대두(Glycine maxim) 추출물이 티로시나아제 프로모터 활성에 미치는 효과 (Effects of Glycine maxim Extract on the Activity of Tyrosinase Promoter)

  • The methanolic extract of Glycine maxim increased the expression of the promoter in B16 mouse melanoma cells harboring a tyrosinase promoter. Extract concentrations of 10 ${\mu}g/m{\ell}$ and 100 ${\mu}g/m{\ell}$ resulted in tyrosinase promoter expression rates of approximately 113% and 184%, respectively, as compared to the control. The fraction layers consisting of butyl alcohol and methylene chloride improved expression effects on the tyrosinase promoter. In particular, the butyl alcohol fraction evidenced a high expression rate at 100 ${\mu}g/m{\ell}$. In the MTT assay, the methanolic extract did not evidence cytotoxicity at concentrations under 500 ${\mu}g/m{\ell}$. Therefore, the results observed with the extract of Glycine maxim showed that the substance exerted a positive effect on the tyrosinase promoter.

  • To estimate the inhibitory effect of Rubus coreanum extract on melanin biosynthesis, we tested its inhibitory effects on tyrosinase promoter in B16 mouse melanoma cells. Rubus coreanum extract by methanol had inhibitory effect approximately 10% on tyrosinase promoter at $100{\mu}g/mL$. In the MTT assay, the same extract exhibited very low cytotoxicity under $100{\mu}g/mL$. The fractions of methylene chloride, butyl alcohol and water did not showed the inhibitory effect on tyrosinase promoter, but the fraction of ethyl acetate exhibited inhibitory effect approximately 11% at $100{\mu}g/mL$.

  • To estimate the inhibitory effect of some mushroom extract on melanin biosynthesis, we tested its inhibitory effects of five mushroom extracts on tyrosinase promoter in Bl6 mouse melanoma cells. In five mushroom extracts, Cordyceps militaris and Poria cocos exhibited low repression effect on tyrosinase promoter. However, Ganoderma lucidum, Paecilomyces japonicus, Phellinus linteus showed high repression effect, Especially, Paecilomyces japonicus and Phellinus linteus extracts had very higher repression effect approximately $81{\sim}83%\;at\;100{\mu}g/mL$. In the MTT assay, Paecilomyces japonicus and Phellinus linteus extracts exhibited high cytotoxicity. Therefore, repression effect of the extracts were closely connected with cytotoxicity.

  • Melanin is specifically produced in melanocytes. The pathway for melanin biosynthesis is mainly controlled by tyrosinase. To estimate the inhibitory effect of melanin biosynthesis from 31 medicinal plants extracts, we tested the inhibitory effects of the tyrosinase promoter on B16 mouse melanoma cells. The result of this study demonstrated that Mori Radicis Cortex and Castena Fractus extracts only in tested medicinal plant extracts have high inhibitory effects on tyrosinase promoters with very low cytotoxicity on B16 mouse melanoma cells. Therefore, extracts of Mori Radicis Cortex and Castena Fractus were evaluated as very effective negative regulators of tyrosinase gene expression.

지치(Lithospermum erythrorhizon) 추출물의 멜라닌 생합성 억제효과 (Inhibitory Effect of Lithospermum erythrorhizon Extracts on Melanin Biosynthesis)

  • 유전자발현 조절 수준에서 멜라닌 색소 생합성 억제물질을 탐색하고자 지치 뿌리로부터 유용성 물질을 추출${\cdot}$분획하여 tyrosinase 프로모터를 지닌 B16 mouse melanoma cell에 처리한 결과 그 메탄을 추출물은 $10{\mu}g/mL$의 농도에서 약 $33\%$ 이상의 tyrosinase프로모터 활성 억제효과를 나타내었으며, 세포 활성능이 $100{\mu}g/mL$의 농도에서 약 $108\%$로 매우 안전하였다. 그리고 methylene chloride, ethyl acetate, butyl alcohol, 물 등의 용매 분획물들도 농도에 따라 차이는 있었지만 $100{\mu}g/mL$또는 $500{\mu}g/mL$의 고농도에서 tyro-sinase 프로모터의 활성을 억제하였다 또한, 지치 메탄올 추출물의 농도를 달리하여 3일 동안 처리하였을 때 멜라닌색소의 생성능은 $10{\mu}g/mL$$100{\mu}g/mL$의 농도에서 대조군 세포에 비 해 각각 약 $11\%$$24\%$로 크게 감소하는 결과를 보여주었다.

In the melanin biosynthetic pathway, tyrosinase is the rate limiting enzyme, and ${\alpha}$-melanocyte stimulating hormone(MSH) or cAMP-elevating agents stimulate melanogenesis and enhance the melanin synthesis and the tyrosinase activity. The author has analyzed the effects of Radix Trichosanthis on the basal melanogenic activities of B16/F10 mouse melanoma cells, and on the ${\alpha}$-MSH or forskolin-induced melanogenesis. Radix Trichosanthis alone markedly suppressed melanin content and tyrosinase activity in a dose-dependent manner. Pretreatment of the cells with Radix Trichosanthis also suppressed the increase of ${\alpha}$-MSH (10 nM) or forskolin (20${\mu}M$)-induced melanin content and tyrosinase activity. The decrease in the tyrosinase activity was paralled by a decrease in the abundance of tyrosinase protein and tyrosinase promoter activity. Pretreatment of the cells with Radix Trichosanthis also inhibited the increase of forskolin($20{\mu}M$) induced the amount of tyrosinase protein and tyrosinase promoter activity. The results of DOPA staining revealed that pretreatment of the cells with Radix Trichosanthis showed less intensity than B16 melanoma cells stimulated with ${\alpha}$- MSH or forskolin. These results suggest that Radix Trichosanthis inhibits melanogenesis and abrogates ${\alpha}-MSH and cAMP-induced melanogenesis in B16 melanoma cells.

  • Chestnut bark extract by methanol repressed the expression of tyrosinase gene of B16 mouse melanoma cell containing tyrosinase promoter. $10{\mu}g/ml{\ell}\;100{\mu}g/m{\ell}$, $1mg/m{\ell}$ of the extract repressed expression of tyrosinase gene about $38\%,\;47\%,\;and\;78\%$, respectively, compared with control. In the MTT assay, the same extract exhibited very low cytotoxicity at $1{\mu}g/m{\ell},\;10{\mu}g/m{\ell},\;100{\mu}g/m{\ell},\;and\;1mg/m{\ell}$, respectively. The fractions of Methylene chloride and ethyl acetate did not showed the repressive effect on the expression of tyrosinase gene, but the fraction of butyl alcohol repressed highly at $10{\mu}g/m{\ell}\;and\;100{\mu}g/m{\ell}$.
