발효울금 50% 에탄올 추출물의 라디칼 소거능 및 지방형성 억제 효과 (Radical Scavenging and Anti-Obesity Effects of 50% Ethanol Extract from Fermented Curcuma longa L.)

  • 본 연구에서는 FCE50을 대상으로 다양한 항산화 실험을 실시한 결과, FCE50이 항산화능을 보유하고 있음을 확인하였다. 또한 FCE50은 3T3-L1 전지방세포의 분화과정 중 세포내 지방축적을 유의적으로 감소시키며 포도당 유입을 감소시켰다. 지방합성과 관련된 ACC 효소의 mRNA 발현량을 측정한 결과, 완전히 분화된 지방세포와 비교하여 FCE50의 처리 시 그 발현량이 현저히 감소됨을 확인하였다. 이상의 결과로부터 FCE50은 항산화 활성을 보유하고 있으며, 이는 FCE50이 지방세포로의 분화과정 중 지방형성을 유의적으로 감소시키는 결과에 영향을 미치는 것으로 사료된다.

울금 분말을 첨가한 식빵의 품질 특성 (Quality Characteristics of Bread Added with Turmeric Powder)

  • Recently, there has been a great deal of public interest in health foods, such as turmeric (Curcuma longa L). In this study, the baking features of turmeric powder were evaluated by making pan bread. To accomplish this, the effects of added turmeric powder on the results of mixograph analysis as well as, the pH and, fermentation rate were measured. In addition, the features of the bread were examined by testing the stickiness of the dough and the TPA of the product using a texture analyzer. Finally, the consumer's preferences were investigated by evaluation of the color difference, crumbScan and sensory evaluation. The results revealed that as the level of turmeric powder increased, the pH decreased. which resulted in the gas possessing capacity of the dough improving and the fermentation persistence of the dough increasing. In addition, the stickiness increased as the turmeric powder content increased. However, there were significant differences among breads produced using different amounts of turmeric powder. Evaluation of the taste revealed that the TP3 group had the higher score than control score and TP7 had the lowest score. In overall preference, TP7 was especially low and the preference decreased as the content of turmeric powder increased.

Antifungal Activity Against Colletotrichum spp. of Curcuminoids Isolated from Curcuma longa L. Rhizomes

  • Methanol extract of the rhizomes of turmeric, Curcuma longa L., effectively controlled the development of red pepper anthracnose caused by Colletotrichum coccodes. In addition three antifungal substances were identified from the methanol extract of C. longa rhizomes as curcumin, demethoxycurcumin, and bisdemethoxycurcumin using mass and $^{1}H-NMR$ spectral analyses. The curcuminoids in a range $0.4-100\;{\mu}g/ml$ effectively inhibited the mycelial growth of three red pepper anthracnose pathogens, C. coccodes, C. gloeosporioides, and C. acutatum. The three curcuminoids inhibited mycelial growth of C. coccodes and C. gloeosporioides to an extent similar to the synthetic fungicide dithianon did, but the synthetic agent was a little more effective against C. acutatum. The curcuminoids also effectively inhibited spore germination of C. coccodes, and bisdemethoxycurcumin was the most active. Among the three curcuminoids, only demethoxycurcumin was effective in a greenhouse test in suppressing red pepper anthracnose caused by C. coccodes.

Evidence for the Participation of ATP-sensitive Potassium Channels in the Antinociceptive Effect of Curcumin

  • Background: It has been reported that curcumin, the main active compound of Curcuma longa, also known as turmeric, exhibits antinociceptive properties. The aim of this study was to examine the participation of ATP-sensitive potassium channels ($K_{ATP}$ channels) and, in particular, that of the L-arginine-nitric oxide-cyclic GMP-$K_{ATP}$ channel pathway, in the antinociceptive effect of curcumin. Methods: Pain was induced by the intraplantar injection of 1% formalin in the right hind paw of Wistar rats. Formalin-induced flinching behavior was interpreted as an expression of nociception. The antinociceptive effect of oral curcumin was explored in the presence and absence of local pretreatment with L-NAME, an inhibitor of nitric oxide synthase, ODQ, an inhibitor of soluble guanylyl cyclase, and glibenclamide, a blocker of $K_{ATP}$ channels. Results: Oral curcumin produced a dose-dependent antinociceptive effect in the 1% formalin test. Curcumin-induced antinociception was not altered by local L-NAME or ODQ, but was significantly impaired by glibenclamide. Conclusions: Our results confirm that curcumin is an effective antinociceptive agent. Curcumin-induced antinociception appears to involve the participation of $K_{ATP}$ channels at the peripheral level, as local injection of glibenclamide prevented its effect. Activation of $K_{ATP}$ channels, however, does not occur by activation of the L-arginine-nitric oxide-cGMP-$K_{ATP}$ channel pathway.

  • Recently, there have been considerable efforts to search for naturally occurring substances for the intervention of carcinogenesis. Curcumin, a yellow coloring ingredient of turmeric (Curcuma longa L., Zingiberaceae), has been shown to inhibit experimental carcinogenesis and mutagenesis, but molecular mechanisms underlying its chemopreventive activities remain unclear. In the present work, we found that curcumin inhibited 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced expression of CPX-2 in female ICR mouse skin when applied topically 30 min prior to TPA as determined by both immunoblot and immunohistochemical analyses.   • Hair coloring before the 1980s was mostly to hide white hair by dying in black. With the introduction of color TV, however, the size of the coloring market has been expanding. Nowadays, artificial synthetic dyes are widely used, which, though advantageous in many points, have problems such as harmful effects on the human body, carcinogenesis, environmental contamination and damage on hair. On the contrary, natural vegetables obtained from Curcuma Longa L., A. catechu, polygonum indigo, henna, etc. are little harmful to hair or the skin of the head and cause few environmental problems. In addition, as they are natural materials collected from nature, they are considered positively by consumers. Thus the present study started from the necessity of research on the convenient use of environment?friendly and side-effect-free natural dyes, coloring technology for reproducing original color, the improvement of adhesion rate, etc. From the present research were obtained yellow color from Curcuma Longa L., brown from A. catechu, blue from polygonum indigo and orange from henna. It is expected that, based on materials from previous researches, there may be more researches on the use of natural dyes as hair colors.

종근 저장 온도 및 충진제가 강황 수량 및 품질에 미치는 영향 (Effect of Temperatures and Fillers on Yield and Quality of Turmeric (Curcuma longa L.) During Postharvest Seed Rhizome Storage)

  • Studies were carried to evaluated the influence of storage method by temperatures and fillers on yield and quality of seed rhizome in turmeric. Seed rhizome was stored at styrofoam box filled with rice hull and sand (3:1) or vermiculite for 30, 60 and 90 days at different temperatures (5, 10 and $15^{\circ}C$. compared to traditional method (rhizome only). Parameters were obtained for weight loss, cold injury, percentage of decayed in stored rhizome during storage periods. Also, the germination, growth pattern and yield from stored rhizome has been investigated. It was confirmed that storage of turmeric in stored with filled with vermiculite helps in prevention of rhizomes from microbial and fungal attack. The storage of rhizomes in styrofoam box without any filler at low temperature below $10^{\circ}C$. is not advocated due to heavy losses weight and decayed in management of postharvest for turmeric rhizome. Germination percentage, growth pattern and yield was maximum for rhizomes stored at styrofoam box filled with vermiculite for 90 days at $15^{\circ}C$. The paper outlines a brief attempt to assess the efficacy of non-chemical methods including optimal storage method (temperature and filler) of control of decay and moisture losses during storage of turmeric.

Dietary turmeric (Curcuma longa L.) supplementation improves growth performance, short-chain fatty acid production, and modulates bacterial composition of weaned piglets

  • In livestock nutrition, natural feed additives are gaining increased attention as alternatives to antibiotic growth promoters to improve animal performance. This study investigated the effects of dietary turmeric supplementation on the growth performance and gut health of weaned piglets. A total of 48 weaned piglets (Duroc × [Landrace × Yorkshire]) were used in a 6-week feeding trial. All piglets were allotted to two dietary treatments: corn-soybean meal basal diet without turmeric (control) and with 1% weight per weight (w/w) turmeric powder (turmeric). The results showed that dietary inclusion of turmeric with the basal diet improved final body weight and total average daily gain (p < 0.05). The concentrations of short-chain fatty acids in the fecal samples, including acetic, butyric, and propionic acids, were higher in the turmeric group (p < 0.05). The villus height-to-crypt depth ratio was higher in the ileum of turmeric-fed piglets (p = 0.04). The 16S rRNA gene sequencing of fecal microbiota indicated that, at the phylum level, Firmicutes and Bacteroidetes were the most predominant taxa in all fecal samples. Bacteroidetes were significantly decreased in the turmeric group compared to the control group (p = 0.021). At the genus level, turmeric showed a decreased abundance of Prevotella (p = 0.021) and an increasing trend of Lactobacillus (p = 0.083). Among the total detected species, nine bacterial species showed significant differences between the two groups. The results of this study indicated that turmeric altered the gut microbiota and shortchain fatty acid production. This suggests that turmeric could be used as a potential alternative growth promoter for piglets.

발효울금 주정추출물부터 분리된 에틸아세테이트 분획물에 대한 3T3-L1 세포에서의 지방 형성 억제 효과 (Anti-Obesity Effect of Ethyl Acetate Fraction from 50% Ethanol Extract of Fermented Curcuma longa L. in 3T3-L1 Cells)

  • FCEE를 3T3-L1 전지방세포의 분화과정 중에 처리한 후, FCEE가 지방의 축적에 미치는 영향을 확인하였다. FCEE($200{\mu}g/mL$)를 세포에 처리한 후 Oil Red O 염색법과 AdipoRed 형광염색법을 이용한 지방 축적과 세포내 중성지방 함량을 측정한 결과, 무처리한 지방세포와 비교하여 각각 10.2%, 13.7% 유의하게 감소하는 것을 확인하였다. 이러한 결과에 대한 기작을 확인하고자 포도당 유입량과 유리 글리세롤 방출량을 측정하였다. FCEE($200{\mu}g/mL$) 처리 시 지방세포와 비교하여 포도당 유입량이 36.6% 유의적으로 감소하였으며, 유리 글리세롤 방출량도 8일째에 유의적으로 감소하였다. 이는 지방세포의 분화과정 중에 FCEE의 처리로 분화가 억제되어 지방구의 형성이 억제되고 배양액으로 배출되는 유리 글리세롤의 총량이 감소한 것으로 판단된다. 이러한 결과가 지방세포 분화의 억제에 의한 것인지 판단하기 위해 지방 형성 관련인자의 mRNA 발현량을 측정하였다. FCEE($200{\mu}g/mL$)의 처리로 AMPK mRNA 발현량은 지방세포와 비교하여 3배 증가하였으며, SREBP-1c, $C/EBP{\alpha}$$PPAR{\gamma}$ mRNA 발현량은 각각 0.6, 0.6 및 0.8배 감소하는 것을 확인하였다. 이상의 결과들로부터 FCEE는 지방 합성을 억제하는 활성을 보유하고 있는 바, 향후 항비만 기능성 식품소재로 활용될 수 있을 것으로 사료된다.
