석곡(石斛)이 흰쥐의 뇌조직출혈에 미치는 영향 (Effects of Dendrobii herba against Intracerebral Hemorrhage in Rats)

  • Objects : This study was performed in order to observe the effects of water extract of Dendrobii herba on intracerebral hemorrhage(ICH), Method : ICH was induced by the stereotaxic intrastriatal injection of bacterial collagenase type IV in Sprague-Dawley rats. After the water extracts of Dendrobii herba were administrated orally once a day for 3 days, hematoma volume, percentage of brain edema, expression of iNOS and MPO were observed using immunohistochemistry. Results : Rats fed with water extracts of Dendrobii herba showed reduction of hematoma volume and percentage of brain edema compared with controls. In addition, Infiltration of myeloperoxidase (MPO) expressing neutrophil and expression of inducible nitric oxide synthatase(iNOS) were significantly reduced in rats fed with water extracts of Dendrobii herba. Conclusion : These results demonstrated that water extracts of Dendrobii herba reduced brain damage of intracerebral hemorrhage(ICH) and subsequent ICH-induced cerebral edema, and inhibited neutrophil infiltration.

Secretion and Expression of Matrix Metalloproteinase-2 and 9 from Bone Marrow Mononuclear Cells in Myelodysplastic Syndrome and Acute Myeloid Leukemia

  • Background: Matrix metalloproteinase -2 (gelatinase-A, Mr 72,000 type IV collagenase, MMP-2) and -9 (gelatinase-B, Mr 92,000 type IV collagenase, MMP-9) are key molecules that play roles in tumor growth, invasion, tissue remodeling, metastasis and stem-cell regulation by digesting extracellular matrix barriers. MMP-2 and -9 are well known to impact on solid cancer susceptibility, whereas, in hematological malignancies, a paucity of data is available to resolve the function of these regulatory molecules in bone marrow mononuclear cells (BM-MNCs) and stromal cells of myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML). Objectives: The present study aimed to investigate mRNA expression and gelatinase A and B secretion from BM-MNCs in vitro and genotypic associations of MMP-2 (-1306 C/T; rs243865), MMP-9 (-1562 C/T; rs3918242), tissue inhibitor of metalloproteinase -1 (TIMP-1) (372T/C; rs4898, Exon 5) and TIMP-2 (-418G/C; rs8179090) in MDS and AML. Results: The study covered cases of confirmed MDS (n=50), AML (n=32) and healthy controls (n=110). MMP-9 mRNA expression revealed 2 fold increased expression in MDS-RAEB II and 2.5 fold in AML M-4 (60-70% blasts). Secretion of gelatinase-B also revealed the MMP-9 mRNA expression and ELISA data also supported these data. We noted that those patients having more blast crises presented with more secretion of MMP-9 and its mRNA expression. In contrast MMP-9 (-1562 C/T) showed significant polymorphic associations in MDS (p<0.02) and AML (p<0.02). MMP-9 mRNA expression of C/T and T/T genotypes were 1.5 and 2.5 fold increased in MDS and AML respectively. In AML, MMP-2 C/T and T/T genotypes showed 2.0 fold mRNA expression. Only MMP-9 (-1306 C/T) showed significant 4 fold (p<0.001) increased risk with chemical and x-ray exposed MDS, while tobacco and cigarette smokers have 3 fold (p<0.04) risk in AML. Conclusions: In view of our results, MMP-9 revealed synergistic secretion and expression in blast crises of MDS and AML with 'gene' polymorphic effects and is significantly associated with increased risk with tobacco, cigarette and environmental exposure. Release and secretion of these enzymes may influence hematopoietic cell behavior and may be important in the clinical point of view. It may offer valuable tools for diagnosis and prognosis, as well as possible targets for the treatments.

황련해독탕(黃連解毒湯)이 Collagenase에 의한 흰쥐의 뇌출혈 및 뇌부종에 미치는 영향 (Effects of Hwangnyeonhaedok-tang on Cerebral Hemorrhage and Edema in Collagenase Induced-ICH Rats)

  • Objectives: Brain edema is brain swelling that occurs due to the accumulation of excess water in the brain parenchyma. AQP4 and AQP9 are water-channel proteins expressed strongly in the brain and control water fluxes into and out of the brain parenchyma. This study was conducted to evaluate effect of Hwangnyeonhaedok-tang on brain edema and intracerebral hemorrhage. Methods: Intracerebral hemorrhage was induced by intrastriatal injection of type IV collagenase(0.23 U/${\mu}l$, 0.1 ${\mu}l$/min) into Sprague-Dawley rat brains. Hwangnyeonhaedok-tang water extract(1,000 mg/kg) was administered orally three times every 20 hours from 4 hours after ICH operation. Then hematoma volume, brain edema percentage, and water content of brain tissue were measured. Immuno-histochemistry was performed for AQP4 and AQP9 expressions in the brain sections and area % of immuno-labeling was analyzed with image analyzing system. Results: 1. Water extract of Hwangnyeonhaedok-tang reduced hematoma volume of ICH induced rat. 2. Water extract of Hwangnyeonhaedok-tang reduced MPO positive neutrophils in the perihematoma of the ICH induced rat. 3. Water extract of Hwangnyeonhaedok-tang reduced brain edema percentage and water content of brain tissue of ICH induced rat. 4. Water extract of Hwangnyeonhaedok-tang reduced AQP4 immuno-positive cells in the perihematoma of the ICH induced rat. 5. Water extract of Hwangnyeonhaedok-tang reduced AQP9 immuno-positive cells in the perihematoma of the ICH induced rat. Conclusions: These results suggest that Hwangnyeonhaedok-tang decreases intracerebral hemorrhage and brain edema by means of downregulating AQP4 and AQP9 expressions in the brain.

선학초(仙鶴草)가 선조체내출혈(線條體內出血) 흰쥐의 뇌조직 손상에 미치는 영향 (Neuroprotective Effects of Agrimoniae Herba against Intrastriatal Hemorrhage in Rats)

  • Objects : This study was performed in order to evaluate the effects of Agrimoniae herba (AH) ethanol extract on intrastriatal hemorrhage (ISH). Method : ISH was induced by the stereotaxic intrastriatal injection of bacterial collagenase type IV in Sprague-Dawley rats. AH was orally given once a day for 3 days after ISH. Hematoma volume and percentage edema were examined. Immunohistochemistry was processed for iNOS, c-Fos, MMP-9, and MMP-12 expressions in the brain sections and each immuno-labeling were calculated with image analysis. Results : results are as follows; 1. AH reduced the hematoma volume and percentage edema of the ISH-induced rat brain. 2. AH swollen apoptotic bodies and neurons in the peri-hematoma regions of the ISH-induced rat brain. 3. AH significantly reduced c-Fos, MMP-9 and MMP-12 positive cells in the peri-hematoma regions of the ISH-induced rat brain. 4. AH swollen iNOS expressions in the peri-hematoma regions of the ISH-induced rat brain. Conclusion : These results suggest that AH plays an anti-apoptotic neuroprotective effect through control of ISH, suppression of c-Fos, and down-regulation of MMP-9 and MMP-12 expressions in the brain tissues.

PMA 유도 제4형 단백분해효소 분비를 차단하는 SP600125에 의한 Jun Aminoterminal Kinase의 억제 (Inhibition of the Jun Aminoterminal Kinase by SP600125 Blocks PMA-Induced 92kDa Type IV Collagenase Secretion)

  • 제 4형 콜라젠을 분해하는 MMP-9은 조직 재생에 중요한 역할을 하기 때문에 주목받아 왔는데, 이전의 문헌들에서 PMA에 의해 이 유전자 발현이 강하게 상승발현된다고 알려져 있다. 비록 MMP-9 발현을 조절하는 PMA의 기전이 잘 밝혀지지는 않았지만, 다른 유전자발현에서의 이 phorbol ester의 효과는 c-raf-1-ERK 신호전달통로의 활성에 관해 연구되어 오고 있다. 하지만 이번 연구에서 저자는 MMP-9 발현에서 PMA의 상승효과에는 대개는 스트레스성 자극과 관련된 JNK1 의존성 신호전달과정이 또한 필요하다는 다른 가능성을 조사하였다. 그 결과 JNK 억제재중 하나인 SP 600125가 UM-SCC-1세포주에서 PMA에 의해 유도된 MMP-9 상승발현을 용량의존적으로 억제하는 것으로 나타났고 72시간동안 전처치를 한 경우에 그의 억제효과가 최대이었다. phorbol ester로 처리한 세포에 GAL4 luciferase reporter와 vector를 주입해서 조사한 결과 PMA가 c-Jun transacting 활성을 상승시켰다. 그뿐 아니라 PMA에 의한 MMP-9 촉진자 활성에는 AP-1 motif가 필요하며 이 motif에 c-Jun이 결합하는 것을 EMSA를 통해 확인하였다. 결론적으로 UM-SCC-1 세포주에서 PMA에 의한 MMP9의 증가된 발현은 이미 밝혀진 ERK 경로뿐 아니라 JNK 경로를 통한 결과임이 밝혀져 이 경로를 차단하는 방법이 또하나의 암치료 방향을 제시해주고 있다.

마이크로 PET을 이용한 고양이 뇌 경색 모델의 평가 (Evaluation of Cat Brain infarction Model Using MicroPET)

  • 목적: PET은 해상도 한계로 인하여 각종 소동물 실험에서 영상화하는데 현실적인 어려움이 많았다. 마이크로 PET의 등장으로 랫트나 마우스와 같은 실험 동물을 보다 나은 해상도로 평가할 수 있으나 실험 동물의 크기가 작기 때문에 영상이 사람에서와 같이 선명하지는 않다. 고양이와 같은 중소형 동물은 뇌가 상대적으로 크기 때문에 마이크로PET을 이용하여 보다 선명한 영상을 얻을 수 있다. 이 연구에서는 고양이 뇌 경색을 구축하고 마이크로PET으로 평가하였으며 시간에 따른 변화도 같이 평가하였다. 대상 및 방법 : 수컷 고양이 2마리를 사용하였으며, 체중은 각각 3.0 kg, 3.3 kg 이었다. Xylazine과 Ketamine HCl으로 마취하였다. 고양이 대천문에서 오른쪽으로 1cm되는 곳에 burr 구멍을 뚫은 후 30 G 바늘을 삽입하여 collagenase type IV 10 ${\mu}l$를 5분에 걸쳐 주사하여 뇌 경색 모델을 만들었다. 경색을 만든 후 1일, 11일, 32일 후에 마이크로 PET R4 scanner (Concorde Microsystems Inc., Knoxville, TN)을 사용하여 $^{18}F$-FDG PET 영상을 얻었다. 추가로 경색 13, 47일에 사람용 PET scanner (Gemini, Philips medical systems, CA, USA)를 사용하여 FDG-PET 촬영을 하였다. 결과: 성공적으로 고양이 뇌출혈경색 모델을 만들 수 있었으며 마이크로 PET으로 얻은 영상에서 병변의 당 대사는 시간이 지남에 따라서 호전되었다. 사람용 PET으로도 병변을 확인할 수 있었다. 결론: 고양이 2마리에 두개골을 통해 collagenase를 주입하여 성공적으로 출혈성 뇌 경색 모델을 만들었으며 $^{18}F$-FDG 마이크로 PET으로 영상화할 수 있었다.

참죽나무에서 분리한 flavonol glycoside의 금속단백분해효소-2 억제 활성 (Inhibition of Matrix Metalloproteinase-2 Activity of Flavonol Glycosides from Cedreia sinensis)

  • 참죽나무는 멀구슬과 나무로서 한국과 중국에 널리 분포하고 있다. 한방에서는 장염(腸炎), 이질(痢疾), 개선(疥癬) 등의 치료에 이용되고 있다. 본 연구는 참죽나무 잎에서 3종의 flavonol glycosides를 분리하여 NMR을 통해 구조를 확인한 결과, kaempferol-3-O-rhamnoside (1), quercetin-3-O-rhamnoside (2)와 quercetin-3-O-glucoside(3)로 구조 동정되었다. 분리된 화합물들의 생리활성은 matrix metalloproteinase-2 저해 활성과 인체암 세포주에 대한 세포독성과 apoptosis실험을 통하여 항암효과를 조사하였다. 그 결과 인체암 세포주에 대한 세포독성과 apoptosis 저해활성은 나타내지 않았으나, MMP-2활성저해 조사에서는 강한 저해 효과를 나타내었다. 특히 quercetin-3-O-glucoside는 암세포에 직접적인 독성을 보이는 것이 아니라 암의 침윤과 전이에 특이적으로 작용하는 물질로 보여진다.

Genome Wide Expression Analysis of the Effect of Woowhangchongshim-won on Rat Brain Injury

  • Objectives : ICH breaks down blood vessels within the brain parenchyma, which finally leads to neuronal loss, drugs to treat ICH have not yet been established. In this experiment, we measured the effect of Woowhangchongshim-won (WWCSW) on intracerebral hemorrhage (ICH) in rat using microarray technology. Methods : We measured the effect of WWCSW on ICH in rat using microarray technology. ICH was induced by injection of collagenase type IV, and total RNA was isolated. Image files of microarray were measured using a ScanArray scanner, and the criteria of the threshold for up- and down-regulation was 2 fold. Hierarchical clustering was implemented using CLUSTER and TREEVIEW program, and for Ontology analysis. GOSTAT program was applied in which p-value was calculated by Chi square or Fisher's exact test based on the total array element. Results : WWCSW-treatment restored the gene expression altered by ICH-induction in brain to the levels of 76.0% and 70.1% for up- and down-regulated genes, respectively. Conclusion : Co-regulated genes by ICH model of rat could be used as molecular targets for therapeutic effects of drug including WWCSW. That is, the presence of co-regulated genes may represent the importance of these genes in ICH in the brain and the change of expression level of these co-regulated genes would also indicate the functional change of brain tissue.
