to lessen gastric acid secretion in vivo. KH 3218 inhibited rabbit gastric H$^{+}$+ $K^{+}$ ATPase in a concentration and time dependent manner. $IC_{50}$/ value was estimated to be about 15 $\mu$M. The inhibition of the H$^{+}$+ $K^{+}$ ATPase by KH 3218 was blocked by sulfhydryl reducing agents, dithiothreitol or $\beta$-mercaptoethanol. The inhibition of the enzyme was not reversible by 50 fold dilution of the incubation mixtures, suggesting the irreversible nature of the inactivation. In the pylorus-ligated rift, KH 3218 reduced the total acid output as compared with the control. In addition, KH 3218 was capable of inhibiting H. pylori urease activity. These data suggest that KH 3218 is a potent inhibitor for H$^{+}$+ $K^{+}$ ATPase activity as well as for gastric acid secretion, and has a potential to be developed as a novel antiulcer agent.

Southern blight caused by Sclerotium rolfsii is a serious soilborne disease in economically important crops including pepper. In this study, we conducted a selection of antagonistic bacterial strains and organic materials to biologically control the disease. Out of 39 strains isolated from soils at Jinju in Korea, strain JJ2-01 showed the highest mycelial growth inhibition; garlic oil among various organic materials significantly reduced disease incidence and severity. When a combination of strain JJ2-01 and garlic oil, or each was drenched into the pepper plants, combined treatment and garlic oil significantly suppressed the disease development, however, acid phosphatase activity in garlic oil-treated plants decreased. In the case of combined treatment, the soil activities did not affect by treatment, while soil urease activity was significantly increased by the combined treatment. Therefore, given soil quality and health for sustainable agriculture, the combination of strain JJ2-01 and garlic acid was an effective application for environmental-friendly control of Southern blight in pepper plants.
The phytochemical investigation of the crude methanolic extracts roots and stem bark of Anthonotha cladantha (Harms) J.Léonard led to the isolation and identification of twelve secondary metabolites: 2,3-dihydroxypropyl hexacosanoate (1), hederagenine (2), cycloeucalenol (3), 2α-hydroxylupeol (4), betulinic acid (5), lupeol (6), heptacosan-2-one (7), triacontanoic acid (8), stigmast-4-en-3-one (9), β-sitosterol (10), stigmasterol (11), and stigmasterol-3-O-β-D-glucopyranoside (12). Their structures were elucidated with the help of their spectroscopic and physical data and by comparison with those reported in the literature. To the best of our knowledge, from all those compounds, 2,3-dihydroxypropyl hexacosanoate (1), hederagenine (2), cycloeucalenol (3), 2α-hydroxylupeol (4), and betulinic acid (5) are being reported for the first time from this genus. In addition, the acetylation of compound 1 afforded a new derivative 3-(hexacosanoyloxy)propane-1,2-diyl diacetate (1a). Compound 1 possessed a moderate α-glucosidase inhibitory activity with an IC50 value of 39.2 ± 0.22 μM; it neither showed antioxidant activity nor inhibition against the enzyme urease. Compound 1a exhibited weak antioxidant activity in the DPPH assay with an IC50 value of 80.3 ± 0.83 μM but was inactive against α-glucosidase and urease. Furthermore, both compounds 1 and 1a were inactive against seven pathogenic bacterial strains.
Objective: The objective of this study was to select an effective in vitro digestion-fermentation model to estimate the effect of decreasing dietary crude protein (CP) on odor emission during pig production and to suggest potential prediction markers through in vitro and in vivo experiments. Methods: In the in vitro experiment, three diet formulations with different CP contents (170 g/kg, 150 g/kg, and 130 g/kg) but containing the same standardized ileal digestible essential amino acids (SID-EAA) were assessed. Each diet was evaluated by two different in vitro gastric-intestinal phase digestion methods (flask and dialysis), combined with fresh pig feces-ferment inoculation. Eighteen growing barrows (31.9±1.6 kg) were divided into three groups: control diet (180 g CP/kg, without SID-EAA adjustment), 170 g CP/kg diet, and 150 g CP/kg diet for 4 weeks. Results: The in vitro digestion results indicated that in vitro digestibility was affected by the gastric-intestinal phase digestion method and dietary CP level. According to the gas kinetic and digestibility results, the dialysis method showed greater distinguishability for dietary CP level adjustment. Nitrogen-related odor compounds (NH3-N, indole, p-cresol, and skatole) were highly correlated with urease and protease activity. The feeding study indicated that both EAA-adjusted diets resulted in a lower odor emission especially in p-cresol and skatole. Both protease and urease activity in feces were also closely related to odor emissions from nitrogen metabolism compounds. Conclusion: Dialysis digestion in the gastric-intestinal phase followed by fresh fecal inoculation fermentation is suitable for in vitro diet evaluation. The enzyme activity in the fermentation and the fecal samples might provide a simple and effective estimation tool for nitrogen-related odor emission prediction in both in vitro and in vivo experiments.
Grifola umbellatus was extracted using methanol, and the extract was further fractionated by water and ethyl acetate. Assay of each fraction with MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium-bromide] revealed significant cytotoxicity effect of the methanol extract of Grifola umbellatus against human gastric cancer cell but not normal human lymphocytes. The methanol extract showed the highest antioxidant activity as well. Antimicrobial activity of Grifola umbellants against Helicobacter pylori was higher in method extract than in other fractions. Grifola umbellatus had a significant inhibitory effect on Helicobacter pylori reducing both its growth and urease activity. These results show that the methanol extract of Grifola umbellatus possesses therapeutic potential on gastric diseases.
Helicobacter pylori (H. pyroli) infection it associated with type B gastritis, peptic uler disease, and gastric cancer. The vacuolation of cells induced by H. pylori is thought to be essential for the initiation and maintenance of gastric infection. The roles of H. pylori cytotoxin, urease, and ammonia in the vacuolation of HeLa cells were determined. H. pylori toxin induced vacuolation of HeLa cells. Korean and Japanese radishes significantly prevented the vacuolation of HeLa cells induced by H. pylori toxin. The urease activity in vacuolated cells was also decreased with Korean and Japanese radishes. H. Pylori toxin-induced vacuolation was inhibited by vacuolar type ATPase inhibitors (bafilomycin and N-ethylmaleimide). However, further investigation is required to determine the mechanisms of radish for the inhibition of vacuole formation of eukaryotic cells in response to the H. pylori toxin.
The effect of oyster shell meal, which is made of a simple crushing and alkaline calcium materials, on soil microbial properties, microbial biomass C, N and P contents, and enzyme activities were evaluated in silt loam soil. The oyster shell meal fertilizer was added at the rates of 0, 4, 8, 12 and $16Mg\;ha^{-1}$. Microbial biomass C, N and P contents were significantly increased with increasing application of oyster shell meal. Soil enzyme activities, such as urease, ${\beta}$-glucosidase and alkaline phosphomonesterase were increased significantly by shell meal application, due to increased soil pH towards neutral range and increased nutrient availability in soil. In particular, the increased microbial biomass P content and phosphomonoesterase activities were strongly correlated with available P content in soil. Conclusively, oyster shell meal fertilizer could be a good supplement to improve soil microbial activities.
