Characteristics of Two Unrecorded Yeasts from Wild Flowers in Ulleungdo, Korea

  • Two unrecorded yeasts, Meyerozyma caribbica UL5-1 and Pichia silvicola UL6-1 were screened from 58 yeasts which were isolated from wild flowers in Ulleungdo in Gyeongsangbuk-do, Korea. The morphological and cultural characteristics of these unrecorded yeasts were investigated. Both yeasts were oval in shape and formed pseudomycelia. P. silvicola UL6-1 formed ascospore, but M. UL5-1 did not. P. silvicola UL6-1 and M. caribbica UL5-1 also grew in vitamin-free medium and 5% NaCl-containing yeast extract-peptone-dextrose medium. The two unrecorded yeasts assimilated glucose, galactose, xylose, cellobiose, trehalose, glycerol and sorbitol, and also fermented glucose, fructose and mannose. The supernatant of both M. caribbica UL5-1 and P. silvicola UL6-1 showed high antihypertensive angiotensin I-converting enzyme inhibitory activity of 84.2% and 82.6%, respectively. Cell-free extract of P. silvicola UL6-1 also showed very high anti-diabetic ${\alpha}$-glucosidase inhibitory activity (85.8%).

Mycological Characteristics of Nine Unrecorded Yeasts from Flowers in the Orchard of Yesan-gun, Chungcheongnam-do and Hanbat Arboretum in Daejeon City, Korea

  • Six unrecorded yeasts, Cryptococcus festucosus 41-3, Cryptococcus heveanensis 56-4, Debaryomyces nepalensis 95-4, Issatchenkia occidentalis 142-1, Dioszegia zsoltii 39-1, and Kwoniella europala 47-2 were screened from 108 yeasts isolated from flowers and fruits in orchards of Yesan-gun, Chungcheongnam-do, Korea. The morphological and cultural characteristics of these unrecorded yeasts were investigated. They had various shapes, including ellipsoidal, globose, and oval, and also had budding mode in vegetable reproduction, except I. occidentalis 142-1 (fission mode). K. europaea 47-2 only formed pseudomycelium. D. zsoliti 39-1 did not grow in yeast extract-malt extract medium, potato dextrose medium, and vitamin-free medium. C. festucosus 41-3 grew well in 5% NaCl-containing yeast extract-peptone-dextrose medium and had a growth pH range of 7.0~10.0. Three unrecorded yeasts Ogataea polymorpha HB45-1, Rhodotonula hinnulla HB62-2, and Cryptococcus rajasthanensis HB80-4 were screened from 51 yeasts isolated from flowers in Hanbat arboretum in Daejeon city, Korea. They were globose in shape and did not form pseudomycelium. In addition, O. polymorpha HB45-1 and C. rajasthanensis HB80-4 had budding mode in vegetable reproduction. All of them grew well in vitamin-free medium and C. rajasthanesis HB80-4 also grew in 50% glucose and 5% NaCl-containing YPD medium.

낙동강과 영산강 담수와 주변 토양으로부터 야생효모의 분리 및 동정 (Isolation and Identification of Wild Yeasts from Freshwaters and Soils of Nakdong and Yeongsan River, Korea, with Characterization of Two Unrecorded Yeasts)

  • 우리나라 주요 강 담수와 주변 토양 중의 야생효모 분포특성을 조사하고자 먼저 낙동강과 영산강 일부지역의 담수와 주변 토양 25점을 수집하여 15균주 9종의 야생효모들을 PCR을 이용한 ITS 부위와 26S rDNA의 D1/D2 부위의 염기서열 상동성 비교법을 이용하여 동정하였다. 이들 가운데 Aureobasidium pullulans와 Cryptococcus besiolae가 각각 3균주로 가장 많이 분리되었고, Lachancea thermotolerans와 Wickerhamomyces anomalus도 각각 2주씩 분리되었다. 특히 Candida ghanaensis와 Meira geulakonigii는 국내 처음 보고되는 미기록 균주들로서 포자를 형성하였고, YPD 배지와 potatoes dextrose 배지 등에서 잘 생육하는 특성을 갖고 있었다. 또한 두 미기록 효모들은 5% NaCl을 함유한 YPD 배지에서 생육하는 내염성 효모들이었다.

Description of unrecorded wild yeasts from soil in Republic of Korea under cold conditions

  • The purpose of this study was to isolate and identify wild yeasts from soil collected in Daegu City and Cheongyang County, Republic of Korea. Among 11 strains isolated in this study, nine strains were previously reported and two strains were unreported in Republic of Korea. To identify wild yeast strains, pairwise sequence comparisons of the D1/D2 region of the 26S rRNA gene sequence were done using Basic Local Alignment Search Tool (BLAST). The cell morphologies were observed by phase contrast microscope and assimilation test are done using API 20C AUX kit. All strains were assigned to the phylum Basidiomycota. Of the two unrecorded yeast strains, CY-9-10C belongs to the genus Mrakia (family Mrakiaceae, order Cystofilobasidiales, class Tremellomycetes) and PG3-4-10C belongs to the genus Slooffia (family Chrysozymaceae, order Microbotryomycetes incertae sedis, class Microbotryomycetes). Both strains had oval-shaped and polar budding cells. This research described the morphological and biochemical properties of the two unreported yeast species that had not officially reported in Korea.

Isolation and characterization of four unrecorded wild yeasts from the soils of Republic of Korea in winter

  • The purpose of this study was to isolate and identify wild yeasts from the soil collected in Gwangju and Pocheon City, Gyeonggi Province, Republic of Korea. Among 10 strains, six strains were already reported, but four strains were unrecorded in Republic of Korea. To identify wild yeast strains, pairwise sequence comparisons of the D1/D2 region of the 26S rRNA gene sequence were performed using Basic Local Alignment Search Tool (BLAST). The cell morphologies were observed by phase contrast microscope and assimilation tests were carried out using API 20C AUX kit. The 10 strains were assigned to the phyla Basidiomycota (8 strains) and Ascomycota (2strains). The unrecorded four yeast strains, NH33, NH19, NH20, and YP416, belong to the phylum Basidiomycota and the genera Buckleyzyma, Leucosporidium, Holtermanniales, and Mrakia, respectively. All strains had oval-shaped and polar budding cells. In this research, the morphological and biochemical properties of four unreported yeast species were characterized intensively, which were not officially reported in Korea.

Isolation and characterization of unrecorded yeasts species in the family Metschnikowiaceae and Bulleribasidiaceae in Korea

  • The goal of this study was to isolate and identify wild yeasts from soil samples. The 15 wild yeast strains were isolated from the soil samples collected in Pocheon city, Gyeonggi Province, Korea. Among them, four yeast stains were unrecorded, and 11 yeast stains were previously recorded in Korea. To identify wild yeasts, microbiological characteristics were observed by API 20C AUX kit. Pairwise sequence comparisons of the D1/D2 domain of the 26S rRNA were performed using Basic Local Alignment Search Tool(BLAST). Cell morphology of yeast strains was examined by phase contrast microscope. All strains were oval-shaped and polar budding and positive for assimilation of glucose, 2-keto-ᴅ-gluconate, N-acetyl-ᴅ-glucosamine, ᴅ-maltose and ᴅ-saccharose (sucrose). There is no official report that describes these four yeast species: one strain of the genus Kodamaea in the family Metschnikowiaceae and three strains of the Hannaella in the family Bulleribasidiaceae. Kodamaea ohmeri YI7, Hannaella kunmingensis YP355, Hannaella luteola YP230 and Hannaella oryzae YP366 were recorded in Korea, for the first time.

Report of two unrecorded yeast species in the class Tremellomycetes

The two unrecorded yeast strains had oval shape and polar budding cells. This research describers the morphological and biochemical properties of the two unreported yeast species that had not officially reported in Korea.

  • The purpose of this study was to isolate and identify wild yeasts from soil samples collected in Seoul and Daejeon, Republic of Korea. To identify wild yeast strains, pairwise sequence comparisons of D1/D2 region of the 26S rRNA gene sequence were done using Basic Local Alignment Search Tool (BLAST). The cell morphologies were observed by phase contrast microscope and carbon source assimilation test were done using API 20C AUX kit. Among the 13 isolated strains, 11 strains were previously reported, but two strains have never been reported from Republic of Korea. The 13 strains were assigned to the phylum Basidiomycota. The two unrecorded yeast strains B2UV-201 and DJ1-5-B-10C belong to the genera Rhodotorula and Rhodosporidiobolus, respectively. The two unrecorded yeast strains are oval shaped and polar budding cells. This research focuses on the morphological and biochemical properties of the two unreported yeast species that have not officially been reported in Korea.

향신료 재배 토양과 주변 산림 토양으로부터 야생효모의 분리 및 국내 미기록 효모들의 특성 (Isolation and Characterization of Unrecorded Wild Yeasts Obtained from Soils of Spice Fields and Mountains)

  • 향신료 재배 토양 내에서 분리한 효모 중 이들의 분포와 국내 미기록 야생효모들의 특성을 조사한 결과, 충청남도 금산군 진산면의 향신료 재배토양에서 35종 58균주의 야생효모들을 분리, 동정하였고 Trichosporon moniliiforme균이 우점균으로 확인되었다. 또한, 대둔산 일대 산림토양에서 46균주의 야생효모들을 분리하였고 이들 향신료 토양들과 산림 토양 등에서 분리한 야생효모들 중 아직까지 국내에 보고되지 않은 Debaryomyces vindobonensis GHY31-3, Pseudozyma alboarmeniaca CD23-5, Filobasidium stepposum SFG1-4, Aureobasidium caulivorum HD17-5, Metschnikowia noctiluminum HD29-2, Papiliotrema anemochoreius NK2-12, Hannaella surugaensis DCH25-5, Microbotryozyma collariae DCH1-4, Rhodosporidiobolus lusitaniae DCH30-2, Wickerhamiella azyma DCH46-3등 10균주의 국내 미기록 효모들을 선별하였다. 이들 미기록 균주들은 구형-타원형으로 출아, 분열법으로 영양 증식하였다. Filobasidium stepposum SFG1-4균주를 포함하는 5균주들은 포자를 형성하였고 Pseudozyma alboarmeniaca CD 23-5등 3 균주들은 비타민을 첨가하지 않은 YPD 배지에서도 모두 생육하였다. Wickerhamiella azyma DCH 46-3는 20% 포도당을함유한 YPD 배지에서, Debaryomyces vindobonensis GHY31-3 균주는 5% NaCl을함유한 YPD 배지에서 잘 자라는 특성을 보였다. 또한, 이들의 무세포추출물들의 항산화 활성은 거의 없거나 10% 미만이었지만, 항통풍성 xanthine oxidase 저해활성은 Filobasidium magnum SFG1-3가 28.6%로 높게 나타났으며, 미백성 tyrosinase 저해활성은 Rhodotorula mucilaginosa GHP40-2 균주의 무세포추출물이 26%로 가장 높게 나타났다.

서울특별시 허브공원 토양으로부터 야생 효모의 분리, 동정 및 미기록 효모의 특성 (Isolation and Identification of Wild Yeasts from Soils of an Herb Park in Seoul Metropolitan City and Characteristics of Unrecorded Yeasts)

  • 우리나라 토양 중의 효모 종 다양성 조사의 일환으로 서울특별시 H동 허브공원 일대 토양 29점을 2015년 5월에 수집하여 9종 15균주의 야생효모들을 polymerase chain reaction 을 이용한 internal transcribed spacer 부위와 26S rDNA의 D1/D2 부위의 염기서열 상동성 비교법을 이용하여 분리, 동정하였다. 이들 가운데 Cryptococcus laurentii 2균주를 포함하는 Cryptococcus속 균이 8균주로 가장 많이 분리되었고 Saccharomyces cerevisiae도 2균주 분리되었다. 이들 가운데 아직까지 우리나라 미생물 관련 학술지에 보고되지 않은 Candida boleticola SU14-2와 Candida novakii SU6-2, Cryptococcus podzolicus SU18-2 등의 미기록 효모들을 선별한 후 이들의 미생물학적 특성을 조사한 결과 Candida novakii SU6-2는 자낭포자와 의균사를 형성하였고 Candida boleticola SU14-2는 5% NaCl을 함유한 YPD 배지에서 생육하는 호염성 효모이었다.
