Chemical Ingredients of Cordyceps militaris

  • Medicinal mushrooms, including Cordyceps militaris, have received attention in Korea because of their biological activities. In the fruiting body and in corpus of C. militaris, the total free amino acid content was 69.32 mg/g and 14.03 mg/g, respectively. In the fruiting body, the most abundant amino acids were lysine, glutamic acid, proline and threonine. The fruiting body was rich in unsaturated fatty acids, which comprised about 70% of the total fatty acids. The most abundant unsaturated acid was linoleic acid. There were differences in adenosine and cordycepin contents between the fruiting body and the corpus. The adenosine concentration was 0.18% in the fruiting body and 0.06% in the corpus, and the cordycepin concentration was 0.97% in the fruiting body and 0.36% in the corpus.

  • The changes of various lipid content and fatty acids composition of mung-bean sprout during 6 days. The results are summarized as follows 1) The total lipid contents of mung-bean sprouts are generally decreased during growing. 2) On T.L.C. gram, triglyceride contents are decreased and free fatty acids and sterol are increased with time. 3) The ratio of unsaturated fatty acids to saturated fatty acids U/S are increased by six times after 4 days of mung-bean germination. 4) The contents of palmitic acid as main fatty acid in the mangbean but oleid, linoleic acid were increased after 2 days, 4 days, 6 days. 5) Total lipids from mung-bean sprouts contained fatty acid in order of linoleic acid, oleic acid. 6) The content of essential fatty acid are rich in 4 days.

  • An attempt was made to investigate free and bound lipid contents and fatty acid compositions of ginseng and its eveven kinds related crude drugs. Contents of free and bound lipids of ginseng were 1.23% and 0.47%, and those of the related crude drugs were $0.31{\sim}13.05%$ and $0.26respectively. Fourteen even-numbered and 4 odd-numbered fatty acids were identified by GLC in those lipids from ginseng, and the major fatty acids were linoleic, palmitic, oleic and linolenic acids. The crude drugs are notably different from ginseng in kinds and compositions of fatty acids of free and bound lipids. Composition ratios of unsaturated fatty acids were higher in free lipids from ginseng than those of the crude drugs except 'Won-ji' Polygalae Radix.

Purification and Characterization of Lipase from Trichosporon sp. Y-11and Its Use in Ester Synthesis of Unsaturated Fatty Acids and Alcohols

  • A 28-kDa extracellular lipase (pI 8.7) was purified to homogeneity from the culture supernatant of Trichosporon sp. Y- 11 by mmonium sulfate precipitation, DEAE-Sephadex A-50, Bio-Gel P-30, CM- Sephadex C-50, and Bio-Gel P- 10 chromatographies. The purified enzyme exhibited a specific activity of $2,741{\;}{\mu}mol/min/mg$ based on the hydrolysis of triolein, and the optimal hydrolysis activity was dentified at pH 8.0 and $40^{\circ}C$. The enzyme activity was inhibited by $Ag^+$ and enhanced by $Fe^{2+}$, $Fe^{3+}$, $Mg^{2+}$, $Mn^{2+}$, and $Li^{+}$. The enzyme activity exhibited for the hydrolysis of both tributyrin and trilinolein. The ester synthesis of unsaturated fatty acids with various alcohols catalyzed by the purified lipase in a nonaqueous medium or microaqueous system was also investigated. The esterification activity of the lipase increased with an increase of the carbon chain length in the alcohol. The synthesis rate of linoleic acid and oleyl alcohol was the highest with an optimal temperature and pH of $40^{\circ}C$ and 8.0, respectively. The water content and agitation also affected the esterification activity of the lipase.

  • This study was designed to elucidate the effects of iron, a well known catalyst of lipid peroxidation, on the contents of phospholipids, unsaturated fatty acids composed in phospholipid molecules and their derivatives, prostaglandins, and the composition changes of fatty acids contained in phospholipids. Iron decreased the contents of phospholipids and its components of unsaturated fatty acids. Catalytic action of iron decreased the composition rates of linoleate and linolenate composed in phospholipid molecules, while that of arachidonate was inclined to increase. The content of arachidonate was increased and that of prostaglandins was decreased without regard to increase the precursor of prostaglandins. It may be concluded that the decreases of prostaglandins and the increase of arachidonate are due to inhibition of the activities of enzyme systems responsible for prostaglandin synthesis by lipid peroxides produced by the catalyst of iron.

The Changes of Malonaldehyde and Fatty Acids Composition of Yellow Corvenia during Gulbi Processing and Storage (굴비의 가공.저장 중 Malonaldehyde 함량 및 지방산 조성 변화)

  • For the improvement of Gulbi processing, yellow corvenia was salted by 2 different time (5 hours, low salted and 5 days, high salted) and then dried in the sun and stored for 5 and 21 days, respectively. Malonaldehyde contents and fatty acid composition were analyzed during Gulbi processing. The moisture contents of yellow corvenia were significantly decreased during processing procedure. The salt contents were 10 folds higher in 5 day-salted sample than in 5 hour-salted one. The salt contents showed reversed tendency to moisture contents. There was little change in pH during storage. The contents of malonaldehyde in yellow corvenia were increased during Gulbi processing and storage. Its contents were higher in 5 day-salted sample and exterior parts than 5 hour-salted sample and interior parts of Gulbi. The fatty acids composition showed higher oleic acid $(C_{18:1})$, palmitic acid $(C_{16:0})$ and docosahexaenoic acid $(C_{22:6})$ than any other fatty acids in Gulbi. Saturated fatty acids were increased but unsaturated fatty acids were decreased during Gulbi processing. After 21 days storage, unsaturated fatty acids remaining ratio in 5 hour-and 5 day-salted sample were 1.39 and 0.99 respectively. The contents of unsaturated fatty acids were dramatically changed as salt concentration increased during storage than in processing.

Investigating Fatty Acid Content Change in Broiler Breast Meat Produced by Supplementing Feed with Tenebrio molitor L Powder (갈색거저리(Tenebrio molitor L) 분말을 활용한 닭가슴살 지방산 변화에 대한 조사)

  • This study was conducted to investigate the effects of adding Tenebrio molitor L Powder to broiler feed on fatty acid profiles in broiler breast meat. In total, 180 broiler chicks (1-day old Arbor Acres) were included. The birds were randomly divided into control and treatment groups, with 3 replicate subgroups each(30 birds per subgroup), and fed a diet for 35 days without (control) or with 1% Tenebrio molitor L powder (treatment). Among individual fatty acids, addition of Tenebrio molitor L. powder resulted in slightly higher C18:1n-9, C20:3n-3 and C20:3n-6 contents, and lower C18:2n-6 content compared withcontrols (p<0.05). No remarkable differences in total SFA and total USFA contents were found between groups. In conclusion, inclusion of Tenebrio molitor L. in broiler diets did not improve overall fatty acid profiles.

Fatty Acid Composition of Adipose Tissues in Obese Mice and SD Rats Fed with Isaria sinclairii Powder

  • Isaria sinclairii (Cicada Dongchunghacho) was studied as a potential crude natural food in powdered form. The role of tissue fatty acids in relation to the anti-obesity effects of I. sinclairii (IS) was examined by feeding the powder to SD rats ad libitum at 0, 1.25, 2.5, 5 and 10% (calculated about 8 g/kg) of the feed for a period of 3 months and 6 months. The fatty acid composition profile as indicated GC-MS, showed significantly slight dose-dependent increases in the levels of unsaturated fatty acids, particularly, arachidonic acid (C20: 4n6), oleic acid, linoleic acid, eicosadienoic acid, eicosapentaenoic acid (EPA) (C20: 5) concentration in the the ad libitum IS-fed groups compared to the control group in SD abdominal fat over 6 month period. Over viewing of the SD and Ob mice treated Isaria sinclairii powder; there were increases in the single (mono) unsaturated fatty acids ratio but decreases in polyunsaturated fatty acid. In IS-fed groups in proportion to the treatment period, this Dongchunghacho also induced an increase in the level of same result of unsaturated fatty acid in C57BL/6 obese (ob/ob) mice over a 6-month period treatment compared to those given 10% dry mulberry leaf powder (ML) or silkworm powder mixed with the standard diet.

A Study on the Changes of Fatty Acid Composition of Phospholipid and Glycolipid during Storage Period of Kaesojoo (저장기간에 따른 개소주의 Phospholipid 및 Glycolipid의 지방산 변화에 관한 연구)

  • This experiment was carried out to investigate the changes in phospholipid and glycolipid of Kaesojoo and Kaesojoo added medicinal herbs during storage(30 days) at 4$^{\circ}C$and -18$^{\circ}C$. Two dogs with 12kg live weight(♀, The Korean Jindo dog Hy-breed, 11∼12 month) were slaughtered to obtain samples. The result obtained were as follows: The saturated fatty acids found in phospholipid of Kaesojoo were palmitic acid, stearic acid, myristic acid, while the unsaturated fatty acid found in phospholipid of Kaesojoo with medicinal herbs were palmitic acid, stearic acid, arachidonic acid, while the unsaturated fatty acid were mostly oleic acid, increased during the period of storage. The saturated fatty acid found in glycolipid obtained from Kaesojoo were mostly oleic acid, linoleic acid and linolenic acid. The saturated fatty acid obtained from Kaesojoo with medicinal herbs were palmitic acid, stearic acid, while the unsaturated fatty acid were mostly oleic acid, linoleic acid and linolenic acid. The rates of changes in unsaturated and saturated fatty acid were higher at 4$^{\circ}C$ than that of -18$^{\circ}C$ during storage.

Two-Stage Microbial Biotransformation for the Production of 6-Dodecen-4-olide (Butter Lactone) from Plant Oils Containing Unsaturated Fatty Acids (불포화 지방산 함유 식물유를 이용한 천연 6-Dodecen-4-oilde (Butter Lactone) 생산을 위한 2-Stage Microbial Biotransformation)

  • Natural 6-dodecen-4-olide (Butte lactone) was produced from plant oils containing high unsaturated fatty acids via two-stage microbial hiotransformation. After unsaturated fatty acids were liberated from plant oil by microbial lipase, these were converted to optically active hydroxyl fatty acid (HFA) by hydroxylation reaction of Pseudomonas sp. NRRLB-2994. When safflower oil containing >75% unsaturated fatty acid, linoleoic acid wasused, Pseudomonas sp. produced 8g/L of 10-hydroxy-12(z)-octadecanoicacid with average of 39.2% bioconversion efficiency during 48 hr biotransformation period. The recovered 10-hydroxy-12-octadecanoic acid was further bioconverted to 4-hydroxy-6-dodecenoic acid via partial ${\beta}-oxidation$ by Yarriowia lipolytica ATCC34088. 4-hydroxy-6-dodecenoic acid in culture was lactonized by lowering pH to 4.0 using $4N\;H_{2}SO_{4}$ and heating for 5 min to 6-dodecen-4-olide (Butter lactone). Natural 6-dodecen-4-olide had characteristic aroma properties when compared to 6-dodecan-4-oilde (dodecalactone) and 4-decen-4-olide (decalactone).
