Pathological Observations of Polycystic Kidney in a Korean Native Calf (한우(韓牛)에 발생(發生)한 Polycystic Kidney)

  • This paper dealt with the macroscopical and histopathological observations on the polycystic kidney occurred in a Korean native calf aged about a year. The results summarized are as follows: 1. In macroscopical findings, numerous cysts in the bilateral kidneys were seen under the renal capsule, and the cysts were various in size and clear or cloudy in their contents. The cysts in the inner area of the renal cortex were smaller than those of the outer area of the renal cortex in size and in number. 2. In microscopical findings, marked dilatations of Bowman's spaces and convoluted tubules were prominant feature. Numerous cysts with or without eosinophilic materials were contained atrophic glomerulus. prolferation of fibrous connective tissue, atrophy of convoluted tubules were also observed. Inner walls of the cysts were surrounded by cuboidal cells, sguamous cells or fibrous connective tissue. 3. This case was regarded as congenital polycystic kidney belonged to type III of Osathanondh and Potter.

Influence of Bradykinin on the tubular function of the dog kidney (신세뇨관기능(腎細尿管機能)에 미치는 Bradykinin의 영향(影響))

  • In this study the influence of Bradykinin, a biogenic polypeptide, on the excretory function of the dog kidney was investigated, utilizing the clearance and stop-flow method. The results are summarized as follows; 1) Bradykinin administered intravenously elicited a marked antidiuresis. 2) When given into the renal artery, however, prompt increase of the urine flow, sodium excretion and free water clearance without significant change in the glomerular filtration rate ensued. 3) It was also effective during an osmotic diuresis with 10% Mannitol infusion, though the response was not so marked. 4) The stop-flow experiment showed that sodium reabsorption in the proximal tubules is inhibited by the infusion of bradykinin into the renal artery. It was thus concluded that the diuretic effect of bradykinin given intra-arterially results from the inhibition of sodium transport in the proximal tubules.

  • Adult male ICR mice were exposed to methylmercuric chloride (CH$_3$HgCI) through drinking water for 80 days. The distribution of mercury in the kidney, liver, spleen and cerebellum of the mouse was examined according to a autometallographic silver-enhancement technique based on a physical development process which renders mercury deposit visible. Grains of mercury traces were located in the proximal convoluted tubules. Lesser staining of the grains was seen in the collecting tubules of medulla. The glomerular basement membrane was void. In the liver, mercury accumulations were present primarily in the hepatocytes around portal area containing interlobular bile duct, artery and portal vein. Also grains of mercury traces were accumulated in the white pulp of the spleen and Purkinje cell layer of the cerebellum.

  • A case of renal candidiasis is reported in a 4-year-old male Masked Palm civet (Paguma larvata). On necropsy, the kidneys were bilaterally swollen, pale and had numerous 1 to 3 mm diameter white foci throughout the parenchyma on cut section. The urinary bladder was filled with opaque and milky exudate. Histologically, severe infiltration of neutrophils and macrophages and necrosis were noted in the interstitial areas of both cortex and medulla and in the lumens of renal tubules and collecting duct often resulting in cystic dilation of the tubules.   • The incidence of spontaneous testicular atrophy and its morphological changes during stage-specific spermatogenesis were investigated in male Sprague-Dawley rats at 10, 19, and 32 weeks of age. The incidence of testicular atrophy was 0.2%(2/90) 7.9%(9/114) and 10%(4/40) in 4, 13 and 26 weeks respectively. The epididymis with testicular atrophy had low sperm density. In the minimally affected tests scattered tubules showed complete depletion of germ cells without stage specificity. Testes with moderate to severe testicular atrophy showed seminiferous tubules lined with dense Sertoli cell population. While Leydig cells in the interstitium appeared hypertrophy they were immunohistochemically negative for proliferating cell nuclear antigen a marker of cell proliferation.

Studies on Renal Action of Atractylodes Rhizoma Alba in Dogs (백출의 신장작용에 관한 연구)

  • The actions of Atractylodes rhizoma alba on renal function of dog were investigated with water and methanol extract of it, utilizing clearance technique. Water extract elicited diuresis while methanol extract induced antidiuresis in the dog when they were given intravenously. The diuresis induced by the intravenous water extract seemed to be closely related to the decrement of reabsorption of electrolytes in renal tubules. On the contrary, the antidiuresis by intravenous methanol extract appeared to be related to the increment of renal reabsorption of sodium. Water and methanol extract, when infused into a renal artery, exhibited identical resposes to the intravenous actions confined only to the infused kidney, inferring that renal actions of water and methanol extract are not mediated by any endognous humoral extract are not mediated by any endogenous humoral agents. From these observations it is concluded that diuretic action of water extract and antidiuretic action of methanol extract were brought about by direct action in the renal tubules.

  • The cobalt capture method of Gomori's modified technique(Gomori, 1952) was employed to study the histochemistry of the developing frog kidney. Alkaline phosphatase activity was observed in association with the brush borders of pronephros and mesonephros. By the stage of transition from larva to tadpole alkaline phosphatase activity was gradually increased in the brush border of pronephros, and as the pronephros begun to degenerate the enzyme activity was decreased and disappeared. By the time of maximum development of the pronephros the mesonephros began to develop and alkaline phosphatase activity of the mesonephric tubules showed highly positive throughout the stage of metamorphosis. No activity was observed in association with the collecting tubules and ductal elements.

  • Polypyrrole (PPy) was chemically synthesized within the pores of nanoporous polycarbonate (PC) Particle Track-etched Membranes (nano-PTM). Hollow tubules are formed because polypyrrole initially deposits on the surface of the pores walls. By running successive syntheses, we have obtained wires (filled tubules). The redox property of PPy nanotubules was investigated by cyclic voltammetry. The redox potential was lowered as much as 0.5V vs. Ag/AgC1, comparing with electrosynthesized PPy film. It suggests that an electron hopping mechanism of PPy nanotubules was improved. Electric conductivity of PPy nanotubules and nanowire was evaluated. We obtained good electric conductivity of PPy nanotubules even in the neutral state. The conductivity and activation energy were $10^1$ order at the room temperature and 25.3 meV respectively.

Effect of Diltiazem on Renal Function in the Dog (딜티아젬의 개 신장기능에 미치는 영향)

  • This study was performed in order to investigate the effect of diltiazem, which is a $Ca^{2+}$ channel blocker of benzothiazepine derivatives, on renal function in the dog. Diltiazem, when infused into the vein or carotid artery, produced the antidiuresis accompanied with the decreased excretion rates of sodium and potassium in urine$(E_{Na},\;E_K)$ and the increased reabsorption rates of sodium and potassium in renal tubules$(R_{Na},\;R_K)$. Diltiazem, when infused into a renal artery, exhibited the diuresis along with the increased renal plasma flow(RPF), osmolar clearance$(C_{osm})$, $E_{Na}$ and $E_K$, and decreased $R_{Na}$ and $R_K$ in only infused kidney. Above results suggest that diltiazem possess both antidiuretic action through central action and diuretic action by direct inhibition of electrolytes reabsorption rates in renal tubules, mainly distal tubule.

Tubulopathy: the clinical and genetic approach in diagnosis

  • Remarkable advances in genetic diagnosis expanded our knowledge about inherited tubulopathies and other genetic kidney diseases. This review suggests a simple categorization of inherited tubular disease, clarifies the concept of autosomal dominant tubulointerstitial kidney disease (ADTKD), and introduces novel therapies developed for tubulopathies. Facing patients with suspicious tubular disorders, clinicians should first evaluate the status of volume and acid-base. This step helps the clinicians to localize the affected segment and to confirm genetic diagnosis. ADTKD is a recently characterized disease entity involving tubules. The known causative genes are UMOD, MUC1, REN, and HNF1β. Still, only half of ADTKD patients show mutations for these four identified genes. Whole exome sequencing is a suitable diagnostic tool for tubulopathies, especially for ADTKD. Genetic approaches to treat tubulopathies have progressed recently. Despite the practical obstacles, novel therapies targeting inherited tubulopathies are currently in development.
