LQG와 LQG/LTR방법을 이용한 헬리콥터의 비행위치 제어 (Control of a Helicopter in Hover Using the LQG and LQG/LTR Methods)

  • The LQG and LQG/LTR compensators have the same structrues in dynamics. The only difference is the values of the design parameters in the two compensators. The design parameters of the LQG and LQG/LTR compensators are selected in the sense of the least-squares error minimi- zation and loop shaping, respectively. In this paper, the LQG and LQG/ LTR design methods are applied to the helicopter in hover which is modeled as a SISO fourth order system. The dynamic characteristics and the perfor- mance of the two control systems are analyzed by the computer simulation. It is found that the LQG/LTR design method is systematic and has good performance in comparision with the LQG design method.

Identification and Phylogeny of the Human Endogenous Retrovirus HERV-W LTR Family in Cancer Cells

  • The long terminal repeats (LTRs) of human endogenous retrovirus (HERV) have been found to be coexpressed with sequences of closely located genes. It has been suggested that the LTR elements have contributed to the structural change or genetic variation of human genome connected to various diseases and evolution. We examined the HERV-W LTR elements in various cancer cells (2F7, A43l , A549, HepG2, MIA-PaCa-2, PC-3, RT4, SiHa, U-937, and UO-31). Using genomic DNA from the cancer cells, we performed PCR amplification and identified twelve new HERV-W LTR elements. Those LTR elements showed a high degree of sequence similarity (88-99%) with HERV-W LTR (AF072500). A phylogenetic tree obtained by the neighbor-joining method revealed that HERV-W LTR elements could be mainly divided into two groups through evolutionary divergence. Three HERV-W LTR elements (RT4-2, A43l-1, and UO3l-2) belonged to Group 1, whereas nine LTR elements (2F7-2, A549-1, A549-3, HepG2-3, MP2-2, PC3-1, SiHa-8, SiHa-10, and U937-1) belonged to Group 11. Taken together, our new sequence data of the HERV-W LTR elements may contribute to an understanding of tissue-specific cancer by genomic instability of LTR integration.

DVD 드라이브의 포커스 서보 시스템 제어기 설계 (Control System Design for the Focus Servo System of DVD Drive)

  • In this paper, two plant models, of which one is newly developed and the other one is the conventional one, of the focus servo system of DVD drive are presented and a two-degree-of freedom controller consisted of Inverse dynamics feedforward and LQG/LTR feedback controller is designed. The newly developed plant model is used to design the feedforward controller and the conventional model is used for the design of feedback controller. The output of newly developed model is the displacement of objective lens and the output of conventional model is the focus error of the DVD focus servo system. The displacement of the objective lens is estimated by the dynamics model of the DVD focus servo system. The disturbance rejection performance of the two-degree-of freedom controller is compared with that of an LQG/LTR one.

Identification and Phylogenetic Analysis of Long Terminal Repeat Elements of the Human Endogenous Retrovirus K Family (HERV-K) from a Human Brain cDNA Library

  • Long terminal repeats (LTRs) of the human endogenous retrovirus K family (HERV-K) have been found to be coexpressed with sequences of genes closely located nearby. We examined transcribed HERV-K LTR elements in human brain tissue. Using cDNA synthesized from mRNA of the human brain, we performed PCR amplification and identified ten HERV-K LTR elements. These LTR elements showed a high degree of sequence similarity (92.4-99.7%) with the human-specific LTR elements. A phylogenetic tree obtained by the neighbor-joining method revealed that HERV-K LTR elements could be divided into two groups through evolutionary divergence. Some HERV-K LTR elements (HKL-B7, HKL-B8, HKL-B10) belonging to the group II from human brain cDNA were closely related to the human-specific HERV-K LTR elements. Our data suggest that HERV-K LTR element are active in the human brain; they could conceivably play a pathogenic role in human diseases such as psychosis.

  • This paper copes with control design for unmanned aerial vehicle transportation system. Moving pendulum dynamics of slung-load system is derived using two methods: Udwadia-Kalaba equation and Newtonian approach. PID controller is applied to Udwadia-Kalaba equation model for structural consistency and linear quadratic Gaussian / Loop Transfer Recovery (LQG/LTR) technique is employed for Newtonian model with minimal state-space realization. Characteristics of PID and LQG/LTR controller are compared, and two controllers are combined to compensate the drawbacks of each other. Numerical simulation is set for two cases and conducted to evaluate performance of designed controllers. The result proves that combination of LQG/LTR and PID control performs stable and robust.

유용버섯 Lentinus tuber-regium이 비만 및 지질대사에 미치는 영향 (Effects of Edible Lentinus tuber-regium on the Obesity and Lipid Metabolism of SD Rats)

  • SD계 랫트를 사용하여 6주 동안 탄수화물 대용으로 50% 및 100%의 Lentinus tuber-regium(LTR)-첨가 투여그룹의 비만 및 지질대사에 미치는 영향을 평가하였다. LTR-50 및 LTR-100 투여그룹의 체중 증가는 대조그룹 대비 각각 3.2%, 29.3%의 억제효과로 나타났는데도 사료 섭취량은 대조그룹 대비 오히려 24.5% 및 40,7%나 증가했다. 이들 LTR-첨가사료의 사료효율이 대조그룹 대비 22% 및 50%나 현저히 감소했는데도 불구하고 에너지효율은 이와는 반대로 두 그룹 모두 약 10%의 GE 증가효과가 인정되었다. LTR-50 및 LTR-100 투여그룹의 대사체중에 대한 사료 섭취량의 비(DI/MBS ratio)는 대조그룹 대비 50.3% 및 136.1%나 매우 유의적인 증가현상이 인정되었다. LTR-50 및 LTR-100 투여그룹은 대조그룹 대비 22.4% 및 32.6%나 현저한 TG 함량의 억제효과가 인정되었고 이들 LTR 투여그룹은 대조그룹 대비 8.3% 및 12.7%나 유의적인 LDL-콜레스테롤의 혈관축적 억제효과가 인정되었다. 이들 LTR투여그룹은 대조그룹 대비 $3{\sim}$10%의 효과적인 HDL-콜레스테롤의 생성효과가 인정되었다. 또한 이들 LTR 투여그룹의 동맥경화지수(Al) 는 대조그룹 대비 $8{\sim}13%$의 상당히 유의적인 Al의 감소효과가 인정되었다. 이상의 결과를 종합해 볼 때 탄수화물 대용으로 LTR의 50%이상 투여에서는 비만을 매우 효과적으로 억제할 수 있을 뿐만 아니라 LTD의 50%이하 부여에서는 가축의 대체사료 개발에 기여할 수 있을 것으로 기대된다. 또한 이들 LTR-첨가사료는 TG, LDL-콜레스테롤 및 동맥경화지수(Al)의 감소, 그리고 HDL-콜레스텔롤의 증가 때문에 성인병을 매우 효과적으로 억제할 수 있을 것으로 기대된다.

Insertional Mutation of the Rice Blast Resistance Gene, Pi-b, by Long Terminal Repeat of a Retrotransposon

  • The Pi-b is the rice gene conferring race specific resistance to the blast fungus Magnaporthe grisea race having a corresponding avirulence gene, AVR-Pi-b. All resistant cultivars have two copies of the Pi-b gene, but susceptible cultivars have a single copy of the gene. About 1 Kbp insertion sequence was detected in the open reading frame of the Pi-b gene from the susceptible cv. Nipponbare. The nature of insertion sequence was identified as a solo long terminal repeat (LTR) of new rice Tyl-copia-like retrotransposon. LTR was widely distributed in the rice genome. Various types of different patterns of restriction fragment length polymorphism of LTR were detected in indica cultivars, whereas a single type was detected from japonica cultivars. The insertion of LTR sequence in the Pi-b gene in the susceptible cultivar suggested that retrotransposon-mediated insertional mutation might played an important role in the resistance breakdown as well as evolution of resistance genes in rice.

Transposable Elements and Genome Size Variations in Plants

  • Although the number of protein-coding genes is not highly variable between plant taxa, the DNA content in their genomes is highly variable, by as much as 2,056-fold from a 1C amount of 0.0648 pg to 132.5 pg. The mean 1C-value in plants is 2.4 pg, and genome size expansion/contraction is lineage-specific in plant taxonomy. Transposable element fractions in plant genomes are also variable, as low as ~3% in small genomes and as high as ~85% in large genomes, indicating that genome size is a linear function of transposable element content. Of the 2 classes of transposable elements, the dynamics of class 1 long terminal repeat (LTR) retrotransposons is a major contributor to the 1C value differences among plants. The activity of LTR retrotransposons is under the control of epigenetic suppressing mechanisms. Also, genome-purging mechanisms have been adopted to counter-balance the genome size amplification. With a wealth of information on whole-genome sequences in plant genomes, it was revealed that several genome-purging mechanisms have been employed, depending on plant taxa. Two genera, Lilium and Fritillaria, are known to have large genomes in angiosperms. There were twice times of concerted genome size evolutions in the family Liliaceae during the divergence of the current genera in Liliaceae. In addition to the LTR retrotransposons, non-LTR retrotransposons and satellite DNAs contributed to the huge genomes in the two genera by possible failure of genome counter-balancing mechanisms.

Frequent Genetic Defects in the HIV-1 5'LTR/gag Gene in Hemophiliacs Treated with Korean Red Ginseng: Decreased Detection of Genetic Defects by Highly Active Antiretroviral Therapy

  • We investigated whether Korean red ginseng (KRG) and highly active antiretroviral therapy (HAART) affect the frequency of gross deletion in 5'LTR/gag in 20 hemophiliacs. This study is a prospective study in 20 hemophiliacs who were infected with Korean subclade B of HIV-1 from two cash-paid plasma donors in 1990. Over a 13-year period, we obtained 436 amplicons of 5'LTR/gag genes by nested polymerase chain reaction using 147 peripheral blood mononuclear cells. Of the 436 amplicons, 92 (21.1%) showed gross deletion in 5'LTR/gag. Despite of a 2.3-fold higher monthly dose of KRG intake, the frequency of gross deletion in 5'LTR/gag (16.4%) was significantly decreased during HAART compared with 28.1% prior to HAART (p<0.01). Gross deletion in 5'LTR/gag was 10% more detected on KRG-therapy than prior to KRG-therapy (p<0.05). In addition, we also obtained 28 amplicons containing premature stop codon or isoleucine at initiation codon of 254 amplicons sequenced on KRG intake (7.5%) or HAART (13.6%) compared with 0% before KRG intake. These findings indicate that high frequency of gross deletion in 5'LTR/gag and genetic defects prior to HAART are significantly associated with KRG intake and the detection of gross deletion in 5'LTR/gag is decreased by HAART.

  • Human endoqenous retroviral long terminal repeats (LTRs) have been found to be coexpressed with sequences of genes closely located nearby. It has been suggested that the LTR elements have contributed to structural changes or genetic variations of human genome connected to various diseases and evolution. Using cDNA library derived from placenta tissue, we performed PCR amplification and identified five new HERV-W LTR elements. Those LTR elements showed a high degree of sequence similarity (98-99%) with HERV-W LTR (AF072500). A phylogenetic tree obtained by the neighbor-joining method revealed that HERV-W LTR elements could be mainly divided into two groups through evolutionary divergence. Five new HERV-W LTR elements (pla-1, 4, 5, 6, 7) belonged to the group I with AX000960, AF072504, and AF072506 from GenBank database. The data suggest that several copy numbers of the HERV-W LTR elements are transcribed in placenta and may contribute to the understanding of biological function such as human placental morphogenesis.
