Backbone NMR Assignments of a Prokaryotic Molecular Chaperone, Hsp33 from Escherichia coli

  • The prokaryotic molecular chaperone Hsp33 achieves its holdase activity upon response to oxidative stress particularly at elevated temperature. Despite many structural studies of Hsp33, which were conducted mainly by X-ray crystallography, the actual structures of the Hsp33 in solution remains controversial. Thus, we have initiated NMR study of the reduced, inactive Hsp33 monomer and backbone NMR assignments were obtained in the present study. Based on a series of triple resonance spectra measured on a triply isotope-[$^2H/^{13}C/^{15}N$]-labeled protein, sequence-specific assignments of the backbone amide signals observed in the 2D-[$^1H/^{15}N$]TROSY spectrum could be completed up to more than 96%. However, even considering the small portion of non-assigned resonances due to the lack of sequential connectivity, we confirmed that the total number of observed signals was quite smaller than that expected from the number of amino acid residues in Hsp33. Thus, it is postulated that peculiar dynamic properties would be involved in the solution structure of the inactive Hsp33 monomer. We expect that the present assignment data would eventually provide the most fundamental and important data for the progressing studies on the 3-dimensional structure and molecular dynamics of Hsp33, which are critical for understanding its activation process.

Backbone assignment and structural analysis of anti-CRISPR AcrIF7 from Pseudomonas aeruginosa prophages

  • The CRISPR-Cas system provides adaptive immunity for bacteria and archaea against invading phages and foreign plasmids. In the Class 1 CRISPR-Cas system, multi-subunit Cas proteins assemble with crRNA to bind to DNA targets. To disarm the bacterial defense system, bacteriophages evolved anti-CRISPR (Acr) proteins that actively inhibit the host CRISPR-Cas function. Here we report the backbone resonance assignments of AcrIF7 protein that inhibits the type I-F CRISPR-Cas system of Pseudomonas aeruginosa using triple-resonance nuclear magnetic resonance spectroscopy. We employed various computational methods to predict the structure and binding interface of AcrIF7, and assessed the model with experimental data. AcrIF7 binds to Cas8f protein via flexible loop regions to inhibit target DNA binding, suggesting that conformational heterogeneity is important for the Cas-Acr interaction.

H-1, C-13, and N-15 resonance assignments of ENOD40B, a plant peptide hormone

  • t ENOD40B, a plant peptide hormone, was doubly labeled with C-13 and N-15 by recombinant production in Escherichia coli. The peptide was prepared by affinity chromatography followed by protease cleavage and reverse-phase chromatography. To elucidate the mode of action against its receptor, sucrose synthase, we proceeded to assign the backbone and side-chain resonances using a set of double and triple resonance experiments. This result will be used to determine the three-dimensional structure of the peptide at its bound state as well as to observe the chemical shift changes upon binding.

Compact Triple-Mode Resonator and Triple-Mode Filter Design (소형의 삼중 모드 공진기와 삼중 모드 필터 설계)

  • In this paper, we present a compact triple-mode resonator. The resonator itself can provide three transmission poles and one transmission zero. The resonance condition of each mode is analyzed theoretically, and the transmission zero is generated by open stub. Using this proposed resonator, a compact three pole bandpass filter for 2.4 GHz WLAN application with one transmission zero is designed and fabricated. The fabricated triple-mode filter shows 3 dB bandwidth of 15.8 % with the center frequency 2.4 GHz and less then 1.17 dB in 2.4~2.5 GHz passband. The size of fabricated triple-mode filter including the feed lines is 15.9 mm${\times}$9.7 mm and very compact compared with previous reported triple-mode filter.

  • Purpose: To correlate the histological differentiation of hepatocellular carcinomas (HCCs) with finding on triple contrast-enhanced MR imaging using gadolinium-chelates, superparamganetic ire oxides (SPIO), and mangafodipir trisodium. Method: Ten patients with proven HCC underwent triple contrast-enhanced MRI befo surgical resection. Subjective ratings of the enhancement pattern and degree were compare with the histological grades determined on surgical specimen. Quantitative measurements signal-to-noise ratio (S/N) of the lesion and the lesion-to-liver contrast-to-noise ratio C/N on the enhanced MR images, and the degree of S/N and C/N changes between the unenhanced and enhanced images were also correlated with the histological grades.

  • Angiogenin is unique among angiogenic molecules in that it is a member of the pancreatic ribonuclease superfamily and, in fact, is a ribonucleolytic enzyme. Its enzymatic activity is extremely weak compared to that of the digestive RNases but is critical for its capacity to induce neovascularization. In this study, we completed the backbone resonance assignment of bovine angiogenin using triple resonance NMR experiments of $^{15}N\;and/or\;^{13}C$ isotope labeled protein and investigated the chemical shift variation upon binding with inhibitor phosphate ion and determine the phosphate binding site.

A Triple-Band Transceiver Module for 2.3/2.5/3.5 GHz Mobile WiMAX Applications

  • A triple-band transceiver module for 2.3/2.5/3.5 GHz mobile WiMAX, IEEE 802.16e, applications is introduced. The suggested transceiver module consists of RFIC, reconfigurable/multi-resonance MIMO antenna, embedded PCB, mobile WiMAX base band, memory and channel selection front-end module. The RFIC is fabricated in $0.13{\mu}m$ RF CMOS process and has 3.5 dB noise figure(NF) of receiver and 1 dBm maximum power of transmitter with 68-pin QFN package, $8{\times}8\;mm^2$ area. The area reduction of transceiver module is achieved by using embedded PCB which decreases area by 9% of the area of transceiver module with normal PCB. The developed triple-band mobile WiMAX transceiver module is tested by performing radio conformance test(RCT) and measuring carrier to interference plus noise ratio (CINR) and received signal strength indication (RSSI) in each 2.3/2.5/3.5 GHz frequency.

1H, 15N, and 13C Resonance Assignments of the Anti-CRISPR AcrIIA4 from Listeria monocytogenes Prophages

  • The CRISPR-Cas system is the adaptive immune system in bacteria and archaea against invading phages or foreign plasmids. In the type II CRISPR-Cas system, an endonuclease Cas9 cleaves DNA targets of phages as directed by guide RNA comprising crRNA and tracrRNA. To avoid targeting and destruction by Cas9, phages employ anti-CRISPR (Acr) proteins that act against host bacterial immunity by inactivating the CRISPR-Cas system. Here we report the backbone $^1H$, $^{15}N$, and $^{13}C$ resonance assignments of AcrIIA4 that inhibits endonuclease activity of type II-A Listeria monocytogenes Cas9 and also Streptococcus pyogenesis Cas9 using triple resonance nuclear magnetic resonance spectroscopy. The secondary structures of AcrIIA4 predicted by the backbone chemical shifts show an ${\alpha}{\beta}{\beta}{\beta}{\alpha}{\alpha}$ fold, which is used to determine the solution structure.

Triple-band Compact Chip Antenna Using Coupled Meander-line Structure for Mobile RFID/PCS/WiBro (결합 미엔더 선로를 이용한 모바일 RFID/PCS/WiBro 삼중 대역 소형 칩 안테나)

  • The proposed triple-band compact chip antenna using coupled meander line and stacked meander structure for mobile RFID/PCS/WiBro. The proposed antenna is designed to operate at 900, 1,800, and 2,350 MHz, and is realized by parasitic coupled and stacked a meander line. Meander lines are using extend length of effective current path more than monopole and contribute miniaturization. The coupled meander line controls the excitations of the mobile RFID and PCS, stacked meander line controls the excitation of the WiBro. The fabricated antenna size is $10.98{\times}22.3{\times}0.98\;mm$. The resonance frequencies are 905 MHz, 1.77 GHz and 2.32 GHz. The impedance bandwidths are 24 MHz, 140 MHz and 92 MHz. The maximum gains of antenna are 0.34 dBi, 2.58 dBi and 0.4 dBi at resonance frequencies.

Triplexer based on Filter Characteristics of CRLH Transmission Line and Triple-Band Amplifier Applications (CRLH 전송선로의 필터 특성을 이용한 트리플렉서와 삼중대역 증폭기에의 응용)

  • In this paper, we proposed the triplexer using unit-cell of CRLH transmission line which has a bandpass characteristic to reduce adjacent channel interference. The input impedance of triplexer with each channel filter is operated open-circuit at the resonance frequencies of other channels. Such property is due to the combination a series and parallel resonance circuits of CRLH-TL unit-cell. The measured triplexer results are showed a measured insertion loss of each channel, less than 1.5 dB, matching at each port, less than 15dB and isolation between channel, better than 25 dB. Also, to validate the triplexer, a small signal amplifier with triple-band is designed and tested. the measured amplifier results show good agreements with prediction.
