LncRNA H19/miR-29b-3p/PGRN Axis Promoted Epithelial-Mesenchymal Transition of Colorectal Cancer Cells by Acting on Wnt Signaling

  • This investigation was aimed at working out the combined role of lncRNA H19, miR-29b and Wnt signaling in the development of colorectal cancer (CRC). In the aggregate, 185 CRC tissues and corresponding para-carcinoma tissues were gathered. The human CRC cell lines (i.e. HT29, HCT116, SW480 and SW620) and normal colorectal mucosa cell line (NCM460) were also purchased. Si-H19, si-NC, miR-29b-3p mimics, miR-29b-3p inhibitor, si-PGRN and negative control (NC) were, respectively, transfected into the CRC cells. Luciferase reporter plasmids were prepared to evaluate the transduction activity of $Wnt/{\beta}-catenin$ signaling pathway, and dual-luciferase reporter gene assay was arranged to confirm the targeted relationship between H19 and miR-29b-3p, as well as between miR-29b-3p and PGRN. Finally, the proliferative and invasive capacities of CRC cells were appraised through transwell, MTT and scratch assays. As a result, overexpressed H19 and down-expressed miR-29b-3p displayed close associations with the CRC patients' poor prognosis (P < 0.05). Besides, transfection with si-H19, miR-29b-3p mimic or si-PGRN were correlated with elevated E-cadherin expression, decreased snail and vimentin expressions, as well as less-motivated cell proliferation and cell metastasis (P < 0.05). Moreover, H19 was verified to directly target miR-29b-3p based on the luciferase reporter gene assay (P < 0.05), and miR-29b-3p also bound to PGRN in a direct manner (P < 0.05). Finally, addition of LiCl ($Wnt/{\beta}-catenin$ pathway activator) or XAV93920 ($Wnt/{\beta}-catenin$ pathway inhibitor) would cause remarkably altered E-cadherin, c-Myc, vimentin and snail expressions, as well as significantly changed transcriptional activity of ${\beta}-catenin/Tcf$ reporter plasmid (P < 0.05). In conclusion, the lncRNA H19/miR-29b-3p/PGRN/Wnt axis counted a great deal for seeking appropriate diagnostic biomarkers and treatment targets for CRC.

Ngugi wa Thiong'o's Pan-Africanism: People's Memory and Alliance to Overcome Postcolonial Nations (응구기와 시옹오의 범아프리카주의 - 포스트식민 국가를 넘어서는 주변부의 기억과 연대)

  • In modern history, there have been several kinds of continental unions or supranational politico-economic unions in the world, such as the United Nations, the European Union, the Union of South American Nations, the African Union, etc. Modern thinkers proposed many pan-isms on their continental base, for example, Pan-Arabism, Pan-Latin Americanism, Pan-Asianism, Pan-Celtism, etc. What is the most common in these pan-isms is that a continental union would be a politico-economic system to overcome the limits of the modern state-nation and to realize a long and happy relationship between member nations and continents. However, the concept of a supranational union differs from that of cosmopolitanism, in that the former presupposes the common cultural and historical heritage in the concerned region or continent. Ngugi wa Thinog'o' Pan-Africanism implies two keywords that are connected to his concepts such as 'decentralization' and 'African languages.' Pan-Africanism supposes that Africa may gain benefits from the union of African nations under the umbrella of anti-colonial efforts to down size the Euro-American influences. Moreover, using African languages enhances self-reliance and self-imagination among the African people. For in the former colonial regimes, the European colonial languages, such as English, French, or Portuguese, were central to the dissemination of European culture and modernity. Ngugi asserts that the African peripheralized languages could reinstate the African cultural heritage and propose an alternative to the Western modernity.

Factors Affecting the Period of Bone Union When Treating Femoral Fractures with a Retrograde Intramedullary Nail (역행성 골수강 내 금속정을 이용한 대퇴골절의 치료 시 골유합 기간에 영향 미치는 인자)

  • Purpose: A retrograde intramedullary nail is commonly used to treat femoral shaft and distal femur fractures. The authors analyzed the treatment results and the factors affecting the period of bone union for thirty-five patients treated with a retrograde intramedullary nail. Materials and Methods: Thirty-five patients who could be followed up for more than twelve months after the treatment with retrograde intramedullary nailing were analyzed retrospectively. The treatment results and the relationship between the period of bone union and the patient's age, sex, comminution of fracture, presence of open fracture, location of the fracture, and accompanying fractures were evaluated. Results: The average bone union time was 4.50 months. The period of bone union was unaffected by the age, sex, location, and presence of open fractures but was affected by the presence of comminution and accompanying fractures. Conclusion: Retrograde intramedullary nailing is effective for distal and shaft of fractures of the femur. The period of bone union is affected by the presence of comminution and accompanying fractures.

Gypenoside XVII protects against myocardial ischemia and reperfusion injury by inhibiting ER stress-induced mitochondrial injury

  • Background: Effective strategies are dramatically needed to prevent and improve the recovery from myocardial ischemia and reperfusion (I/R) injury. Direct interactions between the mitochondria and endoplasmic reticulum (ER) during heart diseases have been recently investigated. This study was designed to explore the cardioprotective effects of gypenoside XVII (GP-17) against I/R injury. The roles of ER stress, mitochondrial injury, and their crosstalk within I/R injury and in GP-17einduced cardioprotection are also explored. Methods: Cardiac contractility function was recorded in Langendorff-perfused rat hearts. The effects of GP-17 on mitochondrial function including mitochondrial permeability transition pore opening, reactive oxygen species production, and respiratory function were determined using fluorescence detection kits on mitochondria isolated from the rat hearts. H9c2 cardiomyocytes were used to explore the effects of GP-17 on hypoxia/reoxygenation. Results: We found that GP-17 inhibits myocardial apoptosis, reduces cardiac dysfunction, and improves contractile recovery in rat hearts. Our results also demonstrate that apoptosis induced by I/R is predominantly mediated by ER stress and associated with mitochondrial injury. Moreover, the cardioprotective effects of GP-17 are controlled by the PI3K/AKT and P38 signaling pathways. Conclusion: GP-17 inhibits I/R-induced mitochondrial injury by delaying the onset of ER stress through the PI3K/AKT and P38 signaling pathways.

  • It has been proved that the union of two linear star-configurations in $\mathbb{P}^2$ of type $t{\times}s$ for $3{\leq}t{\leq}9$ and $3{\leq}t{\leq}s$ has generic Hilbert function. We extend the condition to $t$ = 10, so that it is true for $3{\leq}t{\leq}10$, which generalizes the result of [7].

Plain Fingerprint Classification Based on a Core Stochastic Algorithm

  • We propose plain fingerprint classification based on a core stochastic algorithm that effectively uses a core stochastic model, acquiring more fingerprint minutiae and direction, in order to increase matching performance. The proposed core stochastic algorithm uses core presence/absence and contains a ridge direction and distribution map. Simulations show that the fingerprint classification accuracy is improved by more than 14%, on average, compared to other algorithms.

  • Generalized intuitionistic fuzzy soft set theory, proposed by Park et al. [Journal of Korean Institute of Intelligent Systems 21(3) (2011) 389-394], has been regarded as an effective mathematical tool to deal with uncertainties. In this paper, we prove that certain De Margan's law hold in generalized intuitionistic fuzzy soft set theory with respect to union and intersection operations on generalized intuitionistic fuzzy soft sets. We discuss the basic properties of operations on generalized intuitionistic fuzzy soft sets such as necessity and possibility. Moreover, we illustrate their interconnections between each other.

  • Classification of subjects with unknown distribution in small sample size setup may involve order-restricted constraints in multivariate parameter setups. Those problems makes optimality of conventional likelihood ratio based statistical inferences not feasible. Fortunately, Roy (1953) introduced union-intersection principle(UIP) which provides an alternative avenue. Redescending M-estimator along with that principle yields a considerably appropriate robust testing procedure. Furthermore, conditionally distribution-free test based upon exact permutation theory is used to generate p-values, even in small sample. Applications of this method are illustrated in simulated data and read data example (Lobenhofer et al., 2002)
